(54) Fremgangsmåde til fremstilling af vandopløselige beer er bundne protei® ner.
Den foreliggende opfindelse angår en fremgangsmåde til fremstilling af vandopløselige bærerbundne proteiner, hvor et vandopløseligt carbon-hydrat aktiveres med et cyanhalogenid under alkaliske betingelser, hvorefter det således aktiverede carbonhydrat omsættes med et protein.
I USA patentskrift nr. 3.639.213 og andetsteds har det været foreslået, at forskellige enzymer nyttigt kan bindes til en vandopløselig matrix bestående af carbonhydratmolekyler eller lignende. Det er fastslået, at et enzym eller andre former for proteiner kan stabiliseres på denne måde, og at det matricebundne produkt kan anvendes på forskellige måder. F.eks. kan nogle præparater anvendes som lægemiddel til injektion, hvorved der opnås en formindsket antistofreaktion mod epzymet eller det pågældende protein som følge af, at enzymet er kemisk bundet til det forholdsvis biologisk inerte carbonhydrat.
I USA patentskrift nr. 3.645.852 har det været foreslået at omsætte polysaccharider, såsom cellulose, eller dextrancopolymere med cyan- 2 141292 halogenider i et vandigt medium for at aktivere materialet til reaktion med et enzym eller et andet protein.
Vandopløselige carbonhydrater, såsom dextran, kan aktiveres ved hjælp af de i USA patentskrift nr. 3.645.852 omhandlede metoder til omsætning med enzymer og lignende. Under aktiveringsreaktionen har dele af dextranreaktanterne imidlertid tendens til at blive vanduopløselige formodentlig på grund af tværbindingssidereaktioner. Dette er naturligvis en væsentlig negativ faktor, når man søger at fremstille vandopløselige matricebundne enzymer.
I overensstemmelse med den foreliggende opfindelse tilvejebringes en metode til frembringelse af høje udbytter af vandopløselige matricebundne enzymer, som kan være praktisk taget frie for vanduopløselige biprodukter. Ydermere kan der ved fremgangsmåden ifølge opfindelsen tilvejebringes vandopløselige matricebundne enzymprodukter med en væsentligt forøget enzymaktivitet pr. mg i sammenligning med fra teknikken kendte metoder til vandig aktivering med cyanhalogenider.
Den foreliggende opfindelse angår i overensstemmelse hermed en fremgangsmåde til fremstilling af vandopløselige bærerbundne proteiner, hvor et vandopløseligt carbonhydrat aktiveres med et cyanhalogenid under alkaliske betingelser, hvorefter 1-300 vægtdele af det således aktiverede carbonhydrat i vandig opløsning omsættes med 1 vægtdel af et protein, hvilken fremgangsmåde er ejendommelig ved, at aktiveringen af carbonhydratet med et cyanhalogenid foretages i nærværelse af en absolut alkohol, der ikke indeholder mere end 3 carbonatomer og 1 hydroxylgruppe, eller n-butanol, og i nærværelse af et opløst alkali med formlen MOR i en koncentration, som er tilstrækkelig til at tilvejebringe en pH-værdi på mindst 10, hvis samme koncentration af alkali sattes til deioniseret vand, i hvilken formel M betegner et alkalimetal og R en lavere alkylgruppe eller hydrogen, i et tidsrum, som er tilstrækkeligt til at tillade aktiveringen. Reaktionen bør forløbe i et tidsrum, der er tilstrækkeligt til at tillade fuldstændig aktivering, hvilket tidsrum er forholdsvis kort. Et tidsrum i intervallet fra 10 til 40 minutter er sædvanligvis tilstrækkeligt til, at reaktionen forløber til ende.
Det i det foreliggende anvendte cyanhalogenid er typisk cyanbro-mid, men andre cyanhalogenider, såsom cyaniodid eller cyanchlorid, kan anvendes. Den almindeligt anvendte mængde cyanhalogenid synes ikke at være særligt kritisk, idet der fortrinsvis anvendes fra 0,5 til 1,5 vægtdele cyanhalogenid pr. vægtdel carbonhydrat, som er til stede i reakt ionsblandingen.

Den anvendte alkohol er absolut, dvs. i alt væsentligt fri for vand som urenhed. Nærværelsen af vand synes at fremme uønskede side-reaktioner, som forårsager uopløseliggørelse af det vandopløselige carbonhydrat og en formindskelse af udbyttet eller reaktionsproduktets aktivitet. Der anvendes alkoholer med 1-3 carbonatomer og 1 hydroxy1-gruppe eller n-butanol, da opløsningen af bestanddelene i glycoler eller højere alkoholer ofte er vanskelig, og uønskede reaktioner har vist sig at finde sted med forgrenede alkoholer, såsom isobutanol, således at alkoholerne med de laveste molekylvægte, såsom methanol, foretrækkes. Det foretrækkes også i almindelighed i reaktionsblandingen at anvende fra ca. 20 til ca. 80 ml alkohol pr. g tilstedeværende carbonhydrat.
