und Verfahren zu ihrer Herstellung
Es wurde gefunden, daß N-Acyl-(piperazinoalkyl)-pyrazole
der Formel I,

sowie deren physiologisch unbedenkliche Säureadditionssalze wertvolle pharmakologische Eigenschaften besitzen. Bei guter
Verträglichkeit zeigen die Substanzen bemerkenswerte zentraldepressive Wirkungen. Z.B. treten narkoseverlängernde, narkosepotenzierende,
tranquillierende und/oder neuroleptische Wirkungen auf. Außerdem wurden auch blutdrucksenkende, Noradrenalin-
bzw. Adrenalin-potenzierende oder stimulierende, z.B. thymoanaleptisehe und Tetrabenazin-antagonistische, ferner
antihistaminische und bronchospasmolytisehe Eigenschaften
beobachtet.
Die neuen Verbindungen können dementsprechend als Arzneimittel und auch als Zwischenprodukte zur Herstellung weiterer
Arzneimittel verwendet werden. .
Gegenstand der Erfindung sind somit Verbindungen der Formel I, worin A, Ar, Q und E die oben angegebene Bedeutung haben,
sowie deren physiologisch unbedenklichen Säureadditionssalze.
Bevorzugt sind Verbindungen der Formel I, worin A unverzweigtes Alkylen mit 1-4 C-Atomen ist.
Gegenstand der Erfindung ist ferner ein Verfahren zur Herstellung von Verbindungen der Formel I sowie ihrer physiologisch
unbedenklichen Salze, dadurch gekennzeichnet, daß man eine Verbindung der Formel II

angegebene Bedeutung.
A bedeutet vorzugsweise -(CH2)n- wie -CH2-, -CH2CH2CH2-
oder -CH2CH2CH2CH2-. Ferner kann es bedeuten:
-CH(CH3)-, -CH(CH3)CH2-, -CH2CH(CH3)-, -CH(C2H5)-,
-CH(CH3)CH2CH2-, -CH2CH(CH3)CH2-, -CH2CH2CH(CH3)-,

Bei der Definition von X sind unter reaktiv veresterten OH-Gruppen
solche zu verstehen, die bei der Umsetzung mit einem Amin durch Aminstickstoff ersetzt werden können, vorzugsweise
die folgenden: Acyloxy wie Acetoxy, Methansulfonyloxy, p-Toluolsulfonyloxy;
ferner insbesondere auch Cl, Br oder J.
Als Ausgangsverbindungen der Formel II kommen vor allem 1-(N'-Arylpiperazino)-2,4-pentandioe in Frage, wobei Aryl
die oben angegebenen Bedeutungen haben kann, sowie die entsprechenden -3,5-hexandione, -4,6-heptandione oder -5,7-octandione.
Die Ausgangsverbindungen der Formel II können z.B. durch Umsetzung
von endständig halogenierten 1,3-Dicarbonylverbindungen
wie 1-Brom- oder 1-Chlorpentan-dion-(2,4) mit Arylpiperazinen
der Formel 711 erhalten werden.
Ausgangsstoffe der "Formel III kommen beispielsweise
folgende aliphatisehe, araliphatische oder aromatische Acylhydrazine
in Frage: Acetyl-, Propionyl-, Butyryl-, Isobutyryl-,
Yaleryl-, Isovaleryl-, Hexanoyl-, Heptanoyl-, Octanoyl-, iTonanoyl-, Acryloyl-, Propioloyl-, Phenylacetyl-, Cinnamoyl-,
Benzoyl-, Naphthoyl-, o-, m-, p-Ioluoyl, 3,4,5-Trimethoxybenzoyl-,
p-Aminobenzoyl- oder p-Dimethylaminobenzoylhydrazin,
Man kann aber auch z.B. funktionell abgewandelte Kohlensäurehydrazide
wie Semicarbazid verwenden.
Die Aeylhydrazlne der Formel III erhält man z.B. aus den entsprechenden Carbonsäureester]! und Hydrazinhydrat.
Die Ausgangsverbindungen der Formel IV sind vorzugsweise
3(5)-/lNf-Aryl-piperazino)-äthyl7-5(3)-methyl-pyrazole, wobei

der Arylrest Ar die oben angegebene Bedeutung hat und insbesondere
m-Chlorphenyl oder m-Tolyl ist; es können aber auch die
entsprechenden -methyl7-5(3)-methyl-, -propyl7-5(3)-methyl-
und -buty17-5(3)-methyl-pyrazole verwendet werden, wobei die
Alkylgruppen verzweigt oder unverzweigt sein können.
Die Verbindungen der Formel IV sind bekannt, oder .können
beispielsweise durch Umsetzen eines Pyrazols der Formel ;

