Abstract
Die Erfindung betrifft ein Verfahren zur Synchronisation von Mikroorganismen in kontinuierlichen Kulturen zur biotechnischen Stoffwandlung mit Hilfe wachsender Zellen. Ziel und Aufgabe sind, ein Verfahren zur Synchronisation und anschliessenden Selbstregulation des synchronen Wachstums von Mikroorganismen in kontinuierlichen Kulturen anzugeben. Dies wird erreicht, indem das Wachstum einer kontinuierlichen Kultur durch ein solches Substrat limitiert wird, das in einer bestimmten Phase des Teilungszyklus verstaerkt benoetigt wird. Das limitierende Substrat wird nicht kontinuierlich, sondern portionsweise zugegeben, so dass die Zellen, die im Moment der Zugabe die hoechste Aufnahmegeschwindigkeit fuer dieses Substrat haben, in der darauffolgenden Generationszeit nahezu mit Dmmax wachsen. Der Abstand zwischen den Zugaben muss jeweils gleich der aktuellen Generationszeit dieser Zellen sein. Dieser Abstand wird automatisch eingehalten, indem die Zugaben jeweils dann ausgeloest werden, wenn das b-fache der Naehrmediummenge, die einem Kulturvolumen entspricht, zugeflossen ist. Diese Methode ist bisher nur für die Synchronisation von semikontinuierlichen Kulturen nutzbar. Durch Einsatz dieser Methode bei der Futterhefeproduktion konnte durch eine Reduktion der Kohlenstoffsubstratzufuhr während etwa Vs der Generationszeit der Zellen einer synchronisierten Hefepopulation eine erhöhte Effektivität der Ausnutzung der C-Quelle bei der Biomasseproduktion erzielt werden (Heinritz, B. et al., 1983, Acta Biotechnol. 3, S. 125-131; Kardos, J. et al., Dynamische Grundoperationen der Verfahrenstechnik, Akademie-Verlag Berlin, 1984, S. 107).Thus, in Dawson, P.S.S., 1965, Canad. Die Generationszeit g der Zellen kann damit automatisch aus der Durchflußrate D nach der Beziehung g = In2/D bestimmt werden. Turbidostatkulturen sind bei technisch interessanten Biomassekonzentrationen nur dann stabil, wenn alle essentiellen Nährstoffe im Überschuß angeboten werden. Sie arbeiten daher ökonomisch ineffektiv. Die Synchronisation dieser Kulturen wird durch eine solche zeitweilige Temperatur- oder pH-Änderung erreicht, die zu einem Entwicklungsstillstand der Zellen führt, wenn eine bestimmte Phase.des Teilungszyklus erreicht ist. Die ungünstige Bedingung muß so lange einwirken, bis praktisch alle Zellen mit der unter diesen Bedingungen herabgesetzten Wachstumsgeschwindigkeit bis zu diesem Stadium des Teilungszyklus herangewachsen sind. Erst dann wird wieder der optimale Wert (z. B. pH oder Temperatur) eingestellt, und es schließt sich eine Generationszeit des synchronen Wachstms bei optimalen Bedingungen an, nach der die Kultur wiederum in dieser bestimmten Phase des Teilungszyklus zum Wachstumsstillstand gebracht wird. Aus diesen beiden Gründen ist dieses Verfahren zur Verbesserung der Ökonomie technischer Verfahren ungeeignet. Nach diesem für alle bekannten Formen von Turbidostaten anwendbaren Verfahren wird mindestens eine wachstumsessentielie Komponente des Nährmediums nicht über die der Wachstumsgeschwindigkeit proportionale Regelung, sondern mit konstanter, die Wachstumsrate limitierende Geschwindigkeit zur Kultur zugeführt. Mischlimitationen können bei vorgewählten limitierenden Angeboten der einzelnen Komponenten im Verhältnis zur Biomassekonzentration eingestellt werden. Die dritte prinzipielle Methode zur Synchronisation des Wachstums von Mikroorganismen ist aus Vrana, D., 1976, Biotech. Bioerig. 18, S. 297 bekannt und beinhaltet die Gewinnung von Zeilen in gleicher Phase des Teilungszyklus durch Abtrennung der kleinen Knospen einer wachsenden Hefezellpopulation, z. B. durch Zentrifugation und deren Einsatz als Inokulum.für eine synchronisiert wachsende batch-Kultur. Die Erfindung soll nachfolgend anhand eines Ausführungsbeispieles näher erläutert werden. Aus der Literatur (Jeter, J. R. et al., 1982, Cytobios, 35, S.47-62) ist bekannt, daß z. B. Magnesium in bestimmten Zeitintervallen des Teilungszyklus verstärkt von den Zellen benötigt und aufgenommen wird. Demzufolge wird Magnesium zur Synchronisation der Hefekultur als limitierendes Substrat gewählt. Zunächst wird die zu synchronisierende Hefe nach einem Turbidostatprinzip und anschließend mit Mg++-1imitierender Steuerung im Laborfermentor gezüchtet. Die Kultivierungsbedingungen sollen denen beim geplanten Produktionsprozeß möglichst ähnlich sein. Das betrifft insbesondere die Biomassekonzentration und den O2-Eingang pro Einheit Kulturvolumen. Als Ausgangswerte für die Synchronisation werden dabei folgende Parameterbestimmt: die unter diesen Bedingungen mögliche maximale spezifische VVachstumsrate /x.max, die Ertragskoeffizienten des limitierenden Substrates sowohl bei /xmax, als auch bei mehreren spezifischen Wachstumsraten im Bereich von 0,01 ßmax bis 0,85|u,max. Diese Werte werden in der Form 1/Y = f(/x) graphisch dargestellt. Aus dieser Kurve wird der Mg++-Bedarf bei/xmax für 2-10% der in der Kultur vorhandenen Biomasse errechnet. Diese Menge ist die 1. Portion, die zu der im steady state befindlichen Mg"+-Iimitiert gesteuerten Kultur zugegeben wird. Nach dieser Zugabe wird der kontinuierliche Mg++-Zulauf abgestellt, wenn das limitierende Substrat nicht die C-Quelle ist. Die nächsten Zugaben erfolgen jeweils im Abstand von wenig mehr als I η2/μηΐ3χ Stunden. Die Mg++-Mengen werden von Zugabe zu Zugabe schrittweise erhöht. Bei großer Differenz (/u.max-D) können diese Schritte entsprechend größer gewählt werden. Ist ein ausreichender Synchronisationsgrad erreicht, bleiben die Mg++-Mengen je Zugabe konstant. Zu diesem Zeitpunkt wird die Generationszeit der synchron wachsenden Zellen ga und die Durchflußrate der Gesamtkultur Da bestimmt. Daraus wird der Faktor b = a · I n2 = gaDa berechnet. Bei dem relativ langsamen Übergang dieser Kulturen in den streng limitierten Zustand, der für die Einleitung der Synchronisation erforderlich ist, bedeutet das erhebliche Zeiteinsparung bei den Stufen mit höherem Kulturvolumina. Der Faktor b muß jedoch in jeder Stufe neu ermittelt werden. Entsprechend dem Steuerprinzip muß diese Bestimmung aber im Prinzip nur jeweils für eine Generationszeit durchgeführt werden. Nur wenn die Möglichkeit einer Veränderung von YH+ besteht, durch die sich die Biomassekonzentration und damit der Limitations- und Synchronisationsgrad der Kultur verändern würde, ist eine Überprüfung bzw.
