Enzymer er forbindelser, sædvanligvis proteiner eller glycoproteiner, der specifikt accelererer biokemiske reaktioner, idet de nedsætter aktiveringsenergien. I det følgende betegner udtrykket biokatalysator ikke blot det aktiverende stof selv, men 15 kan eventuelt også omfatte komponenter, der anvendes til immobilisering, holdbarhed, regenerering osv. af det aktiverende stof.
Immobiliserede, dvs. i deres bevægelighed indskrænkede biokatalysatorer er især egnede til industrielle formål.
20 Ved immobiliseringen bliver det muligt at udføre de katalyserede processer kontinuerligt og gentaget, at skabe og opretholde høje biokatalysatortætheder, at opnå høje tids-rum-udbytter og at sænke omkostningerne for udvinding af produktet fra reaktionsopløsningen.
25 Det biokatalytiske materiale kan til immobilisering eksempelvis indesluttes i polymere overtræksstoffer (matrix), i kapsler eller fibre af semipermeable membraner eller bag ultrafiltreringsmembraner, tværbindes med bi- eller multifunktionelle reagenser eller ved adsorption eller 30 ionisk eller covalent binding fikseres på bærere af uorganisk materiale eller naturlige eller syntetiske polymere. Også en kombination af disse metoder er mulig.
Ved immobiliseringen må på den ene side den størst mulige bevaring af den biokatalytiske aktivitet og på den anden 2 DK 1:7 571 2 B1 side høj mekanisk styrke og kemisk: stabilitet, men s-rr-tidig også god permeabilitet for biokatalysatoren sikre s.

Ved inddrypning eller indsprøjtning af en suspension af enzymatisk aktivt materiale og en vandig opløsning af 15 matricestoffet i en vandig tværbindingsopløsning, c er indeholder polyvalente modioner, f.eks. Ca2+, Co2+, Zn:<+,
Fe2 + , Fe^ + , Al-3+ og andre, opnås chelatisering og dan nelse af perleformede biokatalysatorer.
Ved den ionotrope geldannelse af organiske polymere :iås 20 som regel kun relativt bløde, mekanisk ustabile immobil i -sater, der har tendens til kvældning og deformation, således at f.eks. deres anvendelse i fyldlegemereaktorar, således som de hyppigt anvendes industrielt til enzymatiske reaktioner, giver vanskeligheder, Hærdningsmetoder ve3 25 efterbehandling af immobilisaterne eller ved forbehandling af det biologisk aktive materiale før tilsætninga i til geleringsmidlet, i hvert tilfælde med multi -funktionelle tværbindingsreagenser, såsom polyaldehyder, f.eks. glutaraldebyd, eller isocyanater, f.eks. hexama-30 thylendiisocyanat, kan på grund af deres celle- og/ellar enzymbeskadigende virkning kun anvendes meget begrænset.