Säuren der Formel 7 sind beispielsweise aliphatisch^ Carbonsäuren
mit 1-11 C-Atomen; Kohlensäure; araliphatische Carbon-j
säuren mit bis zu 11 C-Atomen; gegebenenfalls substituierte
aromatische Carbonsäuren mit bis zu 11 C-Atomen^
Funktionelle Derivate dieser Carbonsäuren sind vorzugsweise
ihre Halogenide, insbesondere ihre Bromide und Chloride, Ester, Anhydride vor allem auch gemischte Anhydride von Carbonsäuren
und Kohlensäurehalbestern oder Ketene; als Kohlensäurederivate
kommen insbesondere Chlorameisensäureester in Frage. }
Als Ausgangsverbindungen der Formel VI kommen vor allem die i-Acyl-5-methylderivate von 3-Chlormethyl-, 3-(1-Chloräthyl)-,
3-(2-Chloräthyl)-, 3-(3-Chlorpropyl)- oder 3-(4-Chlorbutyl)-pyrazol,
bzw. die i-Acyl-3-methylderivate von 5-Chlormethyl-,
5_(1-Chloräthyl)-, 5-(2-Chloräthyl)-, 5-(3-Chlorpropyl)- oder
5_.(4_Chlorbutyl)-pyrazol, sowie die entsprechenden 3-Bromalkyl-,
3-Jodalkyl-, 5-Bromalkyl- oder 5-Jodalkyl-verbindungen
und die Ester der entsprechenden 1-Acyl-5-methyl-3-hydroxyalkyl-,
bzw. 1-Acyl-3-methyl-5-hydroxyalkylpyrazole, insbesondere
deren Methan- und p-Ioluolsulfonate, in Frage.
Die Verbindungen der Formeln VI sind entweder bekannt oder sie können leicht analog bekannten Verbindungen hergestellt werden.
Beispielsweise kann man 5-Methyl-pyrazol-3-carbonsäureester

katalytisch oder mit Lithiumaluminiumhydrid zu 3-Hydroxymethyl-5-methyl-pyrazol
reduzieren. 2-Hydroxy-5-methyl-4-pyron kann mit Hydrazin zu 5-Methyl~pyrazol-3-essigsäure-hydrazid umgesetzt
v/erden, aus. dem durch Alkoholyse die entsprechenden Ester
und durch deren Reduktion 3-(2-Hydroxyäthyl)-5-methyl-pyrazol
erhältlich ist. 3-(3-Hydroxypropyl)- bzw» 3-'(4-Hydroxyhutyl)-5-methyl-pyrazole
(VI, X=OH, A=C5H6 oder O.HQ) sind zugänglich
durch Kondensation von Carbönylverbindungen R-CO-CH* mit
γ-Butyrolacton Tdzw. /-Yalerolacton zu 2-Hydroxy-2-aeylmethyltetrahydrofuranen
"bzw. -pyranen und nachfolgende Reaktion mit Hydrazin. Die entsprechenden Halogenverbindungen (VI, X=Ol, Br
oder J) werden aus den so hergestellten Alkoholen mit z.B. Thionylchlorid, Brom- oder Jodwasserstoffsäure erhalten, die
entsprechenden Acylate und Sulfonsäureester durch übliche Veresterung, z.B. mit Säureanhydriden oder -halogeniden wie
Acetanhydrid, Methan- oder p-Toluolsulfonylchlorid. Durch
Acylierung der so erhaltenen Produkte kommt man schließlich zu AusgangsverMndungen der Formel VI.
Als Verbindungen der Formel VII sind folgende Piperazine "bevorzugt

Die Piperazine VII sind bekannt oder können durch Umsetzung von Aminen der Formel ArNHp mit Diäthanolamin, Morpholin oder
Bis-(2-chloräthyl)-amin oder auch aus Piperazin und Halogeniden, vorzugsweise Bromiden, der Formel ArX erhalten werden.

