ß-Rhodomycin V
Durch das Patent 851 398 wird .ein Verfahren zur Herstellung antibiotisch wirksamer Stoffe geschützt, das
dadurch gekennzeichnet ist, daß man Streptomyceten, insbesondere Streptomyces purpurascens nov. spec, unter
aeroben Bedingungen auf oder in für die Züchtung von Streptomyceten an sich bekannten Nährmedien wachsen
läßt und aus den so gewonnenen Kulturen die Rhodomycine nach üblichen Arbeitsweisen gewinnt.
Die Rhodomycine sind nahe miteinander verwandte und im allgemeinen als Gemische vorkommende stickstoffhaltige
Basen. Sie stellen rote Farbstoffe dar.
Gegenstand des Patentes 913 813 ist die Gewinnung eines kristallisierten Rhodomycin-hydrochlorids, welches dort
als Rhodomycin-A-hydrochlorid bezeichnet wird.

Dieses neue Rhodomycin unterscheidet sich von den bekannten Rhodomyeinen dadurch, daß es fünf Zuckerreste enthält,
während die bekannten Rhodomycine, welche nunmehr ala ß-Rhodomycine I, bzw. ß-Rhodoiaycine II bezeichnet werden,
nur ein bzw. zwei Zuckerreste enthalten ^Tetrahedron Letters, 1969, Seite 415 bis 4197-
Das neue Rhodomycin wird erfindungsgemäß dadurch hergestellt, daß man Streptomyces purpuraaeena kultiviert,
das Kulturfiltrat und das Mycel neutral extrahiert und
aus den Extrakten ß-Rhodomycin V in an eich bekannter Weise gewinnt.
Das erfindungsgemäße Herstellungsverfahren unterscheidet sich somit von den bekannten Herstellungsverfahren zur
Herstellung der bekannten Rhodomycine dadurch, daß hier neutral extrahiert wird, während in den genannten älteren
Patenten die Extraktion sauer durchgeführt wird.

eignen sieh sämtliche Spezies von Streptomyceten. Besonders geeignet ist Streptomyces purpurascens Ist 313· Unter neutraler
Extraktion wird hier eine Extraktion bei einem pH-Wert von 6,5 bis 7,5 verstanden.
ß-Rhodomyein V zeigt eine ausgezeichnete chemotherapeutische,
insbesondere antibakterielle Wirksamkeit, wie die folgenden in-vitro Versuchsergebnisse, zeigen.
Die Prüfung der minimalen Hemmkonzentration erfolgte für die
Gram-positiven und Gram-negativen Erreger im Klein-Medium ^ (Fleisch-Extrakt, Pepton, Dextrose, pH 7,1) bei einer Be- T
brütungstemperatur von 370G über 24 Stunden; Keimeinsaat

Die Tabellenwerte zeigen, daß ß-Rhodomycin V eine ausgezeichnete und außergewöhnlich breite antibakteriell Aktivität,
besonders bei Mykoplasmen und Gram-positiven Bakterien, aufweist.
Die ausgezeichnete und breite antibakterielle Wirksamkeit ermöglicht ihren Einsatz sowohl in der Human- als auch in
der Veterinärmedizin, wobei sie zur Prophylaxe von bakteriellen Infektionen aber auch zur Behandlung bereits eingetretener
bakterieller Infektionen verwendet werden können.
Als Indikationen für den veterinärmedizinischen Bereich ' ä
seien beispielsweise Infektionen durch Mykoplasmen und i»- testinale Infektionen genannt. Prophylaktisch kann
ß-Rhodomycin V als Beimischung zu Futterstoffen verwendet werden.
Eine 10 Liter-Vorkultur von Streptomyces purpurascens Ist 313 wird auf einer Nährlösung mit 2 Prozent Sojamehl und
2 Prozent Mannit submers 24 Stunden bei 260C bebrütet,
auf 350 Liter einer Nährlösung gleicher Zusammensetzung übergeimpft und 96 Stund ι