Også tørrinj og skrumpning af biokatalysatorperlerne anvendes tit hærdning, da perlerne efter genbefugtning ikke atter kai DK 173712 B1 3 indtage deres oprindelige fugtige rumfang og forbliver hårdere. Tysk offentliggørelsesskrift nr. 2.835.875 beskriver en fremgangsmåde til ionotrop geldannelse under 5 anvendelse af polyanioner, f.eks. natriumalginat, og eksempelvis calciumioner som tværbindingsmiddel, ved hvilken de med enzymatisk stof ladede biokatalysatorperler tørres ved en temperatur indtil 80°C. Sådanne tørringstrin kan dog ikke altid gennemføres, når højfølsomme enzymer 10 eller enzymsystemer, især levende celler, skal immobili-seres, da de høje temperaturer ved tørringen eller den tørre tilstand inaktiverer cellerne eller enzymerne.
Endnu en ulempe ved de ved ionotrop geldannelse fremstillede calciumalginat-immobilisater er foruden de dårlige 15 mekaniske egenskaber også indskrænkningen i mulige reaktionsmedier ved den videre forarbejdning. Sådanne immobi-lisater er f.eks. kemisk ustabile i nærvær af ioner, som de gængse pufferopløsninger indeholder, såsom natrium eller kalium. I sådanne elektrolytopløsninger, især når de 20 er højkoncentreret, fortrænges f.eks. de gelstyrkende calciumioner i calciumalginat-geler, fordi natrium- og kaliumioner konkurrerer om alginatanionen. Også phosphat-ioner fører efterhånden til opløsning af calciumalginat-biokatalysatorperler, da phosphater reagerer med calcium 25 og unddrager alginatet det strukturstyrkende middel. Opløsningen kan hindres, når der som matricestoffer anvendes polykationer, f.eks. chitosan, der tværbindes med polyvalente anioner, såsom [Fe(CN)g]3-, [Fe(CN)g]4-, polyphosphater og lignende. De tyske patentskrifter nr.
30 3.005.632 og 3.005.633 beskriver fremstillingen af biokatalysatorperler under anvendelse af chitosan og K3[Fe(CN)g ]. Fremgangsmåden ifølge tysk patentskrift nr. 3.005.632 indeholder til hærdning af perlerne et tørringstrin ved en temperatur indtil 80°C. For at muliggøre 35 immobiliseringen af høj følsomt enzymatisk stof, indeholder fremgangsmåden ifølge tysk patentskrift nr. 3.005.633 ikke dette tørringstrin, hvilket dog fører til en ringere 4 DK 1 73712 B1 mekanisk styrke af biokatalysatorperlerne.
Det er den foreliggende opfindelses opgave af stills 5 perleformede biokatalysatorer til råd.:kjhed, der har høj mekanisk styrke, hvis stabilitet også er sikret ved anvendelse af da gængse biologiske opløsninger og puffersystemer, og som endvidere sikrer høj følsomt enzymatisk aktivt 10 materiales enzymatiske aktivitet.
Endvidere er det opfindelsens opgave at påpege egneda fremgangsmåder til fremstilling af sådanne biokatalysatorer.

Som særlig egnede til løsning af disse opgaver viser sig perleformede biokatalysatorer, der indeholder enzymatisk aktivt materiale, ©n kationisk polyelektrolyt, fortrinsvis chitosan, polyvalente anioner og kiselsyre. Sådanne bioka-20 talysatorperler fremstilles i fast form ved en skånende immobiliseringsfremgangsmåde ved ionotrop geldannelse og ved anvendelse af kiselsyre og fremstilles ud fra en suspension af det enzymatisk aktive materiale, kiselsyren, pufferopløsning og polyelektrolytten, hvori slutkon-25 centrationen af kiselsyre ligger i et interval mellem 2 og 15 vægt-%.
Opfindelsen angår perleformede biokatalysatorer med høj mekanisk styrke omfattende enzymatisk aktivt materiale, ei 30 kationisk polyelektrolyt og polyvalente anioner, som er ejendommelige ved, at biokatalysatorerne ifølge opfindelsen indeholder kiselsyre og fremstilles ud fra en suspensior af det enzymatisk aktive materiale, kiselsyren, pufferopløsning og polyelektrolytten, hvori slutkoncentrationen cif 35 kiselsyre ligger i et interval mellem 2 og 15 vægt-%.

