Opfindelsen angår hidtil ukendte triazolderivater, som har antifungal aktivitet og er nyttige ved behandling af svampeinfektioner hos dyr, herunder mennesker, og som landbrugsfungicider. Opfindelsen angår endvidere farmaceutiske 5 og landbrugsmæssige fungicide præparater indeholdende disse forbindelser og en fremgangsmåde til behandling af planter eller frø mod svampeinfektion.

Opfindelsen tilvejebringer yderligere et antifungalt præparat til anvendelse inden for landbrug (herunder havebrug), som er særegent ved, at det indeholder en forbindelse med 25 formlen (I) eller en af de ovennævnte estere eller ethere deraf eller et landbrugsmæssigt acceptabelt syreadditionssalt deraf sammen med et landbrugsmæssigt acceptabelt fortyndingsmiddel eller bærestof.
Opfindelsen tilvejebringer desuden en fremgangsmåde til be-30 handling af planter eller frø, som har en svampeinfektion, hvilken fremgangsmåde er særegen ved, at planten eller frøet eller plantens eller frøets voksested behandles med en antifungalt effektiv mængde af en forbindelse med formlen 3 DK 172423 B1 (I) eller en af de ovennævnte estere eller ethere deraf eller et landbrugsmæssigt acceptabelt syreadditionssalt deraf.
Når R er en evt. substitueret phenylgruppe, er den phenyl 5 med 1-3 substituenter, mere foretrukket 1 eller 2 substitu-enter, der hver for sig er valgt blandt F, Cl, Br, I og CF3, og fortrinsvis blandt F og Cl. De foretrukne individuelle grupper repræsenteret ved R er 4-fluorphenyl, 4-chlor-phenyl, 4-bromphenyl, 4-iodphenyl, 4-trifluormethylphenyl, 10 2-chlorphenyl, 2,4-dichlorphenyl, 2,4-difluorphenyl, 2-chlor-4-fluorphenyl, 2-fluor-4-chlorphenyl, 2,5-difluorphe-nyl, 2,4,6-trifluorphenyl, 4-brom-2,5-difluorphenyl og 5-chlor-pyridin-2-yl.
X er fortrinsvis OH. R1 og R2 er hver fortrinsvis H eller 15 CH3.
O-esterne er (C2-C4)alkanoyl- (f.eks. acetyl-) og benzoyl-estere. Phenylringen i benzoylesterne kan være substitueret med 1 eller 2 (Ca-C4)alkylgrupper eller halogenatomer. 0-etherne er (C1-C4)alkyl-, (C2-C4)alkenyl-, phenyl-(C!-C4al-20 kyl)- og phenylethere. Igen kan de nævnte phenylgrupper være ringsubstitueret med 1 eller 2 (Cx-C^alkylgrupper eller halogenatomer.

hvori R, R1 i og R2 har den ovenstående betydning, med tetrazol, typisk i nærvær af bis(tri-n-butylstannyl)oxid.
Bis(tri-n-butylstannyl)oxidet antages at reagere med te-30 trazol til dannelse af 1- og 2-tributylstannyl-(Bu3Sn-)-derivaterne deraf, som letter ringåbningen af epoxidet 4 DK 172423 B1 (II). Omsætningen udføres typisk i et egnet organisk opløsningsmiddel, f.eks. toluen, ved fra stuetemperatur op til ca. 110 °C, og ønskeligt under en nitrogenatmosfære. Reaktionen frembringer en blanding af 1-tetrazolyl- og 2-5 tetrazolylslutprodukterne, som kan adskilles chromatografisk.
Oxiran-udgangsmaterialerne med formlen (II) er enten kendte forbindelser eller kan fremstilles ved metoder analoge med den kendte teknik (se f.eks. EP 0069442, EP 10 0044605, US 4 499 281, EP 0122056, EP 0122693, EP 0120276 og EP 0118245).
De forbindelser med formlen (I), hvori X er F, Cl, eller Br, kan fremstilles ved halogenering af de tilsvarende forbindelser, hvori X er OH.
15 Halogeneringen udføres ifølge konventionelle procedurer, f.eks. under anvendelse af S0C12, SOBr2 eller diethylami-no-svovl-trifluorid (Et2NSF3) (se EP offentliggørelsesskrift nr. 96569).
Ved en typisk procedure under anvendelse af thionylchlo-20 rid eller -bromid omsættes det hydroxylholdige triazolde-rivat (I) i et egnet opløsningsmiddel, f.eks. tørt aceto-nitril, med thionylchlorid eller -bromid ved en temperatur på fra 0 °C til tilbagesvalingstemperaturen, evt. i nærvær af en base, f.eks. imidazol. Det halogenholdige 25 produkt kan derpå isoleres og renses på konventionel måde. Reaktionen under anvendelse af diethylaminosvovltri-fluorid udføres typisk ved en temperatur på fra omkring 0 °C til stuetemperatur, fortrinsvis i methylenchlorid som opløsningsmiddel. Igen kan produktet isoleres og renses 30 konventionelt.
