Anwendungsgebiet der Erfindung:
Die Erfindung betrifft Aerosolzubereitungen und insbesondere wirkstoffhaltige Aerosolzubereitungen mit Fluorchlorkohlenwasserstoff als Treibmittel zur topischen medizinischen Anwendung oder zur endopulmonalen oder nasalen Inhalation sowie für Pesticide.
Charakteristik der bekannten technischen Lösungen: Aerosolzubereitungen enthalten im allgemeinen ein Gemisch aus Fluorchlorkohlenwasserstoffen, z.B. Trichlormonofluormethan (Treibmittel 11), Dichlortetrafluorethan (Treibmittel 114) und Dichlordifluormethan (Treibmittel 12). Der Wirkstoff ist entweder als Lösung in der Aerosolzubereitung oder als Dispersion feiner Teilchen vorhanden. Zur endopulmonalen oder nasalen Inhalation sind Teilchen überwiegend im Größenbereich von 2 bis 5 μΐη erforderlich.
Es gibt sehr wenig Wirkstoffe, die in Fluorchlorkohlenwasserstoff-Aerosoltreibmitteln allein löslich sind. Im allgemeinen ist es notwendig, ein polares Co-Lösungsmittel, wie Ethanol, zu verwenden, um eine Lösung bzw. Solubilisierung des Wirkstoffes zu erreichen. Die erhaltenen Lösungen kön-

nen jedoch chemisch instabil sein aufgrund einer Reaktion zwischen dem Co-Lösungsmittel und dem Wirkstoff oder dem Co-Lösungsmittel und dem Treibmittelsystem.
Darüber hinaus kann, wenn große Anteile an Co-Lösungsmittel, z.B. Ethanol, erforderlich sind, um eine Lösung des Wirkstoffes zu erreichen, die Tropfengröße des erhaltenen Sprays zu groß sein für bestimmte Anwendungen, insbesondere für die endopulmonale Inhalationstherapie.
Eine Suspension des Wirkstoffes in Aerosoltreibmitteln wird erreicht durch Pulverisieren bzw. Zerkleinern des Wirkstoffes auf den gewünschten Teilchengrößebereich und anschließendes Suspendieren der Teilchen in Treibmitteln mit Hilfe' eines grenzflächenaktiven Mittels. Die Nachteile dieser Technik liegen darin, daß Wirkstoffteilchen agglomerieren, in der Größe zunehmen oder an der Oberfläche des Behälters, in dem die Zubereitungen vor dem Dispergieren gelagert werden, adsorbiert werden können. Ferner ist es erforderlich, das Produkt vor der Anwendung zu bewegen bzw. zu schütteln, um die Dispersion der Zubereitung und Gleichmäßigkeit der Dosierung sicherzustellen.
Ziel der Erfindung:
Ziel der Erfindung istt ein alternatives Verfahren zum Einbau von Wirkstoffen in Fluorchlorkohlenwasserstoff-Treibmittel für Aerosole.
Darlegung des Wesens der Erfindung:
Die Erfindung betrifft daher eine Aerosolzubereitung, umfassend ein oder mehrere Fluorchlorkohlenwasserstoff-Treibmittel für Aerosole, ein Glycerinphosphatid und einen Wirkstoff, wobei der Wirkstoff in dem Gemisch gelöst ist.
Das Glycerinphosphatid kann irgendeine der folgenden Verbindungen sein: Phosphatidylcholin (Lecithin), Phosphatidy!ethanolamin (Cephalin), Phosphatidylinositol, Phosphatidylserin, Diphosphatidylglycerin oder Phosphatidsäure.
überraschenderweise hat es sich gezeigt, daß Glycerinphosphatide zu einer vollständigen Lösung bestimmter Wirkstoffe in Fluorchlorkohlenwasserstoff-Treibmitteln führen. Phosphatidylcholin (Lecithin) wurde angewandt als grenzflächenaktives Mittel in Aerosolzubereitungen, enthaltend suspendierte feste Wirkstoffteilchen. Es wurde jedoch nicht erkannt, daß diese spezielle Verbindung die Löslichkeit bestimmter Wirkstoffe in Fluorchlorkohlenwasserstoff-Treibmitteln so erhöhen kann, daß diese dann in gelöster Form im Treibmittel vorhanden sind.
Es hat sich gezeigt, daß Wirkstoffe, die eine zumindest sehr geringe Löslichkeit in Fluorchlorkohlenwasserstoff-Treibmitteln aufweisen, eine verstärkte Löslichkeit in diesen Treibmitteln in Gegenwart von Glycerinphosphatid besitzen. Es wird angenommen, daß diese verstärkte Löslichkeit darauf beruht, daß der Wirkstoff in echter Lösung sich assoziiert mit umgekehrten Micellen (reverse micelles) des Glycerinphosphatids, wodurch es möglich wird, daß weiterer Wirkstoff sich in dem Treibmittel löst. So wird angenommen, daß der Lösungsprozeß entsprechend dem folgenden Schema abläuft: ;

