Abstract
Es werden 14,16β-Ethano-15β,161-cyclo-14β-estra-1,3,5(10)-triene der allgemeinen Formel (I), worin R1 ein Wasserstoffatom, einen C¿1?- bis C9-Alkyl oder C1- bis C9-Acylrest, R?2¿ ein Wasserstoffatom, einen C¿1?- bis C9-Alkyl-, einen C2- bis C9-Alkenyl- oder Alkinylrest sowie R?3¿ eine Hydroxygruppe oder einen C¿1?- bis C9-Acyloxyrest bedeuten, sowie ein Verfahren zu ihrer Herstellung beschrieben. Die neuen Verbindungen besitzen hohe (perorale) Wirksamkeit und sind zur Herstellung von Arzneimitteln geeignet.
Description
14, 16β-ETHANO-15β , 161 -CYCLO- 14β-ESTRA- 1 , 3 , 5 ( 10 )-TRIENE
Die vorliegende Erfindung betrifft 14,16β-Ethano-15β,161-cyclo-14β-estra- 1,3,5(10)-triene der allgemeinen Formel I
worin
R1 ein Wasserstoffatom, einen C1- bis C9-Alkyl oder C1- bis C9-Acylrest,
R2 ein Wasserstoffatom, einen C1- bis C9-Alkyl-, einen C2- bis C9-Alkenyloder Alkinylrest sowie
R3 eine Hydroxygruppe oder einen C1- bis C9-Acyloxyrest
bedeuten,
ein Verfahren zu ihrer Herstellung, pharmazeutische Präparate, die diese Verbindungen enthalten sowie deren Verwendung zur Herstellung von Arzneimitteln.
R2 kann sich sowohl in der 17α- wie auch in der 17β-Position und dementsprechend R3 in der 17β- oder 17α-Position befinden.
Als Acylrest R1 und für den Acylrest in dem Acyloxyrest R3 kommen Reste von organischen Carbonsiuren mit 1-12 Kohlenstoffatomen infrage. Sie leiten sich ab von aliphatischen, cycloaliphatischen, aliphatisch-cycloaliphatischen, cycloaliphatisch-aliphatischen und aromatischen Monocarbonsäuren mit 1 bis 12 Kohlenstoffatomen. Die Anzahl der Kohlenstoffatome im Ring variiert von 3 bis 7. Bevorzugt werden für die Reste R1, R2 und R3 die Acylgruppen der Essig-, Propion-, Butter-, Isobutter-, Pivalin-, Capron-, Acryl-, Croton-, Heptyl-, Capryl-, Pelargon- , Decan-, Undecan-,
Dodecan-, 3-Cyclopentylpropion- und Benzoesäure.
Insbesondere sollen die Acylreste R1, R2 und der Acyloxyrest R3 von solchen Carbonsäuren abstammen, die 2 bis 8 Kohlenstoffatome aufweisen.
Die Acylgruppen R1, R2 und R3 können auch von Dicarbonsauren mit bis zu 6
Kohlenstoffatomen stammen; hier ist insbesondere an die Bernsteinsäure gedacht.
Ist R1 ein Alkylrest, ist vor allem an den Methylrest gedacht; auch dem
Ethyl-, Propyl- und Isopropylrest kommen besondere Bedeutung zu.
Als Cycloalkylrest ist der Cyclopentylrest für R1 bevorzugt.