O-esterne og 0-etherne kan fremstilles konventionelt, typisk ved omsætning af et alkalimetalsalt af forbindelsen 5 DK 172423 B1 med formlen (I) [X = OH] med den passende chlor- eller bromforbindelse, f.eks. et alkanoyl- eller benzoylchlorid eller alkyl-, alkenyl-, benzyl- eller phenylchlorid eller -bromid.
5 Farmaceutisk acceptable syreadditionssalte af forbindelserne med formlen (I) er sådanne, der dannes ud fra stærke syrer, som danner ikke-toxiske syreadditionssalte, såsom saltsyre, hydrogenbromidsyre, svovlsyre, oxalsyre og methansulfonsyre. Sådanne salte er også nyttige til land-10 brugsformål.
Saltene kan opnås ved konventionelle procedurer, f.eks. ved blanding af opløsninger indeholdende tilnærmelsesvis ækvimolære mængder af den frie base og ønskede syre, og det ønskede salt opsamles ved filtrering, hvis det er 15 uopløseligt, eller ved afdampning af opløsningsmidlet.
Forbindelserne med formlen (I) og deres nævnte O-estere, 0-ethere og salte er antifungale midler, der er nyttige til bekæmpelse af svampeinfektioner hos dyr, herunder mennesker. For eksempel er de nyttige til behandling af 20 topiske svampeinfektioner hos mennesket, frembragt af blandt andre organismer arter af Candida Trichophyton, Microsporum eller Epidermophyton, eller til behandling af slimhindeinfektioner forårsaget af Candida albicans (f.eks. trøske og vaginal candidiasis). De kan også an-25 vendes til behandling af systemiske svampeinfektioner forårsaget af f.eks. Candida albicans. Cryptococcus neo-formans, Aspergillus flavus, Aspergillus fumigatus, Coc-cidioides, Paracoccidioides, Histoplasma eller Blastomyces .
30 In vitro bedømmelsen af forbindelsernes antifungale aktivitet kan gennemføres ved bestemmelse af den minimale in-hiberende koncentration (MIC), som er den koncentration af prøveforbindelserne i et egnet medium, hvorved der DK 172423 Bl 6 netop ikke sker vækst af den bestemte mikroorganisme. I praksis podes en række agarplader, som hver har prøveforbindelsen inkorporeret i en bestemt koncentration, med en standardkultur af f.eks. Candida albicans, og hver plade 5 inkuberes derpå i 48 timer ved 37 °C. Pladerne undersøges derefter for forekomst eller fravær af vækst af svampen, og den pågældende MIC-værdi noteres. Andre mikroorganismer, som kan anvendes ved sådanne prøvninger, inkluderer Candida albicans, Aspergillus fumigatus, Trichophyton 10 spp, Microsporum spp, Epidermophyton floccosum, Coccidio-ides immitis og Torulopsis glabrata.
In vivo bedømmelsen af forbindelserne kan udføres ved en række dosisniveauer ved intraperitoneal eller intravenøs injektion eller ved oral indgivelse til mus, som er podet 15 med f.eks. en stamme af Candida albicans eller Aspergillus flavus. Aktiviteten baseres på overlevelsen af en behandlet gruppe af mus efter døden af en ubehandlet gruppe af mus. Det dosisniveau, ved hvilket forbindelsen frembringer 50 % beskyttelse mod infektionens dødelige virk-20 ning ( PD50 ) noteres.
Til human brug kan de antifungale forbindelser med formlen (I) og deres nævnte salte, 0-ethere og O-estere indgives alene, men almindeligvis vil de blive indgivet i blanding med en farmaceutisk bærer udvalgt under hensyn-25 tagen til den påtænkte indgivelsesvej og farmaceutisk standardpraksis. For eksempel kan de indgives oralt i form af tabletter indeholdende sådanne excipienter som stivelse eller lactose, eller i kapsler eller æg enten alene eller i blanding med excipienter, eller i form af 30 eliksirer eller suspensioner indeholdende aroma- eller farvemidler. De kan injiceres parenteralt, f.eks. intravenøst, intramuskulært eller subcutant. Til parenteral indgivelse anvendes de bedst i form af en steril vandig opløsning, som kan indeholde andre stoffer, f.eks. til- 7 DK 172423 B1 strækkeligt salte eller glucose til at gøre opløsningen isotonisk med blod.
Til oral og parenteral indgivelse hos mennesker vil det daglige doseringsniveau af de antifungale forbindelser 5 med formlen (I) og deres nævnte salte, O-ethere og 0- estere være 0,1-10 mg/kg (i opdelte doser). Således vil tabletter eller kapsler med forbindelserne indeholde fra 5 mg til 0,5 g aktiv forbindelse til indgivelse alene eller to eller flere ad gangen efter behov. Lægen vil i 10 hvert tilfælde bestemme den faktiske dosering, som vil være mest egnet for en individuel patient, og den vil variere med den bestemte patients alder, vægt og reaktion.
