Description
- Die vorliegende Erfindung betrifft neue Antitumor- und antivirale Alkylierungsmittel, die mit dem bekannten Antibiotikum Distamycin A verwandt sind: dieses gehört zur Familie der Pyrrolamidin-Antibiotika und es wurde beschrieben, dass es reversibel und selektiv mit DNA-AT-Sequenzen interagiert, so dass es sowohl mit der Replikation als auch mit der Transkription wechselwirkt [Nature, 203, 1064 (1964); FEBS Letters, 7 (1970) 90; Prog. Nucleic Acids Res. Mol. Biol., 15, 285 (1975)].
- DE-A-17 95 539 offenbart die Herstellung von Distamycinderivaten, bei denen die Formylgruppe durch ein Wasserstoffatom oder durch einen Säurerest einer organischen aliphatischen C1-4-Säure oder einer Cyclopentylpropionsäure ersetzt ist.
- EP-B-246 868 beschreibt Distamycinanaloga, bei denen die Distamycinformylgruppe durch einen aromatischen, alicyclischen oder heterocyclischen Teil mit Alkylierungsgruppen ersetzt ist.
- Distamycinanaloga, worin die Formylgruppe durch eine Vielzahl von Gruppen ersetzt ist, einschliesslich aromatischen heterocyclischen Gruppenteilen, sind ebenfalls offenbart in J. Med. Chem. (1989), 32(4), 774–778; WO93/13739; WO94/20463; Bioorg. Med. Chem. Lett. (1994), 4(12), 1467–1472; und WO96/05196.
- Es wurde nun gefunden, dass eine neue Klasse von Distamycinderivaten, wie sie im folgenden definiert sind, nützliche biologische Eigenschaften aufzeigt, wobei die Distamycinformylgruppe ersetzt ist durch einen, eine Alkylierungsgruppe tragenden benzoheterocyclischen Ring.
- Entsprechend betrifft die vorliegende Erfindung neue Distamycinderivate der Formel (I), wie im folgenden definiert, ein Verfahren zur Herstellung dieser, pharmazeutische Zusammensetzungen, enthaltend diese, und deren Verwendung in der Therapie, insbesondere als Antitumor- und antivirale Mittel.
- Daher sind ein Ziel der vorliegenden Erfindung Distamycinderivate der Formel: worin:
n 2, 3 oder 4 ist;
A ist O, S oder NR, wobei R Wasserstoff oder C1-4-Alkyl ist;
B ist CH oder N;
R1 ist Wasserstoff oder C1-4-Alkyl;
T ist ausgewählt aus (i) worin: p 0 oder 1 ist; R2 und R3 sind jeweils unabhängig ausgewählt aus Wasserstoff, C1-4-Alkyl, gegebenenfalls substituiert mit ein oder mehreren Fluoratomen, und C1-4-Alkoxy; R4 ist C1-4-Alkyl oder C1-3-Haloalkyl; X1 ist ein Halogenatom; und (ii) worin: X2 ein Halogenatom ist;
oder pharmazeutisch annehmbare Salze davon. - Die vorliegende Erfindung schliesst weiterhin alle möglichen Isomere ein, die durch die Formel (I) abgedeckt werden, sowohl einzeln als auch als Mischung, genauso wie Metaboliten und pharmazeutisch annehmbare Biovorläufer (auch als Pro-Arzneimittel bekannt) der Verbindungen der Formel (I).
- Die Alkyl- und Alkoxygruppen können gerade oder verzweigte Ketten aufweisen. Eine C1-4-Alkylgruppe ist bevorzugt Methyl, Ethyl oder Propyl, eine C1-4-Alkoxygruppe ist bevorzugt Methoxy oder Ethoxy, während eine C1-3-Haloalkylgruppe bevorzugt 2-Chlorethyl oder 2-Bromethyl ist. Wenn eine C1-4-Alkylgruppe mit ein oder mehreren Fluoratomen substituiert ist, ist sie bevorzugt eine C1-4-Perfluoralkylgruppe, z. B. -CF3. Die Halogenatome X1 und X2 sind bevorzugt Chlor oder Brom. Besonders bevorzugte Werte für n sind 2 und 3.
- Wenn T ein Alkylierungsanteil der Formel (II) gemäss Punkt (i), wie oben aufgeführt, mit p = 1 ist, sind die Carboxamidogruppe und die Aminogruppe am Phenylring bevorzugt in der meta- oder para-Position zueinander, während R2 und R3 an irgendeiner freien Position sein können.
- Pharmazeutisch annehmbare Salze der Verbindung der Formel (I) sind Salze mit pharmazeutisch annehmbaren organischen oder anorganischen Säuren. Beispiele für anorganische Säuren sind Salzsäure, Bromsäure, Schwefelsäure, Salpetersäure; Beispiele für organische Säuren sind Essigsäure, Propionsäure, Bernsteinsäure, Malonsäure, Zitronensäure, Weinsäure, Methansulfonsäure und p-Toluolsulfonsäure.