Alkaliet med formlen MOR kan være et alkalimetalalkoxid eller -hydroxid, såsom natriummethoxid, kaliumethoxid, kaliumbutoxid, lithium-hydroxid eller natriumhydroxid. Tilsvarende rubidium- og cesiumfotbin-delser kan også anvendes. Det foretrækkes i almindelighed at anvende alkoxidforbindelser (dvs. hvor R betegner lavere alkyl), da alkalime-talhydroxider har en stærk tendens til at bibeholde absorberet vand, og således kan give tendens til fremmelse af sidereaktioner, med mindre de er tørret omhyggeligt før anvendelse. Alkalimetalalkoxider kan ligeledes have bedre opløselighed i de ifølge opfindelsen anvendte alkoholer. Alkalimaterialet med formlen MOR sættes til reaktionsblandingen i en sådan koncentration, at den, hvis reaktionsblandingen var deioniseret vand, ville være tilstrækkelig til at bibringe blandingen en pH-værdi på mindst 10. Det er i ikke-vandige systemer naturligvis generelt ukorrekt at regne med en pH-værdi som sådan, men det tilstedeværende alkaliske materiale tilvejebringer ikke-vandige alkaliske betingelser.
Det er især ønskværdigt at tilvejebringe en koncentration af alkali, der er tilstrækkelig til give deioniseret vand en pH-værdi på 11 eller derover. Det kan ligeledes være nødvendigt at tilsætte alkali under de første få minutter af reaktionen, hvis der tilsættes små mængder alkali til at begynde med, da alkali tilsyneladende forbruges under reaktionen.
En beskrivelse af den tilsyneladende mekanisme ved cyanbromids aktivering af carbonhydrat i et vandigt system kan findes i "A Study of the Mechanism of Cyanogen Bromide Activation of Cellulose", pp. 1039--1044, Biotechnology and Bioengineering, bind XIV (1972) .
Det er på nuværende tidspunkt ukendt, hvorvidt reaktionsmekanismen for det ikke-vandige system ifølge opfindelsen svarer til eller ad 4 141292 skiller sig fra den i den umiddelbart ovenfor omtalte publikation omhandlede mekanisme.
De ifølge opfindelsen anvendte carbonhydrater kan være dextran, opløselig stivelse, opløselig glycogen eller lignende, og bør i almindelighed have en molekylvægt på mindst ca. 5000. Det mest foretrukne er at anvende materialer, såsom dextran med en molekylvægt på fra ca. 40.000 til ca. 100.000, især "Dextran 70" (molekylvægt 70.000), der er et ikke-toxisk middel, som allerede anvendes til forskellige former for terapi. Imidlertid kan opløselige dextraner med molekylvægte på 500.000 og derover anvendes.
En typisk foretrukken reaktionsblanding består af fra 1 til 3 vægtdele natriummethoxid, 1 til 3 vægtdele dextran og 1 til 3 vægtdele cyan-bromid samt 100 vægtdele methanol.
Reaktionen ved fremgangsmåden ifølge opfindelsen forløber ved stuetemperatur, men det foretrækkes at opretholde en sænket reaktions-temperatur, typisk fra -20 til +10°C, da en sådan sænket reaktionstemperatur giver bedre resultater.
Efter at reaktionen er forløbet til ende, kan det resulterende aktiverede carbonhydrat, som under de foretrukne reaktionsbetingelser er uopløseligt i det ikke—vandige opløsningsmiddel, udvindes ved simpel filtrering, efterfulgt af udvaskning med et ikke-vandigt opløsningsmiddel og vakuumtørring til fjernelse af opløsningsmidlet.
Det tørrede, aktiverede carbonhydrat, fra ca. 1 til ca. 300 vægtdele, kan derpå opløses i en passende vandig puffer, såsom en 0,1M natriumhydrogencarbonatopløsning, til opretholdelse af en pH-værdi, der i almindelighed ligger på imellem ca. 5 og ca. 9, og blandes med en opløsning eller dispersion af ca. 1 vægtdel enzym eller et andet protein ved formindsket temperatur, typisk fra 0°C til ca. 5°C, i et antal timer eller dage. Reaktionsproduktet mellem carbonhydrat og protein kan isoleres fra fri protein ved kolonnekromatografi eller på anden måde, såsom ved selektiv udfældning under anvendelse af blandinger af alkohol og vand.