Im Rahmen der vorliegenden Erfindung sind folgende Verbindungen hervorzuheben:
(161S)-14,16β-Ethano-15β,161-cyclo-14β-estra-1,3,5(10)-trien-3,17β-diol, (161S)-14,16β-Ethano-15β,161-cyclo-14β-estra-1,3,5(10)-trien-3,17α-diol, (161S)-17β-{Acetyloxy)-14,16β-ethano-15β,161-cyclo-14β-estra-1,3,5(10)- trien-3-ol,
(161S)-17β-(Benzoyloxy)-14,16β-ethano-15β,161-cyclo-14β-estra-1,3,5(10) trien-3-ol,
(161S)-17α-(Acetyloxy)-14,16β-ethano-15β,161-cyclo-14β-estra-1,3,5(10)- trien-3-ol,
(161S)-17α-(Benzoyloxy)-14,16β-ethano-15β,161-cyclo-14β-estra-1,3,5(10) trien-3-ol,
Die erfindungsgemäßen Verbindungen der allgemeinen Formel I weisen eine starke Affinität zum Estrogenrezeptor auf und sie besitzen auch nach peroraler Applikation hohe estrogene Wirksamkeit.
Als Verbindungen mit hoher estrogener Wirksamkeit sind beispielsweise die natürlichen Estrogene Estradiol und Estriol (E. Schröder, C. Rufer und R. Schmiechen, Pharmazeutische Chemie, 1982, Georg Thieme Verlag, StuttgartNew York, S. 568 ff) bekannt. Sie sind aber metabolisch nicht stabil und werden nach oraler Applikation durch Oxidation der 17-Hydroxygruppe zum entsprechenden, unwirksameren Estronderivat abgebaut.
Durch Einführung bspw. einer Ethinylgruppe am 17-C-Atom (Ethinylestradiol, loc. cit., S. 574) kann die Oxidation der 17-Hydroxygruppe verhindert werden und die entsprechenden Derivate verfügen demzufolge über hohe estrogene Wirksamkeit nach peroraler Applikation.
Erst in jüngerer Zeit ist es gelungen, nicht durch Variationen der Substituenten am Steroidgerüst sondern durch Modifikation des Steroidgerüstes selbst estrogene Verbindungen mit hoher peroraler Wirksamkeit zu erhalten. So blockiert die Überbrückung der 14- und 17-Kohlenstoffatome des Estradiols mit einer Etheno- bzw. Ethanobrύcke die Oxidation dser 17β-Hydroxygruppe (J.Chem. Commun., 1986, 451-453 bzw. Internationale Patentanmeldung PCT/DE87/00361).
Offenbar wird auch bei den erfindungsgemäßen Verbindungen durch die doppelte Überbrύckung (trotz Vorhandenseins eines Wasserstoffatoms am 17-C- Atom) die Oxidation der 17-Hydroxygruppe erschwert.
Die estrogene Wirksamkeit der erfindungsgemäßen Verbindungen wird durch die Ergebnisse des Estrogenrezeptor-Bindungstests belegt. Bei diesem bekannten in vitro-Test wird Gewebe aus Ratten-Uteri präpariert sowie als Bezugssubstanz radioaktiv markiertes H-Estradiol verwendet. Die erfindungsgemäßen Verbindungen (161S)-14,16β-Ethano-15β,161-cyclo-14β-estra- 1,3,5(10)-trien3,17β-diol (A) und (161S)-14,16β-Ethano-15β,161-cyclo-14β- estra-1,3,5(10)-trien-3,17α-diol (B) besitzen demnach folgende Kompetitionsfaktoren:
(A): KA = 2,5
(B): KB = 6,5.
Die Erfindung betrifft somit auch die Verwendung der Verbindungen der allgemeinen Formel I zur Herstellung von Arzneimitteln.
Die erfindungsgemäßen Verbindungen können in der gleichen Weise wie Ethinylestradiol, welches das am meisten verwendete Estrogen ist, formuliert und eingesetzt werden. Sie werden mit den in der galenischen Pharmazie üblichen Zusätzen, Trägersubstanzen und/oder Geschmackskorrigentien nach an sich bekannten Methoden zu den üblichen Arzneimittelformen verarbeitet. Für die orale Applikation kommen insbesondere Tabletten, Dragees, Kapseln, Pillen, Suspensionen oder Lösungen infrage. Für die parenterale Applikation kommen insbesondere ölige Lösungen, wie zum Beispiel Sesamöl- oder Rizinusόllόsungen, infrage, die gegebenenfalls zusätzlich noch ein Verdünnungsmittel, wie zum Beispiel Benzylbenzoat oder Benzylalkohol, enthalten können.