De perleformede biokatalysatorer ifølge opfindelsen er karakteristiske ved høj mekanisk styrke, hvilket kan føres tilbage til udviklingen af en Dual-gelmatrice-struktur. De har derfor gode pakningsegenskaber, eksempelvis ved fyld-5 ning af en reaktionsbeholder, og kan anvendes som fyldmateriale til fastiejereaktorer. På grund af de milde , immobiliseringsbetingelser, der ikke omfatter noget tørringstrin eller en behandling med skadelige stoffer, er biokatalysatorerne ifølge opfindelsen navnlig egnede til 10 højfølsomt materiale, især til levende celler. Fremstillingen af de perleformede biokatalysatorer ifølge opfin- . delsen ved anvendelse af kiselsyre i fast form har i forhold til de fra teknikkens standpunkt kendte fremgangsmåder de fordele, at tids- og omkostningskrævende frem- 15 gangsmåder til forberedelse af de ved hærdningstrinnet anvendte tværbindingsmidler undgås. Endvidere kan biokatalysatorerne anvendes i puffersystemer, der indeholder ioner, der fører til opløsning af alginat-geler. Således kan biokatalysatorerne ifølge opfindelsen 20 eksempelvis anvendes i opløsninger, der indeholder, phosphationer, f.eks. i den meget billige og derfor til | industrielle formål fordelagtige tripolyphosphatpuffer. De perleformede biokatalysatorer ifølge opfindelsen egner sig til omsætning af organiske stoffer. Da de kan anvendes til 25 reaktioner, der kræver høje elektrolytkoncentrationer, egner biokatalysatorperler ifølge opfindelsen, der indeholder en bakterie af slægten Proteus eller en oxidoreduktase som enzymatisk aktivt materiale, sig eksempelvis til fremstilling af α-hydroxycarboxylsyrer ud 30 fra α-ketocarboxylsyrer, f.eks. af 2-(R)-hydroxy-4-phenyl-smørsyre ud fra 2-oxo-4-phenylsmørsyre.
De perleformede biokatalysatorer ifølge opfindelsen har 35 høj mekanisk styrke. Ved høj mekanisk styrke forstås eksempelvis høj trykstyrke, dvs. at biokatalysatorperler DK 173712 B1 6 ifølge opfindelsen med en gennemsnitlig størrelse på ca. 3 mm ved en belastning på 0,01 til 0,03 N, isser 0,02 N, efter et til fem minutter har en maksimal deformation på 30 til 50 μπι, især 40 μτη, og efter 10 til 20 minutter, ' ^ især 15 minutter ikke udviser yderligere deformations- tilvækst.

Mikroorganismen kan være af prokaryot eller eukaryot natur. Egnede mikroorganismer er eksempelvis - en bakterie, f.eks. en mælkesyrebakterie (Leuconostoc, 2Q Streptococcus, Lactobacillus), Corynebacterium,
Streptomyces, Pseudomonas, Xanthomonas, Bacillus, Clostridium, Escherichia, Proteus osv., p en svamp, f.eks. Mucor, Aspergillus, Penicillium, især gærarter, såsom Saccharomyces, Candida osv., __ - en alge, f.eks. Chlorella, Curvularia osv., eller Z o - en plantecelle, f.eks. Catharanthus roseus, Daucus carota, Digitalis lanata, Morinda citrifolia osv. .
Anvendelsen af hele celler, cellefragmenter eller celle-30 organeller, dvs. med membraner overtrukne eller af membransystemer bestående celleunderenheder (chloroplaster, thylakoider, mitochondrier osv.), som enzymatisk aktivt materiale er fordelagtig, når den reaktion, der skal katalyseres, kræver et multienzymsystem, f.eks. når 35 cof aktorer (pyridin- eller f lavinnukleotider osv.) er nødvendige, eller når der i stedet for et katabolsk 7 DK 173712 B1 produkt af en biosyntese f.eks. skal fremstilles et sekundært produkt, hvortil komplicerede stofskiftekæder er nødvendige.

15 Som enzymatisk aktivt materiale kan biokatalysatorerne ifølge opfindelsen også omfatte en vilkårlig kombination af de beskrevne arter enzymatisk materiale, eksempelvis en kombination af hele celler af to eller flere arter eller stammer af mikroorganismer eller en kombination af hele 2 0 celler og enzymer. Grundtanken er kompletteringen af en celles biokatalytiske egenskaber med yderligere enzymer, der ikke eller kun i ringe mængde er til rådighed i cellen.

