Den foreliggende opfindelse angår hidtil ukendte 5-hetero-cyclyl-2,4-dialkyl-3H-l,2,4-triazol-3-thioner samt farmaceutisk acceptable salte deraf. Disse forbindelser er anvendelige som antidepressive midler.

Fra Chem. Abstr. 90 (1979), nr. 203.971y, kendes der 3-quin-olyl-5-mercapto-l,2,4-triazoler, som angives at have antikon-vulsiv virkning.
10 Endvidere er forbindelsen 1,4-dimethyl-3-a-pyridyl-l,2,4-triazolin-5-thion beskrevet i Chem. Pharm. Bull. 20 (1972), 2096-2101. Forbindelsen er fremstillet i forbindelse med en påvisning af, at tilsvarende 1- eller 4-monosubstituerede forbindelser optræder i tautomere former, og der er ikke 15 nævnt noget om biologisk aktivitet af denne forbindelse.
Den foreliggende opfindelse angår forbindelser med formel (I) N—N-R2 20 *n “(I) R4 hvor R betyder halogen, alkyl med 1-6 carbonatomer, alkoxy med 1-6 carbonatomer, hydroxy eller trifluormethyl, n er 0, 1 eller 2, R2 og R4 uafhængigt betyder alkyl med 1-6 carbonatomer, og 5 Het betyder 3- eller 4-pyridyl, 2- eller 3-furyl, 2- eller 3-thienyl, pyrrol-2-yl, N-Ci_g-alkylpyrrol-2-yl, 2-, 3- eller 4-piperidinyl eller deres N-C^.g-alkylsubstituerede homologe, 6-isoquinolyl, 6-quinolyl eller 3-quinolyl, samt farmaceutisk acceptable salte deraf.
Halogen er fortrinsvis chlor eller fluor, og de foretrukne alkylgrupper er methyl og ethyl, selv om alle ligekædede, forgrenede og cycliske former deraf, såsom n-propyl, cyclo- 10 DK 171234 B1 2 pentyl, cyclohexyl og cyclopropyl, er omfattet deraf. Alkoxy-grupper omfatter ethere med alkyldele, der er parallelle med alkylgruppen med 1-6 carbonatomer. Kendte salte dannet med de heterocycliske grupper anvendes 15 sædvanligvis, idet hydrochloridet er bekvemt og alment anvendt. Sådanne salte fremstilles ved standardmetoder, der er kendte i teknikken.
Forbindelserne med formel (I) kan let fremstilles under 20 anvendelse af analoge metoder og processer, der er kendte i teknikken, som det kan ses af det efterfølgende reaktionsskema :

I trin A udføres fremstillingen af thiosemicarbaziderne med 5 formel (IV) let ved omsætning af en hydrazin med formel (II) med et isothiocyanat med formel (III) ved at bringe reaktanterne i kontakt i et egnet opløsningsmiddel. Reaktionen er ganske hurtig og kan udføres fra 0eC til stuetemperatur.
Selv om reaktionen skrider hurtigt frem, kan blandingen 10 henstå i op til 24 timer uden signifikant fald i udbytte. Tilbagesvalingsbetingelser kan anvendes, men er ikke foretrukket. Næsten alle opløsningsmidler, undtagen vand og organiske syrer, kan anvendes. Vandfri alkoholer, fortrinsvis ethanol eller methanol, foretrækkes, selv om DMF, CHCI3, 15 CH2CI2, THF og Et20 også kan anvendes. De nødvendige hydraziner og isothiocyanater er let tilgængelige, men kan fremstilles ved hjælp af teknik, der er helt indlysende for en fagmand.
20 I trin B kan de ønskede heterocyclylsubstituerede thiosemi-carbazider med formel (VI) fremstilles ved omsætning af thiosemicarbaziderne med formel (IV) med et R-substitueret heterocyclylchlorid med formel (V) i et aprotisk opløsningsmiddel, såsom pyridin, CHC13 og THF. Acyleringen skrider 25 ret let frem ved temperaturer fra 0°C til stuetemperatur i løbet af 3-24 timer, selv om der kan arbejdes ved forhøjede temperaturer, f.eks. tilbagesvalingstemperaturer. I praksis skal thiosemicarb-5 aziderne med formel (VI) ikke renses til anvendelse på trin C, således at endog 1:1 blandinger med pyridin-hydrochlorid kan anvendes.
De følgende specifikke eksempler gives til illustration af 10 fremstillingen af de her omhandlede forbindelser, selv om omfanget af de eksemplificerede forbindelser ikke er begrænset hertil, idet dette er indlysende på grund af den lethed, hvormed forbindelserne med formel (I) kan fremstilles.

