Abstract
Die Erfindung betrifft ein Verfahren zur Herstellung von substituierten und disubstituierten Pyridin-2,3-dicarboxylatestern, die nuetzliche Zwischenverbindungen fuer die Herstellung von Herbiziden, und zwar von 2-(2-Imidazolin-2-yl)nicotinsaeuren, -estern und -salzen darstellen. Beispielsweise werden Ethacrolein, Diethyl-3-chlor-2-oxo-butandioat und Ammoniumsulfat in Ethanol miteinander umgesetzt, um nach Extraktion mit Ethylacetat und Reinigung Diethyl-5-ethylpyridin-2,3-dicarboxylat zu erhalten.
Description
Die Erfindung betrifft ein Verfahren zur Herstellung von substituierten und disubstituierten Pyridin-2,3-dicarboxylat-estern.
In der Literatur beschriebene Verfahren zur Herstellung von 5,6-Dialkyl- und 5,6-Alkylarylpyridin-2,3-dicarboxylaten sind begrenzt, oftmals ist die Oxydation von Alkyl- oder Arylsubstituenten an den Positionen 2 und 3 erforderlich, um Diaeide zu erhalten. R.Jones, J. Am. Chem. Soc. 73,4380 (1951) beschreibt ein Verfahren, bei dem die Reaktion eines primären Enamins 6-Alkylpyridin 2,3-dicarboxylate ergibt, die elektronenanziehende Substituenten wie COCH3, CN oder CO2Et in der 5-Stellung enthalten. Dieses Verfahren kann zur Herstellung von 5,6-Dialkyl-oder Alkylarylpyridin 2,3-dicarboxylaten nicht angewandt werden, weil primäre Enamine ohne elektronenanziehende Substituenten nicht ohne weiteres hergestellt werden können, d. h. die Reaktion von Ammoniak mit aliphatischen Ketonen erzeugt Imine, die die Enamine nicht tautomerisieren und die, wenn sie nicht in situ festgehalten werden, polymerisieren. Durch andere Verfahren, bei denen Malononitrile vewendet werden, wie z. B. den JA-PS 7869.835 beschriebenen und dem von J. I. DeGrawJ. Het. Chem. 19,1461 (1982), können 5-Alkyl- und 5,6-Dialkylpyridine gewonnen werden, aber nicht direkt mit der verlangten 2,3-Dicarboxylat-Substitution. Pyridin-2,3-dicarboxylate sind nützliche Zwischenverbindungen für die Herstellung von herbiziden 2-(2-lmidazolin-2-yl)nicotinsäuren,-estern und-salzen, wie sie in der EU-PA Nr. 81103638.3, eingereicht am I.Dezember 1981, dargelegt werden, und wie in dem folgenden Fließschema I gezeigt wird,

Die Kondensation mit a, ^-ungesättigten Systemen durch Michael-Addition einiger Halogenmethylen-Kohlenstoffnucleophile, die durch elektronenanziehende Gruppen aktiviert sind, ist allgemein bewiesen, aber die entsprechende Reaktion von a-Halogen-/3-ketoestern, wie die von Diethyl 3-chlor-2-oxo-butandioat mit Acrolein hat nach den Berichten von P. Bouvier und H. Gault, Bull. Soc. Chim. Fr. 711 (1963) zu Gemischen geführt.
Eine Aufgabe der Erfindung betrifft die Schaffung eines Verfahrens zur Herstellung von substituierten und disubstituierten Pyridin-2,3-dicarboxylaten und 2-Alkylnicotinaten unter Anwendung von a-Halogen-/3-ketoestern und α, ^-ungesättigten Aldehyden oder Ketonen in Gegenwart eines Ammoniumsalzes.

Die Erfindung stellt ein neuartiges Verfahren zur Herstellung substituierter und disubstituierter Pyridin-2,3-dicarboxylate der 7

worin R3 Wasserstoff, Halogen, gerades oder verzweigtes C1-C6-AIkYl, Alkenyl, Phenyl oder substituiertes Phenyl ist; R4 und R7Je Wasserstoff, gerades oder verzweigtes C1-C6-Alkyl, Alkenyl, Phenyl oder substituiertes Phenyl sind, während R5 und R6JeC1-C4-Alkyl bedeuten und R8 — CO2R6ISt, worin R6 wie oben definiert ist, das in der Umsetzung einer der Formel Il .

worin R6 und R8 die oben erläuterte Bedeutung haben und X Halogen ist, mit einem a, ^-ungesättigten Aldehyd oder Keton der Formel III .

worin R3, R4 und R7 die für Formel I beschriebene Bedeutung haben, in Gegenwart eines Minim ums von 2 Mol äquivalenten eines Ammoniumsalzes in einem organischen Lösungsmittel, in einem zwischen Umgebungstemperatur und dem Siedepunkt des Lösungsmittels liegenden Temperaturbereich besteht, bis die Reaktion im wesentlichen abgeschlossen ist, wie in dem folgenden Fließschema I dargestellt wird.