Den kationiske polyelektrolyt er en naturlig eller syntetisk polymer eller en naturlig polymer efter kemisk modifikation, eksempelvis en polymer med protoniserede aminogrupper, fortrinsvis chitosan ([ 2-amino-2-deoxy- 5 1-+4 )-(?-D-glucopyranan; poly-( 1,4-0-D-glucopyranosainin ]).
Chitosan er en lines;r polymer med høj nolvægt af glucos-amin-underenheder, der fås ud fra chitin ved delvis deacetylering ved hjælp af koncentreret base og varme.
Chitin er vidt udbredt i naturen, f.eks. i marine! 10 invertebrater, insekter og svampe. Til produktion af! chitosan anvendes sædvanligvis chitin fra krabber, rejer, 1 hummere osv.. De perleformede biokatalysatorer ifølge opfindelsen omfatter fortrinsvis chitosan med en molvægt fra ca. 50.000 til ca. 3.000.000.

Som polyvalente anioner er polyvalente uorganiske eller polyvalente organiske anioner egnede. De uorganiske anioner er fortrinsvis orthophosphat, metaphosphater, f.eks. hexametaphosphat, pyrophosphater, polyphosphater, 20 f.eks. tri-, tet'ra- eller octapolyphosphat, eller cyano-ferrater, f.eks. [Fe(CN)gJ4- eller [Fe(CN)g]^“· Især foretrækkes tripolyphosphat. De organiske anioner er fortrinsvis polymere organiske carboxylater, sulfonater eller, hydroxyforbindelser„ 25
Sotn kiselsyre egner sig fortrinsvis en fældet kiselsyre, især en fældet kiselsyre med en gennemsnitlig kornstørrelse i området fra. 5 til 50 μπι og en gennemsnitlig rumvægt i området fra 5 til 100 g pr. 100 ml, især fra ca. 20 30 til 50 g pr. 100 ml.

