Det skal forstâs, at udtrykket halogen som anvendt i det fore-liggende omfatter fluor, brom, iod og fortrinsvis chlor, mens udtrykkene "Cj^alkyl" og "C^^alkoxy" refererer til bâde ligekædede og forgrenede carbonkæder med fra 1 til 4 carbonatomer inklusive. Eksempler pâ disse 15 grupper er carbonkæder, som kan være methyl, ethyl, propyl, isopropyl, 1-butyl, 1-methylpropyl og 2-methylpropyl.
De farmaceutisk acceptable syreadditionssalte af de omhandlede forbindelser er sâdanne, hvori anionen ikke væsentligt indvirker pà toxiciteten eller den farmakologiske virkning af saltet og som sâdanne 20 er de farmakologisk ækvivalente med baserne med formel (I). De er i almindelighed foretrukne til medicinsk brug. I nogle tilfælde har de fysiske egenskaber, som gpr dem mere pnskelige til farmaceutisk formu-lering, sâsom opliselighed, mange! pâ hygroskopicitet, kompressibilitet med hensyn til tabletdannelse og forenelighed med andre bestanddele, 25 hvormed stoffet kan anvendes til farmaceutiske formâl. Saltene frem-stilles rutinemæssigt ved blanding af basen med formel (I) med den udvalgte syre, fortrinsvis ved kontakt i oplpsning under anvendelse af et overskud af sædvanligt anvendte inerte oplpsningsmidler, sâsom vand, ether, benzen, éthanol, ethylacetat og fortrinsvis acetonitril. De kan 30 ogsâ fremstilles ved methatese eller behandling med en ionbytterharpiks under betingelser, hvor anionen af ét sait af en forbindelse med formel (I) erstattes af en anden anion under betingelser, som tillader adskillelse af de pnskede stoffer, sâsom ved fældning fra opl0sning eller ekstraktion i et opl0sningsmiddel eller eluering fra eller 35 tilbageholdelse pâ en ionbytterharpiks. (II) - (III) I ovenstâende formler (II og III) har symbolet "Z" samme betydning som i formel (I) og "X" betegner syreresten af en reaktiv estergruppe, 15 sâsom chlorid, bromid, iodid, fluorid, sulfat, phosphat, tosylat eller mesylat.
Fremgangsmâden udfpres konventionelt under reaktionsbetingelser, som anvendes til fremstilling af tertiære aminer ved alkylering af sekundære aminer. Sâledes opnâs forbindelserne med formel (I) ved 20 omsætning af en 3-(4-X-butyl)spiro-2,4-thiazo1idindion med formel (II) i et inert reaktionsmedium ved temperaturer fra ca. 50° til ca. 200°C med en "Z-piperazin" med formel (III) i nærværelse af en base, der er egnet til anvendelse som et syrebindende middel. Anvendelige uorganiske og organiske syrebindende baser omfatter tertiære aminer, alkali- og 25 jordalkalimetalcarbonater, -bicarbonater eller -hydrider med natrium-carbonat og kaliumcarbonat som særligt foretrukne. Som anvendt i det foreliggende betyder udtrykket "inert reaktionsmedium" ethvert prot eller aprot oplpsningsmiddel eller fortyndingsmiddel, der ikke indgâr i reaktion i nogen væsentlig grad. I denne henseende er acetonitril et 30 særligt foretrukkent oplpsningsmiddel med reaktinen hensigtsmæssigt udfprt ved tilbagesvalingstemperatur. Tilfredsstillende udbytter af de omhandlede forbindelser opnâs med reaktionsperioder varierende fra ca.
2-24 timer. Forbindelserne med formel (I) kan renses ved krystalli-sationsteknik fra standard oplpsningsmedier, sâsom acetonitril, 35 isopropanol, éthanol og lignende og ved hjælp af andre konventionelle metoder, sâsom chromatografering under anvendelse af en silikagel-kolonne med blandinger af chloroform og alkanoler, sâsom methanol og éthanol, som elueringsmiddel.