- Eine bevorzugte Klasse von Distamycinderivaten gemäss der vorliegenden Erfindung ist eine der Formel (I), worin:
n 2 oder 3 ist;
A ist O, S, NH oder NCH3;
B ist CH oder N;
R1 ist Wasserstoff;
T ist eine Gruppe der Formel (II) gemäss Punkt (i), worin X1 Chlor oder Brom ist; R4 ist Ethyl, 2-Chlorethyl oder 2-Bromethyl; R2 und R3 sind jeweils unabhängig Wasserstoff oder Methyl; p ist 0 oder 1; oder
T ist eine Gruppe der Formel (III) gemäss Punkt (ii), wobei X2 Brom oder Chlor ist. - Beispiele für spezifische Verbindungen gemäss der vorliegenden Erfindung, insbesondere in Form von Salzen, bevorzugt mit Salzsäure, sind die folgenden:
- 1. 3-[1-Methyl-4[1-methyl-4[1-methyl-4[5-N,N-bis(2-chlorethyl)aminoindol-2-carboxamido]pyrrol-2-carboxamido]pyrrol-2-carboxamido]pyrrol-2-carboxamido]propionamidin;
- 2. 3-[1-Methyl-4[1-methyl-4[1-methyl-4[1-methyl-5-N,N-bis(2-chlorethyl)-aminoindol-2-carboxamido]pyrrol-2-carboxamido]pyrrol-2-carboxamido]-pyrrol-2-carboxamido]propionamidin;
- 3. 3-[1-Methyl-4[1-methyl-4[1-methyl-4[5-N,N-bis(2-chlorethyl)aminobenzofuran-2-carboxamido]pyrrol-2-carboxamido]pyrrol-2-carboxamido]-pyrrol-2-carboxamido]propionamidin;

- Ein weiteres Ziel der vorliegenden Erfindung ist ein Verfahren zur Herstellung der Verbindungen der Formel (I) und deren pharmazeutisch annehmbaren Salzen, umfassend: (1)(a) Reagieren einer Verbindung der Formel (IV) worin n 2, 3 oder 4 ist, mit einer Verbindung der Formel: worin:
A O, S oder NR ist, wobei R Wasserstoff oder C1-4-Alkyl ist;
B ist CH oder N;
R1 ist Wasserstoff oder C1-4-Alkyl;
T ist ausgewählt aus: (i) worin p 0 oder 1 ist; R2 und R3 sind unabhängig voneinander ausgewählt aus Wasserstoff, C1-4-Alkyl, gegebenenfalls substituiert mit ein oder mehreren Fluoratomen, und C1-4-Alkoxy; R4 ist C1-4-Alkyl oder C1-3-Haloalkyl; X1 ist ein Halogenatom; und (ii) worin X2 ein Halogenatom ist;
Y ist Hydroxy oder eine Abgangsgruppe;
um eine Verbindung der Formel (I), wie oben definiert, zu erhalten; oder: (b) Reagieren einer Verbindung der Formel worin n, A, B und R1 wie oben definiert sind, mit einer Verbindung worin X1, R2, R3 und R4 wie oben definiert sind, und Y1 ist Hydroxy oder eine Abgangsgruppe,

um eine Verbindung der Formel (I), wie oben definiert, zu erhalten, wobei T eine Gruppe der Formel (II) gemäss Punkt (i) ist, mit p = 1, oder eine Gruppe der Formel (III) gemäss Punkt (ii);
oder: (c) Reagieren einer Verbindung der Formel worin p, n, A, B, R1, R2 und R3 wie oben definiert sind, und R5 ist Wasserstoff oder C1-4-Alkyl, mit Ethylenoxid, um eine Verbindung der Formel: zu erhalten, worin p, n, A, B, R1, R2 und R3 wie oben definiert sind, und R6 ist gleich R5, wenn R5 C1-4-Alkyl ist, oder R6 ist gleich -CH2CH2-OH, wenn R5 Wasserstoff ist;
und dann Reagieren der Verbindung der Formel (IX) mit einem Halogenierungsmittel, um eine Verbindung der Formel (I) zu erhalten, wie oben definiert, worin T eine Gruppe der Formel (II) gemäss Punkt (i) mit p = 0 oder 1 ist; und
(2) wenn notwendig, Umwandeln der so erhaltenen Verbindung der Formel (I) in ein pharmazeutisch annehmbares Salz davon. - Die Reaktion der Verbindung der Formel (IV) mit einer Verbindung der Formel (V) (Verfahren (a)) kann gemäss bekannten Verfahren durchgeführt werden, z. B. solchen, beschrieben in EP-B-246 868.
- Die Reaktion zwischen einer Verbindung der Formel (IV) und einer Verbindung der Formel (V), worin Y Hydroxy ist, wird bevorzugt in einem molaren Verhältnis von (IV) : (V) von 1 : 1 bis 1 : 2 durchgeführt, in einem organischen Lösungsmittel, wie z. B. Dimethylsulfoxid, Dimethylacetamid, Dimethylformamid, Ethanol, Benzol oder Pyridin, in Anwesenheit einer organischen oder anorganischen Base, wie z. B. Triethylamin, N,N'-Diisopropylethylamin oder Natrium- oder Kaliumcarbonat oder -bicarbonat, und einem Kondensationsmittel, wie z. B. N-Ethyl-N'-(3-dimethylaminopropyl)carbodiimid, N,N'-Dicyclohexylcarbodiimid oder 1-Hydroxybenzotriazolhydrat. Die Reaktionstemperatur kann von –10 bis ca. 100°C und die Reaktionszeit von ca. 1 bis 24 Stunden variieren.