Undgåelse af skumdannelse ved fremstilling af suspensionen 10 af enzymatisk aktivt materiale, fældet kiselsyre i fast form, vandig pufferopløsning og en vandig opløsning af en kationisk polyelektrolyt er af afgørende betydning, da der ellers sotn følge opstår katalysatorperler med dårligere^ mekanisk® egenskaber. Fortrinsvis undgås skumdannelse ved, 15 at blandingen af de forskellige komponenter sker i nærværelse af kiselsyren. Ellers er den rækkefølge, i hvil-; ken stofferne bringes sammen, ligegyldig. Fortrinsvis' blandes det enzymatiske materiale først med kiselsyren i fast form, først derpå optages i vandig pufferopløsning, 20 og derefter blandes med opløsningen af den kationiske polyelektrolyt.
Ved fremstillingen ifølge opfindelsen af biokatalysator-25 perler anvendes enzymatisk materiale som beskrevet ovenfor, dvs. hele celler, cellefragmenter eller celleorga-neller af en mikroorganisme eller enzymer eller en kombination deraf. Hele celler kan derved foreligge levende, døde eller eksempelvis lyofiliserede eller dehydrerede.
30 Dyrkningen af celler i et næringsmedium, der indeholder alle til opretholdelse af cellestofskiftet og til formeringen nødvendige organiske og uorganiske bestanddele (C-kilde, N-kilde, sporelementer, vækststoffer osv.), og' cellefragmenteringen til fremstilling af cellefragmenter 35 eller isolering af celleorganellerne sker ved i og for sig kendte metoder. Som vandig pufferopløsning egner sig eksempelvis biole -15 giska puffere i et pH-interval fra pH 3 til 7, fortrins -vis ca. 5, f.eks. phosphat-, citrat- eller acetatpuffer.
De kationiske polyelektrolytter, der kan anvendes v=c fremgangsmåden ifølge opfindelsen, er beskrevet i dsi 20 foregående. Chitosan foretrækkes. Chitosan må til arvendelse til fremstilling af biokatalysatorperlerne før:= t bringes i vandig opløsning for at udgøre integrerende bestanddel af immobilisatet. Dertil optages chitosan, der i ikke-protoniseret: form er vanduopløselig, i fortyndere 25 vandige, især organiske, syrer, eksempelvis i myresyre, eddikesyre, pyrodruesyre, æblesyre, citronsyre eg lignende, fortrinsvis i eddikesyre, ved en pH-værdi i , fortrinsvis pH 4 til 5,5, og opvarmes eventuelt til ce .
60°C for at støtte opløsningsprocessen. Chitosanopløs-30 ningen filtreres derefter til fjernelse af uopløselice bestanddele. De anvendte stoffer anvendes fortrinsvis således, at vægtforholdt mellem kiselsyre og kationisk polyelektrolyt ligger i et område fra 1,5-50:1, især ved ca. 15:1. Slut-koncentrationen af kiselsyre i den ovenfor beskrevne 5 suspension ligger i et område fra 2 til 15 vægtprocent, f.eks. 10%. Foretrukne slutkoncentrationer af kationisk polyelektrolyt i den ovenfor beskevne suspen- sion er 0,3 til 1,5 vægtprocent, især ca. 0,7%.
10 Den kiselsyre, der anvendes til fremstilling af biokatalysatorperlerne ifølge opfindelsen, er beskevet ovenfor.
Den anvendes i fast form.
Ved tværbindingsbadet drejer det sig om en vandig opløs-15 ning af en polyvalent anion. De mulige anioner er beskrevet i det foregående. Dé^anvendes til tværbindingsbadet" i form af syrer eller som salte, f.eks. som alkalimetalsalte, eksempelvis natrium- eller kaliumsalte, endvidere også som ammoniumsalte. En vandig opløsning fore- 20 trækkes, til hvilken tripolyphosphorsyre eller et salt deraf, f.eks. natrium- eller kaliumtripolyphosphat, anvendes. I tværbindingsbadet foreligger de nævnte forbindelser eksempelvis i en koncentration i området fra 0,5 til 10% (v/r), fortrinsvis fra 1,5 til 3% (v/r).
25 Tværbindingsopløsningens pH-værdi ligger i et område fra pH 5 til 10, fortrinsvis på ca. pH 8.
Suspensionen af enzymatisk aktivt materiale, kiselsyre, pufferopløsning og polykation dryppes under omrøring til 30 tværbindingsopløsningen, hensigtsmæssigt således, at tværbindingsbadets rumfang udgør mindst det tredobbelte af suspensionens rumfang. Den dråbevise tilsætning sker under anvendelse af en dyse med en lille åbning, eksempelvis en injektionssprøjte, en sprøjtelignende injek-35 tionsindretning, en porøs plade eller et lignende egnet middel. Dråberne blæses eventuelt af dysens spids under DK 17:ϊ712 B1 anvendelse af komprimeret luft, komprimeret nitrogen eller lignende. Suspensionen kan også indsprøjtes i tværbindingsopløsningen, eksempelvis under anvendelse af ni; atomiseringsappetrat, fortrinsvis et trykatomisering: s-apparat.
De ved den ovenfor beskrevne fremgangsmåde fremstilletU 5 biokatalysatorperler forbliver mindst ca. en time, f.eks. ! natten over i tværbindingsbadet til indskrumpning og hærd- ! ning. Eventuelt skilles de fra tværbindingsbadet vue. gængse, til adskillelse af faste og flydende faser egneik metoder, såsom dekantering, filtrering osv., og underkas- 10 tes før den yderligere anvendelse en fysiologisk vaskeproces, eksempelvis med fysiologisk natriumchloridopløsning.