Typisk omsættes fra ca. 10 til ca. 80 vægtdele aktiveret carbonhydrat, og fortrinsvis fra ca. 15 til ca. 40 vægtdele carbonhydrat med 1 vægtdele protein.
Specielt opnås ved omsætning af det aktiverede carbonhydrat med et enzym ved fremgangsmåden ifølge opfindelsen de bedste resultater, når der anvendes fra 10 til 80 vægtdele af det aktiverede carbonhydrat for hver vægtdel enzym.
Enestående og medicinsk interessante materialer er fremstillet 5 141292 ved at omsætte carbonhydrat med streptokinase ved fremgangsmåden ifølge opfindelsen. Sådant opløseligt omsat materiale, der har været afprøvet på dyr, udviser store terapeutiske fortrin i forhold til det frie enzym. Det har vist sig, at der forekommer formindsket antistofdannelse ved indgivelse af den matricebundne streptokinase fremstillet i overensstemmelse med opfindelsen, selv i sammenligning med indgivelsen af større doser fri streptokinase.
Den omhandlede fremgangsmåde kan anvendes til binding af mange forskellige store molekyler, som typisk har en molekylvægt på over 1000. Typiske proteiner, som kan anvendes til reaktionen med de aktiverede carbonhydrater ved fremgangsmåden ifølge opfindelsen, er i almindelighed enzymer, skønt antistoffer, hormoner og lignende også kam bindes. Ligeledes kan proteiner, som indeholder visse ikke-proteinag- tige bestanddele, såsom metalioner, polysaccharider, porphyringrupper, adenindinucleotid, ribonukleinsyre, phospholipider og lignende, bindes i overensstemmelse med den foreliggende opfindelse. Sådanne materialer betragtes i det foreliggende som faldende ind under definitionen "protein" , da de indeholder en større del polypeptid- eller proteinsubsti-tuenter, og da de kemisk kan bindes til carbonhydratstøttemedier af den ovenfor beskrevne type. Proteinpolypeptidfragmenter, såsom aktive dele af enzymmolekyler, kan også bindes i overensstemmelse med den foreliggende opfindelse.
Eksempler på proteinholdige materialer, som ifølge opfindelsen kemisk kan bindes, er enzymer, såsom phosphorpyrudruesyretransphos-phorylase, transmethylase, aminoxidase, glutaainsyre-oxaleddikesyre-transaminase, glutaminsyre-pyrudruesyretransarainase, lipoxidase, peroxidase, catalase, glucoseoxidase, urease, glutaminase, urokinase, arginiase, phosphatase, lacithinase, cholinesterase, lipase, histidase, pectinase, maltase, a-amylase, amyloglucosidase, keratinase, bromelin, chymotrypsin, trypsin, pepsin, ficin, papain og chymopapain. Andre egnede enzymer omfatter de rennin-lignende proteaser, der fremstilles af organismen Mucor miehei og Mucor pusillus, hvilke proteaser er nyttige ved fremstillingen af ost, alkaliske proteaseenzymer, såsom de af organismen Bacillus subtilis varietet licheniformis fremstillede, amylaseenzymer, såsom de af Bacillus amyloliquefaciens fremstillede, og enzymerne L-asparaginase eller serindehydratase, der kan anvendes mod visse typer tumorceller ved at berøve dan asparagin eller serin, en essentiel aminosyre for sådanne tumorceller.
Andre proteinmaterialer, som kan bindes til de aktiverede carbonhydrater ved fremgangsmåden ifølge opfindelsen, omfatter hormoner, såsom insulin og de forskellige hypofysehormoner, proteiner fra gammaglobulinfraktionen, f.eks. antistoffer af klasserne G, M, A, D
U1292 6 og E, og andre blodfaktorer, såsom den antihæmofiliske faktor, blodkoagulationsfaktorerne, specifikke antistoffer, såsom hepatitis-, influenza-, poliomyelitis-, mæslinge-, fåresyge- og rabiesantistoffer, passende antigener, såsom støv, pollen fra planter af slægten ambrosia, eller mugantigen, eller hepatitis-, poliomyelitis-, mæslinge-, fåresyre-, influenza- eller rabiesantigen til rensning eller til eksperimentel undertrykkelse af antistofreaktioner, samt almindelige legemsproteiner, såsom hæmoglobin eller albumin.
Opfindelsen belyses i det følgende ved eksempler.
Eksempel 1 1 g natriummethoxid sattes til 50 ml absolut methanol. Til denne opløsning sattes 1 g cyanbromid efterfulgt af 1 g "Dextran 70" (molekylvægt 70.000) , som ikke opløstes i alkoholblandingen. Reaktionsblandingen omrørtes i 20 minutter ved stuetemperatur. Det resulterende faste produkt udvandtes ved simpel filtrering efterfulgt af udvaskning med absolut methanol og lufttørring.
