Abstract
Beschrieben werden 3-Alkoxypyridin-2-carbonsäureamidester der Formel (I), in welcher R?1, R2 und R3¿ Wasserstoff, R4 (C1-C6)-Alkyl, A eine -CH2-Gruppe bedeutet, in der ein Wasserstoff durch eine Methylgruppe ersetzt sein kann, und B -CO2G bedeutet, wobei G den Rest eines Alkohols GOH bedeutet, einschließlich der physiologisch wirksamen Salze; ihre Herstellung, ihre Verwendung zur Hemmung der Kollagenbiosynthese und ihre Verwendung als Arzneimittel zur Behandlung von fibrotischen Erkrankungen.
Description
3-Alkoxypyridin-2-carbonsäureamidester, ihre Herstellung und ihre Verwendung als Arzneimittel
Die Erfindung betrifft 3-Alkoxypyridin-2-carbonsäureamidester, ihre Herstellung und ihre Verwendung zur Hemmung der Kollagenbiosynthese und ihre Verwendung als Arzneimittel zur Behandlung von fibrotischen Erkrankungen.
Verbindungen, die die Enzyme Prolyl- und Lysylhydroxylase inhibieren, bewirken eine sehr selektive Hemmung der Kollagenbiosynthese durch Beeinflussung der kollagenspezifischen Hydroxylierungsreaktionen. In deren Verlauf wird proteingebundenes Prolin oder Lysin durch die Enzyme Prolyl- bzw. Lysylhydroxylase hydroxyliert. Wird diese Reaktion durch Inhibitoren unterbunden, so entsteht ein nicht funktionsfähiges, unterhydroxyliertes Kollagenmolekül, das von den Zellen nur in geringer Menge in den extrazellulären Raum abgegeben werden kann. Das unterhydroxylierte Kollagen kann außerdem nicht in die Kollagenmatrix eingebaut werden und wird sehr leicht proteolytisch abgebaut. Als Folge dieser Effekte verringert sich insgesamt die Menge des extrazellulär abgelagerten Kollagens.
Inhibitoren der Prolylhydroxylase sind deshalb geeignete Substanzen in der Therapie von Erkrankungen, in denen die Ablagerung von Kollagenen maßgeblich zum Krankheitsbild beiträgt. Hierzu gehören u. a. Fibrösen der Lunge, Leber und Haut (Skleroderma und Vernarbungen nach Verbrennungen, Verletzungen und chirurgischen Eingriffen) sowie die Artherosklerose.
Es ist bekannt, daß das Enzym Prolylhydroxylase durch Pyridin-2,4- und -2, 5- dicarbonsäure effektiv gehemmt wird (K. Majamaa et al., Eur. J. Biochem. 138 ( 1 984) 239-245). Diese Verbindungen sind in der Zellkultur allerdings nur in sehr hohen Konzentrationen als Hemmstoffe wirksam (Tschank, G. et al., Biochem. J. 238 ( 1 987) 625-633).
Auch Prodrugs der Pyridin-2,4(5)-dicarboxylate sind bekannt. Diese sind in den
ORIGINAL UNTERLAGEN
Patentanmeldungen EP-A-0 590 520 und DE-A-42 38 506 und EP-A-0 562 51 2 beschrieben.
N-Oxalylglycine als Inhibitoren der Prolyl-4-hydroxylase sind aus J. Med. Chem. 1 992, 35, 2652-2658 (Cunliffe et al. ) und EP-A-0 457 1 63 bekannt.
Weiterhin ist 3-Hydroxypyridin-2-carbonsäure(glycylethylester)amid in J. Org. Chem. 31 , 636-638 ( 1966) beschrieben.
Es bestand daher die Aufgabe, Verbindungen bereitzustellen, die sich durch eine besonders hohe in vivo und/oder in vitro-Aktivität auszeichnen, insbesondere bei ihrer systemischen und/oder lokalen Anwendung.