Für die Verwendung bei dem erfindungsgemäßen Verfahren geeingnete Lösungsmittel umfassen: Wasser, Alkohole, chlorierte Kohlenwasserstoffe, Kohlenwasserstoffe, aromatische Kohlenwasserstoffe, Ether, organische Säuren, Ester und aprotische Lösungsmittel wie Acetonitril.
So können Pyridin-2,3-dicarboxylate, die Substituenten in den Positionen 4, 5 und 6 tragen, zweckmäßig hergestellt werden, indem im wesentlichen äquimolare Mengen eines a-Halogenketoesters der Formel Il und eines α, ß-ungesättigten Aldehyds oder Ketons der Formel III mit einem Minimum von 2-Moläquivalenten eines Ammoniumsalzes in einem geeigneten Lösungsmittel vermischt werden und das resultierende Reaktionsgemisch bei einer im Bereich von Umgebungstemperatur bis zur Siedetemperatur des Lösungsmittels liegenden Temperatur, und vorzugsweise unter Rückfluß, gerührt wird, bis die Reaktion im wesentlichen abgeschlossen ist, und das auf diese Weise gebildete 4-substituierte, 4,5-disubstituierte, 5-substituierte, 6-substituierte oder 5,6-disubstituierte Pyridin-2,3-dicarboxylat durch Standard-Labortechniken wie Extraktion, Eindampfung oder Säulenchromatographie isoliert wird.
Nicotinate der Formel I sind auch nützliche Zwischenverbindungen für die Herstellung von herbiziden 2-(2-lmidazolin-2-yl)-nicotinsäuren, -estern und -salzen, und zwar durch Umsetzung mit einem Aminocarboxamid in Gegenwart von mindestens Äquivalenten Schwefel, wie in US-PS 4474962 beschrieben wird, oder durch Oxydation von R8, zum Beispiel mit Hilfe des in US-PS 4459409 beschriebenen Verfahren, um die Pyridin-2,3-dicarbonsäureverbindungen der obigen Formel I zu gewinnen.
Um die Erfindung leichter verstehen zu können, werden die folgenden Beispiele hauptsächlich zur Erläuterung bestimmter spezifischer Einzelheiten gebracht, durch die die Erfindung aber in keiner Weise eingeschränkt werden soll.
a Ein gerührtes Gemisch von Ethacrolein (4,2g, 0,05 Mol), Diethyl 3-chlor-2-oxo-btatandioat (11,2g, 0,05 Mol) und Ammoniumsulfamat (15,4g, 0,135 Mol) in Ethanol (37ml) wird am Rückfluß gekocht. Nach 15 Stunden wird das Gemisch auf Raumtemperatur gekühlt und das Lösungsmittel durch Destillation unter reduziertem Druck entfernt. Der Rückstand wird mit Wasser behandelt und mit Ethylacetat extrahiert. Die organische Phase wird abgetrennt und unter Vakuum eingeengt, und der Rückstand wird mit Hilfe der Säulenchromatographie auf Silicagel unter Verwendung von 4:1 Hexan-Ethylacetat als Eluierungsmittei gereinigt und ergibt 10,8g (Ausbeute 75%) des Titelproduktes in Form eines Öls, von dem durch Gaschromatographieanalyse gezeigt wird, daß es zu 95% rein ist.
b Wird die obige Reaktion mit Essigsäure als Lösungsmittel und unter Verwendung von Diethyl 3-brom-2-oxo-butandioat anstelle der Chlorbindung durchgeführt, entsteht das gleiche Produkt in einer Ausbeute von 48%.
Durch Anwendung der Verfahrensweise von Beispiel 1 und Verwendung des entsprechend substituierten a, /3-ungesättigten Aldehyds oder Ketons von Formel III anstelle von Ethacrolein entstehen die in der folgenden Tabelle I aufgeführten Pyridin-2,3-dicarboxylate.

Ammoniumsalz Lösungsmittel
Äquimolare Mengen von Ethacrolein und Diethyl S-chlor^-oxo-butandioat in verschiedenen Lösungsmitteln werden in Gegenwart von 2,7 Moläquivalenten verschiedener Ammoniumsalze 15 Stunden lang unter Rühren am Rückfluß gekocht. Die Produkte werden wie in Beispiel 1 isoliert und mit Hilfe der Gaschromatographie analysiert. Die Ergebnisse dieser Versuche, die inderfolgenden Tabelle Il zusammengefaßt sind, zeigen die Wirksamkeit verschiedener Ammoniumsalze und Lösungsmittel bei dem erfindungsgemäßen Verfahren.