. 500 mg af det resulterende aktiverede produkt opløstes i 5 ml af en 0,1M natriumhydrogencarbonatpufferopløsning. Samtidig opløstes 19,6 mg streptokinase i 3 ml af en 0,1M natriumhydrogencarbonatopløsning.
Begge opløsningsprøver dialyseredes ved 4°C i 2-3 timer imod en 0,1M natriumhydrogencarbonatopløsning.
De to dialyserede opløsninger forenedes, og 2 ml af en 0,1M natri-umhydrogencarbonatopløsning tilsattes for at bringe det samlede volumen op på 10 ml. Reaktionsblandingen omrørtes ved stuetemperatur i 1 time og fik derpå lov til at henstå i 41 timer ved 4°C.
Det opløselige bundne produkt isoleredes ved kolonnekromatografi. Porøse glasmikroperler ("Corning CPG beads" solgt af Corning Glass Company, Corning, New York) kan anvendes som et passende separationsmedium i kolonnen. Fraktionerne indeholdende dextran-streptokinase lyofiliseredes til dannelse af et fnugagtigt faststof med en enzymaktivitet på 748 streptokinaseaktivitetsenheder pr. mg.
Eksempel 2
Til 50 ml absolut methanol sattes 4 ml af en 25% opløsning af na-triummethoxid i methanol. Bægeret indeholdende denne blanding pakkedes i tøris til afkøling deraf på vandfri måde, og derefter tilsattes 1 g cyanbromid efterfulgt af 1 g "Dextran 70" (molekylvægt 70.000). Reaktionsblandingen omrørtes i 20 minutter ved den af tørisen bibragte formindskede temperatur på fra ca. 0° til ca. -10°C. Produktet, et 7 141292 hvidt faststof, udvandtes ved filtrering efterfulgt af udvaskning med methanol og tørring i 1 time i vakuum i en beholder indeholdende et tørringsmiddel (calciumchlorid) adskilt fra den aktiverede dextranprøve.
Den ovenstående reaktion gentoges efter behov adskillige gange til tilvejebringelse af tilstrækkeligt aktiveret dextran til de nedenstående bindingsreaktioner.
Portioner af det ovenfor fremstillede aktiverede dextran og prøver af streptokinaseenzym sattes til særskilte portioner af vandig natriumhydrogencarbonatpuffer og forenedes derefter under omrøring ved en temperatur på ca. 4°C til tilvejebringelse af en reaktionsprøve med et opløsningsvolumen som angivet i tabellen nedenfor og koncentrationer af dextran og streptokinase ligeledes som angivet nedenfor. Vægtene af nærværende streptokinase beregnedes ud fra en streptokinaseaktivitet på 80.000 streptokinaseenheder pr. mg tilsat enzym. Reaktionsblandingerne holdtes ved 4°C i det nedenfor angivne antal timer.
Efter reaktionens afslutning isoleredes dextran-streptokinasen ved kolonnekromatografi igennem en glasperlekolonne, og fraktionerne indeholdende det bundne enzym lyofiliseredes til dannelse af et fast" produkt med det angivne antal streptokinaseenheder pr. mg. Det maksimale teoretiske udbytte af streptokinaseenheder pr. mg er også angivet, ligesom den ved analyse af nogle produkter fundne nitrogenprocent.

Det kan ses, at en procentvis meget høj del af den i denne reaktionsblanding anvendte streptokinase bliver kemisk bundet til dextra-nen uden tab af enzymaktivitet. Med hensyn til forsøget med referencenummer 2200-13 viser det sig faktisk, at den specifikke aktivitet for det matricebundne produkt udtrykt i enheder streptokinaseaktivitet pr. mcg kemisk bundet nitrogen er højere end for rent streptokinase.
Eksempel 3
Ca. 7,8 mg (1560 internationale enheder) L-asparaginase dialyseredes i og imod en 0,1M natriumphosphatpuffer ved en pH-værdi på ca.
7,3. Derefter fortyndedes asparaginaseopløsningen til 25 ml i samme puffer.
1 g af den aktiverede dextran fra eksempel 1 sattes til denne opløsning under omrøring og fik lov at reagere ved 35°C i 41 timer. Det resulterende produkt fraktioneredes ved hjælp af en glasperlekolonne, og de fraktioner, som gav kemisk bundet dextran-asparaginase, lyofili-seredes til udvinding af 1,19 g af produktet med en enzymaktivitet på 0,717 internationale asparaginaseaktivetsenheder pr. mg - et udbytte på ca. 55% bundet enzym.
