Diesteren og aspartylesteren skal fjernes ved rensningstrin til o.pnåelse af det ønskede produkt.
Tidligere enzymatiske esterificeringsmetoder har været rettet mod esterificeringen af enkelte aminosyrer. De har ikke 5 angået de selektive esterificeringsproblemer, som forekommer ved dipeptider, især α-L-aspartyl-L-phenylalanin, som har mere end en carboxylgruppe.
F.eks. har Ingalls, et al., BIOTECHNOLOGY AND BIOENGINEERING 17, 1627-1637 (1975), beskrevet esterificeringen 10 af en enkelt aminosyre, N-acetyltyrosin, under anvendelse af ubevægeliggjort chymotrypsin eller subtilisin Carlsberg i den frie, umodificerede form i et opløsningsmiddelsystem af vand, ethanol og glycerol. En ulempe ved denne metode var, at der dannedes væsentlige mængder af glycerolesteren af aminosy-15 ren.
Klibanov, et al., BIOTECHNOLOGY AND BIOENGINEERING 19, 1351-1361 (1977) , beskriver esterificeringen af en enkelt aminosyre, N-acetyltryptophan, under anvendelse af ubevægeliggjort chymotrypsin i et tofasesystem af vand og et med vand 20 ikke-blandbart opløsningsmiddel, chloroform.
Det .har nu overraskende vist sig, at carboxylgruppen i phenylalanindelen i α-L-aspartyl-L-phenylalanin kan al-‘ kyleres selektivt ved enzymatisk esterificering i et enfa-sesystem til opnåelse af a-L-aspartyl-L-phenylalanin-al-25 kylester uden at frembringe den uønskelige diester og aspartylester. Dette opnås ifølge opfindelsen ved en fremgangsmåde, som er ejendommelig ved, at eventuelt N-beskyttet α-L-aspartyl-L-phenylalanin bringes i kontakt med en alkohol i nærværelse af en effektiv mængde af en alkalisk 30 serinproteinase, en esterase, som foretrækker aromatiske aminosyrer, i et vandigt alkoholmedium ved en pH-værdi på 4-7, hvori alkoholkoncentrationen er mindst 10%, hvorpå en eventuelt tilstedeværende N-beskyttelsesgruppe fjernes.
En alkalisk serinproteinase er et proteolytisk 33 enzym, som har specifik esteraseaktivitet i et vand-al-kohol-opløsningsmiddelsystem, hvori alkoholkoncentrationen er tilstrækkelig til at reversere enzymets hydrolytiske 3

virkning. Specifik esteraseaktivitet refererer til det pro-teolytiske enzyms evne til selektiv alkylering af carboxyl-gruppen i phenylalanindelen. De således fremstillede a-L-aspartyl-L-phenylalanin-alkylestere er værdifulde som 5 sødemidler. Denne anvendelighed er beskrevet i US-patent-skrift nr. 3 492 131.
Foretrukne alkoholer er repræsenteret af formlen R-OH
10 hvor R betyder alkyl med 1-7 carbonatomer. Disse alkylgrup-per eksemplificeres af methyl, ethyl, propyl, butyl, pentyl, hexyl,, heptyl og isomerene deraf med forgrenet kæde.
Aminogruppen i a-L-aspartyl-L-phenylalanin kan beskyttes af de almindeligvis anvendte beskyttelsesgrupper.

Aminogrupperne kan også beskyttes ved dannelse af enami-ner, opnået ved omsætning af aminogruppen med 1,3-diketoner, f.eks. benzoylacetone eller acetylacetone.
Beskyttelsesgrupper fjernes bekvemt ved sådanne reaktio-5 ner som hydrogenolyse (f.eks. i nærværelse af en palladium-sortkatalysator), behandling med en hydrogenhalogenidsyre (såsom hydrogenbromidsyre, hydrogenfluoridsyre eller saltsyre) i eddikesyre eller behandling med trifluoreddikesyre.
Den omsætning, ved hvilken esterbindingen dannes, kan 10 gennemføres i en vandig puffer-alkohol-opløsning med en pH-værdi på 4-7. Typiske pufferopløsninger omfatter natriumpyrophos-phatpufferopløsning, citronsyrepufferopløsning, eddikesyrepuf feropløsning eller tris-HCl-pufferopløsning.
Den esterase, som anvendes ifølge opfindelsen, er en 15 alkalisk serinproteinase. Disse enzymer udviser høj esterase-aktivitet og virker selektivt på aromatiske aminosyrer.
Typiske alkaliske serinproteinaser omfatter subtilisin og alkaliske proteinaser fra forskellige stammer af Bacillus, Aspergillus, Streptomyces, Penicillium og Arthrobacter. En 20 foretrukket alkalisk serinproteinase er subtilisin Carlsberg, som er kommerciel tilgængelig. En katalytisk mængde enzym anvende*s ved reaktionen, fortrinsvis 10-500 mg pr. 1 mmol α-L-aspartyl-L-phenylalanin eller N-beskyttet a-L-aspartyl--L-phenylalanin. Et calciumsalt kan også tilsættes for at 25 fremme enzymaktiviteten.
Enzymet kan være til stede i fri tilstand eller kan være ubevægeliggjort ved binding til en egnet bærer, såsom porøst glas, polyacrylamidgel, carboxymethylcellulose eller aminoethylcellulose. "IMMOBILIZED ENZYMES", bind 44, METHODS 30 IN ENZYMOLOGY, udgiver Klaus Mosback, diskuterer forskellige metoder til at gøre enzymer ubevægelige.

