Abstract
Se explica un método altamente eficiente para la generación de anticuerpos humanos, en particular, que sean especificos para ser proteina de fusión RSV que combina células de bazo humano in vitro primario y la carga de antígenos en ratones SCID. Este método proporciona concentraciones muy altas de anticuerpo humano con predominancia del isotipo igG que contiene anticuerpos de alta especificidad y afinidad con los antígenos deseados. Este método es muy adecuado para la generación de anticuerpos monoclonales humanos para aplicaciones diagnósticos y terapéuticas asi como para la recuperación de células humanas para la generación de bibliotecas genéticas combinacionales de anticuerpos humanos. Se explican dos anticuerpos monoclonales humanos, RF -1 y RF-2, cada uno de los cuales posee una afinidad para la proteína RSV de >2 X10 -molar, asi como sus correspondientes amino ácidos y secuencias de ADN. Estos anticuerpos van a ser usados terapéutica y profilácticamente para el tratamientoo prevención de la infección RSV, asi como para la dignosis de RSV en analitos.A highly efficient method for the generation of human antibodies is explained, in particular, which are specific to be RSV fusion protein that combines human spleen cells in primary in vitro and antigen loading in SCID mice. This method provides very high concentrations of human antibody with a predominance of the igG isotype that contains antibodies of high specificity and affinity with the desired antigens. This method is very suitable for the generation of human monoclonal antibodies for diagnostic and therapeutic applications as well as for the recovery of human cells for the generation of combinational genetic libraries of human antibodies. Two human monoclonal antibodies, RF-1 and RF-2, each possessing an affinity for the RSV protein of> 2 X 10 -molar, as well as their corresponding amino acids and DNA sequences, are explained. These antibodies are to be used therapeutically and prophylactically for the treatment or prevention of RSV infection, as well as for RSV dignosis in analytes.
