Definitions
- the invention relates to a device and a method for the continuous and reversible measurement of the concentration of a chemical species with the aid of a fluorescence-optical indicator substance.
- the object of the invention is to avoid the disadvantages mentioned and to provide a measuring device or a measuring method. propose, which combines the advantages of the decay time measurement with the technical advantages of the intensity measurement when using fluorescence-optical indicator substances.
- the indicator substance is excited with polarized excitation radiation of a wavelength corresponding to the absorption of the indicator substance, that the intensity of the fluorescent radiation of the indicator substance is measured in two mutually normal polarization directions, that from the ratio of the intensities obtained the fluorescence anisotropy of the radiation is measured and the concentration of the chemical species is determined therefrom with the aid of a previously determined calibration curve.
- the depolarization of the fluorescent radiation of an indicator which is dependent on the concentration of the chemical species, is used for the measurement.
- Fluorescence depolarization is to be understood as the fact that after excitation of a molecule with radiation of a certain polarization direction, the. emitted fluorescence also contains radiation of the normal polarization component. The reason for this is the thermal movement of the molecules, which leads to the spatial direction of the excited molecular dipole changing during the decay time ⁇ of the excited state.
- the fluorescence anisotropy r is defined as
- the fluorescence anisotropy r has the following dependence on the decay time r of the excited state:
- a prerequisite for the use of this measurement method is the use of media with low light scatter as a matrix for the indicator, since light scatter falsifies the anisotropy values.
- a variant of the measuring method according to the invention therefore provides that a fluorescence indicator is used to measure the concentration of the chemical species, the fluorescence of which is quenched dynamically by the species to be measured, which shortens the fluorescence decay time T of the dye and the fluorescence anisotropy r according to equation (4 ) is reduced.
- Efficient dynamic quenching of the fluorescence of organic fluorophores occurs when their fluorescence decay time is long (to> 100 ns). In order to achieve anisotropy sensitivity that is sufficient for sensor purposes, the time required for the rotation of the molecule must not be shorter.
- V volume of the indicator molecules (cultivation of large side groups) or by a high microviscosity in the environment of the indicator.
- Dynamic quenching takes place, for example, through oxygen, various anesthetic gases (eg halothane), halogenated hydrocarbons, halide and pseudohalide ions, sulfur dioxide, etc.
- f (pH) means a function to be determined empirically. With this relationship there is a pH dependence of the anisotropy
- a further embodiment of the invention therefore provides that a fluorescence indicator is used to measure the concentration of hydronium ions (pH measurement), the fluorescence decay time T of which is influenced by the concentration of the hydronium ions (by the pH value), as a result of which the luminescence anisotropy r changes according to equation (6).
- a fluorescent dye with a pH-dependent fluorescence decay time serves as an indicator, for example acridine, salicylic aldehyde, indole-3-acetic acid, 9 (10H) -acridinone, 2-naphthol, etc.
- a matrix material of low viscosity for example a hydrogel.
- the measuring method according to the invention is also suitable for determining the concentration of, for example, potassium, sodium, lithium.
- Indicator molecules to which the ions in question bind with high selectivity for example crown ether derivatives
- the hydrodynamic volume of the indicator molecule changes as a result of the binding. Depending on the prevailing ion concentration, different proportions of the total number of indicator molecules present are present. If the hydrodynamic volume of the unbound indicator is designated as V 0 and that of the bound indicator as V j -, then the effective hydrodynamic volume applies
- Another embodiment of the invention provides that a fluorescence indicator is used to measure the concentration of cations, the hydrodynamic volume V of which changes with the binding of the cations to be measured, as a result of which the fluorescence anisotropy r changes in accordance with equation (8).
- the anisotropy depends not only on the properties of the indicator (fluorescence decay time, hydrodynamic molecular volume), but also on the microviscosity n of the surrounding matrix. This fact can be used e.g. to measure the viscosity of cell membranes. The microviscosity of a given matrix can in turn depend on the concentration of a certain chemical species S. Then it applies
- n Q is the microviscosity in the absence of species S and f [S] is an empirically determined function. This then results for an indicator with a constant fluorescence decay time ⁇ 0
- a fluorescence indicator present in a matrix substance can be used to measure the concentration of a chemical species in a sample, the fluorescence decay time of which is not influenced by the chemical species, but whose matrix substance depends on the concentration of the chemical species Micro viscosity n changes, whereby the fluorescence anisotropy r changes according to equation (10).
- One process that can change the viscosity n of a matrix is e.g. swelling by including e.g. a liquid.
- the specified measurement method is thus also usable for sensors of the kind which are of particular importance in environmental measurement technology.
- the chemical species can also be water.
- the indicator serves e.g. a hydrophobic dye molecule which does not interact with water to change the photophysical properties, e.g. a rhodamine.
- the matrix used is e.g. PVC with plasticizer, which is known to have a state of swelling that depends on the water content.
- a device for the continuous and reversible measurement of the concentration of a chemical species, with a fluorescent optical indicator substance that is at least indirectly in contact with the chemical species, which is possibly dissolved or immobilized in a matrix substance, with a radiation source for providing the excitation radiation, a measuring device for Detection of the fluorescence radiation as well as an evaluation device are provided for carrying out the measuring method according to the invention, in its simplest execution it is distinguished by the fact that a polarization filter is arranged between the radiation source and the indicator substance.
- the measuring device for detecting the fluorescence radiation has two detectors, which are connected upstream of normal polarization filters, and that the signal outputs of the two detectors are connected to the evaluation unit in which the calibration functions required for determining the concentration are stored.
- the measuring device prefferably has only one detector with an upstream polarization filter which can be switched into two positions which are normal to one another, the signal output of the detector being connected to the evaluation unit.
- Fig. 1 shows a device according to the invention
- Fig. 2 shows an embodiment of this.
- An advantageous embodiment of the invention is to be illustrated using the example of an oxygen sensor in FIG. 1.
- An indicator known per se for example an organic rudenium complex, pyrenebutyric acid or a polycyclic aromatic, is used.
- the indicator I is dissolved or immobilized in a transparent, non-scattering carrier material or matrix substance T of high viscosity (for example a polymer).
- the excitation takes place with a light source L of a suitable wavelength (for example light-emitting diode, laser or lamp with narrow-band filter).
- the excitation light is linearly polarized by a polarization filter P ⁇ .
- the fluorescence radiation is registered by two detectors D 1 # D (photodiodes) of the measuring device M, in front of which there are polarization filters P, P3, one of which is oriented in parallel P and the other normal P 3 to the direction of polarization of the excitation light.
- the oxygen concentration is determined by comparison with a stored calibration curve and displayed digitally. Any filters in the beam path for separating excitation and fluorescent radiation are designated F ⁇ to F 3 .
- the measuring device M has only one detector O j ⁇ with a polarization filter P 2 which can be switched to two positions that are normal to each other.