Überraschenderweise wurde nun gefunden, daß 3-Alkoxypyridin-2- carbonsäureesteramide der Formel I besonders stark wirksame Inhibitoren der Kollagenbiosynthese sind. Die erfindungsgemäßen Verbindungen stellen eine Auswahl aus den in der EP-A-0 650 960 beschriebenen Verbindungen dar. Sie zeichnen sich im Vergleich zu den in der europäischen Patentanmeldung genannten Verbindungen durch eine besonders hohe in-vivo- und in-vitro- Aktivität aus, insbesondere bei ihrer systemischen und/oder lokalen Verwendung gegen fibrotische Erkrankungen. Hierzu gehören beispielsweise Fibrösen der Lunge, der Leber, der Niere, des Herzens, des Auges sowie der Haut und bei Artherosklerose.
Die Verbindungen der Formel I führen zu einer starken Hemmung der Kollagenbiosynthese in den verschiedenartigsten Zellen (z.B. normale humane Hautfibroblasten, primäre Fettspeicherzellen aus der Rattenleber, Rattenleberepithelzellen und in Organkulturen von Calvarien) .
Sie sind besonders geeignet zur lokalen Anwendung als Inhibitoren der Kollagenbiosynthese, zur lokalen Anwendung als antlfibrotische Wirkstoffe und
zur lokalen Anwendung bei Krankheitsformen, die von einer erhöhten Bildung von Bindegewebe (Kollagen) verursacht werden. So sind die Verbindungen der Formel I z.B. geeignet zur lokalen Anwendung zur Vermeidung/Verringerung von Vernarbungen nach chirurgischen Eingriffen am menschlichen Körper und zur lokalen Anwendung bei der postoperativen Behandlung von Augen¬ erkrankungen, z.B. der nachoperativen Behandlung des Glaukoms und bei strahleninduzierter oder durch Chemotherapie induzierter Fibröse, insbesondere an der Lunge.
Die erfindungsgemäßen Verbindungen sind Ester-Prodrugs der entsprechenden Carbonsäuren der Formel I, in denen B eine Carboxylgruppe bedeutet.
Die Verbindungen der Formel I werden im lebenden Organismus (in vivo) und in Zellkulturen (in vitro) zu Verbindungen der Formel I, in denen B eine Carboxylgruppe oder deren Salze bedeutet, gespalten.
Nach der Applikation der Verbindungen der Formel I bewirken sie die in vivo und in vitro zu beobachtende Hemmung der Kollagenbiosynthese, wobei sich die Verbindungen der Formel I, in denen B eine Carboxylgruppe oder deren Salze bedeutet, bilden. Diese Verbindungen inhibieren die Prolyl-4-hydroxylase und führen deshalb zu einer Hemmung der Kollagenbiosynthese.

Besonders bevorzugt sind solche Verbindungen der Formel I, in denen G einen linearen (CrC1 6)-Alkyl-Rest bedeutet, einschließlich der physiologisch wirksamen Salze.
Die Erfindung umfaßt weiterhin Salze der Verbindungen der allgemeinen Formel I.
Die Salzbindung mit sauren Reagenzien kann am Pyridin-N-Atom erfolgen.
Zur Anwendung kommende Reagenzien sind beispielsweise Toluolsulfonsäure, Methansulfonsäure, HCI, H2SO4, H3PO4 und Arzneimittel, die eine saure Gruppe enthalten.
Die Erfindung betrifft die Verbindungen der allgemeinen Formel I sowie die physiologisch verträglichen Salze zur Anwendung bei der Hemmung der Kollagenbiosynthese, insbesondere in verschiedenen Zellen.
Die Erfindung betrifft die Verbindungen der allgemeinen Formel I sowie die physiologisch verträglichen Salze zur Anwendung bei der Hemmung der Prolyl- 4-hydroxylase in vivo und in vitro.
Weiterhin betrifft die Erfindung die Verbindungen der allgemeinen Formel I sowie die physiologisch verträglichen Salze zur Anwendung bei fibrotischen Erkrankungen der Lunge, der Leber, der Niere, des Herzens, des Auges und der Haut. Die Verbindungen können auch Anwendung finden bei Artherosklerose. Hierbei werden systemische und/oder lokale Applikationen angewandt.