Die Wirkstoffkonzentration in den pharmazeutischen Zusammensetzungen ist abhängig von der Applikationsform und dem Anwendungsgebiet. So können zum Beispiel Kapseln oder Tabletten zur Behandlung von Estrogenmangelerscheinungen 0,001 bis 0,05 mg Wirkstoff, ölige Lösungen zur intramuskulären Injektion pro 1 ml etwa 0,01 bis 0,1 mg Wirkstoff und Vaginalsalben etwa 0,1 bis 10 mg pro 100 ml Salbe enthalten. Zur Kontrazeption bei der Frau können die erfindungsgemäßen Estrogene in Kombination mit 6estagenen ange
wandt werden. Tabletten oder Dragees zur täglichen Einnahme einer Tablette oder eines Dragees sollen vorzugsweise 0,003 bis 0,05 mg des erfindungsgemäßen Estrogens und 0,05 bis 0,5 mg eines Gestagens enthalten.
Die erfindungsgemäßen Verbindungen können bei Estrogenmangelerscheinungen der Frau, wie zum Beispiel Amenorrhoe, Dysmenorrhoe, Sterilität, Endometritis, Kolpitis und klimakterischen Beschwerden und zur Prävention der Osteoporose verwendet werden. Ferner können die Verbindungen als estrogene Komponente in hormoneilen Kontrazeptiva (Einphasen- und Mehrphasen- und Mehrstufenpräparate) eingesetzt werden. Außerdem sind sie in Verbindung mit anderen Wirkstoffen zur Verwendung in hormontragenden Intrauterinpessaren, implantierbaren Wirkstoffträgem sowie in transdermalen Applikationssystemen geeignet.
Die neuen Verbindungen der allgemeinen Formel I

Als typische Vertreter für das zur Reduktion der Verbindung der allgemeinen Formel II zu verwendende komplexe Hydrid seien beispielsweise Lithiumaluminiumhydrid oder Natriumborhydrid genannt. Bei der Reduktion entstehen die C-17-Alkohole als Gemisch der α,β-Isomeren, die durch fraktionierte Kristallisation oder Chromatographie getrennt werden (können).
Die Spaltung der 3-Alkylether der allgemeinen Formel Ia gelingt mit DiisobutyIaluminiumhydrid in einem inerten Lösungsmittel wie beispielsweise Toluol unter erhöhter Tempera tur ( i . a . unter Rύckflußsieden ) . Die Abspaltung der Alkylgruppe in den 3-Alkylethern der allgemeinen Formel II geht unter den gleichen Bedingungen vonstatten. Man kann auch eine Lewissäure wie zum Beispiel Trimethylsilyljodid in einem polaren Lösungsmittel wie Acetonitril zur Etherspaltung verwenden.
Die Einführung einer C1- bis C9-Alkyl-, einer C2- bis C9-Alkenyl- oder Alkinylseitenkette in die 17-Position der Verbindung der Formel III gelingt durch nucleophile Addition einer entsprechenden Alkalialkyl-, -alkenyl- oder -alkinylverbindung (Alkali = Lithium, Natrium, Kalium) an die 17-Ketogruppe nach Standardverfahren. Auch hier erhält man im allgemeinen beide C-17-Isomeren, die sich durch Chromatographie oder andere gängige Verfahren trennen Iassen.