Med hensyn til bivirkninger fremkalder denne forbindelse ikke signifikant katalepsi i rotten, 5 hvilket indikerer en relativ mangel pâ ekstrapyramidale reaktioner og yderligere reverserer den tidligere etableret phenothiazin-induceret katalepsi i rotten. Den sidstnævnte virkning indikerer, at 2-[4-[4-(7,9-dioxo-6-thia-8-azaspiro[4.4]nonan-8-yl)butyl]-l-piperazinyl]pyridin-3-carbonitril er et effektivt middel med hensyn til behandling af ekstra-10 pyramidale symptomer og at den er potentielt nyttig til behandling af tardiv dyskinesia.
Som tidligere nævnt har de omhandlede forbindelser psychotrope egenskaber, der især er egnede til deres anvendelse som neuroleptiske eller anxiolytiske midler. De omhandlede forbindelser kan derfor 15 anvendes til forbedring af en psychotisk eller nervos tilstand i et pattedyr med behov for sâdan behandling, ved hvilken man systemisk til dyret administrerer en effektiv dosis pâ fra ca. 0,01 til 40 mg/kg legemsvægt af en forbindelse med formel (I) eller et farmaceutisk acceptabelt syreadditionssalt deraf.
20 Udtrykket systemisk administrering som her anvendt refererer til orale, rektale og parenterale (dvs. intramuskulære, intravenpse og subkutane) veje. I almindelighed vil det vise sig, at nâr en af de omhandlede forbindelser administreres oralt, som er den foretrukne vej, kræves en stprre maengde af den aktive bestanddel til at fremkalde den 25 samme virkning som en mindre mængde indgivet parentéralt. I overens-stemmelse med god klinisk praksis foretrækkes det at administrere de omhandlede forbindelser ved et koncentrationsniveau, som vil fremkalde effektive neuroleptiske (antipsychote) eller anxiolytiske virkninger uden at bevirke nogen skadelige eller udnskede bivirkninger.
30 Terapeutisk indgives de omhandlede forbindelser i almindelighed som farmaceutiske præparater omfattende en effektiv antipsychotisk eller anxiolytisk mængde af en forbindelse med formel (I) eller et farmaceutisk acceptabelt syreadditionssalt deraf og en farmaceutisk acceptabel bærer. Farmaceutiske præparater, som giver fra ca. 1 til 500 35 mg af den aktive bestanddel pr. enhedsdosis, foretrækkes og fremstilles konventionelt som tabletter, halspastiller, kapsler, pulvere, vandige eller olieagtige suspensioner, sirupper, eliksirer og vandige oplpsninger.

Foretrukne orale præparater er i form af tabletter eller kapsler, og de kan indeholde konventionelle excipienter, sâsom bindemidler (f.eks. sirup, acacia, gélatine, sorbitol, tragacanth eller polyvinyl-pyrrolidon), fyldstoffer (f.eks. lactose, sukker, majsstivelse, 5 calciumphosphat, sorbitol eller glycin), smdremidler (f.eks. magne-siumstearat, talkum, polyethylenglycol eller siliciumoxid), disintegre-ringsmidler (f.eks. stivelse) og befugtningsmidler (f.eks. natrium-laurylsulfat). Sâdanne præparater med den pnskede klarhed, stabilitet og egnethed til parentéral brug opnâs ved oplpsning af fra 0,1 til 10 vægt% af den aktive bestanddel i vand eller en vehikel bestâende af en polyvalent 15 alifatisk al kohol, sâsom glycerin, propylenglykol og polyethylenglykoler eller blandinger deraf. Polyethylenglykolerne bestâr af en blanding af ikke-flygtige, normalt flydende polyethylenglykoler, der er oplpselige i bâde vand og organiske væsker, som har molekylvægte fra ca. 200 til 1500.