Man kann auch in Gegenwart von Katalysatoren, vorzugsweise sauren Katalysatoren, beispielsweise Mineralsäuren wie HCl·
oder HpSO., Lewissäuren wie B3?- oder AlCl, oder organischen
Säuren wie p-Toluolsulfonsäure, Essigsäure oder Trifluoressigsäure
arbeiten. Die Reaktionstemperaturen liegen zwischen -10° und 120°, vorzugsweise zwischen 0° und 100°, insbesondere
bei der Siedetemperatur des verwendeten Lösungsmittels»
Die Acylierung einer Verbindung der Formel IV mit einer Säure "der Formel V oder vorzugsweise einem ihrer funktioneilen
Derivate zu einer Verbindung der Formel I gelingt nach den in der Literatur beschriebenen Verfahren unter den verschiedensten
Reaktionsbedingungen, die mit dem verwendeten Acylierungsmittel variieren. Verwendet man ein Acylhalogenid,

vorzugsweise ein Acylchlorid oder -bromid, so arbeitet man
vorteilhaft in Lösung bzw. Suspension. Man verwendet eines der üblichen, gegen Acylierungsmittel inerten, Lösungsmittel, meist
in Gegenwart eines basischen Katalysators. Die Reaktions- ·
temperaturen liegen zwischen -20° und +100°, vorzugsweise f
zwischen -5° und +40°. Besonders vorteilhaft ist es, unter j wasserfreien Bedingungen zu arbeiten und z.B. das Acylhalogenid
bei 0 zu einer wasserfreien Lösung des Pyrazols in Pyridin zu | tropfen. Die Reaktionszeiten variieren zwischen 1 Stunde und
A Tagen.
Verwendet man ein Säureanhydrid als Acylierungsmittel, so kann man meist unter den oben angeführten Reaktionsbedingungen
arbeiten. Man kann aber beispielsweise auch ein Hydrochlorid des eingesetzten Pyrazols, nach Zugabe der berechneten Menge
Hatriumaeetat, in wässeriger Lösung bzw. Suspension bei 40 60° mit dem Säureanhydrid acylieren. Verwendet man überschüssiges
Säureanhydrid als Lösungsmittel, so wird die Reaktion durch einen Zusatz von wenig konzentrierter H2SO2,
oft beschleunigt.
Die Acylierung kann auch mit Estern, vorzugsweise Methylestern, der Säuren der Formel V durchgeführt werden. Man arbeitet
mit überschüssigem Ester, gegebenenfalls unter Zusatz eines der üblichen Lösungsmittel. Die Reaktionstemperatüren liegen
vorzugsweise zwischen -20° und 100°.
Verwendet man Ketene als Acylierungsmittel, so kann man ebenfalls unter den oben angeführten Reaktionsbedingungen arbeiten,
beispielsweise kann das Keten unter Eiskühlung zu einer wässerigen Suspension des Pyrazols getropft werden. Es ist
aber vorteilhafter, in einem organischen Lösungsmittel bei erhöhter Temperatur zu acylieren, z.B. durch Zutropfen des
Ketene zu einer siedenden Lösung des Pyrazole in Äthylenchlorid.

Die Umsetzungen der Verbindungen VI und VII verlaufen nach
Methoden, wie sie für die Alkylierung von Aminen aus der Literatur bekannt sind. Man arbeitet z.B. in einem der üblichen
Lösungsmittel gegebenenfalls unter Zusatz eines säurebindenden Mittels oder eines Überschusses der Verbindung der Formel VII.
Die Reaktionszeiten liegen je nach den angewendeten Bedingungen
zwischen einigen Minuten und 14 Tagen, die Reaktionstemperatur zwischen 0 und 200°, gewöhnlich bei 80 - 130°. Es ist mitunter
ein 12 - 24stündiges Erhitzen notwendig, um gute Ausbeuten zu erzielen.
Bei manchen der vorstehend beschriebenen Umsetzungen können zwei Isomere entstehen, die sich durch die Lage der Doppelbindungen
und der N-Acylgruppe im Pyrazolring unterscheiden; diese Isomeren entsprechen den Verbindungen der Formel· Ia bzw.

