Claims (10)

1. Monoklonalt anti-synuclein-antistof 1H7, eller humaniseret eller kimært 1H7, hvor antistoffet 1H7 er produceret af hybridom JH17.1 H7.4.24.34 deponeret under ATCC-accessionsnummer PTA-8220.
2. Monoklonalt anti-synuclein-antistof ifølge krav 1, hvilket antistof er humaniseret 1H7 produceret ved podning af CDR’er af 1H7 på humane framework- og konstante regioner, hvor udvalgte framework-rester fra den humane variable region eventuelt er substitueret, som følger: en udvalgt framework-rest fra den humane variable region er substitueret med en tilsvarende framework-aminosyre fra 1 H7-museantistoffet, når aminosyren (1) ikke-kovalent binder antigen direkte, (2) støder op til en CDR-region, (3) interagerer på anden måde med en CDR-region eller (4) deltager i VL-VH-interfacet; eller en human framework-aminosyre, der er usædvanlig for et humant immunglobulin ved denne position, er substitueret med en aminosyre fra den tilsvarende position af musedonorantistoffet eller fra den tilsvarende position af et mere typisk humant immunglobulin.
3. Fremgangsmåde til humanisering af monoklonalt antistof 1H7 produceret af hybridomet deponeret under ATCC-accessionsnummer PTA-8220, hvilken fremgangsmåde omfatter: bestemmelse af aminosyresekvensen fra CDR-regioner af det monoklonale antistof; udvælgelse af et acceptorantistof og fremstilling af et humaniseret antistof omfattende CDR’erne fra det monoklonale antistof og framework fra den variable region af acceptorantistoffet.
4. Fremgangsmåde til fremstilling af en kimær form af monoklonalt antistof 1H7 produceret af hybridomet deponeret under ATCC-accessionsnummer PTA-8220, hvilken fremgangsmåde omfatter: bestemmelse af aminosyresekvensen fra de variable let- og tungkæde-regioner af det monoklonale antistof; udvælgelse af en konstant tung- og letkæde-region; fremstilling af et kimært antistof opfattende en letkæde omfattende den variable letkæde-region kondenseret til den konstante letkæde-region, og en tungkæde omfattende den variable tungkæde-region kondenseret til den konstante tungkæde-region.
5. Antistof ifølge krav 1 eller krav 2 til anvendelse i profylakse eller behandling af en sygdom kendetegnet ved Lewy-legemer eller alpha-synuclein aggregering i hjernen.
6. Antistof ifølge krav 5 til anvendelse ifølge dette krav, hvor sygdommen er Parkinsons sygdom.
7. Antistof ifølge krav 5 eller krav 6 til anvendelse ifølge dette krav, hvor antistoffet er af human lgG1-isotype.
8. Antistof ifølge et hvilket som helst af kravene 5 til 7 til anvendelse ifølge dette krav, hvor antistoffet administreres i talrige doseringer over mindst seks måneder.
9. Antistof ifølge et hvilket som helst af kravene 5 til 7 til anvendelse ifølge dette krav, hvor antistoffet administreres ad en perifer vej.
10. Celle af hybridom JH17.1H7.4.24.34 deponeret under ATCC-accessionsnummer PTA-8220.
