Claims (17)
1. Isoleret proteinkompleks i form af et syntetisk kimært protein, der omfatter en aminosyresekvens for: (i) keratinocytvækstfaktor (KGF) eller mindst et domæne af KGF, som er i stand til at binde til en keratinocytvækstfaktorreceptor; og (ii) et eller flere domæner af vitronectin (VN), som omfatter mindst et integrinbindende domæne af VN.
2. Isoleret proteinkompleks ifølge krav 1, hvor det ene eller de flere domæner af VN, der omfatter mindst et integrinbindende domæne af VN, ikke inkluderer et C-terminalt heparinbindende domæne (HBD).
3. Isoleret proteinkompleks ifølge krav 2, der omfatter aminosyrerne 1-346 af en moden VN-sekvens som vist i SEQ ID NO: 2 .
4. Isoleret proteinkompleks ifølge et hvilket som helst af kravene 1-3, hvor det integrinbindende domæne er et cxv- integrinbindende domæne, såsom et oo^3-integrinbindende domæne eller et oo^s-integrinbindende domæne.
5. Isoleret proteinkompleks ifølge krav 1, hvor det ene eller de flere domæner af VN, der omfatter mindst et integrinbindende domæne af VN, omfatter aminosyrerne 1-311 af en moden VN-sekvens som vist i SEQ ID NO: 2; eller aminosyrerne 1-125 af en moden VN-sekvens som vist i SEQ ID NO: 2; eller aminosyrerne 1-64 af en moden VN-sekvens som vist i SEQ ID NO: 2; eller aminosyrerne 1-52 af en moden VN-sekvens som vist i SEQ ID NO: 2.
6. Isoleret proteinkompleks ifølge krav 5, hvor det ene eller de flere domæner af VN, der omfatter mindst et integrinbindende domæne af VN, ikke omfatter en polyanionisk aminosyresekvens, der svarer til resterne 53-64 af et modent VN-protein som vist i SEQ ID NO: 2.
7. Isoleret proteinkompleks ifølge et hvilket som helst af ovennævnte krav, der yderligere omfatter et ekstra fragment af VN.
8. Isoleret proteinkompleks ifølge et hvilket som helst af ovennævnte krav, der yderligere omfatter mindst én linkersekvens, som eventuelt kan omfatte et proteasespaltningssted og eventuelt er udvalgt fra gruppen, der består af: (i) Gly4Ser (SEQ ID NO: 4); (ii) Gly4Ser3 (SEQ ID NO: 5) ; (iii) (Gly4Ser)3 (SEQ ID NO: 6); (iv) (Gly4Ser)4 (SEQ ID NO: 7); (v) Leu Ile Lys Met Lys Pro (SEQ ID NO: 8); og (vi) Gin Pro Gin Gly Leu Ala Lys (SEQ ID NO: 9).
9. Isoleret proteinkompleks ifølge krav 1, hvor det syntetiske kimære protein omfatter en aminosyresekvens: VNi_459 : (Gly4Ser) 4: KGFi_i63 : Gly4SerGly4His6 som vist i SEQ ID NO: 10; VNi-3h : (Gly4Ser) 4: KGFi_i63 : Gly4SerGly4His6 som vist i SEQ ID NO: 11; VNi-125 : (Gly4Ser) 4: KGFi_i63 : Gly4SerGly4His6 som vist i SEQ ID NO: 12; VNi_64 : (Gly4Ser) 4: KGFi_i63 : Gly4SerGly4His6 som vist i SEQ ID NO: 13; VNi-i64: (Gly4Ser) 4: VN343-376 : (Gly4Ser) 4: KGFi_i63 : Gly4SerGly4His6 som vist i SEQ ID NO: 14 .
10. Isoleret nukleinsyre, der koder for det isolerede proteinkompleks ifølge et hvilket som helst af ovennævnte krav.
11. Genkonstruktion, der omfatter den isolerede nukleinsyre ifølge krav 10, der er operabelt koblet til en eller flere regulatoriske nukleotidsekvenser i en vektor, hvor den regulatoriske nukleotidsekvens kan være en promotor.

13. Farmaceutisk sammensætning, der omfatter det isolerede proteinkompleks ifølge et hvilket som helst af kravene 1-9 og et farmaceutisk acceptabelt bæremateriale, fortyndingsmiddel eller excipiens.
14. Sår- eller forbrændingsbandage, kirurgisk implantat, gittersystem eller protese, der er imprægneret med, beklædt med eller på anden vis omfatter det isolerede proteinkompleks ifølge et hvilket som helst af kravene 1-9.
15. Isoleret proteinkompleks ifølge et hvilket som helst af kravene 1-9 til anvendelse til fremme af cellemigration og/eller proliferation, hvor komplekset binder til både en keratinocytvækstfaktorreceptor og en integrinreceptor, der udtrykkes af en celle, til derved inducering, forøgelse eller på anden vis fremme af migration og/eller proliferation af cellen.
16. Isoleret proteinkompleks ifølge et hvilket som helst af kravene 1-9 til anvendelse til fremme af sårheling in situ hos et dyr, hvor det isolerede proteinkompleks administreres til et dyr, såsom et menneske, til binding til både en keratinocytvækstfaktorreceptor og en integrinreceptor, der udtrykkes af en celle, til derved fremme af cellemigration og/eller proliferation in situ, for eksempel til profylaktisk eller terapeutisk inducering, forøgelse eller på anden vis fremme af epitelcellemigration og/eller proliferation.
17. Isoleret proteinkompleks til anvendelse ifølge krav 16, hvor det isolerede proteinkompleks administreres til en eller flere celler eller væv in vitro.