Während die erfindungsgemäßen Mittel für das Auge den Anschein echter Lösungen haben, da keine disperse Phase sichtbar ist, sind es korrekter micellare Lösungen.
Die erfindungsgemäßen Zubereitungen können hergestellt werden durch Bildung eines Konzentrats von Glycerinphosphatid mit einem Wirkstoff und Treibmittel 11. Das Konzentrat kann durch einfaches Vermischen unter Bewegen (Rühren und/oder Schütteln) und gegebenenfalls unter Wärmen (beispielsweise auf 5O0C), bis eine vollständige Lösung des Wirkstoffes eingetreten ist, hergestellt werden. Das Konzentrat kann dann mit dem Rest der Treibmittelzubereitung, z.B. mit den Treibmitteln 12 und 114, vermischt werden.
Phosphatidylcholin ist das geeignetste Glycerinphosphatid zur Anwendung im Hinblick auf seine geringe Toxizität und hohe Wirksamkeit bei dem Lösen des Wirkstoffs. Phosphatidylcholin, das aus Sojabohnenlecithin gewonnen und gereinigt wird, :ist leicht kommerziell zugänglich und geeignete Produkte sind u.a. Epikuron 200 (Lucas-Meyer) und Lipoid SlOO (Lipoid KG). Beide Produkte besitzen einen Phosphatidylcholingehalt von mehr als 95 %.
Es hat sich gezeigt, daß bestimmte Wirkstoffe, die in Fluorchlorkohlenwasserstoff-Treibmitteln allein praktisch unlöslich sind, in dem System Treibmittel/Glycerinphosphatid löslich gemacht werden können durch Zusatz einer kleinen Menge eines Co-Lösungsmittels, wie Ethanol.
Es wird angenommen, daß das Co-Lösungsmittel die anfängliche Lösungsstufe bei dem Lösungsprozeß verstärkt. Bestimmte kommerziell erhältliche Formen von Lecithin enthalten neben dem Phosphatidylcholin als Verunreinigung Ethanol. Bei derartigen Verbindungen, z.B. Lipoid S45, kann das Ethanol ebenfalls die Löslichkeit des Wirkstoffs erhöhen.
Geeignete Wirkstoffe, die erfindungsgeinäß angewandt werden können, umfassen solche Verbindungen, die zumindest eine sehr geringe Löslichkeit in Fluorchlorkohlenwasserstoff-Treibmitteln besitzt. Im allgemeinen liegt der Wirkstoff in Form eines Esters, einer Base oder eines freien Alkohols vor. Stark polare ionische Salze von Wirkstoffen sind weniger geeignet, da es unter Umständen nicht möglich ist, den Wirkstoff in ausreichender Menge in Lösung zu bringen, selbst in Gegenwart einer kleinen Menge eines Co-Lösungsmittels.
Beispielhafte Wirkstoffe umfassen Pesticide sowie Arzneimittel, wie Steroide, z.B. Beclomethason-dipropionat, Betamethason-dipropionat, -acetat, -valerat und freien Alkohol. Andere Arzneimittel sind u.a. Salbutamol-base, Atropin-base, Prednisolon, Formoterol-base, -hydrochlorid, -fumarat und -hemisulfat. j

