Abstract
Die Erfindung betrifft ein Verfahren zur Herstellung bisher nicht zugaenglicher Aroxymethylchinoxaline (Formel IV), die als Zwischenprodukte fuer die Zubereitung biologisch aktiver Praeparate anwendbar sind. Die Synthese erfolgt durch Umsetzung von 2-(Halogenmethyl)chinoxalinen mit den aus den Phenolen ArOH gebildeten Phenolaten, wobei Ar einen beliebigen aromatischen Rest, bevorzugt unsubstituiertes bzw. ein- oder mehrfach substituiertes Phenyl oder Naphthyl, darstellt.
Description
Erfindungsgemäß wird die Aufgabe dadurch gelöst, daß 2-Halogenmethyl)chinoxaline der allgemeinen Formel I, in der Hai = Halogen, bevorzugt Cl oder Br, und R1 = H, Alkyl, Aryl bedeuten, mit den Phenolaten, die durch Einwirkung geeigneter Basen, bevorzugt Alkalihydroxid,-carbonat oder-hydrogencarbonat, auf die Phenole ArOH gebildet werden, wobei Ar bevorzugt der allgemeinen Formel Il oder III entspricht, in denen R2... R7 gleich oder verschieden sein können und unabhängig voneinander H, Hal, NO2, Alkyl, Alkoxy, CHO, CO-Alkyl, CO-Aryl bedeuten können, umgesetzt werden, wobei unter Bildung eines anorganischen Salzes, gegebenenfalls Alkalihalogenid, die Aroxymethylchinoxaline der allgemeine Formel IV, in der R1 die gleiche Bedeutung wie in der allgemeinen Formel I besitzt und Ar einen beliebigen aromatischen Rest darstellt, der bevorzugt der allgemeinen Formel Il oder 111 mit der gleichen Bedeutung für R2... R7 wie im eingesetzten Phenol entspricht, entstehen. ·

Die Umsetzungen werden bei Temperaturen von —80°C bis 25O0C, bevorzugt aber im Bereich von 1O0C bis 1000C, in der Schmelze, in Lösung oder in Suspension durchgeführt, wobei als Lösungsmittel bevorzugt handelsübliches Ethanol oder ein anderes Ethanol-Wasser-Gemisch benutzt wird. Die Reaktionspartner kommen entweder in äquimolarer Menge zum Einsatz, oder die billigere Komponente wird im geringfügigen Überschuß, bevorzugt in 1,1- bis 1,5facher molarer Menge, eingesetzt. Die Herstellung der benötigten Ausgangsstoffe mit der allgemeinen Formel I ist in der Literatur beschrieben (z. B. Z. obsc. chim. 29 [1959] 2763; J. org. Chemistry 31 [1966] 2613; Z. Chem. 22 [1982] 300; J. Amer. Chem. Soc. 86 [1964] 1830; HeIv. chim. Acta 29 [1946] 1247).

Aus 11,3Tl. Phenol und der äquimolaren Menge Alkalihydroxid bereitet man eineethanolische Phenolatlösung,die mit der warmen Lösung von 23,7Tl. 2-(Brommethyl)-3-methyl-chinoxalin in 350Tl. Ethanol versetzt wird. Unter Rühren wird das Gemisch 2 Stunden unter Rückfluß gekocht. Das bei der Reaktion entstehende Alkalibromid kann durch Filtration der heißen Reaktionslösung und/oder Zugabe von Wasser zum Reaktionsgemisch entfernt werden. Man engt die Lösung im Vakuum ein und saugt das Reaktionsprodukt kalt ab. Die Reinigung des Rohprodukts erfolgt durch Umkristallisation aus einem polaren und/oder einem unpolaren Lösungsmittel. Ausbeute 66%, Schmelztemperatur 80,5-82,50C (aus Hexan).

Nach dem Abkühlen saugt man das Produkt ab, wascht mit Wasser und kristallisiert aus Propanol bzw Toluen um Ausbeute 82%, Schmelztemperatur 151,5-152,5°C

3-Methoxy-4-(3-methyl-chinoxahn-2-ylmethoxy)benzaldehyd Analog Beispiel 1 mit18,5TI Vanillin, Ausbeute 84%, Schmelztemperatur 166-167,5°C (aus Ethanol).
2-(3-Methyl-chinoxahn-2-ylmethoxy)acetophenon Analog Beispiel 1 mit 16,3Tl 2-Hydroxy-acetophenon, Ausbeute 35%, Schmelztemperatur 102-1030C (aus Heptan)
2-(3-Methyl-chinoxalin-2-ylmethoxy)-5-methyi-benzophenon Analog Beispiel 1 mit 23Tl. г-НусІгоху-б-теШуІ-ЬепгорІіепоп, Ausbeute 64%, Schmelztemperatur 147-149°C (aus Ethanol).
2-(3-Methyl-chinoxalin-2-ylmethoxy)-4-methoxy-benzophenon Analog Beispiel 1 mit 25Tl. г-Нусігоху^-теіпоху-ЬепгорІіепоп, Ausbeute 60%, Schmelztemperatur 127-1280C (aus Tetrachlormethan).
2-Methyl-3-(naphth-1-yloxymethyl)-chinoxalin Analog Beispiel 1 mit 17,3Tl. a-Naphthol, Ausbeute 68%, Schmelztemperatur 133-1360C (aus Octan).
2-Methyl-3-(naphth-2-yloxymethyl)-chinoxalin Analog Beispiel 1 mit 17,3Tl. ß-Naphthol, Ausbeute 63%, Schmelztemperatur 120-1230C (aus Octan).
2-(4-Chlor-phenoxymethyl)-3-phenyl-chinoxalin Analog Beispiel 1 mit 15,4Tl. 4-Chlor-phenol und einer Lösung von 30Tl. 2-(Brommethyl)-3-phenyl-chinoxalin in 400Tl. Ethanol, Ausbeute 65%, Schmelztemperatur 101-102,50C (aus Hexan).
Claims (7)
1. Verfahren zur Herstellung von Aroxymethylchinoxalinen, gekennzeichnet dadurch, daß 2-(Halogenmethyl)chinoxaline mit Phenolaten zu den Aroxymethylchinoxalinen umgesetzt werden.
2. Verfahren nach Punkt 1, gekennzeichnet dadurch, daß die als Ausgangsstoffe eingesetzten 2-(Halogenmethyl)chinoxaline der allgemeinen Formel I entsprechen, wobei Hai = Halogen, bevorzugt Cl oder Br, und R1 = H, Alkyl oder Aryl bedeuten.

