Abstract
El invento se refiere a un procedimiento para purificar y aislar un nucleósido enriquecido en el anómero beta que comprende: a) proporcionar una mezcla que contiene R¿¿ y un nuclésido enriquecido en el anómero beta, de la fórmula (gráfico), en la que cada X se selecciona independientemente entre grupos protectores de hidroxi y R¿es un núcleo base de la fórmula (gráfico), en la que W es un grupo protector de amino; y R¿¿ es un núcleo base de la fórmula (gráfico), en la que W es un protecyor de amino o hidrógeno; en un disolvente de alta ebullioción; b) diluir la mezcla con un disolvente orgánico seleccionado entre el grupo compuesto por éteres, esteres y nitrilos; c) añadir la mezcla de reacción diluida en ácido acuoso; y d) mantener la mezcla ácida asi preparada a una temperatura de 70oC a 100oC hasta que haya precipitado el producto de fórmula IB, en el que W es ahora W.The invention relates to a process for purifying and isolating a nucleoside enriched in the beta anomer comprising: a) providing a mixture containing R¿¿ and a nucleoside enriched in the beta anomer, of the formula (graph), in which each X is independently selected from hydroxy protecting groups and R¿ is a base nucleus of the formula (graph), wherein W is an amino protecting group; and R¿¿ is a base nucleus of the formula (graph), in which W is a protecyor of amino or hydrogen; in a high boiling solvent; b) diluting the mixture with an organic solvent selected from the group consisting of ethers, esters and nitriles; c) adding the reaction mixture diluted in aqueous acid; and d) maintaining the acidic mixture thus prepared at a temperature of 70oC to 100oC until the product of formula IB, where W is now W., has precipitated.