Nach Fermentationsabbruch wird die rotbraune Kulturbrühe mit 20 kg Kieselgur verrührt und filtriert. Kultur-Filtrat
und Mycel werden getrennt aufgearbeitet. Das Mycel rührt man dreimal mit je 60 Litern Aceton aus, dampft die vereinigten,
orangeroten Acetonauszüge bei 50 C im Vakuum auf 30 Liter ein, extrahiert dreimal mit je 15 Litern
Butanol und engt die vereinigten Butanolauszüge im Vakuum

bei 5O0C bis auf etwa 3 Liter ein. Das hinterbleibende
rotbraune öl wird in 10 Liter Chloroform aufgenommen
(Lösung A).
Das KuIturfiltrat (350 Liter, pH 7,0 bis 7,5) wird mit
150 Litern Butanol extrahiert und die tiefrote Butanolphase im Vakuum bei 5O0C eingeengt, bis etwa 3 Liter
eines tiefroten Öles hinterbleiben, das in 10 Liter Chloroform aufgenommen wird (Lösung B).
Die vereinigten Chloroformlösungen A und B verteilt man auf
sechs 100 χ 10 cm-Säulen aus Carboxymethylcellulose (mit 2n Salzsäure eingeschlämmt;
ausgewaschen
1.) mit Wasser bis zur neutralen Reaktion,
2.) mit Aceton und 3.) mit Chloroform)
Nachwaschen mit je 5 bis 10 Litern Chloroform pro Säule bringt Fette, Anthracyclinone und nichtbasische Anthracycline
ins Piltrat.
Der auf der Säule verbleibende Anteil wird mit 10-%igem
wäßrigen Kaliumhydrogenphosphat eluiert und das tiefrote,
mit verdünntem Ammoniak auf pH 8,0 gebrachte Eluat (pro Säule 5 Liter) dreimal mit je drei Litern Chloroform
extrahiert. Die vereinigten Chloroformphasen verdampft man im Vakuum zur Trockene, nimmt den Rückstand in wenig
Chloroform auf und fällt das rote ß-Rhodomycin V mit Petroläther (40 bis 500C) aus. Ausbeute 150 bis 170 mg
pro Liter Kulturbrühe.

An einer 100 χ 4 cm Cellulosesäule chromatographiert man
2 g des wie vorstehend bereiteten ß-Rhodomycin V Präparates im System Butanol-0,7 m Phosphatpuffer pH 5,8 (1 : 1). Die
Hauptzone (am schnellsten wandernd) bringt man durch Nachwaschen mit Oberphase ins Filtrat, versetzt dieses mit verdünntem
Ammoniak bis pH 8,0, schüttelt mit Chloroform durch und verdampft die mit Natriumsulfat getrocknete Chloroformphase
im Vakuum zur Trockene. Das als roter Lack hinterbliebene ß-Rhodomycin V wird in wenig Chloroform aufgenommen
und in der Siedehitze mit Cyclohexan als rotes Pulver oder auch in roten Nadeln ausgefällt. Schmelzpunkt 173 bis

Claims (5)
- Patentansprüchefür einen Tetraglykoeidrest steht, der aus 2 Mol Rhodinose, 1 Mol 2-Desoxyfucose und 1 Mol Rhodosamin besteht.
- 2. Verfahren zur Herstellung von ß-Rhodomycin V, dadurch gekennzeichnet, daß man Streptomyces purpurascens kultiviert, das Kulturfiltrat und das Mycel neutral extrahiert und aus den Extrakten ß-Rhodomycin V in an sich bekannter Weise gewinnt .
- 3· Verwendung von Streptomyces purpurascens zur Gewinnung von ß-Rhodomycin V.
- 4. Verwendung von ß-Rhodomycin V als Prophylacticum und/ oder Therapeutikum gegen Infektionskrankheiten in der Human- und/oder Veterinärmedizin.
- 5. Verwendung von ß-Rhodomycin V als Beimischung zu Futterstoffen.Le A 12 840109848/1925