- Die Reaktion zwischen einer Verbindung der Formel (IV) und einer Verbindung der Formel (V), wobei Y eine Abgangsgruppe ist, wie oben definiert, kann mit einem molaren Verhältnis von (IV) : (V) von ca. 1 : 1 bis ca. 1 : 2 durchgeführt werden, in einem organischen Lösungsmittel, wie z. B. Dimethylformamid, Dioxan, Pyridin, Tetrahydrofuran oder Mischungen davon mit Wasser, gegebenenfalls in Anwesenheit einer organischen Base, z. B. N,N'-Diisopropylethylamin, Triethylamin, oder einer anorganischen Base, z. B. Natrium- oder Kaliumbicarbonat, bei einer Temperatur von ca. 0 bis ca. 100°C und über einen Zeitraum von ca. 2 bis ca. 48 Stunden.
- Die Verbindungen der Formel (IV) sind bekannte Verbindungen oder können gemäss bekannten Verfahren aus bekannten Verbindungen hergestellt werden, siehe z. B. Arcamone et al., Gazzetta Chim. Ital. 97, 1097 (1967).
- Die Verbindungen der Formel (V), worin Y Hydroxy ist und T eine Gruppe der Formel (II gemäss Punkt (i) mit p = 1 oder eine Gruppe der For mel (III) gemäss Punkt (ii) ist, können hergestellt werden durch Reagieren einer Aminoverbindung der Formel: worin A, B und R1 wie oben definiert sind, mit einer Carbonsäure oder einem Derivat davon der Formel (VIIa) oder (VIIb), wie oben definiert.
- Die Verbindungen der Formel (V), worin Y Hydroxy ist und T eine Gruppe der Formel (II) gemäss Punkt (i) mit p = 0 ist, können durch Reagieren einer Verbindung der Formel: worin A, B, R1 und R5 wie oben definiert sind, mit Ethylenoxid und anschliessend mit einem Halogenierungsmittel, analog zu dem für Verfahren (c) beschriebenen, hergestellt werden. Vor Durchführung der Reaktion wird die Carboxylgruppe bevorzugt mit einer geeigneten Schutzgruppe gemäss bekannten Techniken geschützt.
- Die Verbindung der Formel (V), worin Y eine Abgangsgruppe ist, können ausgehend von den entsprechenden Säuren durch bekannte Reaktionen hergestellt werden.
- Die Carbonsäuren der Formeln (VIIa) und (VIIb) oder deren Derivate sind kommerziell erhältliche Produkte oder können durch Reaktionen, wie sie in der organischen Chemie allgemein bekannt sind, hergestellt werden (siehe z. B. Tetrahedron Letters 31, 1299 (1990), Anti-cancer Drug Design 9, 511 (1994), JACS 62, 3495 (1940), J. Org. Chem. 26, 4996–97 (1961) oder Synth. Commun. 24, 3129–3134 (1994)).
- Die Verbindungen der Formeln (X) und (XI) sind kommerzielle Produkte oder sie können durch bekannte Verfahren hergestellt werden (siehe z. B. J. Am. Chem. Soc. 80, 4621 (1958), Helv. Chim. Acta 31, 75 (1948), Synth. Commun. 21, 959 (1991), Anti-cancer Drug Design 10, 25 (1995), J. Org. Chem. 26, 4996–97 (1961) oder Synth. Commun. 24, 3129–3134 (1994)).
-
- Die Nitroderivate der Formel (XII) können entsprechend erhalten werden durch Reagieren einer Verbindung der Formel (VI), wie oben definiert, mit einer Verbindung der Formel: worin A, B und R1 wie oben definiert sind und Y3 ist Hydroxy oder eine Abgangsgruppe. Die Verbindungen der Formel (VIII) sind bekannte Verbindungen und können durch bekannte Verfahren erhalten werden (siehe z. B. Tetrahedron Letters 31, 1299 (1990), Anti-cancer Drug Design 9, 511 (1994), JACS 62, 3495 (1940), J. Org. Chem. 26, 4996–97 (1963) oder Synth. Commun. 24, 3129–3134 (1994)).
- Die Reaktion gemäss Verfahren (c) kann analog zu den in US-PS 4 738 980 beschriebenen durchgeführt werden. Das Halogenierungsmittel kann z. B. reines Halogenid, wie Chlor oder Brom, oder ein Thionylhalogenid, wie Thionylchlorid, sein. Die Ausgangsverbindungen der Formel (VIII) können gemäss bekannten Reaktionen erhalten werden, z. B. durch Monoalkylierung der Aminoverbindungen der Formel (VI), wie oben definiert, gegebenenfalls zuvor kondensiert mit entsprechender Aminobenzoesäure oder einem Derivat davon.
- In den obigen Formeln sind die Gruppen Y, Y1, Y2 und Y3 Hydroxy oder Abgangsgruppen, ausgewählt z. B. aus Chlor, 2,4,5-Trichlorphenoxy, 2,4-Dinitrophenoxy, Succinimido-N-oxy, Imidazolyl und dergleichen.
- Die Versalzung der Verbindung der Formel (I), genauso wie die Herstellung der freien Verbindung aus einem Salz, können gemäss bekannten Standardverfahren durchgeführt werden. Allgemein bekannte Verfahren, wie z. B. fraktionierte Kristallisation oder Chromatografie, können genutzt werden zur Trennung einer Isomerenmischung der Formel (I) in die einzelnen Isomere.
- Die Verbindungen der Formel (I) können durch herkömmliche Techniken gereinigt werden, wie z. B. Silicagel- oder Aluminiumoxid-Säulenchromatografie, und/oder durch Rekristallisation aus einem organischen Lösungsmittel, wie z. B. einem niederaliphatischen Alkohol, z. B. Methyl- Ethyl- oder Isopropylalkohol oder Dimethylformamid.