Insbesondere betrifft die Erfindung Verbindungen der allgemeinen Formel I sowie der physiologisch verträglichen Salze zur lokalen Anwendung, insbesondere als Inhibitoren der Kollagenbiosynthese, als antifibrotische Wirkstoffe und bei Krankheitsformen, die von einer erhöhten Bildung von
Bindegewebe (Kollagen) verursacht werden. Hierunter zählen die Anwendungen zur Vermeidung/Verringerung von Vernarbungen nach chirurgischen Eingriffen am menschlichen Körper und bei der postoperativen Behandlung von Augenoperationen, z.B. bei Glaukomoperationen und bei strahleninduzierter oder durch Chemotherapie induzierter Fibröse, insbesondere an der Lunge.
Schließlich betrifft die Erfindung die Verbindungen der allgemeinen Formel I zur Verwendung als Arzneimittel.
Insbesondere betrifft die Erfindung Verbindungen der Formel I zur lokalen Anwendung bei Fibrösen der Haut, der Lunge und des Auges, insbesondere zur nachoperativen Behandlung des Glaukoms.
Weiterhin betrifft die Erfindung ein Verfahren zur Herstellung von Verbindungen der allgemeinen Formel I.
Die Herstellung der Verbindungen der Formel I, in denen A eine -CH2-Gruppe, in der ein Wasserstoff durch eine Methylgruppe ersetzt sein kann und B CO2G bedeuten, erfolgt, indem
i 1 .) Pyridin-2-carbonsäuren der Formel II (R5 = H) mit den Aminoestern
Formel lll oder deren Salze zu den Amidestern der Formel I umgesetzt werden; oder i2.) Pyridin-2-carbonsäureester der Formel II (R5 = (CrC1 6)-Alkyl) unter den
Bedingungen der Aminolyse zu den Verbindungen der Formel I umgesetzt werden; oder
ii) die Verbindungen der Formel IV mit einem Alkohol GOH verestert werden; oder
iii) die Verbindungen der Formel V mit R4X alkyliert werden, worin X eine Abgangsgruppe, insbesondere Halogen, -OSO2Me, -OSO2Phenyl u.a. bedeutet.

Geeignete Verfahren zur Amidbildung (Umsetzung i1 ) sind die Methoden der Carbonylaktivierung und die aus der Peptidchemie bekannten Kondensationsreaktionen.
An Reagenzien zur Carbonsäureaktivierung können die dem Fachmann bekannten Substanzen, wie Thionylchlorid, Oxalylchlorid, Pivaloylchlorid, Chlorameisensäureester-Derivate oder N,N'-Carbonyldimidazol Verwendung finden. Die aktivierten Derivate der Verbindungen der Formel II werden nach Herstellung in situ mit den Amidderivaten der Formel lll umgesetzt.
Ein geeignetes Kondensationsmittel ist beispielsweise die Kombination von N,N'-Dicyclohexylcarbodiimid/N-Hydroxy- 1 H-benzotriazol und N-Ethylmorpholin.
Geeignete Lösungsmittel sind Dichlormethan, Tetrachlormethan, Butylacetat, Ethylacetat, Toluol, Tetrahydrofüran, Dimethoxyethan, 1 ,4-Dioxan, Acetonitril, N,N-Dimethylformamid, N,N-Dimethylacetamid, Dimethylsulfoxid, Nitromethan und/oder Pyridin.
Die erfindungsgemäßen Verbindungen der Formel I besitzen wertvolle pharmakologische Eigenschaften und zeigen insbesondere antifibrotische Wirksamkeit.