Fremgangsmåder til fremstilling af a-hydroxycarboxylsyrer ud fra cx-ketocarboxylsyrer ved hjælp af biokatalysatorer ifølge opfindelsen, hvis enzymatisk aktive materiale indeholder en oxidoreduktase, er en oxidoreduktase eller 5 består af hele celler af en bakterie af slægten Proteus, især arterne Proteus vulgaris og/eller Proteus mirabilis, fortrinsvis Proteus vulgaris, foretrækkes. De redu-15 cerede nukleotider opstår atter f.eks. ved elektronoverførsel over naturlige eller syntetiske elektronmediatorer med egnet redoxpotentiale, såsom ferredoxin eller bipyri-dyliumderivater, f.eks. 4,4'-bipyridyliumderivater (Violo-gen) eller 2,2'-bipyridyliumderivater (eksempelvis diquat-20 kationer, såsom diquat-dibromid eller diquat-dichlorid).
Der kendes også finale reduktaser, der kan optage elektronerne direkte fra mediatorerne.
25 Især foretrækkes en fremgangsmåde til fremstilling af 2-hydroxy-4-phenylsmørsyre, fortrinsvis af 2-(R)-hydroxy- 4-phenylsmørsyre, ud fra 2-oxo-4-phenylsmørsyre ved hjælp af biokatalysatorerne ifølge opfindelsen, hvis enzymatisk aktive materiale indeholder en oxidoreduktase eller er en 30 oxidoreduktase eller består af hele celler af en bakterie af slægten Proteus, især af arterne Proteus vulgaris og/-eller Proteus mirabilis, fortrinsvis Proteus vulgaris.
2-(R)-hydroxy-4-phenylsmørsyre er et værdifuldt mellemprodukt ved fremstillingen af ACE, angiotensinomdannende 35 enzym-("angiotensin converting enzyme")-inhibitorer eller deres forstadier. Dennes klasse aktive stoffer har i de 14 DK 1 7 Ϊ712 B1 forløbne år fundet voksende interesse:. Den udvider de t:.l rådighed stående antihypertensivas potentiale og dermed de terapeutiske muligheder ved bekæmpelsen af højt blodtryk.
Af særlig interesse er derved fremstillingen af ACE-ha;n-5 meren 3-{[ l-ethoxycarbonyl-3-phenyl-(lS)-propyl ]amino'>- 2,3,4,5-tetrahydro-2-oxo-lH-l-(3S)-ben:iazepin-l-eddike-syre-monohydrochlorid (= Benazepril).
Opfindelsen angår især en fremgangsmåde til fremstilliiu'j 10 af 2-(R)-hydroxy-4-phenylsmørsyre ud fra 2-oxo-4-pheny L··' smørsyre, som er ejendommelig ved, at perleforme· kil biokatalysatorer ifølge opfindelsen, hvis enzymati sk: aktive materiale består af hele celler af en bakterie r.:\ slægten Proteus, især af arterne Proteus vulgaris og/eller.· 15 Proteus mirabilis, fortrinsvis Proteus vulgaris, anvend i en fastleje- eller hvirvellagsreaktor, fortrinsvis i sn! fastlejereaktor, gennem hvilken der kontinuerligt ledes en vandig opløsning af substratet 2-oxo-4-phenylsmørsyrϊ eksempelvis i en koncentration i området fra 50 til 2)0 20 mM, af formiat, eksempelvis et alkalimetalformiat, såson kalium- eller natriumformiat, eksempelvis i en koncentration i området fra 100 til 500 mM, og en elektronirid-diator, f.eks. et 4,4'-bipyridyliumderivat, såson methylviologen, carbamoylmethylviologen eller benzylvio-25 logen, eller et 2,2'-bipyridyliumderivat, såsom diquatd:-kation, eksempelvis i en koncentration i området fra 0, ϊ til 10 mM. Reduktionen af substratet katalyseres eksempelvis af den i Proteus vulgaris indeholdte 2-oxocarboxy:-syrereduktase (2-hydroxycarboxylat-viologen-oxidoreduk-30 tase). Da dette enzym har en høj enantiomerselektivite:, og produktet opstår med et enantiomeroverskud på mere ene 99,8% ligger der en særlig fordel ved anvendelse af d<d ved fremgangsmåden ifølge opfindelsen fremstillede produki i, at der ved syntesen af ACE-hæmmere, som forløber ovci 35 talrige trin, allerede i et relativt tidligt stadium ni DK 173712 B1 15 syntesen kan anvendes en enantiomerren syntese. Fremstilling af perleformede biokatalysatorer med Proteus 20 vulgaris.
Proteus vulgaris (ATCC 9484) dyrkes i et fuldstændigt medium (gærekstrakt 5 g/1, glucose-monohydrat 5 g/1, K2HPO4 5 g/1, pepton fra kød 20 g/1, pH 7,0) i 24 timer ved 37eC 25 under let gasning med nitrogen. De således fremkomne celler høstes ved 12°C i en CEPA-centrifuge type Z 41 G (gennemløb 300 1/time) under skyldning med nitrogen ved et omdrejningstal på 20.000 rpm (16.950 g). Ud fra 400 1 kulturvæske fås 580 g bakteriesediment med et fugtigheds-30 indhold på ca. 80%. 1 Vægtdel fugtig bakteriemasse op- slæmmes sammen med 1 vægtdel kiselsyre (Baker produkt nr.