Das Verhältnis Wirkstoff zu Glycerinphosphatid zu;;Co-Lösungsmittel (soweit erforderlich) zu Fluorchlorkohlenwasserstoff-Treibmittel hängt von zahlreichen Kriterien ab:
1. Erforderliche Konzentration an Wirkstoff in der fertigen Zubereitung.
2. Löslichkeit des Glycerinphosphatids in dem speziellen Gemisch von Fluorchlorkohlenwasserstoff-Treibmitteln.
3. Erforderliche Tropfengröße und Verdampfungseigenschaften des ausgestoßenen Sprays. Für Inhalationszwecke sind die optimalen Gehalte an Glycerinphosphatid und Treibmittel 11 die geringstmöglichen Gehalte, um eine stabile Lösung zu erzielen. Höhere Gehalte dieser Komponenten führen zu einer Zunahme der Tropfengröße des Sprays beim Freisetzen aufgrund einer Verringerung der Flüchtigkeit der Zubereitung.
4. Löslichkeit des Wirkstoffes in den Treibmitteln oder Treibmittel/Co-Lösungsmittel.
Ein weiter Bereich von Treibmitteln kann· in den erfindungsgemäßen Zubereitungen verwendet werden, wie z.B.: Treibmittel 11 Trichlormonofluormethan Treibmittel 12 Dichlordifluormethan Treibmittel 13 Monochlortrifluormethan Treibmittel 21 Dichlormonofluormethan Treibmittel 22 Monochlordifluormethan Treibmittel 113 Trichlortrifluorethan Treibmittel 114 Dichlortetrafluorethan

Außer dem Fluorchlorkohlenwasserstoff-Treibmittel für Aerosole kann die Zubereitung anderer Treibmittel, z.B. DME (Dimethylether), enthalten.
Im allgemeinen können die Mittel, enthaltend Wirkstoff, Glycerinphosphatid und Treibmittel, so hergestellt werden, daß sie die einzelnen Bestandteile in den folgenden allgemeinen Gewichtsverhältnissen enthalten:
Wirkstoff : Glycerinphosphatid 1 bis 500 : 100
Glycerinphosphatid : Treibmittel 0,01 bis 20 : 100
Für viele Wirkstoffe kann das Gewichtsverhältnis Wirkstoff zu Glycerinphosphatid allgemein im Bereich von 1 bis 30 : 100 liegen und das Gewichtsverhältnis von Glycerinphosphatid zu Treibmittel im Bereich von 0,01 bis 10 : 100. Vorzugsweise liegt das Gewichtsverhältnis von Wirkstoff zu Glycerinphosphatid im Bereich von 2 bis 10 : 100 und das Gewichtsverhältnis von Glycerinphosphatid zu Treibmittel im Bereich von 0,01 bis 3 : 100.
Ausführungsbeispiele:
Die Erfindung wird durch die folgenden Beispiele näher erläutert.

Das Mittel wurde hergestellt durch Vermischen der Komponenten (a) bis (c) unter etwa 10 Minuten langem Rühren bei einer Temperatur von 25°C. Anschließend wurde das Konzentrat mit der Komponente (d) bei einer Temperatur vermischt, die der Fülltechnik entsprach, allgemein im Bereich von -60 bis +200C. Die erhaltene Zubereitung war eine stabile Lösung.

Die Zubereitung wurde wie in Beispiel 1 hergestellt und ergab eine stabile Lösung.
Es wurde eine Reihe von stabilen Lösungen hergestellt, die geeignet waren als Konzentrate zur Herstellung von Aerosolzubereitungen. Jedes Konzentrat enthielt die folgenden Bestandteile in den angegebenen Gewichtsverhältnissen Wirkstoff zu Epikuron 200 zu Treibmittel 11 1:14:270. Die angewandten Wirkstoffe waren Prednisolon, Betamethason-acetat, Betamethason-valerat, Betamethason-dipropionat und Betamethason als freier Alkohol.

