Opfindelsen angår et cremelignende præparat indeholdende β-methasondipropionat.
En β-methasondipropionatcreme har været solgt af Schering Corporation. Produktet består af β-methason-5 dipropionat i et cremegrundlag bestående af renset vand, mineralolie, hvid vaseline , polyethylenglycol 1000-monocetylether, cetostearylalkohol, monobasisk natriumphosphat og phosphorsyre med 4-chlor-m-cresol og propylenglycol som konserveringsmidler.
10 Præparatet ifølge den foreliggende opfindelse er bemærkelsesværdigt simpelt i forhold til denne kendte creme, idet det indeholder et glycolopløsningsmiddel som hovedkomponent, medens såvel andre konserveringsmidler som overfladeaktive stoffer kan udelades.
15 Der er således mindre sandsynlighed for, at det forårsager allergiske reaktioner på grund af disse additiver. Ikke desto mindre er præparatet ifølge opfindelsen meget effektivt og stabilt. Faktisk er det overraskende mere effektivt end den kendte creme. Desuden 20 har det foreliggende middel, selv om der ikke er standardkonserveringsmidler til stede, vist sig at være selvkonserverénde, når det testes ved standardiserede forsøg til bestemmelse af væksten af Eschericia coli,
Candida albicans, Staphylococcus aureus, Aspergillus 25 niger og Pseudomonas aeruginosa.
US patentskrift nr. 4.008.321 omtaler en topisk sammensætning, der indeholder et steroid, crotamiton, propylenglycol og carbcmer 940.
Som steroid i sammensætningen er β-methason kun 30 foreslået anvendt uden denne sammensætning faktisk er fremstillet. Det særegne i nævnte US patentskrift er anvendelsen af crotamiton som opløsningsmiddel for steroidet, crotamiton anvendes ikke i nærværende opfindelse.
US patentskriftet adskiller sig også fra den foreliggende 35 opfindelse derved, at alle præparateksemplerne er indstillet til pH 6,5, mens pH i den foreliggende opfindelse er indstillet på en værdi mellem 4,0 og 5,5. Hidtil har det været anset for uhensigtsmæssigt at fremstille et

2 creme-præparat med så lav et pH, fordi det formodedes, at et sådant præparat ville forårsage hudirritation. Men præparatet ifølge opfindelsen har ved standardundersøgelser af dermal irritation hos kaniner vist sig at være 5 mindre irriterende end den tidligere β-methasondipropio-natcreme solgt af Schering Corporation.
Præparatet ifølge opfindelsen er nyttigt ved topisk behandling af dermatologiske lidelser, som kan påvirkes ved hjælp af corticosteroider. Lidelser såsom psoriasis, 10 kontakt-dermatitis, atopisk dermatitis og eksem er omfattet af denne kategori. Behandling med præparatet ifølge opfindelsen gennemføres sædvanligvis ved at påføre præparatet, således at det dækker det angrebne område fuldstændigt. Den sædvanlige påføringsfrekvens er to gange 15 dagligt, skønt der for nogle patienter kan opretholdes en tilstrækkelig terapeutisk virkning med et mindre antal påføringer.
Præparatet ifølge opfindelsen indeholder 0,02 til 10 vægt% β-methasondipropionat, 0 til 0,6 vægt% titandioxid, 20 1,0 til 2,5- .vægt%-.Garbexner -940, en tilstrækkelig mængde base til at holde pH mellem 4,0 og 5,5, 60 til 80 vægt% propylenglycol og vand indtil 100 vægt% .
Ved en foretrukken udførelsesform af opfindelsen 25 anvendes 0,06 til 0,08 vægt% β-methasondipropionat og 65 til 75 vægt% propylenglycol. Det er desuden nyttigt, at præparatet indeholder 0,4 til 0,6vægt% titandioxid.
Både organiske og uorganiske baser er egnede baser ifølge opfindelsen, dog foretrækkes uorganiske 30 baser, især natriumhydroxid.
Carbomer 940 er en acrylsyrepolymer med en om— . trentlig molekylvægt på 4.000.000. Den kan fås fra B.F. Goodrich Chemical Company som Carbopol 940 harpiks .
35 Formuleringen af præparatet resulterer i en særde les fin cremelignende gel, når det indeholder fra 0,4 til 6,0 vægt% titandioxid. Hvis der ikke anvendes titandioxid, fås der et klart materiale. Anvendelsen af

3 det førstnævnte præparat er især fordelagtiq, idet et sådant præparat har det ønskede udseende for en creme og er lettere at påføre end kendte geler, medens det er overraskende mere effektivt end den ovenfor nævnte, 5 kendte creme.
Præparatet ifølge opfindelsen fremstilles på sædvanlig måde ved omhyggelig blanding af ingredienserne ved stuetemperatur eller ved forhøjede temperaturer.
Fortrinsvis sættes β-methasondipropionatet, der 10 er opløst i en del af propylenglycolen, til cremegrundlaget. Ingredienserne blandes omhyggeligt, således at produktet er homogent. Om ønsket kan yderligere mekanisk omrøring anvendes som et mellem- eller sluttrin ved fremstillingen for at gøre produktet mere homogent 15 eller for at forbedre konsistensen. Procesudstyr, der egner sig til disse trin, er kendt og omfatter varmevekslere, propelomrørere, kolloidmøller, homogenisato-rer, rullemøller og lignende.

Claims (5)
1. Topisk præparat til behandling af inflammation, kendetegnet ved, at det indeholder 0,02 til 0,10 vægt% β-methasondipropionat, 0 til 0,6 vægt% titan-dioxid, 1,0 til 2,5 vægt% Carbomer 940, en tilstrækkelig mængde base til at indstille pH på en værdi mellem 4,0 10 og 5,5, 60 til 80 vægt% propylenglycol og vand indtil 100 vægt%.
2. Præparat ifølge krav 1, kendetegnet ved, at det indeholder 0,06 til 0,08 vægt% β-methason-dipropionat, 0,4 til 0,6 vægt% titandioxid, 1,0 til 2,5 15 vægt% Carbomer 940, en tilstrækkelig mængde base til at indstille pH på en værdi mellem 4,0 og 5,5, 65 til 75 vægt% propylenglycol og vand indtil 100 vægt%.