Schließlich können die Alkohole der Formeln Ia, Ib oder Ic nach üblichen
Verfahren, z.B. durch Umsetzung mit dem entsprechenden Carbonsäurechlorid oder -anhydrid (je nach letztendlich gewünschten R1, R2/R3) in Gegenwart einer Base wie 4-DimethyIaminopyridin verestern. Durch Beachtung der unterschiedlichen Reaktivitäten der beiden freien Hydroxygruppen in den Verbindungen der Formel Ib bzw. Ic sowie durch die verwendete Menge des Veresterungsreagenz kann die partielle, totale oder sukzessive Veresterung der Ausgangsverbindungen erreicht werden. Soll R ein Wasserstoffatom sein, kann die 3-Acyloxygruppe nach vollständiger Veresterung von Ib oder Ic wieder selektiv verseift werden.
Die Herstellung der Ausgangsverbindung der allgemeinen Formel II (siehe nachstehendes Schema) geht aus von dem bekannten Dienolacetat 1, das in einer Diels-Alder-Cycloaddition mit Chloracrylnitril umgesetzt wird. Die Reaktion wird vorzugsweise unter erhöhtem Druck (2-20 bar) in einem inerten organischen Lösungsmittel wie Benzol, Toluol, Dichlormethan bei Temperaturen zwischen 50-120ºC
durchgeführt. Die Behandlung des Cycloaddukts 2 mit starken Basen wie Alkalimetallhydroxyden, Lithiumdiisopropylamid, Natriumamid ergibt das Ketonitril 4. Ketalisierung zu 6, Reduktion von 6 mit Diisobutylaluminiumhydrid zum Aldehyd 7, Rhodium-katalysierte Decarbonylierung von 7 zu 8 und Deketalisierung ergibt das zentrale Ausgangsprodukt 9.
Die vorliegende Erfindung erstreckt sich auch auf die Ausgangsverbindungen der allgemeinen Formel II (=9).
Die nachfolgenden Beispiele dienen der näheren Erläuterung der Erfindung:
Herstellung der Ausgangsverbindung (9):
16β-Chlor-16α-cyano-3-methoxy-14,17α-ethenoestra-1,3,5(10)-trien-17β-yl-acetat (2)
Eine Mischung aus 3-Methoxyestra-1,3,5(10),14,16-pentaen-17-yl-acetat (1) (2 g; 6.2 mmol) und 1-Chloro-1-cyano-ethylen (1 ml; 12.6 mmol) in Benzol (7 ml) wird unter Rühren bei 80°C in einem verschlossenen Glasdruckrohr gehalten. Nach 7 Tagen wird das Reaktionsgemisch über Celite filtriert und unter vermindertem Druck eingedampft. Kristallisation des Rückstandes aus Chloroform-Methanol ergibt das reine Cycloaddukt (2) (1.67 g; 66%)

Alkalische Behandlung des Cycloaddukts
a) Wäßrige 2M-Kaliumhydroxid-Lösung (1.1 ml; 2,2 mmol) wird unter Rühren zu einer Lösung des Cycloaddukts (2) (360 mg;0.87 mmol) in Tetrahydrofuran (7.2 ml) und Dimethylsulfoxid (7.2 ml) unter Stickstoff bei 0°C gegeben. Nach 3 Stunden wird gesättigte wäßrige Ammoniumchlorid-Lösung zugegeben und das Gemisch mit Chloroform extrahiert. Der Extrakt wird mit Kochsalzlösung und Wasser gewaschen, getrocknet (MgSO4) und unter vermindertem Druck eingeengt. Umkristallisation des Rohprodukts aus Chloroform/Methanol ergibt (161R)-3-Methoxy-17-oxo-14,16β-ethano- 15β,161-cyclo-14β-estra-1,3,5(10)-trien-161-carbonitril (4) (262 mg; 90%).