Die antifibrotische Wirkung kann in folgenden Tiermodellen bestimmt werden:
Bildung einer bindegewebigen Kapsel um eine subkutan implantierte osmotische Minipumpe, z.B. Unemori, E.N. et al., J. Invest. Dermatol . , 1 01 : 280-5, 1 993,
Kollagengehalt von subkutan implantierten "cotton pellets" oder Polyvinylschwämmchen, z.B. Boyle, E., Mangan, F.R. , Br. J . Ep. Pathnol. 61 : 351 -60, 1980; und
Kollagengehalt der Lunge nach strahleninduzierter oder chemisch induzierter Fibröse, Radiat. z.B. Ward, H.E. et al., Res., 1 36, 1 5-21 , 1993, sowie Santana, A. et al. Am. J. Respir. Cell. Mol. Biol. 1 3: 34-44, 1995.
Hemmung der Prolyl-4-hydroxylase in Zellkulturen: Normale humane Hautfibroblasten (Normal human fibroblasts, NHDF), Rattenleber-Epithelzellen (rat liver epithelial cells, Ref. 1 ) und Fettspeicherzellen aus der Leber (fat storing cells, Ref. 2) werden als Zellmodelle zur Substanztestung verwendet. Dazu werden die Zellen in Gegenwart von Hemmstoffen kultiviert. Gleichzeitig wird das in dieser Zeit neu synthetisierte
Kollagen durch 4-3H-L-Prolιn und 1 4C-Prolιn metabolisch markiert. Der Einfluß der Testsubstanzen auf den Hydroxylierungsgrad des Kollagens wird anschließend entsprechend der Methode von Chojkier et al (Ref. 3) bestimmt. Als Referenzsubstanz wird 2,2'-Dipyridyl eingesetzt. ( 1 .: Schrode, W., Mecke, D., Gebhard, R. 1 990. Hemmung der Prolyl-4-hydroxylase in vitro (in Hühnchenembryo - Calvarien) gemäß Biochem. J. ( 1994) 300, 525-300:
1 . Metabolische Markierung
Calvarien werden aus 1 5 Tage alten Hühnchenembryonen präpariert und 3 min bei 37°C in Hanks' balanced salt solution minimum essential medium Eagle (HMEM, BioWhittaker, Walkersville, MD, USA) gewaschen. Je 4 Calvarien werden in Glasgefäßen mit 1 .5 ml HMEM unter Zusatz von 2 mM Glutamin und 1 μO [U- 14C]Prolιn sowie verschiedenen Inhibitorkonzentrationen 2.5 h bei 37°C inkubtert. Die Inkubation wird durch Einbringen der Probengefäße in Eis beendet. Das Medium wird entfernt und die Calvarien werden kurz mit 1 ml bidest H2O gewaschen. Danach werden die Calvarien mit 3 ml 0.5 M Essigsäure und 1 8 μg Phenylmethan-Sulfonylfluoπd versetzt und 1 6 h extrahiert. Die Extraktionslösung wird bis zum Entfernen des freien [U- 14C]Prolιn bei 4°C gegen 0.5 M Essigsäure dialysiert und anschließend in Aliquots gefriergetrocknet.
2. Hydroxyprolin - Analyse
Ein Aliquot der gefriergetrockneten Probe wird in 2 ml 6 N HCI aufgelöst und 24 h bei 105 °C hydrolysiert. Danach wird die Salzsäure abgedampft. Der Rückstand wird in 300 μ\ bidest H2O resuspendiert, in Eppendorf-Gefäße überführt und erneut getrocknet. Die verbleibenden Hydrochloride werden durch Lösen in 60 μ\ einer Ethanol/H2O/Triethylamin 2:2: 1 (v:v:v) und erneutem Trocknen neutralisiert. Anschließend werden die Aminosäuren 20 min bei Raumtemperatur in einer Lösung von Ethanol/Triethylamin/Phenylisothiocyanat/ H2O 7: 1 : 1 : 1 (v:v:v:v) derivatisiert und erneut getrocknet. Zur Analyse mit HPLC werden die Proben in 1 50 μl eines Phosphatpuffers (5 mM Na2HPO4, pH 7.4/ Acetonitril 95:5 (v:v)) gelöst und 5 min bei 10000 g zentrifugiert. 50 μ\ werden zur Analyse verwendet. Die HPLC-Chromatographie erfolgt auf einer C 1 8 reverse phase Säule Ultrasphere ODS 3 μm, 4.6 nim x 7.5 cm (Beckman) bei 50°C mit folgendem Gradientensystem:

3. SDS - Polyacrylamid - Gelelektrophorese
Ein Aliquot der gefriergetrockneten Proben aus 1 . wird für die SDS- Polyacrylamid - Gelelektrophorese mit 10 mM Tris/HCI, pH 8 / 1 mM EDTA / 1 % SDS und 5 % Mercaptoethanol gelöst und 5 min bei 95 °C denaturiert. Die Elektrophorese erfolgt auf einem linearen Gradientengel (5-15 %). Nach Fixierung in Methanol/Eisessig/Wasser 3: 1 :6 (v:v:v) werden die Gele getrocknet und zur Belichtung von Kodak X-Omat Film bei -70°C verwendet.