Durch Substitution der äquivalenten Menge von Dipropyl3-chlor-2-oxo-butandioat für Diethyl 3-chlor-2-oxo-butandioat und die Verwendung von Propanol als Lösungsmittel wird Dipropyl 5-ethylpyridin-2,3-dicarboxylat gewonnen. Dimethyl 3-ch|or-2-oxobutandioat ergibt Dimethyl 5-ethylpyridin-2,3-dicarboxylat, wenn die Reaktion in Methanol vorgenommen wird.
Ein Gemisch von Ethacrolein (12,8g, 0,152 Mol) und Ammoniumacetat (24,4g, 0,304 Mol) in 50cm3 Acetonitril wird bei Raumtemperatur gerührt, und eine Lösung von Ethyl 2-chloracetoacetat (25 g, 0,152 Mol) in 30cm3 Acetonitril wird im Laufe von 15 Minuten tropfenweise zugegeben. Das Reaktionsgemisch wird 16 Stunden la ng unter Rückfluß gekocht, gekühlt und zwischen Wasser und Ethylacetat aufgeteilt. Die organische Phase wird unter Volumen eingeengt und auf Silicagel unter Verwendung von 9:1 Hexan-Ethylacetatchromatographiert und ergibt 12,7'g der Titelverbindung.
Claims (11)
- Patentansprüche:1. Verfahren zur Herstellung von substituierten und disubstituierten Pyridin-2,3-dicarboxylaten der Formel I (worin R3 Wasserstoff, Halogen, gerades oder verzweigtes Ci-C6-Alkyl, Alkonyl, Phenyl oder substituiertes Phenyl ist; R4 und R7 je Wasserstoff, gerades oder verzweigtes CvC6-AIkyl. Alkenyl, Phenyl oder substituiertes Phenyl sind; R5 und R6 je CvC4-Alkyl sind, und R8 CO2 R6 bedeutet, worin R6 wie oben definiert ist, gekennzeichnet dadurch, daß ein a-Halogen-/3-ketoester der Formel Il. HX-C-CO2R6O=C-R
(II)worin X Halogen ist und R6 und R8 die oben erläuterte Bedeutung haben, mit einem a, ßungesättigten Aldehyd oder Keton der Formel lllf
R3-C=CHR7R4-C=O
(III)worin R3, R4 und R7 die für Formel I erläuterte Bedeutung haben, in Gegenwart eines Minimums von 2 Moläquivalenten eines Ammoniumsalzes in einem organischen Lösungsmittel, in einen zwischen Umgebungstemperatur und dem Siedepunkt des Lösungsmittels liegenden Temperaturbereich umgesetzt wird, bis die Reaktion im wesentlichen abgeschlossen ist. - 2. Verfahren nach Anspruch 1, gekennzeichnet dadurch, daß es sich bei dem Lösungsmittel um Wasser, einen Alkohol, Kohlenwasserstoff, chlorierten Kohlenwasserstoff, aromatischen Kohlenwasserstoff, Ether, organische Säure oder Ester, Dimethylsulfoxid, Dimethylformamid oder Acetonitril handelt.
- 3. Verfahren nach Anspruch 2, gekennzeichnet dadurch, daß die Reaktion in einem zwischen 30°C und 140°C liegenden Temperaturbereich vorgenommen wird.
- 4. Verfahren nach Anspruch 3, gekennzeichnet dadurch, daß es sich bei der Verbindung der Formel Il um Diethyl 3-chlor-2-oxobutandioat handelt.
- 5. Verfahren nach Anspruch 3, gekennzeichnet dadurch, daß 5-substituierte, 6-substituierte und 5,6-disubstituierte Pyridin-2,3-dicarboxylate hergestellt werden.
- 6. Verfahren nach Anspruch 3, gekennzeichnet dadurch, daß Diethyl 5-ethylpyridin-2,3-dicarboxylat hergestellt wird.
- 7. Verfahren nach Anspruch 3, gekennzeichnet dadurch, daß Diethyl 5-methylpyridin-2,3-dicarboxylat hergestellt wird.
- 8. Verfahren nach Anspruch 3, gekennzeichnet dadurch, daß Diethyl 5,6-dimethylpyridin-2,3-dicarboxylat hergestellt wird.
- 9. Verfahren nach Anspruch 3, gekennzeichnet dadurch, daß Diethylpyridin-2,3-dicarboxylat hergestellt wird.
- 10. Verfahren nach Anspruch 1, gekennzeichnet dadurch, daß Ethyl 5-ethyl-2-methylnicotinat hergestellt wird.
- 11. Verfahren nach Anspruch 1, gekennzeichnet dadurch, daß Ethyl 2,5-dimethylnicotinat hergestellt wird.