Die Bildung eines Gemisches dieser Isomeren - welches ebenso zum Gegenstand dieser Erfindung gehört wie die reinen Isomeren ist
insbesondere zu erwarten bei der Umsetzung einer Verbindung der Formel II mit einer Verbindung der Formel III und bei der
Acylierung eines Pyrazols der Formel IV mit einer Säure der Formel V, oder einem ihrer funktionellen Derivate. Werden die
Verbindungen der Formel I durch Umsetzung von Verbindungen der Formel VI mit Verbindungen der Formel VII, so sind auch hier
Gemische von Isomeren zu erwarten, sofern schon die Ausgangsprodukte
der Formel VI Gemische waren.

Die reinen Isomeren der Formel Ia bzw. Ib können auch durch Erwärmen ineinander umgewandelt werden, wobei sich das thermodynamisch
stabilere Isomere bzw. wieder Gemische bilden können. Man kann umgekehrt durch Erwärmen auch ein reines Isomeres,
vorzugsweise das thermodynamisch stabilere, aus dem Gemisch erhalten.
Gemische aus Verbindungen der FormelnIa und Ib können in bekannter
Weise auf Grund ihrer unterschiedliche Löslichkeit, gegebenenfalls auch durch chromatographische Methoden getrennt \

unbedenkliche Salze liefern. Es können organische und anorganische Säuren, wie z.B. aliphatische, alicyclische,
araliphatische, aromatische oder heterocyclische ein- oder
mehrbasige Carbon- oder Sulfonsäuren, wie Ameisensäure, Essigsäure,
Propionsäure, Pivalinsäure, Diäthylessigsäure, Oxalsäure,
Malonsäure, Milchsäure, Bernsteinsäure, Pimelinsäure, Fumarsäure, Maleinsäure, Weinsäure, Apfelsäure, Aminocarbonsäuren,
SuIfaminsäure, Benzoesäure, Salicylsäure, Phenylpropionsäure,
Citronensäure, Gluconsaure, Ascorbinsäure, Nicotinsäure, Isonicotinsäure,
Methansulfonsäure, A'thandisulf onsäure, ß-Hydroxyäthansulfonsäure,
p-Toluolsulfonsäure, Naphthalin-mono- und
-disulfonsäuren, Schwefelsäure, Salpetersäure, Halogenwasserstoffsäuren,
wie Chlorwasserstoffsäure oder Bromwasserstoffsäure, oder Phosphorsäuren, wie Orthophosphorsäure, verwendet
werden.
Die neuen Verbindungen können im Gemisch mit üblichen Arzneimittelträgern
in der Human- oder Veterinärmedizin eingesetzt werden. Als Trägersubstanzen kommen solche organische oder
anorganische Stoffe in Frage, die für die parenterale oder enterale Applikation geeignet sind und die mit 'den neuen Ver-