Eine Mischung des Carbonitrils (4) (540 mg; 1.6 mmol), Ethylenglycol (2.0 ml) und para-Toluolsulfonsäure (50 mg; 0.29 mmol) in Toluol (54 ml) wird langsam in einer Dean-und-Stark-Apparatur destilliert. Nach 7,5 Stunden, nachdem sich das Volumen auf ca. 30 ml verringert hat, wird das Gemisch unter Rückführung des Kondensats über Molekular-Sieb (4A) für weitere 17 Stunden am RύckfluB erhitzt. Wäßrige Natriumhydrogencarbonat-Lösung wird zugegeben und das Gemisch mit Toluol extrahiert. Der Extrakt wird mit Wasser und Natriumhydrogencarbonat-Lösung gewaschen, getrocknet (MgSO4) und unter vermindertem Druck eingedampft. Der kristalline Rückstand (632 mg) wird aus Chloroform/Methanol umkristallisiert, wobei das Ketal (6) (565 mg; 92%) erhalten wird.

Zur abgekühlten Lösung wird Ethanol zugegeben und das Gemisch filtriert. Das Filtrat wird unter vermindertem Druck eingedampft und der Rückstand an Kieselgel (62 g) mit EthyIacetat/Chloroform (1:99) chromatographiert, wobei zuerst der 17β-Alkohol (10) (97 mg) und nachfolgend der 17α-Alkohol (11) (116 mg) erhalten werden.

(161S)-14, 16β-Ethano-15ß,161-cyclo-14β-estra-1 ,3,5( 10)-trien-3, 17β- diol (14)
Diisobutylaluminiumhydrid (20%ige Lösung in Hexan; 3.9 ml) wird unter Rühren zu einer Lösung von (10) (150 mg; 0,47 mmol) in Toluol (27 ml) unter Stickstoff zugegeben, das Gemisch 23,5 Stunden am Rückfluß erhitzt, auf Raumtemperatur abgekühlt und die Reaktion durch die Zugabe von wäßriger Ammoniumchlorid-Lösung abgebrochen. Anschließend wird Ethylacetat und danach Wasser zugefügt. Das Gemisch wird mit Salzsäure angesäuert und mit Ethylacetat extrahiert. 241 mg (161S)-14,16β-Ethano-15β,161-cyclo-14β-estra-1,3,5(10)-trien-3,17β-diol (14) werden in 3 ml absolutem Dimethylformamid gelöst Man addiert 173 mg Imidazol und 184 mg Dimethyl-(1,1-dimethylethyl)silylchlorid und läßt 1,5 Stunden bei Raumtemperatur nachrühren. Anschließend wird die Reaktionslösung auf Eiswasser gegossen. Man extrahiert mit Ethylacetat, wäscht die organische Phase mit gesättigter Natriumchloridlösung, trocknet über Natriumsulfat, filtriert und engt am Vakuum ein. Das Rohprodukt wird an Kieselgel mit einem Gemisch aus Hexan/Ethylacetat Chromatographien. 100 mg der unter a) beschriebenen Verbindung werden in 2,5 ml absolutem Dichlormethan gelöst Man addiert 0,05 ml Triethylamin, eine Spatelspitze 4-Dimethylaminopyridin sowie 0,035 ml Acetanhydrid und läßt 1 Stunde bei Raumtemperatur nachrühren. Anschließend wird die Reaktionslösung mit Wasser versetzt Man rührt weitere 30 Minuten nach und extrahiert dann mit Ethylacetat Die organische Phase wird mit 2 normaler wäßriger Salzsäure, gesättigter Natriumhydrogencarbonatlösung sowie gesättigter Natriumchloridlösung gewaschen, über Natriumsulfat getrocknet filtriert und am Vakuum eingeengt Man erhält 104 mg (17), welches ohne Reinigung in die Folgestufe eingesetzt wird.
c) (161S)17β-(Acetyloxy)-14,16β-ethano-15β,161-cyclo-14β-estra-1,3,5(10)-trien-3-ol (18)
104 mg der unter b) beschriebenen Verbindung werden in 1 ml absolutem Tetrahydrofuran gelöst Man versetzt mit 182 mg Tetrabutylammoniumfluorid-Trihydrat und läßt 30 Minuten bei Raumtemperatur nachrühren. Anschließend wird die Reaktionslösung auf eiskalte gesättigte Natriumhydrogencarbonatlösung gegossen. Man extrahiert mit Ethylacetat wäscht die organische Phase mit gesättigter Natriumchloridlösung, trocknet über Natriumsulfat, filtriert und engt am Vakuum ein. Das Rohrodukt wird aus Diisopropylether umkristallisiert Man erhält 45 mg (18) als weiße Kristalle.