- PHARMAKOLOGIE:
- Die erfindungsgemässen Verbindungen der Formel (I) sind als antineoplastische Mittel und als Antivirusmittel nützlich. Insbesondere zeigen sie cytostatische Eigenschaften gegenüber Tumorzellen auf, so dass sie nützlich zur Hemmung des Wachstums von verschiedenen Tumoren in Säugern, einschliesslich Menschen, sind, wie z. B. Karzinomen, z. B. Mammakarzinom, Lungenkarzinom, Blasenkarzinom, Kolonkarzinom, Eierstock- und Gebärmuttertumoren. Andere Neoplasien, bei denen die erfindungsgemässen Verbindungen Anwendung finden können, sind z. B. Sarkome, z. B. Weichgewebe- oder Knochensarkome, und hämatologische Erkrankungen, wie Leukämien.
- Die in vitro-Antitumoraktivität wurde durch Cytotoxizitätsstudien unter Verwendung der Maus L1210-Leukämiezellen untersucht. Die Zellen stammen aus in vivo-Tumoren und wurden in der Zellkultur etabliert. Die Zellen wurden bis zur zehnten Passage verwendet und die Cytotoxizität wurde durch Zählen der überlebenden Zellen nach 48-stündiger Behandlung gezählt.
- Der Prozentsatz an Zellwachstum in den behandelten Kulturen wurde mit denen der Kontrollen verglichen. IC50-Werte (Konzentration, die 50% des Zellwachstums im Vergleich zur Kontrolle hemmt) wurden aufgrund der Dosisantwort berechnet.
- Die erfindungsgemässen Verbindungen wurden ebenfalls in vivo mit L1210 Maus-Leukämien und Maus-Retikulosarkomen M5076 getestet, sie zeigten eine sehr gute Antitumoraktivität bei dem folgenden Verfahren.
- L1210 Maus-Leukämie wurde in vivo erhalten durch serielle i. v.-Transplantation. Für die Experimente wurden 105 Zellen i. p. in weibliche CD2F1 Mäuse, erhalten von Charles River, Italien, injiziert. Die Tiere waren zu Beginn der Experimente 8 bis 10 Wochen alt. Die Verbindungen wurden i. v. am Tag +1 nach Tumorzellinjektionen verabreicht.
- M5076-Retikulosarkom wurde in vivo durch serielle i. m.-Transplantation erhalten. Für die Experimente wurden 5 × 105 Zellen pro Maus i. m. in weibliche C57B16-Mäuse, erhalten von Charles River, Italien, injiziert. Die Tiere waren zu Beginn der Experimente 8 bis 10 Wochen alt. Die Verbindungen wurden i. v. an den Tagen 3, 7 und 11 nach Tumorinjektion verabreicht.
- Die Überlebenszeit der Mäuse und das Tumorwachstum wurden berechnet und die Aktivität wurde ausgedrückt als T/C % und T. I. %.
- T. I. = % Hemmung des Tumorwachstums in bezug auf die Kontrolle
- Tox = Zahl der Mäuse, die aufgrund von Toxizität gestorben sind
- Die Tox-Bestimmung wurde durchgeführt, wenn die Mäuse vor der Kontrolle verstarben und/oder sich bei der Überprüfung ein signifikanter Körpergewichtsverlust und/oder Milz- und/oder Lebergrössenreduktion zeigten.
- Die erfindungsgemässen Verbindungen zeigen ebenfalls eine deutliche Wirksamkeit in der Wechselwirkung mit der Reproduktionsaktivität von pathogenen Viren und schützen Gewebezellen vor viralen Infektionen. Zum Beispiel zeigen sie eine Aktivität gegenüber DNA-Viren, wie z. B. Herpes, z. B. Herpes simplex- und Herpes zoster-Viren, Vaccinaviren, RNA-Viren, wie z. B. Rhinoviren und Adenoviren, und gegenüber Retroviren, wie z. B. Sarkomaviren, z. B. Maus-Sarkomavirus, und Leukämieviren, wie z. B. Friend Leukemia Virus.
- Die Wirksamkeit gegenüber Herpes-, Coxsackie- und Atemwegssyncytiaviren wurde z. B. in einem flüssigen Medium wie folgt getestet. Serielle zweifache Verdünnungen der Verbindungen von 200 bis 1,5 mcg/ml wurden zweifach mit 0,1 ml/Well in 96 Well-Mikroplatten für die Gewebekultur gegeben. Die Zellsuspensionen (2 × 105 Zellen/ml), infiziert mit ca. 5 × 10–3 TClD50 Virus/Zelle, wurden sofort mit 0,1 ml/Well hinzugegeben.
- Nach 3- bis 5-tägiger Inkubation bei 37°C in 5% CO2 wurden die Zellkulturen mikroskopisch untersucht und die minimale Hemmungskonzentration (Minimum Inhibiting Concentration (MIC)) wurde bestimmt, die MIC ist die minimale Konzentration, die eine Reduktion des cytopathologischen Effekts im Vergleich mit den infizierten Kontrollen aufzeigt.
- Die erfindungsgemässen Verbindungen können bei Säugern, einschliesslich Menschen, über übliche Wege verabreicht werden, z. B. parenteral, z. B. durch intravenöse Injektion oder Infusion, intramuskulär, subkutan, topisch oder oral. Die Dosierung hängt vom Alter, Gewicht und Zustand des Patienten und dem Verabreichungsweg ab. Eine geeignete Dosierung zur Verabreichung an den erwachsenen Menschen liegt z. B. im Bereich von ca. 0,1 bis ca. 150 bis 200 mg pro Dosis 1 bis 4 mal am Tag.