bindungen nicht in Reaktion treten, wie "beispielsweise Wasser,
pflanzliche Öle, Polyäthylenglykole, Gelatine, Milchzucker, Stärke, Magnesiumstearat, Talk, Vaseline, Cholesterin usw.
Zur parenteralen Applikation dienen insbesondere lösungen, vorzugsweise ölige oder wäßrige lösungen, sowie Suspensionen,
Emulsionen oder Implantate. Pur die enterale Applikation eignen sich ferner Tabletten oder Dragees, die gegebenenfalls sterilisiert
oder mit Hilfsstoffen, wie Konservierungs-, Stabilisierungs-
oder iTetzmitteln oder Salzen zur Beeinflussung des osmotischen
Druckes oder mit Puffersubstanzen versetzt sind.
Die erfindungsgemäßen Substanzen werden vorzugsweise in einer Dosierung von 2 bis 100 mg pro Dosierungseinheit appliziert.
Zu einer Lösung von 10,1 g 6-(N'-m-Chlorphenyl-piperazino)-2,4-hexandion
(erhältlich aus 6-Brom-2,4-hexandion und üT-m-Chlor- '
phenyl-piperazin) in 200 ml Äthanol gießt man unter Kühlung eine Lösung von 2,5 g Acetylhydrazin in einem Gemisch aus ·
20 ml Äthanol und 1 ml konzentrierter HCl, erwärmt 4 Stunden auf 60°, entfernt das Lösungsmittel im Vakuum, nimmt den Rückstand
in 100 ml 5 $iger NaOH auf, extrahiert 3x mit 40 ml
Äther, trocknet die vereinigten Ätherphasen mit MgSO., entfernt den Äther im Vakuum und erhält 1-Acetyl-3(5)-/5-(N'-m-Chlorphenyl-piperazino)-äthyl/-5(3)-methylpyrazol;
Maleat Έ.=145-147°:
Man gibt zu 9,12 g 3(5)-/"2-(N'-m-Chlorphenyl-piperazino)-äthyl7-5(3)-methyl-pyrazol,
gelöst in 100 ml absolutem Tetrahydrofuran und 4,75 ml wasserfreiem Pyridin, unter Eiskühlung 3,53 ml
Acetylchlorid, rührt unter Wasserausschluß 2 Tage bei Raumtemperatur,
destilliert das Lösungsmittel ab, macht den Rück-

stand mit wässeriger NaOH alkalisch, extrahiert mit Äther,
trocknet die Ätherphase über Na2SO^, destilliert das Lösungsmittel
ab und erhält 1-Acetyl-3(5)-Z2-(Nf-m-chlorphenylpiperazino)-äthyl7~5(3)-methyl-pyrazol;
Maleat F.= 145 - 147°.
5 g 3-/2-(N·-m-Chlorphenyl-piperazino)-äthyl/^-methyl-pyrazol
werden in 50 ml trockenem Tetrahydrofuran gelöst. Man gibt 1,25 g NaH zu, kocht 1 Stunde unter Stickstoff, fügt nach dem
Erkalten 1,85 g Ν,Ν-Dimethylcarbamoylchlorid zu, kocht eine ι *
weitere Stunde und rührt anschließend 2 Tage bei Raumtemperatur.; Man filtriert, dampft das Filtrat ein, nimmt in Äthanol auf und *
fällt mit ätherischer HCl. Nach Umkristallisieren aus Äthanol/ ι
Äther erhält man I-Dimethylaminocarbonyl-, 3-/2-(N'-m-chlorphenyl-piperazino)-äthyl7-5-methyl-pyrazol,
Hydrochlorid, F. 198 - 200°. Nach Eindampfen der Mutterlauge erhält man 1-Dimethylaminocarbonyl-S-/^-(N'-m-chlorphenyl-piperazino)-äthyl7~3-methyl-pyrazol,
Hydrochlorid-monohydrat, F.= 145-147° (aus Isopropanol/Äther).
Man löst 14,2 g 3-/2-(N'-m-Tolyl-piperazino)-äthyl7-5-methylpyrazol
in einem Gemisch aus 120 ml absolutem Tetrahydrofuran und 1 ml konzentrierter HgSO^, tropft während 30 Minuten 10 ml
Buttersäureanhydrid zu, rührt 2 Tage bei Raumtemperatur, gießt in überschüssiges Eiswasser, gibt unter Rühren wässerige NaOH
zu, bis ein piH-Wert von ca. 8 erreicht ist, extrahiert mit Äther, trockr|et die Ätherphase, destilliert das Lösungsmittel
ab und erhälij 1-Butyryl-3(5)-/2-(N'-m-tolyl-piperazino)-äthyl7-5-methyl-pyr4zol,
Pikrat, F.= 200 - 202°.