Man extrahiert mit Ethylacetat, wäscht die organische Phase 2 mal mit gesättigter Natriumhydrogencarbonatlösung und 1 mal mit gesättigter Natriumchloridlösung, trocknet über Natriumsulfat filtriert und engt am Vakuum ein. Das Rohprodukt wird an Kieselgel mit einem Gemisch aus Hexan/Ethylacetat Chromatographien. Man erhält 100 mg (19) als weißen Schaum.
b) (161S)-17β-(Benzoyloxy)-14,16β-ethano-15β,161-cyclo-14β-estra-1,3,5(10)-trien3-ol (20)
Analog zu Beispiel 5c) werden 100 mg der unter a) dargestellten Verbindung in 1 ml Tetrahydrofuran mit 154 mg Tetrabutytemmoiniumfluorid-Trihydrat umgesetzt. Das Rohprodukt wird aus Diisopropylether umkristallisiert Man erhält 61 mg (20) als weiße Kristalle.
Smp:= 186°C

(161S)-17α-(Acetyloxy)-14,16β-ethano-15β,161-cyclo-14β-estra-1,3,5(10)-trien-3-ol (23)
a) (161S)-3-[[ Dimethyl(1,1-dimethylethyl)silyl]oxy]-14,16β-ethano-15β,161-cyclo-14β-estra-1,3,5(10)-trien-17α-ol (21)
Analog zu Beispiel 5a) werden 384 mg (161S)-14,16β-Ethano-15β,161-cyclo-14β-estra-1,3,5(10)-trien-3,17α-diol (15), 275 mg Imidazol und 250 mg Dimethyl-(1,1-dimethylethyl)silylchlorid in 5 ml absolutem Dimethylformamid umgesetzt Man erhält nach Säulenchromatographie 380 mg (21) als weißen Schaum.
1H-NMR (CDCl3): δ= 7,14 dbr (J=9 Hz,1H,H-1); 6,62 dd (J=9, 2.5 Hz,1H,H-2); 6,55 dbr (J=2.5 Hz,1H,H-4); 4,18 m (1H,H-17); 0,97 s (9H,t-Bu); 0,90 s (3H,C-18); 0,10 s (6H,SiMe2)
b) (161S)-17α-(Acetyloxy)-3-[[dimethyl(1,1-dimethylethyl)silyl]oxy]-14,16β-ethano-15β,161-cyclo-14β-estra-1,3,5(10)-trien (22)
Analog zu Beispiel 5b) werden 100 mg der unter a) dargestellten Verbindung mit 0,05 ml Triethylamin, 0,04 ml Acetanhydrid sowie einer Spatelspitze 4-Dimethylaminopyridin in 2 ml Methylenchlorid umgesetzt Man erhält 90 mg 22),
welches als Rohprodukt in die Folgestufe eingesetzt wird. c) (161S)-17α-(Acetyloxy)-14,16β-ethano-15β,161-cyclo-14β-estra-1,3,5(10)-trien-3-ol (23)
Analog zu Beispiel 5c) werden 90 mg der unter b) dargestellten Verbindung und 160 mg Tetrabutylammoniumfluorid-Trihydrat in 1 ml Tetrahydrofuran umgesetzt Das Rohprodukt wird aus Diisopropylether umkristallisiert Man erhält 50 mg 23) als weiße Kristalle.