En blanding af 3-(4-brombutyl)-5-spirocyclopentyl-2,4-thiazoli-dindion (1,62 g, 0,005 mol), l-(3-chlorphenyl)piperazin (1,04 g, 10 0,005 mol), kaliumcarbonat (0,8 g, 0,006 mol) og kaliumiodid (0,09 g, 0,0006 mol) i 50 ml acetonitril opvarmes under tilbagesvaling i et tidsrum af 16 timer. Reaktionsblandingen afkdles, filtreres og filtratet koncentreres i vakuum. Residualt materiale opldses i chloroform, filtreres og filtratet koncentreres til opnâelse af 2,35 g af produktet 15 som fri base, der, omdannet til hydrochloridsaltet i acetonitril, giver 1,29 g (46%) 8-[4-[4-(3-chlorphenyl)-l-piperazinyl]butyl]-6-thia-8-azaspiro[4.4]-7,9-dion, dihydrochlorid, smeltepunkt 166,5-171°C.

Omsætnî ng af 3-(4-brombutyl)-5-spi rocyclopentyl-2,4-thi azolidindion (4,96 g, 0,016 mol) med l-(3-cyano-2-pyridinyl)piperazin (3,04 g, 0,016 5 mol), kaliumcarbonat (2,46 g, 0,018 mol) og kaliumiodid (0,3 g, 0,0018 mol) i 125 ml acetonitril i overensstemmelse med fremgangsmâden i eksempel 1(c) giver det fri base produkt, der omdannes til hydrochlorid-saltet i ethylacetat med ethanolisk saltsyre. Krystallisation af hydrochloridsaltet fra acetonitril giver et 42% udbytte af 2-[4-[4-(7,9-10 dioxo-6-thia-8-azaspiro[4.4]nonan-8-yl)butyl]-l-piperazinyl]pyridin-3-carbonitril, hydrochlorid, smeltepunkt 207-208°C.
Analyse beregnet for Cgjf^NgOgS.HCl (procent): C, 56,05; H, 6,27; N, 15,56. Fundet (procent): C, 55,92; H, 6,24; N, 15,51.

Claims (5)
1. Analogifremgangsmâde til fremstilling af spirothiazolidinyl-piperazinderivater den almene formel (I) CR / V n-(ch2),.-h j-z ®—( N ' 10 0 (I) hvori Z enten betegner en Rj-substitueret phenylgruppe med formlen 15 /S, l+h 20 hvori Rj betegner hydrogen, halogen, Cj^alkyl, Cj^alkoxy eller trifluormethyl, eller en Rg-substitueret 2-pyridinylgruppe med formlen » -O- hvori R2 betegner hydrogen eller cyano, eller et farmaceutisk ac-30 ceptabelt, non-toxisk syreadditionssalt deraf, KENDETEGNET ved, AT man omsætter en spiro-2,4-thiazolidindion med den almene formel (II) £K 35 n-(CH2)4-x (II); X~i O DK 156219B hvori X er syreresten af en reaktiv estergruppe, med en piperazin med den almene formel (III) 5 (III) hvori Z har den ovenfor anfprte betydning, i et for reak-10 tionen inert medium med eller uden en base ved en temperatur fra ca. 50°C til ca. 200°C i et tidsrum, som er tilstrækkeligt til at give den resulterende forbindelse med formel (I) og/eller om pnsket om-danner den resulterende forbindelse med formel (I) til et sait eller et resulterende sait omdannes til den frie forbindelse eller til et an-15 det sait.

Fremgangsmâde ifplge krav 1, KENDETEGNET ved, AT man ud fra 3-(4-brombutyl)-5-spirocyclopentyl-2,4-thiazolidindion og l-(3-35 chlorphenyl)piperazin fremstiller 8-[4-[4-(3-chlorphenyl)-l-piperazinyl]butyl]-6-thia-8-azaspiro[4.4]nonan-7,9-dion eller et farmaceutisk acceptabelt syreadditionssalt deraf.