Man rührt ein Gemisch aus 6,2 g 1-Acetyl-3(5)-(2-chloräthyl)-5(3)-methyl-pyrazol
/erhältlich durch Umsetzen von 3-(2-Chloräthyl)-5-methyl-pyrazol mit Acetylchlorid/, 6,5 g N-(m-Chlorphenyl)-piperazin,
5 g wasserfreiem K2CCU und 100 ml trockenem
Dimethylformamid 24 Stunden bei 100 - 110°,' filtriert nach dem Erkalten, destilliert das Lösungsmittel ab, nimmt in wenig
Äthanol auf, versetzt mit überschüssiger äthanolischer Maleinsäure, läßt 2 Stunden bei 0° stehen, saugt die ausgeschiedenen
Kristalle ab und erhält 1-Acetyl-3(5)-/2-(N'-m-chlorphenylpiperazino)-äthyl7-5(3)-methyl-pyrazol-maleat,
F.= 145 - 147°.
Man kocht ein Gemisch aus 9,7 g 1-p-Aminobenzoyl-3(5)-(2-bromäthyl)-5(3)-methyl-pyrazol
/erhältlich durch Umsetzen von 3-(2-Bromäthyl)-5-methyl-pyrazol mit Benzoylbromid/ und. 5,9 g
N-(m-Tolyl)-piperazin 32 Stunden in 120 ml trockenem Pyridin, destilliert das Lösungsmittel ab, nimmt den Rückstand in
trockenem Äther auf, filtriert, sättigt das Filtrat mit trockenem HCl-Gas, läßt 3 Stunden bei 0° stehen, saugt die
ausgeschiedenen Kristalle ab und erhält 1-p-Aminobenzoyl-3(5)-/2-(N
· -m-tolyl-piperazino) -äthyl7-5 (3) -methyl-pyrazol-hydrochlorid,
Erweichen ab 200°.

Analog Beispiel 4 erhält man durch Umsetzen mit den Anhydriden !
der in Beispiel 9 erwähnten Säuren die entsprechenden 1-Acyl- j
3(5)-/2-(N'-m-Tolyl-piperazino)-äthyl7-5-niethyl-pyrazole. |
Zu einer Lösung aus Butyryl-äthyl-carbonat in absolutem Tetrahydrofuran
(hergestellt aus einem Gemisch von 1,8 g Buttersäure, 1,5 g trockenem Diäthylamin und 40 ml absolutem Tetrahydrofuran,
durch Zugabe von 2,2 g Chlorameisensäureäthylester bei -250C)
gibt man bei -250C 5,6 g 3-Z2-(N'-m-Tolyl-piperazino)-äthyl7-5-methyl-pyrazol,
gelöst in 40 ml absolutem "Tetrahydrofuran, läßt 6 Stunden bei Zimmertemperatur stehen, filtriert, wäscht
mit Tetrahydrofuran nach, dampft das Filtrat im Vakuum ein, nimmt in Äther auf, wäscht mit verdünnter H2SO^, wässeriger
5 %iger Na2C05-Lösung und. Wasser, trocknet über Na2SO^ und
destilliert den Äther ab. Man erhält 1-Butyryl-3(5)-/2-N!-m-Tolyl-piperazino)-äthyl7-5(3)-methyl-pyrazol;

Verfahren zur Herstellung von pharmazeutischen Präparaten, dadurch gekennzeichnet, daß man mindestens eine Verbindungm der allgemeinen Formel I, gegebenenfalls zusammen mit mindestens einem festen, flüssigen oder halbflüssigen Hilfs- oder Trägerstoff und gegebenenfalls zusammen mit mindestens einem weiteren Wirkstoff in eine geeignete Dosierungsform bringt.
- 26. Pharmazeutische Zubereitung, enthaltend eine wirksame Dosis einer Verbindung der Formel I neben mindestens einem festen, flüssigen oder halbflüssigen Träger- oder Zusatzstoff.
- 27. Pharmazeutische Zubereitung, enthaltend 0,1 bis 100 mg einer Verbindung der Formel I neben mindestens einem festen, flüssigen oder halbflüssigen Träger- oder Zusatzstoff.209837/1203
- 28. Verfahren zur Erzielung einer narkoseverlängernden und/oder narkosepotenzierenden und/oder tranquillierenden und/oder neuroleptischen Wirkung in Lebewesen, dadurch gekennzeichnet, daß man eine wirksame Dosis einer Verbindung der Formel I verabreicht.209837/1203