- Eine weitere Aufgabe der vorliegenden Erfindung sind pharmazeutische Zusammensetzungen, die eine Verbindung der Formel (I) als Wirkstoff umfassen, in Verbindung mit einem oder mehreren pharmazeutisch annehmbaren Trägern und/oder Verdünnern.
- Die erfindungsgemässen pharmazeutischen Zusammensetzungen werden üblicherweise gemäss herkömmlichen Verfahren hergestellt und werden in einer pharmazeutisch annehmbaren Form verabreicht. Zum Beispielen können Lösungen zur intravenösen Injektion oder Infusion als Träger z. B. steriles Wasser enthalten, oder sie können bevorzugt in Form steriler wässriger isotonischer Salzlösungen sein.
- Suspensionen oder Lösungen für intramuskuläre Injektionen können zusammen mit dem Wirkstoff einen pharmazeutisch annehmbaren Träger enthalten, z. B. steriles Wasser, Olivenöl, Ethyloleat, Glykole, z. B. Propylenglykol, und, wenn gewünscht, eine geeignete Menge an Lidocain-hydrochlorid.
- In den Formen zur topischen Verabreichung, z. B. bei Cremes, Lotionen oder Pasten zur dermatologischen Behandlung, kann der Wirkstoff mit herkömmlichen öligen oder emulgierenden Exzipienten vermischt sein.
- Die festen oralen Formen, z. B. Tabletten und Kapseln, können neben dem Wirkstoff Verdünner, z. B. Lactose, Dextrose, Saccharose, Cellulose, Maisstärke und Kartoffelstärke; Schmiermittel, z. B. Silica, Talk, Stearinsäure, Magnesium oder Calciumstearat und/oder Polyenglykole; Bindemittel, z. B. Stärke, Gummi arabicum, Gelatine, Methylcellulose, Carboxymethylcellulose, Polyvinylpyrrolidon; Zersetzungsmittel, z. B. Stärke, Algininsäure, Alginate, Natriumstärkeglykolate; Brausemischungen; Färbemittel; Süssstoffe; Benetzungsmittel, z. B. Lecithinpolysorbate, Laurylsulfate; und allgemein nicht-toxische und pharmakologisch inaktive Substanzen, wie sie in pharmazeutischen Formulierungen verwendet werden, enthalten. Diese pharmazeutischen Präparate können durch bekannte Techniken hergestellt werden, z. B. durch Mischen, Granulieren, Tablettieren, Zuckerbeschichtungs- oder Filmbeschichtungsverfahren.
- Eine weitere Aufgabe der vorliegenden Erfindung sind Verbindungen der Formel (I) zur Verwendung in einem Verfahren zur therapeutischen Behandlung des menschlichen oder tierischen Körpers.
- Weiterhin stellt die vorliegende Erfindung ein Verfahren zur Behandlung von Tumoren und viralen Infektionen in einem Patienten, der dieses benötigt, bereit; dies umfasst die Verabreichung einer erfindungsgemässen Zusammensetzung an diesen Patienten.
- Eine weitere Aufgabe der vorliegenden Erfindung ist ein kombiniertes Verfahren zur Behandlung von Krebs oder zur Minderung von Zuständen bei Säugern, einschliesslich Menschen, die an Krebs leiden; dieses Verfahren umfasst die Verabreichung einer Verbindung der Formel (I) oder eines pharmazeutisch annehmbaren Salzes davon und eines weiteren Antitumormittels, zeitnahe genug und in ausreichender Menge, um eine therapeutisch nützliche Wirkung aufzuzeigen.
- Die vorliegende Erfindung stellt weiterhin Produkte, enthaltend eine Verbindung der Formel (I), oder ein pharmazeutisch annehmbares Salz davon bereit, und ein weiteres Antitumormittel als Kombinationspräparat zur gleichzeitigen, getrennten oder aufeinanderfolgenden Verwendung in der Antikrebstherapie.
- Der Begriff "Antitumormittel" umfasst sowohl einzelne Antitumor-Arzneimittel als auch "Cocktails", d. h. eine Mischung von solchen Arzneimitteln, gemäss der klinischen Praxis. Beispiele für Antitumormittel, die mit der Verbindung der Formel (I) formuliert sein können oder alternativ in einem kombinierten Behandlungsverfahren verabreicht werden können, schliessen ein: Doxorubicin, Daunomycin, Epirubicin, Idarubicin, Etoposid, Fluoruracil, Melphalan, Cyclophosphamid, 4-Desmethoxydaunorubicin, Bleomycin, Vinblastin und Mitomycin oder Mischungen davon.
- Die folgenden Beispiele dienen dem besseren Verständnis der Erfindung.
- BEISPIEL 1
- 3-[1-Methyl-4[1-methyl-4[1-methyl-4[5-N,N-bis(2-chlorethyl)aminoindol-2-carboxamido]pyrrol-2-carboxamido]pyrrol-2-carboxamido]pyrrol-2-carboxamido]propionamidin
- Schritt I: Zwischenprodukt 5-N,N-Bis(2-chlorethyl)aminoindol-2-carbonsäure
- Zu einer Lösung von 200 mg Ethyl-5-aminoindol-2-carboxylat (hergestellt wie in J. Am. Chem. Soc. 80, 4621 (1958) beschrieben), in 10 ml Methanol auf –10°C gekühlt, wurde kaltes Ethylenoxid (2,5 ml) zugefügt. Das Reaktionsgefäss wurde verschlossen und konnte über nacht Raumtemperatur annehmen. Methanol und überschüssiges Ethylenoxid wurden durch Verdampfen entfernt und der Rest wurde durch Flash-Chromatografie auf gereinigt, wobei 230 mg Ethyl-5-N,N-bis(2-hydroxyethyl)aminoindol-2-carboxylat erhalten wurden, dieses wurde auf Eis gekühlt und 2 ml Phosphoroxychlorid wurden hinzugegeben. Die Lösung wurde auf 100°C für 1 Stunde erwärmt, anschliessend wurde das Lösungsmittel unter Vakuum abgedampft, der Rest in 7 ml 23%-iger Salzsäure gelöst und für 2 Stunden auf 100°C erwärmt.
