Description
- Die vorliegende Erfindung bezieht sich auf eine therapeutische Zusammensetzung zum Bekämpfen des erworbenen Immundefektsyndroms (AIDS).
- Obwohl im Hinblick auf die Entwicklung einer therapeutischen Zusammensetzung zum Bekämpfen von AIDS Versuche durchgeführt worden sind, ist gegenwärtig keine wirksame Zusammensetzung gefunden worden.
- Demgemäss ist es eine Aufgabe der vorliegenden Erfindung, eine ausgezeichnete therapeutische Zusammensetzung zum Bekämpfen von AIDS zur Verfügung zu stellen.
- Die zuvor genannte Aufgabe ist durch Verwendung eines karzinostatischen Mittels, ausgewählt aus Chlormethin-N-oxid und Nimustin-HCl, für die Herstellung eines Medikamentes für die Behandlung des erworbenen Immundefektsydroms (AIDS) gelöst worden.
- Die vorliegende Erfindung bezieht sich auf eine therapeutische Zusammensetzung zum Bekämpfen von AIDS, umfassend: (A) eine pharmazeutisch wirksame Menge mindestens eines karzinostatischen Mittels, ausgewählt aus verschiedenen bestimmten Alkylierungsmitteln, und (B) einen pharmazeutisch geeigneten Träger.
- Beispiele für verschiedene bestimmte Alkylierungsmittel sind Chlormethin-N-oxid und Nimustin-HCl.
- Diese Mittel können in Kombination verwendet wrden.
- Die bei der vorliegenden Erfindung verwendeten karzinostatischen Mittel sind kommerziell erhältlich.
- Die karzinostatischen Mittel werden in geeigneter Weise mit einem pharmazeutisch geeigneten Träger unter Herstellung pharmazeutischer Zusammensetzungen gemischt.
- Im Falle der oralen Verabreichung umfassen Beispiele eines verwendeten, pharmazeutisch geeigneten Trägers Lactose, Stärke, Sucrose, Glucose, kristalline Zellulose, Kalziumcarbonat, Kaolin, Stearinsäure, Magnesiumstearat, Talkum, Gelatine, Natriumcarboxymethylzellulose, Methylzellulose, Siliziumdioxid.
- Im Falle der Injektion wird das karzinostatische Mittel in einer pharmazeutisch geeigneten Lösung, wie beispielsweise destilliertem Wasser für die Injektion, physiologischer Kochsalzlösung, einer injizierbaren zuckerhaltigen Lösung (beispielsweise Sucrose, Fructose, Xylit), Ringer's Lösung, einer elektrolytischen Lösung, einer zuckerhaltigen elektrolytischen Lösung, gelöst.
- Die Antivirusaktivität jedes karzinostatischen Mittels kann in vitro innerhalb dessen cytotoxischer Begrenzung festgelegt werden. Somit kann die therapeutische Zusammensetzung zum Bekämpfen von AIDS gemäss der vorliegenden Erfindung mit einer Dosis bis zum maximalen Niveau, d.h. cytotoxische Grenze, jedes karzinostatischen Mittels verabreicht werden.
- Insbesondere kann die therapeutische Zusammensetzung gemäss der vorliegenden Erfindung in einer Dosierung von beispielsweise 10 bis 100 mg/Tag (intravenöse Injektion), in bezug auf die Menge des Chlormethin-N-oxids, und 2 bis 3 mg/kg Körpergewicht/Tag (intravenöse Injektion) in bezug auf die Menge von Nimustin-HCl verabreicht werden.
- Die erfindungsgemässe therapeutische Zusammensetzung zum Bekämpfen von AIDS kann auf konventionelle Weise, wie sie für jedes karzinostatische Mittel verwendet wird, verabreicht werden. Beispielsweise wird jede Zusammensetzung, die ein karzinostatisches Mittel gemäss der vorliegenden Erfindung enthält, auf folgende Art verabreicht: Injektion in die Vene, Pleurahöhle, Bauchfellhöhle oder Tumorgewebe bei der Zusammensetzung, die Chlormethin-N-oxid enthält, und intravenöse oder intraarterielle Injektion bei der Zusammensetzung, die Nimustin-HCl enthält.
