R für Wasserstoff, eine Acylgruppe einer Alkynsäure mit bis zu 8 C-Atomen oder für den Säurerest der Bernstein-
oder Maleinsäure, gegebenenfalls in Form des Alkalimetall- oder Ammoniumsalzes steht.
Als gesättigte geradkettige und verzweigtkettige Reste von Alkynsäuren mit bis zu 8 Kohlenstoffatomen seien der Acetyl-, Propionyl-, Isopropionyl-, n-Butyryl-, Isobutyryl-, Capryl- und der Önanthsäurerest genannt. Geeignet sind aber auch die Säurereste der Bernstein- und Maieinsäure.
Als gesättigte geradkettige und verzweigtkettige Reste von Alkynsäuren mit bis zu 8 Kohlenstoffatomen seien der Acetyl-, Propionyl-, Isopropionyl-, n-Butyryl-, Isobutyryl-, Capryl- und der Önanthsäurerest genannt. Geeignet sind aber auch die Säurereste der Bernstein- und Maieinsäure.
Im Fall eines Esters dieser zweibasischen Säuren können die erfindungsgemäßen Verbindungen der Formel I
auch in Form ihrer Alkalimetall- oder Ammoniumsalze vorliegen.
Die erfindungsgemäßen Verbindungen besitzen wertvolle pharmakologische Eigenschaften. Sie sind u. a.
Diuretika vom Typ der Aldosteron-Antagonisten, d. h., sie kehren die Wirkung von Desoxycorticosteron auf die
Natrium- und Kaliumausscheidung um. Die trfmdungsgemäßen Verbindungen wie das 17/?-Hydroxy-17a-(3-aceioxypropylWa-thioacetyl-oa-methyM-androsten-S-on
erweisen sich im Testmodell von Hollmann [G. Hollmann et. al., Tubuläre Wirkungen und renale Elimination von Spirolactonen, Naunyn-Schmiedebergs
Arch. Exp. Path. Pharmak. 247 (1964) 419; P. Marx, Renale Wirkungen des d-Aldosterons und seines Antagonisten
Spironolacton, Diss. Med. Fak. FU Berlin 1966] als dem bekannten Spironolacton in ihrer Wirkung überraschenderweise
überlegen.
Die Erfindung betrifft des weiteren ein Verfahren zur Herstellung von Verbindungen der allgemeinen Formel
I, welches dadurch gekennzeichnet ist, daß man in an sich bekannter Weise 4-Androsten-3-on mit der Formel