Reaktionen gennemføres i et medium af vand og alkohol, hvori alkoholkoncentrationen er tilstrækkelig til at reversere enzymets esteraseaktivitet. Alkoholkoncentrationen kan være 10-90% efter rumfang. En foretrukket alkoholkoncentra-5 tion er 30-70% efter rumfang. En især foretrukket alkoholkoncentration er 50-60% med 60% alkohol efter rumfang som det optimale.
Ved en især foretrukket udførelsesform for opfindelsen er den alkaliske serinproteinase subtilisin Carlsberg, en 10 proteinase med specifik esteraseaktivitet, og alkoholen er methanol, som er til stede i en koncentration på 60% efter rumfang. Vand-alkohol-blandingen indeholder 5,0 mmol calcium-chlorid og har en pH-værdi på 5,(3, og reaktionen gennemføres ved en temperatur på 25°C.
15 Omsætningen skrider jævnt fremad under disse betingelser, indtil den er afsluttet. En foretrukket reaktionstid er 1-260 timer. En især foretrukket reaktionstid er 1-90 timer. Reaktionsproduktet skilles hensigtsmæssigt fra reaktionssystemet ved standardchromatografiteknik eller andre metoder, som er 20 kendte.
Opfindelsen er yderligere Illustreret i de eksempler, som følger. I eksemplerne er mængderne af materialer angivet i vægtdele, medmindre andet er angivet. Forholdet mellem vægtdele og rumfangsdele er det samme som mellem 25 gram og milliliter.
Eksempel 1
Subtilisin Carlsberg (Protease Type VIII, Sigma Chemical Co.) opløses i destilleret vand i en koncentration på 40 mg/ml 30 0g dialyseres imod destilleret vand ved 5°C. 0,1 rumfangsdele af enzymopløsningen blandes med 0,9 rumfangsdele af en vandig methanolopløsning indeholdende 0,0056 vægtdele a-L-aspartyl--L-phenylalanin. Den vandige methanolopløsning fremstilles ved blanding af 0,6 rumfangsdele methanol med 0,075 rumfangs-35 dele 0,33 M natriumacetat, 0,225 rumfangsdele 0,33 M eddikesyre og 0,002 rumfangsdele 2,5 M calciumchlorid. i 1 minut til dispergering af a-L-aspartyl-L-phenylalaninet.
Den samlede pH-værdi indstilles på en pH-værdi på 5,0 med fortyndet saltsyre. Den færdige reaktionsblanding indeholdende 4 mg/ml enzym, 5,0 mmol calciumchlorid, 0,1 M eddikesyrepuf-5 fer, 60% methanol og 0,02 M a-L-aspartyl-L-phenylalanin får lov at reagere ved ca. 25°C under kontinuerlig omrøring i et tidsrum på 4 døgn. Reaktionsblandingen ledes gennem en kolonne af "Sephadex LH-20" for at skille enzymet fra dipep-tidet. Fremgangsmåden ifølge eksempel 1 gentages, bortset fra 25 at der som katalysator anvendes ubevægeliggjort enzym. Enzymet er gjort kovalent ubevægeligt til DEAE-cellulose, som er aktiveret med cyanogenbromid. DEAE-cellulosen (præ-kvældet DE52, Whatman) aktiveres ved, at 0,075 vægtdele vaskes med 0,1 M natriumbicarbonat og derefter omsættes med 3 rumfangs-30 dele cyanogenbromidopløsning (25 mg/ml). pH-Værdien indstilles og holdes på 11,0 med 2 M natriumhydroxid, og der omrøres svagt i 6 minutter ved 25°C. pylenrør (under argon) ved 5°C i 17 timer. Derefter vaskes det bundne enzym med destilleret vand og omsættes med 5 rumfangsdele 1 M ethanolamin (pH-værdi 9,0) under svag omrøring i 2 timer ved 25°C. Gelen vaskes to gange med destilleret vand 5 og suspenderes igen i 0,3 rumfangsdele destilleret vand.
Denne suspension af ubevægeliggjort enzym blandes med 0,7 rumfangsdele af en vand-methanol-blanding, således at den færdige reaktionsblanding indeholder samme koncentration af materialer som i eksempel 1. Reaktionen gennemføres som i eksempel 1, 10 bortset fra at det ubevægeliggjorte enzym fjernes ved centrifugering forud for rensningen af produktet.

Anvendelse af 0,0125 vægtdele N-carbobenzoxy-a-L-aspar-30 tyl-L-phenylalanin i stedet for a-L-aspartyl-L-phenylalaninet i eksempel 1 og 2 og i det væsentlige gentagelse af de deri detaljeret beskrevne metoder giver N-carbobenzoxy-a-L-aspar-tyl-L-phenylalanin-methylester. Reduktion af denne forbindelse ved den af M. Bergmann og L. Zervas, Ber. 65, 1192 (1932), 35 beskrevne metode fjerner den N-beskyttende carbobenzoxygruppe til dannelse af α-L-aspartyl-L-phenylalanin-methylester.
Claims (3)
1. Fremgangsmåde til esterificering af a-L-aspartyl--L-phenylalanin til de tilsvarende alkylestere esterifi-ceret i L-phenylalaningruppen, kendetegnet ved, 5 at eventuelt N-beskyttet a-L-aspartyl-L-phenylalanin bringes i kontakt med en alkohol i nærværelse af en effektiv mængde af en alkalisk serinproteinase i et vandigt alkoholmedium ved en pH-værdi på 4-7, hvori alkoholkoncentrationen er mindst 10%, hvorpå en eventuelt tilste-10 deværende N-beskyttelsesgruppe fjernes,
2. Fremgangsmåde ifølge krav 1, kendetegnet ved, at den alkaliske serinproteinase er subtilisin Carlsberg.
