Claims (9)
1. In vitro-fremgangsmåde til screening for et stof, der påvirker gelatinase-medieret Eph-receptor-A4-spaltning, som omfatter trinene: (a) etablering af kontakt mellem en første biologisk sammensætning, der indeholder gelatinase, og en anden biologisk sammensætning, der indeholder EphA4, ved tilstedeværelse og fravær af et kandidatstof; (b) måling af tilstedeværelse eller mængden af EphA4-ektodomæne- og/eller endodomænefragmentet; og (c) selektion af kandidatstoffet som et stof, der påvirker gelatinase-medieret EphA4-spaltning, hvis resultaterne i trin (b) målt i nærvær af kandidatstoffet er ændret i forhold til resultaterne i trin (b) målt i fravær af kandidatstoffet.
2. Fremgangsmåde ifølge krav 1, hvor trin (c) omfatter identificering af kandidatstoffet som et stof, der fremmer gelatinase-medieret EphA4-spaltning, hvis EphA4-endodomæne-og/eller ektodomænefragmentet målt i nærvær af kandidatstoffet i trin (b) er forøget sammenlignet med EphA4-endodomæne-og/eller ektodomænefragmentet målt i fravær af kandidatstoffet i trin (b).
3. Fremgangsmåde ifølge krav 1, hvor trin (c) omfatter identificering af kandidatstoffet som et stof, der hæmmer gelatinase-medieret EphA4-spaltning, hvis EphA4-endodomæne-og/eller ektodomænefragmentet målt i nærvær af kandidatstoffet i trin (b) er reduceret sammenlignet med EphA4-endodomæne-og/eller ektodomænefragmentet målt i fravær af kandidatstoffet i trin (b).
4. Fremgangsmåde ifølge krav 2, hvor stoffet, der fremmer gelatinase-medieret EphA4-spaltning, yderligere evalueres som et stof, der fremmer γ-secretase-medieret EphA4-spaltningsreaktion.
5. Fremgangsmåde ifølge krav 3, hvor stoffet, der hæmmer gelatinase-medieret EphA4-spaltning, yderligere evalueres som et stof, der hæmmer γ-secretase-medieret EphA4-spaltningsreaktion.
6. Fremgangsmåde ifølge krav 2, hvor stoffet, der fremmer gelatinase-medieret EphA4-spaltning, yderligere evalueres som et stof, der fremmer spina-dannelsesreaktion medieret af det intracellulære EphA4-fragment.
7. Fremgangsmåde ifølge krav 3, hvor stoffet, der hæmmer gelatinase-medieret EphA4-spaltning, yderligere evalueres som et stof, der hæmmer spina-dannelsesreaktion medieret af det intracellulære EphA4-fragment.
8. Anvendelse af gelatinase til spaltning af ektodomænet af EphA4.
9. Anvendelse af et analysekit, der omfatter en første biologisk sammensætning, der indeholder gelatinase, og en anden biologisk sammensætning, der indeholder EphA4, til måling af gelatinase-medieret EphA4-spaltning.
