Description
Die Erfindung betrifft ein Verfahren zur in-vitro-Spaltung von refraktären DNA-Species. Es ist somit anwendbar auf dem Gebiet der Gentechnologie.The invention relates to a method for the in vitro cleavage of refractory DNA species. It is thus applicable in the field of genetic engineering.
Die ortsspezifische Spaltung von DNA-Molekülen mit Hilfe von Restriktionsendonukleasen ist eine Basismethode der Gentechnik. Auf diese Weise lassen sich definierte DNA-Fragmente gewinnen, die weiter analysiert oder gentechnisch neu rekombiniert werden. Das normale Vorgehen besteht in der in vitro-lnkubation von gereinigter DNA mit der Restriktionsendonuklease bei 370C (oder in einigen Fällen bei höheren Temperaturen) in Gegenwart von für jede Restriktionsendonuklease charakterischen Salz- und Kofaktorkonzentrationen bei entsprechendem pH-Wert (MANIATIS et al., Molecular Cloning — A Laboratory Manual. Cold Spring Harbor Laboratory Press, Cold Spring Harbor, 1982). Voraussetzung für die ortsspezifische Spaltung der DNA ist, daß die jeweiligen DNA-Moleküle die entsprechenen Erkennungsorte (spezifischen Basensequenzen) für die verwendete Restriktionsendonuklease besitzen. Es ist jedoch bekannt, daß DNA-Moleküle trotz des Besitzes solcher Erkennungsorte gegen die Restriktionsendonuklease resistent sein können (KRÜGER und BICKLE, Microbiol. Reviews 47,345-360 [1983]). Ein bekanntes Verfahren zur Spaltung solcher zunächst resistenten DNA mit Restriktionsendonukleasen existiert in den Fällen, wo die Resistenz der Erkennungsorte durch Methylierung oder anderweitige Abänderung der DNA-Basen (modifizierte oder sogenannte „seltene" Basen) in oder in der Nähe der Erkennungssequenz bedingt ist. In solchen Fällen läßt sich spaltbare DNA gewinnen, indem man diese vor der Isolierung und Restriktionsendonuklease-Behandlung in solchen Zellen vermehrt, in denen die Synthese von veränderten DNA-Basen nicht möglich ist. Die auf diese Weise vermehrte DNA, die nur die üblichen Basen Adenin, Guanin, Cytosin und Thymin enthält, läßt sich dann normal mit Restriktionsendonukleasen spalten (KRÜGER und BICKLE, siehe oben).The site-specific cleavage of DNA molecules using restriction endonucleases is a basic method of genetic engineering. Die molekularen Ursachen dafür sowie praktische Möglichkeiten zu ihrer Überwindung waren bisher nicht bekannt (KRÜGER und BICKLE, s.o.). Aufgabe der Erfindung ist es, ein Verfahren zur Spaltung resistenter DNA mit Restriktionsendonukleasen zu entwickeln, die sich bisher nicht spezifisch durch Restriktionsendonukleasen spalten lassen. Erfindungsgemäß wird diese Aufgabe gelöst, indem die Spaltung der resistenten DNA (DNA I) durch die Restriktionsendonuklease in Gegenwart einer zweiten, empfindlichen DNA-Species (DNA II) durchgeführt wird. Sowohl DNA I als auch DNA Il besitzen Erkennungsorte für die Restriktionsendonuklease, wobei unter den üblichen experimentellen Bedingungen (MANIATIS et al., siehe oben) bei Inkubation der Restriktionsendonuklease mit jeweils einer der beiden DNA-Species die DNA I resistent ist, die DNA Il aber normal ortsspezifisch spaltbar ist.The object of the invention is to develop a method for cleaving resistant DNA with restriction endonucleases which can not be cleaved specifically by restriction endonucleases. Das mengenmäßige Verhältnis von DNA Il zu DNA I ist so zu gestalten, daß die Gesamtzahl von Restriktionsendonuklease-Erkennungsorten, die in der DNA Il lokalisiert sind, mindestens so hoch oder höher ist als die Gesamtzahl der Erkennungsorte in der eingesetzten DNA I. Als DNA Il können sowohl biologisch replizierte DNA's als auch synthetisch hergestellte Oligonukleotid-Duplices eingesetzt werden. Nach Beendigung der Koinkubation beider DNA-Species und der Restriktionsendonuklease werden die entstandenen DNA-Fragmente durch Agarose- oder Polvacrylamidgelelektrophorese aufgetrennt.