- Die Lösung wurde auf Raumtemperatur abgekühlt, mit 30 ml Wasser verdünnt und zweimal mit Ethylacetat extrahiert (2 × 50 ml). Die organischen Phasen wurden im Vakuum abgedampft und der Rest wurde durch Flash-Chromatografie unter Verwendung einer Methylenchlorid/Methanol-Mischung aufgereinigt, was 220 mg des Zwischenprodukts ergab.
- Durch analoge Verfahren und unter Verwendung der entsprechenden Ausgangsmaterialien wurden die folgenden Produkte erhalten:

- Eine Lösung von 263 mg N-Desformyldistamycin A-dihydrochlorid (hergestellt wie in J. Med. Chem. 32, 774–778 (1989) beschrieben) in 5 ml wasserfreiem Dimethylformamid (DMF) wurde auf 5°C gekühlt und mit 86 ml N,N'-Diisopropylethylamin versetzt. Nach 10 Minuten wurden 175 mg des Zwischenprodukts, erhalten in Schritt (I), und 192 mg N-Ethyl-N'-(3-dimethylaminopropyl)carbodiimid (EDCI) hinzugefügt. Die Reaktion wurde bei Raumtemperatur für 10 Stunden gerührt und anschliessend wurde 2 N Salzsäure zugegeben, um einen pH von 4 zu erhalten. Das Lösungsmittel wurde unter reduziertem Druck entfernt und der Rest wurde durch Flash-Chromatografie auf gereinigt (Methylenchlorid/Methanol 8 : 2), um 210 mg der Titelverbindung als gelben Feststoff zu ergeben.

- 3-[1-Methyl-4[1-methyl-4[1-methyl-4[5(α-bromacrylamido)benzofuran-2-carboxamido]pyrrol-2-carboxamido]pyrrol-2-carboxamido]pyrrol-2-carboxamido]propionamidin-hydrochlorid
- Schritt I: Zwischenprodukt 5-α-Bromacrylamidobenzofuran-2-carbonsäure
- Zu einer Lösung von 500 mg kommerzieller α-Bromacrylsäure in 5 ml Acetonitril wurde eine Lösung von 343 mg N,N-Dicyclohexylcarbodiimid in 15 ml Acetonitril langsam zugefügt. Nach 1 Stunde wurde die erhaltene Lösung nach Filtration des Präzipitats zu einer Lösung von 294 mg 5-Amino-2-benzofuransäure (hergestellt gemäss Helv. Chim. Acta 31, 75 (1948)) und 229 mg Natriumbicarbonat in 20 ml Wasser hinzugefügt. Die Reaktion wurde bei Raumtemperatur für 1 Stunde gerührt, dann wurde 2 N Salzsäure hinzugefügt bis der pH 4 war. Die Lösung wurde mit Ethylacetat extrahiert (3 × 10 ml), über Natriumsulfat getrocknet und zur Trockene im Vakuum abgedampft und der Rest wurde durch Flash-Chromatografie mit einer Methylenchlorid/Methanol-Mischung aufgereinigt, um 500 mg des Zwischenprodukts als blassgelben Feststoff zu erhalten.
- Durch analoge Verfahren und unter Verwendung der entsprechenden Ausgangsmaterialien wurden die folgenden Produkte erhalten:
5-α-Bromacrylamidobenzothiophen-2-carbonsäure;

- Eine Lösung von 263 mg N-Desformyldistamycin A-dihydrochlorid (hergestellt wie in J. Med. Chem. 32, 774–778 (1989) beschrieben) in 5 ml wasserfreiem DMF wurde auf 5°C gekühlt und mit 0,086 ml N,N'-Diisopropylethylamin versetzt. Nach 10 Minuten wurden 180 mg des Zwischenprodukts, erhalten in Schritt I, und 192 mg N-Ethyl-N'-(3-dimethylaminopropyl)carbodiimid (EDCI) hinzugegeben. Die Reaktion wurde bei Raumtemperatur für 16 Stunden gerührt, anschliessend wurde 2 N Salzsäure bis zu einem pH von 4 zugegeben. Das Lösungsmittel wurde im Vakuum abgedampft und der rohe Rest wurde durch. Flash- Chromatografie (Methylenchlorid/Methanol 8 : 2) aufgereinigt, um ein gelbes Öl zu erhalten, das aus Methanol/Diethylether präzipitiert wurde, um 240 mg der Titelverbindung als blassgelben Feststoff zu erhalten.