Verfahren nach Punkt 1, gekennzeichnet dadurch, daß als Reaktionspartner Phenolate dienen, die durch Einwirkung geeigneter Basen, bevorzugt Alkalihydroxid, -carbonat oder -hydrogencarbonat, auf die Phenole ArOH gebildet werden, wobei Ar bevorzugt der allgemeinen Formel Il oder III entspricht, in denen R2... R7 gleich oder verschieden sein können und unabhängig voneinander H, Hal, NO2, Alkyl, Alkoxy, CHO, CO-Alkyl oder CO-Aryl bedeuten.
4. Verfahren nach Punkt 1, gekennzeichnet dadurch, daß die beiden Reaktionspartner in äquimolarer Menge eingesetzt werden oder die billigere Komponente im geringfügigen Überschuß, bevorzugt in 1,1- bis 1,5facher molarer Menge, eingesetzt wird.
5. Verfahren nach Punkt 1, gekennzeichnet dadurch, daß die Umsetzungen bei Temperaturen von -800C bis 2500C, bevorzugt aber im Bereich von 10°C bis 1000C, durchgeführt werden.
6. Verfahren nach Punkt 1, gekennzeichnet dadurch, daß die Umsetzungen in der Schmelze, in Lösung oder in Suspension durchgeführt werden, wobei als Lösungsmittel bevorzugt handelsübliches Ethanol oder ein anderes Ethanol-Wasser-Gemisch benutzt wird.
7. Verfahren nach Punkt 1, gekennzeichnet dadurch, daß die als Endprodukte entstehenden Aroxymethylchinoxaline der allgemeinen Formel IV entsprechen, in der R1 die gleiche Bedeutung wie in der allgemeinen Formel I besitzt (s. Punkt 2!) und Ar einen beliebigen aromatischen Rest darstellt, der bevorzugt der allgemeinen Formel Il oder III mit der gleichen Bedeutung für R2... R7 wie im eingesetzten Phenol (s. Punkt 3!) entspricht.
Anwendungsgebiet der Erfindung
Die Erfindung betrifft ein Verfahren zur Herstellung von Aroxymethylchinoxalinen, die als Zwischenprodukte zur Zubereitung biologisch aktiver Präparate anwendbar sind.
Charakteristik der bekannten technischen Lösungen
Zur Darstellung von Aroxymethylchinoxalinen kommen insbesondere zwei Synthesewege in Betracht. Entweder wird eine reaktive Verbindung mit vorgefertigter Etherstruktur zum Chinoxalin cyclisiert, oder es wird ein geeignetes Chinoxalinderivat in einen Ether umgewandelt. Auf dem ersten Weg wurden bisher 2-Methyl-3-(2-phenoxy-ethyl)-bzw. -3-(ethoxymethyl)-chinoxalin synthetisiert (J. Chem.Soc. 1953,2822; 1956,2052); Aroxymethylchinoxaline sind auf diese Weise bisher nicht zugänglich; nachteilig für die Anwendung dieses Prinzips ist die schlechte Zugänglichkeit der reaktiven Ausgangsverbindung. Auf dem zweiten Weg wurden bisher Alkoxymethylchinoxaline hergestellt, und zwar entweder aus Chinoxalinmethanol und Alkylhalogenid (J. Amer. Chem. Soc. 58 [1936] 1894) — diese Reaktion gelingt mit Arylhalqgeniden nicht — oder aus Halogenmethylchinoxalinen und Alkoholat (Ger. Offen. 2242783 vom 8.3.73; Ger. Offen. 2722777 vom 8.12.77). Um auf diese Weise die Aroxy-Verbindungen zu synthetisieren, müßten an Stelle der Alkoholate Phenolate eingesetzt werden. Phenolate besitzen aber eine bedeutend geringere Nucleophile als die Alkoholate. So blieb dieses Syntheseprinzip bisher auf den Sonderfall des 3-(4-Nitro-phenoxymethyl)chinoxalin-2-ons beschränkt (Bull. Chem. Soc. Jpn. 56 [1983] 326). Aroxymethylchinoxaline mit unveränderten Ringstickstoff-Funktionen sind bisher unbekannt, es existiert bisher noch kein Verfahren zu ihrer Herstellung.
Ziel der Erfindung
Ziel der Erfindung ist es, ein Verfahren zur Herstellung von Aroxymethychinoxalinen, in denen beide Ringstickstoff-Funktionen unverändert vorliegen, zu finden, da derartige Verbindungen bisher nicht zugänglich sind.
Darlegung des Wesens der Erfindung
Aufgabe der Erfindung ist es, ein Verfahren zu finden, nach dem die bisher nicht zugänglichen Aroxymethylchinoxaline mit unveränderten Ringstickstoff-Funktionen hergestellt werden können.