De ovenstående doseringer er eksempler på gennemsnitstilfælde; der kan selvfølgelig være individuelle tilfælde, 15 hvor højere eller lavere doseringsområder kan være af værdi.
Alternativt kan de antifungale forbindelser med formlen (I) indgives i form af et suppositorium eller pessar, eller de kan påføres topisk i form af en lotion, opløsning, 20 creme, salve eller pudder. For eksempel kan de inkorpore res i en creme bestående af en vandig emulsion af poly-ethylenglycoler eller flydende paraffin, eller de kan inkorporeres i en koncentration mellem 1 og 10 % i en salve bestående af en hvid voks eller hvid blød paraffinbase 25 sammen med sådanne stabiliserings- og konserveringsmidler, som måtte være nødvendige.
Forbindelserne med formlen (I) og deres nævnte 0-ethere, 0-estere og salte har også aktivitet over for en række forskellige plantepatogene svampe, herunder f.eks. for-30 skellige rustarter, meldug og skimmelsvampe, og forbindelserne er således nyttige til behandling af planter og frø for at udrydde eller forhindre sådanne sygdomme.

In vitro bedømmelsen af forbindelsernes aktivitet over for plantesvampe kan bestemmes ved måling af deres minimale inhiberende koncentrationer på samme måde som tidligere beskrevet, undtagen at pladerne inkuberes ved 30 °C 5 i 48 timer eller længere, før de undersøges for forekomst eller fravær af vækst.
Mikroorganismer, som anvendes ved sådanne prøvninger, inkluderer Cochliobolus carbonum, Pyricularia oryzae, Glo-merella cingulata, Penicillium digitatum, Botrytis cine-10 rea og Rhizoctonia solani.
Til landbrugs- og havebrugsformål anvendes forbindelserne og deres landbrugsmæssigt acceptable salte fortrinsvis i form af et præparat sammensat efter den bestemte anvendelse og det ønskede formål. Således kan forbindelserne 15 påføres i form af pudder, granulat, frøbehandlingsmidler, vandige opløsninger, dispersioner eller emulsioner, dyppemidler, sprøjtemidler, aerosoler eller røg. Præparater kan også leveres i form af dispergerbare pulvere, granulater eller korn eller som koncentrater til fortynding 20 før brugen. Sådanne præparater kan indeholde sådanne konventionelle bærere, fortyndingsmidler eller tilsætningsmidler, som er kendte og acceptable inden for landbrug og havebrug, og de fremstilles i overensstemmelse med konventionelle procedurer. Præparaterne indeholder typisk 25 0,01-95 vægt%, fortrinsvis 0,01-1 vægt% af den aktive in grediens. Præparaterne kan også inkorporere andre aktive ingredienser, f.eks. forbindelser med herbicid eller insekticid aktivitet eller et yderligere fungicid.
Forbindelserne og præparaterne kan påføres på et stort 30 antal måder, f.eks. kan de påføres direkte på plantelø-vet, stænglerne, grenene, frøene eller rødderne eller på jorden eller andet voksemedium, og de kan anvendes ikke 9 DK 172423 B1 blot til at udrydde sygdom, men også profylaktisk til at beskytte planterne eller frøene mod angreb.
Til anvendelse i marken er sandsynlige påføringsmængder af den aktive ingrediens 0,5-50 g/ha.

En blanding af 2-(2,4-dichlorphenyl)-2-(1,2,4-triazol-l-ylmethyl)oxiran (2,4 g) (se eksempel 4 i EP 0044605), te-trazol (750 mg) og bis(tri-n-butylstannyl)oxid (306 mg) i tørt toluen (50 ml) blev opvarmet til 100-110 °C under en 25 nitrogenatmosfære i 62 timer. Derpå tilsattes eddikesyre 10 DK 172423 B1 (0,5 ml) til den afkølede opløsning, som blev filtreret, og filtratet blev efter inddampning opløst i acetonitril (150 ml) og derpå ekstraheret med n-hexan (3 x 80 ml). De kombinerede acetonitrilfaser blev koncentreret, og rema-5 nensen blev lynchromatograferet på siliciumdioxid (partikelstørrelse svarende til maskevidde 37-62 μπι) under elu-ering med en blanding af dichlormethan, methanol og ammoniakvand med relativ massefylde (vand) 0,88 (96:4:0,5).
De første fraktioner gav noget ureageret epoxid (424 mg) 10 efterfulgt af den i overskriften angivne tetra-zol-2-yl-forbindelse og derefter den i overskriften angivne tetrazol-l-yl-forbindelse.
Fraktionerne indeholdene tetrazol-2-yl-isomeren blev derpå inddampet, remanensen blev udrevet med hexan, og det 15 resulterende faste stof krystalliseret fra ethylace-tat/hexan, hvilket gav den i overskriften angivne tetra-zol-2-yl-forbindelse, snip. 170 °C (680 mg). [Massespektrum m/e 340 (M*)].