- Die vorliegende Erfindung wird detailliert unter Bezugnahme auf das nachfolgende, nicht-einschränkende Beispiel beschrieben.
- (1) Spezimen: Die in der nachfolgenden Tabelle dargestellten Alkylierungsmittel.
- (2) Virus: HIV-I (HTLV-III-Stamm).
- (3) Zielzelle: MT-4-Zellen (Jpn. J. Concer. Res., 28, 219-228 (1982)).
- (4) Bestimmung der Wirkungen des karzinostatischen Mittels:
- (i) Vorbehandlung:
- 50ul/Vertiefung Anteile der Zielzellen (5 x 10&sup6; Zellen/ml) wurden in Proben mit zweifacher Serienverdünnung jedes karzinostatischen Mittels (50ul/Vertiefung) pipettiert. Man liess diese Mischungen bei 37ºC unter CO&sub2;-Atmosphäre 18 bis 24 Stunden reagieren und beimpfte anschliessend mit 100ul/Vertiefung (50 bis 500 TCID&sub5;&sub0;) einer HIV-I-Viruslösung.
- (ii) Gleichzeitige Behandlung:
- 50ul/Vertiefung Anteile der Zielzellen (5 x 10&sup6; Zellen/ml) wurden in Proben mit zweifacher Serienverdünnung jedes karzinostatischen Mittels (50ul/Vertiefung) pipettiert und anschliessend mit 100 ul/Vertiefung (50 bis 500 TCID&sub5;&sub0;) einer HIV-I-Viruslösung beimpft.
- (iii) Die unter (i) und (ii) erhaltenen Lösungen wurden 3 Tage bei 37ºC inkubiert. Anschliessend wurde das Kulturmedium erneuert und die Inkubation wurde weiterhin 3 bis 4 Tage fortgesetzt. Die Anti-HIV-I-Virusaktivität jeder Probe wurde auf der Grundlage von dessen anticytopathogener Aktivität bestimmt.
- Die nachfolgende Tabelle zeigt die Wachstumsinhibierungskonzentration jedes karzinostatischen Mittels auf den HIV-I-Stamm.
- In der nachfolgenden Tabelle bedeutet der Ausdruck "Vorbehandlung" den Fall, bei dem die Zielzellen mit einem karzinostatischen Mittel vor Beimpfen mit dem HIV-I-Stamm behandelt wurden, während der Ausdruck "gleichzeitige Behandlung" den Fall bezeichnet, bei dem die Behandlung mit dem karzinostatischen Mittel gleichzeitig mit der Einimpfung des Virus durchgeführt wurde. Der Ausdruck "cytotoxische Wirkung" bedeutet die Dosis, bei der jedes karzinostatische Mittel einen cytotoxischen Effekt zeigte. - Aus den in der Tabelle dargestellten Ergebnissen ist ersichtlich, dass von den karzinostatischen Mitteln Alkylierungsmittel Anti-HIV-I-Aktivität ausüben und somit wirksam in einer therapeutischen Zusammensetzung zum Bekämpfen von AIDS sind.
- Während die Erfindung detailliert und unter Bezugnahme auf ihre bestimmten Ausführungsformen beschrieben worden ist, ist es für den Fachmann offensichtlich, dass verschiedene Änderungen und Modifikationen gemacht werden können, ohne vom Umfang der Erfindung abzuweichen.
Claims (1)
- Verwendung eines karzinostatischen Mittels, ausgewählt aus Chlormethin-N-oxid und Nimustin-HCl, für die Herstellung eines Arzneimittels zur Behandlung des erworbenen Immundefektsyndroms (AIDS).