- 3-[1-Methyl-4[1-methyl-4[1-methyl-4[1-methyl-5-[4-N,N-bis(2-chlorethyl)aminobenzol-1-carboxamido]indol-2-carboxamido]pyrrol-2-carboxamido]-pyrrol-2-carboxamido]pyrrol-2-carboxamido]propionamidin-hydrochlorid:
- Schritt I: Zwischenprodukt 1-Methyl-5-[4-N,N-bis(2-chlorethyl)aminobenzol-1-carboxamido]indol-2-carbonsäures
- Zu einer Lösung von 495 mg 4-N,N-Bis(2-chlorethyl)aminobenzoesäure (hergestellt wie in Beispiel 1, Schritt I, beschrieben) in 20 ml Benzol wurde 1 ml Thionylchlorid hinzugefügt. Die Mischung wurde für 2 Stunden unter Rückfluss gehalten, das Lösungsmittel unter Vakuum abgedampft, der feste Rohstoff in 15 ml Dioxan gelöst und tropfenweise zu einer Lösung von 167 mg 1-Methyl-5-aminoindol-2-carbonsäure (hergestellt wie in J. Am. Chem. Soc. 80, 4621 (1958) beschrieben) und 239 mg Natriumbicarbonat in 20 ml Wasser zugefügt.
- Die Mischung wurde für 1 Stunde bei Raumtemperatur gerührt und anschliessend mit 2 N Salzsäure auf einen pH von 4 gebracht. Das Lösungsmittel wurde im Vakuum abgedampft und der Rest wurde durch Flash-Chromatografie auf Silicagel mit Methylenchlorid/Methanol-Mischung aufgereinigt, um 400 mg des Zwischenprodukts zu ergeben.
- Durch analoge Verfahren und unter Verwendung der entsprechenden Ausgangsmaterialien können die folgenden Produkte hergestellt werden:

- Zu einer Lösung von 320 mg Zwischenprodukt (hergestellt wie in Schritt I beschrieben) in 10 ml Benzol wurden 0,5 ml Thionylchlorid hinzugefügt. Die Mischung wurde für 2 Stunden unter Rückfluss gehalten, das Lösungsmittel unter Vakuum abgedampft, der feste Rohstoff in 15 ml Dioxan gelöst und portionsweise zu einer Lösung von 200 mg N-Desformyldistamycin A-dihydrochlorid (hergestellt wie in J. Med. Chem. 32, 774–778 (1989) beschrieben), 95 mg Natriumbicarbonat in 10 ml Wasser hinzugefügt.
- Die Mischung wurde für 1 Stunde gerührt und anschliessend mit 2 N Salzsäure auf einen pH von 4 gebracht. Das Lösungsmittel wurde im Vakuum abgedampft und der Rest durch Flash-Chromatografie (Methylenchlorid/Methanol 8 : 2) aufgereinigt, um 250 mg der Titelverbindung als blassgelben Feststoff zu ergeben.

- Schritt I: Zwischenprodukt 3-[1-Methyl-4[1-methyl-4[1-methyl-4[4-nitrobenzothiophen-2-carboxamido]pyrrol-2-carboxamido]-pyrrol-2-carboxamido]pyrrol-2-carboxamido]propionamidindihydrochlorid
- Zu einer Lösung von 156 mg 4-Nitrobenzothiophen-2-carbonsäure (hergestellt wie in Synth. Commun. 21, 959 (1991) beschrieben) in 10 ml Benzol wurden 0,5 ml Thionylchlorid hinzugefügt. Die Mischung wurde für 2 Stunden unter Rückfluss gehalten, das Lösungsmittel wurde unter Vakuum abgedampft, der feste Rohstoff wurde in 15 ml Dioxan gelöst und portionsweise zu einer Lösung von 200 mg N-Desformyldistamycin A-dihydrochlorid (hergestellt wie in J. Med. Chem. 32, 774–778 (1989) beschrieben), 95 mg Natriumbicarbonat in 10 ml Wasser gegeben.
- Die Mischung wurde für 1 Stunde bei Raumtemperatur gerührt und anschliessend wurde 2 N Salzsäure bis zu einem pH von 4 hinzugefügt. Das Lösungsmittel wurde im Vakuum abgedampft und der Rest wurde durch Plash-Chromatografie mit Methylenchlorid/Methanol-Mischung aufgereinigt, um 220 mg der Titelverbindung als Feststoff zu erhalten.
- Durch analoge Verfahren und unter Verwendung der entsprechenden Ausgangsmaterialien können die folgenden Produkte erhalten werden:

- Das Derivat (220 mg), das in Schritt I erhalten wurde, wurde in 10 ml DMF gelöst und über Pd-Katalysator (10% auf Kohle) unter reduziertem Druck (50 psi; 3,45 bar) in einer Parr-Vorrichtung reduziert. Die erhaltene Lösung wurde nach Abfiltrieren des Katalysators im Vakuum abgedampft und der feste Rest wurde in 5 ml wasserfreiem DMF gelöst, auf 5°C gekühlt und mit 0,055 ml N,N'-Diisopropylethylamin versetzt. Nach 10 Minuten wurden 100 mg 4-N,N-Bis(2-chlorethyl)aminobenzoesäure (hergestellt wie in Beispiel 1, Schritt I beschrieben) und 123 mg N-Ethyl-N'-(3-dimethylaminopropyl)carbodiimid wurden zugefügt. Die Lösung wurde für 12 Stunden bei Raumtemperatur gerührt, anschliessend wurde 2 N Salzsäure bis zu einem pH von 4 hinzugegeben. Das Lösungsmittel wurde abgedampft und der Rohstoff wurde durch Flash-Chromatografie (Methylenchlorid/Methanol 8 : 2) gereinigt, um 200 mg der Titelverbindung als gelben Feststoff zu ergeben.