Eksempel 3
Fremstilling af_2-(R)-hydroxy-4-phenylsmørsyre ved hja1p 10 af perleformede biokatalysatorer i 1aboratoriem&lestok 48 ml (rysterumfang) ,,10%'s kiselsyre-biokatalysatoi-perler”, der fremstilles ifølge eksempel 1 af 60 g suspension, skilles fra natriumtripolyphosphat-tværbindinys - .
15 badet og sættes til 200 ml af en med nitrogen forud g;s-behandlet opløsning af 50 mM (1,84% v/r) natriumtripcty -phosphatpuffer med pH 7,0, 100 mM kaliumformiat og 1 nM
carbamoylmethylviologen (1,1' -dicarbamoylmethyl-4,4' - <l: -pyridiniumdikation). I denne reduktionsopløsning f <>i -20 bliver perlerne under vakuum til afgasning indtil fuldstændig violetfarvning, der beror på reduktion af cart-amoylmethylviologenet ved den i Proteus vulgaris indeholdte formiatdehydrogenase. Derefter anbringes perlerne i en enzymfastlejereaktor, dvs. i en med kappe forsynet 25 glassøjle (indvendig diameter 1,6 cm, lejehøjde 24 mi, 250Ο, Den med nitrogen forud gasbehandlede substratopløsning (50 mM natriumtripolyphosphatpuffer pH 6,7, 1C0

Forløbet af reduktionen af 2-oxo-4-phenylsmørsyie til 2-(R)-hydroxy-4-phenylsmørsyre, der katalyseres af (Un i Proteus vulgaris indeholdte 2-oxocarboxylsyrereduktaie 35 (2-hydroxycarboxylat-viologén-oxidoreduktase), måles '^d HPLC-analyse (søjle: Nucleosil C18, partikkelstørrels« 5 19 DK 173712 B1 μΐη, længde 12,5 cm, indvendig diameter 4,6 mm; gennemløb 1,0 ml/min.; løbemiddel: 3 rumfangsdele acetonitril/8 rumfangsdele 100 mM kaliumphosphatpuffer pH 3,0).
5 På tilsvarende måde anvendes "kontrol-biokatalysator-perlerne" (rysterumfang 49 ml ud fra 60 g suspension ifølge eksempel 1) til reduktion af 2-oxo-4-phenylsmør-syre. Derved er det nødvendigt at indskrænke perlelejet ' 10 med søjleadapteren, fordi perlerne overvejende svømmer j ovenpå, og der derfor dannes tomme zoner i søjlen.
Resultaterne af produktivitetsbestemmelsen er sammenfattet i tabel 2.

Af tabel 2 fremgår, at "kontrol-biokatalysatorperlen" har en klart ringere produktivitet end de "10%'s kiselsyre-biokatalysatorperler".

Fremstillingen af de "10%’s kiselsyre-biokatalysator- .
15 perler" sker ud fra 573 g bakteriesediment ifølge eksesi-pel 1, idet der dog i stedet for sprøjten anvendes ir-indretning bestående af en tandhjulspumpe (gennemløb 1,4 l/time; tryk ca. 0,5 bar) og en 7-strålet douche med 0,6 mm kapillærer til dråbedannelsen. Til forøgelse af afdryp-20 ningsfrekvensen eller til påvirkning af dråbestørrelser har hvert kapillær en separat luftudstrømningskanal. Efter indskrumpning og hærdning i en 3%'s (v/r) natriumtripoly-phosphatopløsning fås ud fra 573 g bakteriesedikent c; .