Smp.= 189,5°C
[α]20 D= +94,3° (CHCl3; c=0,510)

(161S)-17α-(BenzoyIoxy)-14,16β-ethano-15β,161-cycIo-14β-estra-1,3,5(10)trien-3-ol
(25) a) (161S)-3-[[Dimemyl( 1,1-dimethylethyl)silyl]oxy]-17α-(benzoyloxy)-14,16β-ethano- 15β-161-cyclc-14β-estra-1,3,5(10)-trien (24)
Analog zu Beispiel 6a) werden 100 mg der unter Beispiel 7a) dargestellten Verbindung und 0,07 ml Benzoylchlorid in 0,6 ml absolutem Pyridin umgesetzt Man erhält nach Chromatographie 95 mg (24) als weißen Schaum.
b) (161S)-17α-(Benzoyloxy)-14,16β-ethano-15β,161-cyclo-14β-estra-1,3,5(10)-trien3-ol (25)
Analog zu Beispiel 5c) werden 95 mg der unter a) dargestellten Verbindung und 150 mg
Tetrabutylammoniumfluorid-Trihydrat in 1,5 ml absolutem Tetrahydrofuran umgesetzt
Das Rohprodukt wird aus Diisopropylether umkristallisiert Man erhält 53 mg 25) als weiße Kristalle.
Smp.= 170,0°C

Claims
1. 14,16ß-Ethano-15ß,161-cyclo-14ß-estra-1,3,5(10)-triene der allgemeinen Formel I
worin
R1 ein Wasserstoffatom, einen C1- bis C9-Alkyl oder C1- bis C9-Acylrest,
R2 ein Wasserstoffatom, einen C1- bis C9-Alkyl-, einen C2- bis C9-Alkenyl- oder Alkinylrest sowie
R3 eine Hydroxygruppe oder einen C1- bis C9-Acyloxyrest
bedeuten.
2. (161S)-14,16β-Ethano-15β,161-cyclo-14β-estra-1,3,5{10)-trien-3,17β-diol (161S)-14,16β-Ethano-15β,161-cyclo-14β-estra-1,3,5(10)-trien-3,17α-diol
3. Verfahren zur Herstellung von 14,16β-Ethano-15β,161-cyclo-14β-estra- 1,3,5(10)-triene der allgemeinen Formel I
worin
R1 ein Wasserstoffatom, einen C1- bis C9-Alkyl oder C1- bis C9-Acylrest,
R2 ein Wasserstoffatom, einen C1- bis C9-Alkyl-, einen C2- bis C9-Alkenyl-oder Alkinylrest sowie
R3 eine Hydroxygruppe oder einen C1- bis C9-Acyloxyrest
bedeuten,
dadurch gekennzeichnet, daB entweder eine Verbindung der allgemeinen
Formel II
mit einem komplexen Hydrid zu einer Verbindung der allgemeinen Formel Ia
reduziert,
die C-17-Isomeren gegebenenfalls getrennt,
gegebenenfalls der 3-Alkylether unter Erhalt der Verbindungen der Formel
Ib
gespalten ,
oder in der Verbindung der allgemeinen Formel I I der 3-Alkylether gegebenenfalls gespalten und die erhaltene Verbindung der Formel I II
mit einer Verbindung der allgemeinen Formel IV
(IV), worin R für einen C1- bis C9-Alkyl- oder einen C2- bis C9- Alkenyl- oder Alkinylrest und X für ein Li-, Na- oder K-Atom steht,
zu einer Verbindung der allgemeinen Formel Ic
worin R die oben angegebene Bedeutung hat, umgesetzt wird und gegebenenfalls die C-17-Alkohole der Formeln Ia. Ib oder Ic durch Umsetzung mit einem entsprechenden C1- bis C9-Carbonsäurechlorid- oder -anhydrid partiell, total oder sukzessive verestert werden.

5. Pharmazeutische Präparate, die eine oder mehrere Verbindung(en) der allgemeinen Formel I sowie einen pharmazeutisch verträglichen Träger enthalten.
6. Verwendung der Verbindungen der allgemeinen Formel I zur Herstellung von Arzneimitteln.