Vitamin-E-acetat und Ascorbyl-palmitat wurden als Antioxidantien zugegeben und verschlechterten die physikalischen Eigenschaften der Lösungen nicht.
Die Zubereitungen wurden hergestellt durch Vermischen des Wirkstoffs, grenzflächenaktiven Mittels, Treibmittels 11 und Antioxidans (soweit vorhanden) unter Rühren bis zu 6
ι Stunden bei ,einer Temperatur von 45 bis 5 00C. Anschließend wurde die erhaltene Lösung bei einer für die Abfüllmethode geeigneten Temperatur mit Treibmittel 12 vermischt unter Bildung einer Lösung.
Es wurde eine Reihe stabiler Lösungen hergestellt, die geeignet waren zur Verwendung >als Konzentrate zur Herstellung von Aerosolzubereitungen. Jedes Konzentrat enthielt Wirkstoff Lipoid SlOO und Treibmittel 11 in einem Gewichtsverhältnis von 1:7:135. Die Wirkstoffe waren:
Diazepam
Lorazepam
Propranolol-hydrochlörid
Hydrocortison
Fluocinolon-acetonid
Triamcinolon-acetonid.
Es wurden in allen Fällen klare stabile Lösungen erhalten. Wenn entsprechende Zubereitungen unter Weglassung des Lipoid SlOO hergestellt wurden, verblieb jeder Wirkstoff in Suspension.

Verwendung von Co-Lösungsmittel zur Unterstützung
des Inlosungbringens
Es wurde eine Zubereitung hergestellt, bestehend aus XyIometazolin-hydrochlorid, Lipoid SlOO und Treibmittel 11 in einem Gewichtsverhältnis von 1:7:135. Eine entsprechende Zubereitung wurde hergestellt, bei der das Lipoid SlOO weggelassen wurde. Nach 4-stündigem Bewegen und Erhitzen auf 500C war eine beträchtliche Menge Wirkstoff der beiden Zubereitungen noch in Suspension. Dann wurden 4 Gew.-% Ethanol zu beiden Zubereitungen gegeben. Nach 15 Minuten war die Lipoid SlOO enthaltende Zubereitung eine klare Lösung. Bei der Zubereitung, bei der das Lipoid SlOO weggelassen worden war, war keine sichtbare Änderung eingetreten. Dieses Ergebnis zeigt die Wirksamkeit einer kleinen Menge Co-Lösungsmittel zur Beschleunigung der anfänglichen Lösungsstufe bei dem Phospholipid-Lösungsprozeß.

Die Auflösung des Konzentrats, enthaltend Lorazepam, Lipoid SlOO und Treibmittel 113, wurde erreicht durch 10 Minuten langes Erwärmen auf 500C. Treibmittel 115 und Treibmittel 22 wurden dann mit dem Konzentrat zusammengegeben, wobei eine physikalisch stabile Lösung entstand.
Verwendung von Treibmittel 500 (Azeotrop)
in solubilisierter Zubereitung Es wurde die folgende Zubereitung hergestellt:

Bei den Konzentraten Treibmittel 11/Lecithin/Wirkstoff trat bei Raumtemperatur schnell Lösung ein. Die beiden Lösungszubereitungen waren bei -600C stabil und erlaubten das kalte Einfüllen bei der Herstellung der unter Druck stehenden Abgabepackungen (Spraydosen).
Inlösungbringen von Lacicorton Es wurden die folgenden Zubereitungen hergestellt;

Bei den Konzentraten Treibmittel 11/Lecithin/Wirkstoff trat bei Raumtemperatur leicht Lösung ein. Die Zubereitung (a) war bei -600C stabil und die Zubereitung (b) war bei -500C stabil, was die Anwendung des Kalteinfüllverfahrens bei der Herstellung der Spraydosen erlaubte.
Verwendung von Glycerinphosphatiden Es wurden die folgenden Zubereitungen hergestellt:

Die Zubereitungen waren jeweils stabile klare Lösungen, die geeignet waren zur Verwendung als Konzentrat bei der Herstellung von Aerosolzubereitungen.
Claims (28)
- lAo-60 025 -. a,Riker Laboratories, Inc.Erfindungsanspruch1. Aerosolzubereitung, umfassend ein oder mehrere Fluorchlorkohlenwasserstoff-Treibmittel sowie einen Wirkstoff, gekennzeichnet dadurch, daß sie zusätzlich Glycerinphosphatid enthält.
- 2. Zubereitung nach Punkt !,gekennzeichnet dadurch, daß das Glycerinphosphatid ausgewählt ist aus Phosphatidylcholin, Phosphatidylethanolamin, Phosphatidylinositol, Phosphatidylserin, Diphosphatidylglycerin, Phos- . phatidsäure und deren Gemischen.
- 3. Zubereitung nach Punkt 2, gekennzeichnet dadurch, daß das Glycerinphosphatid Phosphatidylcholin ist.
- 4. Zubereitung nach einem der Punkte 1 bis 3, gekennzeichnet dadurch, daß das Glycerinphosphatid gereinigt ist.
- 5. Zubereitung nach einem der Punkte 1 bis 4 , gekennzeichnet dadurch, daß sie Trichlormonofluormethan, Glycerinphosphatid und einen Wirkstoff enthält.
- 6. Zubereitung nach einem der vorangehenden Punkte, gekennzeichnet dadurch, daß das Verhältnis von Glycerinphosphatid zu Trichlormonofluormethan 0,01 bis 20:100 ist.
- 7. Zubereitung nach einem der vorangehenden Punkte, gekennzeichnet dadurch, daß das Verhältnis von Glycerinphosphatid zu Trichlormonofluormethan 0,01 bis 10:100 ist.22tÄAi1986*2dS*:i24 1AirlAo-60 025
- 8. Zubereitung nach einem der vorangehenden Punkte, gekennzeichnet dadurch, daß das Verhältnis von Glycerinphosphatid zu Trichlormonofluormethan 0,01 bis 3:100 ist.
- 9. Zubereitung nach einem der vorangehenden Punkte, gekennzeichnet dadurch, daß sie ein oder mehrere Treibmittel, ausgewählt aus Trichlormonofluormethan, Dichlordifluormethan, Monochlortrifluormethan, Dichlormonofluormethan, Monochlbrdifluormethan, Trichlortrifluorethan, Dichlortetrafluorethan, Monochlorpentafluorethan und ein Azetrop aus 73,8 % Dichlordiefluormethan und 2 6,2 % 1,1-Difluorethan, enthält.
- 10. Zubereitung nach einem der vorangehenden Punkte, gekennzeichnet dadurch, daß das Verhältnis von Wirkstoff zu Glycerinphosphatid 1 bis 500:100 ist.
- 11. Zubereitung nach einem der vorangehenden Punkte, gekennzeichnet dadurch, daß das Verhältnis von Wirkstoff zu Glycerinphosphatid 1 bis 30:100 ist.
- 12. Zubereitung nach einem der vorangehenden Punkte, gekennzeichnet dadurch, daß das Verhältnis von Wirkstoff zu Glycerinphosphatid 2 bis 10:100 ist.
- 13. Zubereitung nach einem der vorangehenden Punkte, gekennzeichnet dadurch, daß sie zusätzlich eine kleine Menge eines Co-Lösungsmittels zur Erleichterung des Inlösungbringens enthält.
- 14. Zubereitung nach einem der vorangehenden Punkte, gekennzeichnet dadurch, daß der Wirkstoff ausgewählt ist aus Beclomethason-dipropionat, Betamethasondipropionat, -acetat, -valerat und der freien Base davon, Salbutamol-base, Atropin-base und Prednisolon.ΊΑο-60 025 -S-
- 15. Zubereitung nach einem der Punkte 1 bis 13, gekennzeichnet dadurch, daß der Wirkstoff ausgewählt ist aus Formoterol-base, -hydrochlorid, -hemisulfat und -fumarat.