Fremstilling af R2.R^-disubstituerede thiosemicarbazider 20
Eksempel 1 2.4-Dimethvlthiosemicarbazid
Til en omrørt opløsning af 16,0 ml (3,00 x 10-1 mol) methyl-25 hydrazin og 50 ml molekylsigtetørret ethanol sættes dråbevis en opløsning af 22,0 g (3,00 x 10"1 mol) methylisothiocyanat og 30 ml molekylsigtetørret ethanol. Reaktionen er exotermisk og tilbagesvaler svagt, efterhånden som isothiocyanatet tilsættes. Der dannes hurtigt et bundfald. Efter omrøring 30 natten over afkøles reaktionen i et isbad. Bundfaldet opsamles derefter ved filtrering, vaskes med en smule koldt iso-propanol og tørres ved sugning, hvilket giver 26,7 g (75%) af et bleggult pulver. Dette materiale krystalliseres to gange fra vand og to gange fra isopropanol, hvilket giver 35 14,7 g (41%) små, farveløse nåle, smp. 135-137°C.

Fremstilling af l-Rn-heterocvclovl-R2.R^-disubstituerede thiosemicarbazider
Eksempel 2 5 2.4-Dimethvl-l-(4-Pvridovll-thiosemicarbazid
Til en omrørt opløsning af 6,7 g (5,6 x 10“2 mol) 2,4-dimeth-ylthiosemicarbazid og 150 ml pyridin sættes dråbevis 10,0 g (5,62 x 10”2 mol) 4-pyridoylchlorid-HCl. Efter omrøring i 10 17 timer afdampes opløsningsmidlet til tørhed, hvilket giver en blanding af det ønskede 1-(4-pyridoyl)-2,4-dimethylthio-semicarbazid og pyridin-hydrochlorid. Almindeligvis anvendes denne blanding uden yderligere rensning på det efterfølgende ringslutningstrin. Hvis der ønskes rent 1-(4-pyridoyl)-2,4-15 dimethylthiosemicarbazid, behandles ovennævnte blanding med vand, og det ikke-opløselige materiale opsamles ved filtrering. Efter tørring ved sugning krystalliseres dette materiale.
20 Eksempel 3 2.4-Dimethvl-l-f2-thienovl)-thiosemicarbazid
Til en en omrørt opløsning af 3,75 g (3,15 x 10"2 mol) 2,4-dimethylthiosemicarbazid og 30 ml pyridin sættes ved stue-25 temperatur dråbevis 3,20 ml (2,99 x 10-2 mol) 2-thiophen-carbonylchlorid. Efter omrøring natten over afdampes pyridi-net ved formindsket tryk, og koncentratet behandles med H2O. Det resulterende, halvfaste produkt ekstraheres over i ethylacetat. Ethylacetatekstrakten vaskes med mættet NaCl 30 og tørres derefter over vandfrit Na2S04· Tørringsmidlet fjernes ved frafiltrering, og filtratet inddampes ved formindsket tryk, hvilket efterlader et svagt gulligt faststof, som behandles med ether, og faststoffet opsamles ved filtrering, hvilket giver 4,7 g (68%) farveløst pulver, smp. 176-35 178’C.

En 1:1 blanding fremstillet ifølge eksempel 2 og 100 ml l M (1,00 x 10"1 mol) vandigt NaHC03 omrøres og opvarmes til tilbagesvaling. Efter tilbagesvaling i 15 timer får reaktionen lov til af afkøle i et isbad. Det resulterende bund-10 fald opsamles ved filtrering og tørres ved sugning. Det ønskede produkt krystalliseres fra ethylacetat:hexan til opnåelse af 4,1 g (35%) store, farveløse plader, smp. 150-152 * C.
15 Eksempel 5 2.4-Dimethvl-5-f 2-thienvl^-3H-1.2.4-triazol-3-thion
En omrørt blanding af 4,7 g (2,0 x 10"2 mol) 2,4-dimethyl-1-(2-thienoyl)-thiosemicarbazid fremstillet ifølge eksempel 20 3 og 200 ml 1 M (2,0 x 10”2 mol) vandigt NaHC03 opvarmes til tilbagesvaling i 6 timer. Derefter anbringes reaktionsblandingen i køleskab i nogle timer, før bundfaldet opsamles ved filtrering. Krystallisation fra isopropanol giver 3,6 g (83%) lange, farveløse nåle, smp. 128-129“C.
25 På tilsvarende måde fremstilles ved ombytning af reaktanterne ifølge eksemplerne 1-3 med egnede R2,R4“disubstituerede reaktanter og ved i alt væsentligt at følge metoderne ifølge eksempel 4 og 5 2,4-dimethyl-5-heterocyclyl-3H-l,2,4-triazol-30 3-thioner med formel (I) , hvor den heterocycliske forbindelse har den ovenfor nævnte betydning for Rn-Het.
Andre forbindelser med formel (I) kan fremstilles på lignende måde under anvendelse af metoderne ifølge eksemplerne 1-5.
Under anvendelse af standardlaboratoriemetodik kan de far- 35 DK 171234 B1 7 makologiske egenskaber og deres relative styrke let bestem mes. Ved sammenligning med andre midler, der klinisk vides at være anvendelige som antidepressive midler, kan dosissystemet let bestemmes af fagfolk.