durch Enaminierung oder Enolierung zur 43-5-Verbindung isomerisiert, an die Δ ^Doppelbindung Formaldehyd
anlagert, von der erhaltenen 6-Hyuroxymethylverbindung Wasser abspaltet, das so erhaltene 6-Exomethylensteroid
mit Cyclohexan in Gegenwart eines Edelmetallkatalysators zum 6-Methyl-46-steroid isomerisiert und
gegebenenfalls an die /^-Doppelbindung Thioessigsäure addiert, gegebenfalls die 22-Hydroxygruppe verestert
und im Falle des Bernstein- oder Maleinsäureesters gegebenenfalls das Alkalimetall- oder Ammoniumsalz herstellt.
Das Ausgangsmaterial 17/?-Hydroxy-17a-(3-hydroxypropyl)-4-androsten-3-on kann beispielsweise nach dem
Verfahren der DE-OS 23 27448 aus 3-Keto-4-androsten-17-on hergestellt werden.
Zur Durchführung des erfindungsgemäßen Verfahrens isomerisiert man die 3-Keto-44-Gruppierung entweder
durch Enaminierung oder Enolisierung zur 3-Enamin- bzw. 3-Enol-4 3-5-Gruppierung. Zur Enaminierung
erwärmt man das Ausgangssteroid der Formel II beispielsweise mit Pyrrolidin oder Morpholin in einem geeigneten
Lösungsmittel wie Methanol, Methylenchlorid oder Benzol, gegebenenfalls unter Zusatz von p-Toluolsulfonsäure.
Zur Enolisierung erwärmt man zweckmäßigerweise das Ausgangssteroid z. B. mit Acetondimethylketal,
Orthoameisensäureester oder Benzylalkohol in einem geeigneten Lösungsmittel wie Benzol, Dimethylformamid
oder Dioxan in Gegenwart von p-Toluolsulfonsäure oder Schwefelsäure. Die Bildung der entsprechenden
Thionole mit Monothioglykol oder Benzylmercaptan in Pyridin/Ethanol ist auch möglich.
An die so gebildete ^-Doppelbindung wird Formaldehyd angelagert. Hierzu gibt man Formaldehyd bei
Raumtemperatur langsam zu dem in einem geeigneten Lösungsmittel gelösten A 5-Steroid. Geeignete Lösungsmittel
sind aliphatische Alkohole wie Methanol und Ethanol, aromatische Kohlenwasserstoffe wie Benzol und
Toluol sowie cycloaliphatische Kohlenwasserstoffe wie Cyclohexan.
Anschließend wird von der so gebildeten 6-Hydroxymethylverbindung Wasser abgespalten. Hierzu behandelt
man das 6-Hydroxymethyl-steroid in einem protischen Lösungsmittel mit einer Säure. Geeignet ist beispielsweise
die Kombination Dioxan oder Tetrahydrofuran und Salzsäure oder p-Toluolsulfonsäure bzw. Methansulfonsäurechlorid
in Pyridin ggf. unter Zusatz von Lithiumbromid unter Wärmeeinwirkung.
Das so erhaltene 6-Exomethylensteroid wird anschließend zum 6-Methyl-4'-steroid isomerisiert. Hierzu wird
das 6-Exomethylensteroid tiit Cyclohexan in Gegenwart eines Edelmetallkatalysators wie Palladium über längere
Zeit erwärmt. In der Siedehitze ist die Reaktion bereits nach einigen Stunden beendet. Ein Zusatz von
Natrium- oder Kaliumacetat verkürzt die Reaktion, die zweckmäßigerweise in einem protischen Lösungsmittel
wie Methanol oder Äthanol ausgeführt wird.
• Zur Einführung der Acetylthiogruppierung wird das 6-Methyl-46-steroid mit Thioessigsäure in einem protischen
Lösungsmittel wie Methanol oder Ethanol, gegebenenfalls unter Zusatz von Wasser umgesetzt, wobei die
Temperatur nicht allzuweit über Raumtemperatur gebracht, auf jeden Fall unterhalb der Siedetemperatur des
Lösungsmittels gehalten wird.
Die Veresterung der primären 22-Hydroxygruppe wird nach den gängigen Methoden vorgenommen. Hierzu
löst man das Steroid beispielsweise in Pyridin und gibt das entsprechende Säurederivat wie das Anhydrid oder
das Säurechlorid hinzu.
Falls die primäre 22-Hydroxygruppe mit Bernstein- oder Maleinsäure verestert wurde, kann das 1 lemiacy IaI in
das gewünschte Alkalisalz durch Umsetzung beispielsweise mit einer methanolischen Kalium- oder Natriummethylat-Lösung
erreicht werden. Zur Herstellung des Ammoniumsalzes verwendet man :zweckmäßigerwei.se
eine Lösung von Ammoniak in Methanol.
Nach beendeter Reaktion wird das Reaktionsgemisch in üblicher Weise wie Fällung, Extraktion, Urhkri.stallisation
und/oder Chromatographie aufgearbeitet.
Die pharmakologisch wirksamen, erfindungsgemäßen Verbindungen der allgemeinen Formel I können nach
an sich bekannten Methoden def Galenik zu Arzneimitteln, insbesondere zur oralen Applikation, verwendet
werden. Gegenstand der Erfindung sind somit auch Arzneimittel auf Basis der Verbindunger, gemäß Anspruch
ίο 1 und 2.
D'je Dosierung der erfindungsgemäßen Verbindung liegt beim Menschen bei 20-500 mg/Tag*
Die nachfolgenden Beispiele sollen die Erfindung erläutern.
15,0 g 17yS-Hydroxy-17a-(3-hydroxypropyl)-4-androsten-3-on werden in 75 ml Methanol und 7,5 ml Pyrrolidin
15 Minuten am Rückfluß erhitzt. Es wird dann im Eisbad abgekühlt, der auskristallisierte Niederschlag abgesaugt
und mit wenig eiskaltem Methanol nachgewaschen. Es werden 12,0 g 17a-(3-HydroxypropyI)-3-pyrrolidino-3,5-androstadien-17/?-ol
erhalten.