- 16. Zubereitung nach einem der Punkte 1 bis 13, gekennzeichnet dadurch, daß der Wirkstoff ausgewählt ist aus Diazepam, Lorazepam, Propranolol-hydrochlorid, Hydrocortison, Fluocinolon-acetonid, Triamcinolonacetonid, Xylometazolin-hydrochlorid, Bitolterol-mesylat und Lacicorton.
- 17. Zubereitung nach einem der vorangehenden Punkte, gekennzeichnet dadurch, daß sie in einer unter Druck stehenden Spraydose enthalten ist.
- 18. Verfahren zum Inlösungbringen eines schwerlöslichen Wirkstoffes in einem Fluorchlorkohlenwasserstoff-Treibmittel zur Herstellung einer Aerosolzubereitung nach einem der Punkte 1 bis 17 durch Vermischen des Wirkstoffes mit einem Fluorchlorkohlenwasserstoff-Treibmittel, gekennzeichnet dadurch, daß das Vermischen in Gegenwart einer wirksamen Menge eines Glycerinphosphatids durchgeführt wird.
- 19. Verfahren nach Punkt 18, gekennzeichnet dadurch, daß das Glycerinphosphatid ausgewählt ist aus Phosphatidylcholin, Phosphatidy!ethanolamin, Phosphatidylinositol, Phosphatidylserin, Diphosphatidylglycerin und Phosphatidsäure.
- 20. Verfahren nach Punkt 18, gekennzeichnet dadurch, daß das Glycerinphosphatid Phosphatidylcholin ist.
- 21. Verfahren nach einem der Punkte 18 bis 20, gekennzeichnet dadurch, daß das Glycerinphosphatid gereinigt ist.Z41lAo-60 025 -X-
- 22. Verfahren nach einem der Punkte 18 oder 19, gekennzeichnet dadurch, daß Trichlormonofluormethan, Glycerinphosphatid und ein Wirkstoff unter Rühren miteinander vermischt werden.
- 23. Verfahren nach Punkt 21, gekennzeichnet dadurch, daß das Verhältnis von Glycerinphosphatid zu Trichlormonofluormethan 0,01 bis 20:100 beträgt.
- 24. Verfahren nach einem der Punkte 19 bis 21, gekennzeichnet dadurch, daß ein oder mehrere Treibmittel, ausgewählt aus Trichlormonofluorethan, Dichlordifluormethan, Monochlortrifluormethan, Dichlormonofluormethan, Monochlordxfluormethan, Trichlortrifluorethan, Dichlortetrafluorethan, Monochlorpentafluorethan und ein Azetrop aus 73,8 % Dichlordifluormethan und 25,2 % 1,1-Difluorethan, verwendet werden.
- 25. Verfahren nach einem der Punkte 18 bis 24, gekennzeichnet dadurch, daß zusätzlich eine kleine Menge eines Co-Lösungsmittels zur Erleichterung des Inlösungbringens verwendet wird.
- 26. Verfahren nach einem der Punkte 18 bis 25, gekennzeichnet dadurch, daß der Wirkstoff ausgewählt ist aus Beclomethason-dipropionat, Betaine tha son-dipropionat, -acetat, -valerat und der freien Base davon, Salbutamol-base, Atropin-base und Prednisolon.
- 27. Verfahren nach einem der Punkte 18 bis 25, gekennzeichnet dadurch, daß der Wirkstoff ausgewählt ist aus Formoterol-base, -hydrochlorid, -hemisulfat und -fumarat.
- 28. Verfahren nach einem der Punkte 18 bis 25, gekennzeichnet dadurch, daß der Wirkstoff ausgewählt ist aus Diazepam, Lorazepam, Propranolol-hydrochlorid, Hydrocortison, Fluocinolon-acetonid, Triamcinolon-acetonid, Xylometazolin-hydrochlorid, Bitolterol-mesylat und Lacicorton.η η τ,.« η i«(η 0 C * «3 Λ ß Λ il Π