Forsøgsafprøvning til forebyggelse af reserpinfremkaldt ptosis hos mus og rotter er således et standardforsøg. I disse forsøgsgrupper anbringes vejede mus eller rotter individuelt i trådbure og får indgivet forsøgsforbindelse 10 eller grundmasse. På et bestemt tidspunkt derefter indgives reserpin, fremstillet som 4 mg/ml opløsning i fortyndet eddikesyre, subkutant til rotterne i en dosis på 4 mg/kg og til mus som en 0,2 mg/ml opløsning i fortyndet eddikesyre intravenøst i en dosis på 2 mg/kg i en halevene. I hvert 15 forsøg undersøges dyrene individuelt i en plexiglascylinder 90 minutter senere. Forebyggelse eller forsinkelse i ptosis betragtes som signifikant, hvis den gennemsnitlige lukning af begge øjne er mindre end 50% efter iagttagelse i 30 sekunder. ED50 for forebyggelse af ptosis defineres som den 20 dosis af forsøgsforbindelsen, der signifikant forhindrer ptosis hos 50% af forsøgsdyrene.
Ved disse forsøg har imipramin en ED50 på 2,6 mg/kg (ved en præ-behandlingstid på 30 minutter) hos rotter. Hos mus har 25 imipramin en ED50 på 4,1 mg/kg ved en præ-behandlingstid på 60 minutter. For forbindelsen fremstillet ifølge eksempel 5 er den tilsvarende værdi mellem 0,25 og 0,5 mg/kg hos begge dyrearter ved en præ-behandlingstid på 30 minutter. 1 2 3 4 5 6
Et andet forsøg til vurdering af den antidepressive aktivitet 2 er afprøvning for antagonisme over for RO-4-1284-fremkaldt 3 hypotermia (Niemegeers, J.E. Carlos, "Antagonism of Reserpine 4 - Like Activity", udgivet af S. Fielding og Lal, publiceret 5 af Futura, side 73-98). Ved dette forsøg vejes grupper af 6 hanmus og anbringes individuelt i tremmebure. Den rektale temperatur hos hver mus noteres, og derefter indgives for- DK 171234 B1 8 søgsforbindelsen eller grundmassen. På et udvalgt tidspunkt derefter indgives RO-4-1284, fremstillet som en 2 ml/mg opløsning i destilleret vand, i en dosis på 20 mg/kg i.p. Derefter anbringes mus i et koldt rum (ca. 2eC) i 30 minutter 5 og føres derefter tilbage til stuetemperatur i 30 minutter.
På dette tidspunkt (60 minutter efter indgift af RO-4-1284) noteres den rektale temperatur igen hos musene. Under disse betingelser forårsager RO-4-1284 et fald i den rektale temperatur på 10-12‘C. Sluttemperaturerne hos kontrolgrupperne 10 på 10 mus behandlet med RO-4-1284 fra et antal forsøg kombineres til dannelse af en "historisk kontrol" på 100 mus. Denne kontrol opdateres periodisk ved erstatning af de ældste data. Ethvert dyr, der er behandlet med lægemiddel, og som har en sluttemperatur (efter RO-4-1284) , som er højere end 15 gennemsnittet +2 standardafvigelser af RO-4-1284-historisk kontrol, anses for at udvise signifikant antagonisme over for den hypotermiske virkning af RO-4-1284. ED50 for antagonisme defineres som den dosis af forsøgsforbindelsen, som hos 50% af forsøgsdyrene signifikant antagoniserer RO-20 4-1284-fremkaldt hypotermia.
Ved anvendelse af en præ-behandlingstid på 60 minutter og disse kriterier for vurdering af virkninger viser desipramin sig at have en ED50 på 0,1 mg/kg i.p., imipramin en ED50 på 25 1,8 mg/kg i.p. og "Catron" en ED50 på 0,7 mg/kg i.p. Den ifølge eksempel 5 fremstillede forbindelse har efter en præ-behandlingstid på 30 minutter en ED50 på mellem 0,5 og 1,0 mg/kg ved samme forsøg. 