11,5 g 17a-(3-Hydroxypropyl)-3-pyrrolidino-3,5-androstadien-17/?-ol werden in 794 ml Äthanol und 397 ml
Benzol gelöst, dann 20,8 ml 40prozentige Formaldehydlösung zugetropft und 1 Stunde bei Raumtemperatur
nachgerührt. Es wird dann im Vakuum weitgehend eingeengt und der Rückstand an Silicagel chromatographiert.
Es werden 5,25 g 17/?-Hydroxy-6/?-hydroxymethyl-17a(3-hydroxypropyl)-4-androsten-3-on erhalten.

5,25 g 17/?-Hydroxy-6/?-hydroxymethyl-17o:(3-hydroxypropyl)-4-andiosten-3-on werden in 260 ml Dioxan mit
15,2 ml 5 η Salzsäure 2,5 Stundenbei Raumtemperatur gerührt. Die Reaktionslösung wird dann mit überschüssigem
Natriumhydrogencarbonat versetzt, vom ungelösten Rückstand abfiltriert und das Filtrat im Vakuum zur
Trockne eingedampft. Der Rückstand wird an Silicagel Chromatographien. Es werden 3,8 g 17/?-Hydroxy-l 7«(3-hydroxypropyl)-6-methylen-4-androsten-3-on
erhalten. Eine aus Aceton umkristallisierte Probe schmilzt bei 178-179,5°C.

3,5 g 17/?-Hydroxy-17a(3-hydroxypropyl)-6-methylen-4-androsten-3-on werden in 122 ml Äthanol mil i,75 g
wasserfreiem Natriumacetat und 257 mg Palladium auf Kohle 5%ig versetzt und in der Siedehitze über 8 Stunden
0,7 ml Cyclohexen in 20 ml Äthanol portionsweise zugegeben. Es wird dann vom Katalysator abfiltriert und das
Filtrat im Vakuum weitgehend eingeengt. Der Rückstand wird in Methylenchlorid aufgenommen, mit Wasser
gewaschen, getrocknet, eingedampft und an Silicagel chromatographiert. Es werden 1,9 g 17/S-Hydroxy-17£z(3-hydroxypropyl)-6-methyl-4,6-androstadien-3-on
erhalten. Eine aus Diisopropyläther/Aceton umkrislallisierte Probe schmilzt bei 197-2020C.
UV: £291 =21100.
500 mg 17jö-Hydroxy-17ir(3-hydrcxypropyl)-6-rnethyl4,6-androstadien-3-on werden in 5 ml Thioessigsäure
6,5 Stunden bei 100° C gerührt. Es wird dann mit Äther verdünnt, mit Wasser und gesättigter Natriumhydrogencarbonatlösung
gewaschen, getrocknet und eingedampft. Der Rückstand wird überpräparative Schichlchromatographie
aufgereinigt. Es werden 330 mg 7a-Acetylthio-17a-(3-acetoxypropyl)-17/?-hydroxy-'6a-methyl-4-androsten-3-on
als Öl erhalten.

1,0 g 17/?-Hydroxy-17a(3-hydroxypropyl)-6-methyl-4,6-androstadien-3-on werden in 10 ml Methanol mit 2 ml
p 60 Wasser und 2 ml Thioessigsäure versetzt und 18 Stunden bei 50° C gerührt. Es wird dann fn it Äther verdünnt, mit
■ Wasser und gesättigter Natriumhydrogencarbonatlösung gewaschen, getrocknet und eingedampft. Nach Chro^
matographie an Silicagel wird aus Diisopropyläther/Aceton umkristallisiert. Es werden 650 mg 7a-Acetylthio-17y5-hydroxy-17a(3-hydroxypropyl)-6a-methyl-4-androsten-3-on
vom Schmelzpunkt 175-175,50C erhalten.

250 mg Ta-Acetylthio-n/^hydroxy-na-O-hydroxypropyO-oa-methyM-androsten-S-on werden in 1 ml Pyridin
mit 0,5 ml Buttersäureanhydrid 24 Stunden bei Raumtemperatur stehengelassen; Nach Eiswasserfallung
wird das ausgeschiedene Öl abfiltriert, in Äther aufgenommen, mit Wasser gewaschen und getrocknet. Der nach
dem Eindampfen erhaltene Rückstand wird über präparative Schichtchromatographie aufgereinigt und es
werden 210 mg Ta-Acetylthio-na-Q-butyryloxypropyO-n^hydroxy-oa-methyM-androsten-S-on als Öl erhalten.