Claims (10)
- Verbindung eines benzoheterocyclischen Distamycinderivats der Formel: worin: n 2, 3 oder 4 ist; A ist O, S oder NR, wobei R Wasserstoff oder C1-4-Alkyl ist; B ist CH oder N; R1 ist Wasserstoff oder C1-4-Alkyl; T ist ausgewählt aus (i) worin: p 0 oder 1 ist; R2 und R3 sind jeweils unabhängig ausgewählt aus Wasserstoff, C1-4-Alkyl, gegebenenfalls substituiert mit einem oder mehreren Fluoratomen, und C1-4-Alkoxy; R4 ist C1-4-Alkyl oder C1-3-Haloalkyl; X1 ist ein Halogenatom; und (ii) worin: X2 ein Halogenatom ist; oder ein pharmazeutisch annehmbares Salz davon.
- Verbindung gemäss Anspruch 1, worin: n 2 oder 3 ist; A ist O, S, NH oder NCH3; B ist CH oder N; R1 ist Wasserstoff; T ist ein Gruppe der Formel (II) gemäss Punkt (i), wobei X1 Chlor oder Brom ist; R4 ist Ethyl, 2-Chlorethyl oder 2-Bromethyl; R2 und R3 sind unabhängig voneinander Wasserstoff oder Methyl; p ist 0 oder 1; oder T ist eine Gruppe der Formel (III) gemäss Punkt (ii), wobei X2 Brom oder Chlor ist.

3-[1-Methyl-4[1-methyl-4[1-methyl-4[1-methyl-5-[4-N-ethyl-N(2-chlorethyl)aminobenzol-1-carboxamido]benzoimidazol-2-carboxamido]pyrrol-2-carboxamido]pyrrol-2-carboxamido]pyrrol-2-carboxamido]propionamidin; und die pharmazeutisch annehmbaren Salze hiervon.
- Verfahren zur Herstellung einer Verbindung gemäss Anspruch 1, das Verfahren umfasst: (1)(a) Reagieren einer Verbindung der Formel (IV) worin n 2, 3 oder 4 ist, mit einer Verbindung der Formel: worin: A O, S oder NR ist, wobei R Wasserstoff oder C1-4-Alkyl ist; B ist CH oder N; R1 ist Wasserstoff oder C1-4-Alkyl; T ist ausgewählt aus: (i) worin p 0 oder 1 ist; R2 und R3 sind unabhängig voneinander ausgewählt aus Wasserstoff, C1-4-Alkyl, gegebenenfalls substituiert mit ein oder mehreren Fluoratomen, und C1-4-Alkoxy; R4 ist C1-4-Alkyl oder C1-3-Haloalkyl; X1 ist ein Halogenatom; und (ii) worin X2 ein Halogenatom ist; Y ist Hydroxy oder eine Abgangsgruppe; um eine Verbindung der Formel (I), wie oben definiert, zu erhalten; oder: (b) Reagieren einer Verbindung der Formel worin n, A, B und R1 wie oben definiert sind, mit einer Verbindung der Formel: worin X1, R2, R3 und R4 wie oben definiert sind, und Y1 ist Hydroxy oder eine Abgangsgruppe, oder mit einer Verbindung der Formel: worin X2 wie oben definiert ist, und Y2 ist Hydroxy oder eine Abgangsgruppe; um eine Verbindung der Formel (I), wie oben definiert, zu erhalten, wobei T eine Gruppe der Formel (II) gemäss Punkt (i) ist, mit p = 1, oder eine Gruppe der Formel (III) gemäss Punkt (ii); oder: (c) Reagieren einer Verbindung der Formel worin p, n, A, B, R1, R2 und R3 wie oben definiert sind, und R5 ist Wasserstoff oder C1-4-Alkyl, mit Ethylenoxid, um eine Verbindung der Formel: zu erhalten, worin p, n, A, B, R1, R2 und R3 wie oben definiert sind, und R6 ist gleich R5, wenn R5 C1-4-Alkyl ist, oder R6 ist gleich -CH2CH2-OH, wenn R5 Wasserstoff ist; und dann Reagieren der Verbindung der Formel (IX) mit einem Halogenierungsmittel, um eine Verbindung der Formel (I) zu erhalten, wie oben definiert, worin T eine Gruppe der Formel (II) gemäss Punkt (i) mit p = 0 oder 1 ist; und (2) wenn notwendig, Umwandeln der erhaltenen Verbindung der Formel (I) in ein pharmazeutisch annehmbares Salz davon.
- Verbindung gemäss einem der Ansprüche 1 bis 3 zur Verwendung in einem therapeutischen Behandlungsverfahren des menschlichen oder tierischen Körpers.
- Verbindung gemäss Anspruch 5 zur Verwendung als antineoplastisches Mittel.
- Verbindung gemäss Anspruch 5 zur Verwendung als antivirales Mittel.
- Verwendung einer Verbindung gemäss einem der Ansprüche 1 bis 3 bei der Herstellung eines Medikaments zur Verwendung bei der Behandlung von Krebs.
- Verwendung einer Verbindung gemäss einem der Ansprüche 1 bis 3 zur Herstellung eines Medikaments zur Verwendung bei der Behandlung einer viralen Infektion.
- Pharmazeutische Zusammensetzung, die eine wirksame Menge einer Verbindung gemäss einem der Ansprüche 1 bis 3 als Wirkstoff in Verbindung mit ein oder mehreren pharmazeutisch annehmbaren Trägern und/oder Verdünnern aufweist.