1 2 3 4 5 6 På grundlag af standardlaboratoriemetodik og sammenlignings 2 undersøgelser med kendte midler forventes det, at forbindel 3 serne ifølge den foreliggende opfindelse har farmakologiske 4 virkninger, der almindeligvis tilskrives antidepressive 5 midler, og forbindelserne ifølge den foreliggende opfindelse 6 vil således højne sindsstemningen hos patienter, der lider af depressioner, og vil derfor have en slutanvendelse til DK 171234 B1 9 behandling af patienter, der lider af endogene depressioner, et udtryk, der er synonymt med psykotisk eller involutionel depression. Ved denne anvendelse viser forbindelsen med formel (I) en relativt hurtig start af virkning og har for-5 længet varighed af aktivitet. Almindeligvis forventes det, at forbindelserne udviser deres antidepressive virkninger ved dosisniveauer på ca. 0,25-25 mg/kg legemsvægt pr. dag, selv om graden af alvorligheden af sygdomstilstanden, patientens alder og andre faktorer bestemt af den behandlende 10 diagnostiker naturligvis vil påvirke det nøjagtige tidsskema og dosisskema, der er egnet for hver enkelt patient. Almindeligvis er parenteralt indgivne doser ca. 1/4 til 1/2 af den oralt indgivne dosis.
15 Til oral indgift kan forbindelserne formuleres i faste eller flydende præparater, såsom kapsler, piller, tabletter, pastiller, pulvere, opløsninger, suspensioner eller emulsioner.
De faste dosisenhedsformer kan være en kapsel, som kan være af den gængse gelatinetype, der f.eks. indeholder smøremidler 20 og indifferente fyldstoffer, såsom lactose, saccharose eller majsstivelse. I en anden udførelsesform kan forbindelserne med den almene formel (I) være tabletteret med konventionelle tabletgrundmasser, såsom lactose, saccharose og majsstivelse; i kombination med bindemidler, såsom acacia, majsstivelse 25 eller gelatine, disintegreringsmidler, såsom kartoffelstivelse eller algininsyre, og et smøremiddel, såsom stearinsyre eller magnesiumstearat.
Til parenteral indgift kan forbindelserne indgives som in-30 jicerbare doser af en opløsning eller suspension af forbindelsen i et fysiologisk acceptabelt fortyndingsmiddel med et farmaceutisk acceptabelt bærestof, som kan være en steril væske, såsom vand, alkoholer, olier eller andre acceptable, organiske opløsningsmidler med eller uden tilsætning af et 35 overfladeaktivt middel og andre farmaceutisk acceptable hjælpestoffer. Eksempler på olier, der kan anvendes i disse DK 171234 B1 10 præparater, er olier af mineralsk, animalsk, vegetabilsk eller syntetisk oprindelse, f.eks. jordnøddeolie, soyabøn-neolie og mineralolie. Almindeligvis er vand, en fysiologisk saltopløsning, vandig dextrose og beslægtede sukkeropløsnin-5 ger, ethanol og glycoler, såsom propylenglycol eller poly-ethylenglycol, eller 2-pyrrolidon foretrukne flydende bærestoffer, især til injicerbare opløsninger.
Forbindelserne kan indgives i form af en depotinjektion 10 eller et implantationspræparat, som kan formuleres på en sådan måde, at det tillader forsinket frigivelse af den aktive ingrediens. Den aktive ingrediens kan sammenpresses til pellets eller små cylindre og implanteres subkutant eller intramuskulært som depotinjektioner eller implanta-15 tioner. I implantationer kan være anvendt indifferente materialer, såsom biosønderdelelige polymere eller syntetiske siliconer, f.eks. "Silastic"®, en siliconegummi fremstillet af The Dow-Corning Corporation.

- 2. Forbindelse ifølge krav 1, kendetegnet ved, at Rn-(Het)- er 4-pyridyl.
- 3. Forbindelse ifølge krav 1 eller 2, kendete g-15 net ved, at R2 og R4 begge er methyl.
- 4. Forbindelse ifølge krav 1 eller 3, kendetegnet ved, at n er 1.
- 5. Forbindelse ifølge krav 2 eller 3, kendeteg net ved, at den er 5-(4-pyridyl)-2,4-dimethyl-3H-l,2,4-triazol-3-thion.