Den med nitrogen forud gasbehandlede substratopløsning (100 mM natriumtripolyphosphatpuffer pH 6,7, 112,2 mM 2-oxo-4-phenylsmørsyre, 300 mM kaliumf ormiat, 1 mM carbamoylmethylviologen) sterilfiltreres og ledes kontinu-5 erligt gennem søjlen ved hjælp af en doseringspumpe med overtryk 2,0-3,0 bar med et gennemløb på først 2,5 1/time.
Den ved HPLC målte omsætning er >99,5%. Søjlen drives dag og nat uden afbrydelse ved stuetemperatur, dvs. ved ca. 23‘ til 26,5°C.

Omsætningen kontrolleres ved daglige HPLC-analyser og holdes ved omsætningsværdier >99,5% ved regulering af pumpehastigheden. Efter 25 dage er i alt ca. 800 l reak-; tionsopløsning omsat (gennemsnitlig omsætning 99,6%), og' 15 gennemløbet udgør endnu 1 1/time.
1160 1 omsat reaktionsopløsning blandes med 250 1 ethyl-acetat og indstilles på pH 2,6 med 114 1 42,5%'s ortho-phosphorsyre. 2-(R)-hydroxy-4-phenylsmørsyren ekstraheres 2^ i ethylacetat med et udbytte på 94%. Den i vandfasen resterende 2-(R)-hydroxy-4-phenylsmørsyre ekstraheres endnu to gange med hver 120 1 ethylacetat. Efter de tre ekstraktionstrin er ekstraktionen af 2-(R)-hydroxy-4-phenylsmør-syre fuldstændig (99,8%). Efter samling af de tre ethyl-25 acetatfaser (samlet rumfang 450 1), tilsætning af 250 1 ethylacetat og afdestillering af 470 1 af opløsningsmidlet klarfiltreres opløsningen gennem et enpladefilter. 50 1 ethylacetat anvendes til skyldning af filteret og sættes til den klare opløsning. Efter afdestillering af 200 1 af 30 opløsningsmidlet udgør opløsningens slutrumfang 80 1. Til! denne opløsning sættes 400 1 cyclohexan, og 200 1 af opløsningsmidlet afdestilleres. Efter centrifugering af det udkrystalliserede produkt og tørring i vakuum ved 20-25*0 indtil konstant vægt fås 23,4 kg krystallinsk

Til kontrol af enantiomerrenheden opløses en prøve af den krystallinske syre i absolut ethanol og bringes til omsætning med hydrogenchloridgas i 24 timer ved stuetemperatur. Efter afdestillering af alkoholen og kort afgasning 1 5 højvakuum resterer en lysegul olie, der analyseres ve i HPLC ved 25*C/32 bar på en chiral søjle (250 x 4,6 tom i.d., gennemløb 1 ml/min., stationær fase chiralcel OD [Stehelin, Basel] type OD-5-15-2092,5, mobil fase: 90fr hexan/10% isopropanol/0,1% diethylamln). De stoffer, der; 1° skal analyseres, foreligger i en koncentration på 1 mg/ml i løbemidlet (indsprøjtet mængde 10 //1). Scanningen sker ved en bølgelængde på 210 nm, bedømmelsen ved areaisam-menligning med ekstern standard. Den bestemte ee-værdi er >99,8%.

Claims (10)
1. Perleformet biokatalysator med høj mekanisk styrke omfattende enzymatisk aktivt materiale, en kationisk 5 polyelektrolyt og polyvalente anioner, kendetegnet ved, at biokatalysatoren indeholder kiselsyre og fremstilles ud fra en suspension af det enzymatisk aktive materiale, kiselsyren, pufferopløsning og polyelektrolytten, hvori slutkoncentrationen af kiselsyre ligger i et interval 10 mellem 2 og 15 vaegt-%.
2. Perleformet biokatalysator ifølge krav 1, kendetegnet ved, at det enzymatisk aktive materiale består af . hele celler af en bakterie eller af en gærart. 15