Im folgenden werden die überraschenden Vorteile der erfindungsgemäßen Verbindungen gegenüber Verbindungen aus EP-A-0 650 960 anhand von Vergleichsversuchen über die Hemmung der Prolyl-4-hydroxylase (Kollagenbiosynthese) in Rattenleberzellen verdeutlicht:

Die Vergleiche zeigen, daß die erfindungsgemäßen Verbindungen der vorliegenden Auswahlerfindung gegenüber den aus der EP-A-0 650 960 schon bei überraschend geringen Konzentrationen (≤ 10 μM, je nach Modellversuch) hohe Hemmungsraten (-50%) zeigen.
Die Verbindungen der Formel I können als Medikamente in Form pharmazeutischer Präparate Verwendung finden, welche sie gegebenenfalls mit verträglichen pharmazeutischen Trägern enthalten. Die Verbindungen können als Heilmittel, z.B. in Form pharmazeutischer Präparate Verwendung finden, welche diese Verbindungen in Mischung mit einem für die enterale, perkutane, parenterale und inhalative Applikation geeigneten pharmazeutischen, organischen oder anorganischen Träger, wie z.B. Wasser, Gummi arabicum, Gelatine, Milchzucker, Stärke, Magnesiumstearat, Talk, pflanzliche Öle, Polyalkylenglykole, Vaseline usw. enthalten.
Sie können zu diesem Zweck oral in Dosen von 0, 1 bis 25 mg/kg/Tag, vorzugsweise 1 bis 5 mg/kg/Tag oder parenteral in Dosen von 0,01 bis 5 mg/kg/Tag, vorzugsweise 0,01 bis 2,5 mg/kg/Tag, insbesondere 0, 5 bis 1 ,0 mg/kg/Tag, appliziert werden. Die Dosierung kann in schweren Fällen auch erhöht werden. In vielen Fällen genügen jedoch auch geringere Dosen. Diese Angaben beziehen sich auf einen Erwachsenen von etwa 75 kg Gewicht.
3-Methoxypyridin-2-carbonsäure ist aus J. Org. Chem. 36, 2002 ( 1 971 ) von E.V. Brown, M.B. Shambhu bekannt (vgl. Formel II; R1-R3 = H, R4 = Me, R5 = H).
Weiterhin ist 3-Methoxypyridin-2-carbonsäuremethylester (vgl. Formel II, R5 = CH3) aus Acta Chem. Scand. 23, 1 791 ( 1 969) bekannt.
Die in den folgenden Beispielen als Ausgangsverbindung eingesetzte 3-Methoxypyridin-2-carbonsäure wurde ausgehend von 4-Chlor-3-methoxy-2- methylpyridin, das aus Maltol erhalten wurde, erhalten (vgl. EP-A-0 208 452 und EP-A-0 304 732).
Unter den im folgenden beschriebenen Beispielen werden die erfindungsgemäßen Verbindungen der Formel I als substituierte Pyridin-2- carbonsäure(glycylester)amide bezeichnet.
Unter dieser Bezeichnungsweise werden substituierte Pyridin-2-carbonsäure-N-
((alkoxycarbonyl)methyUamide verstanden.
Die Klassifizierung als substituierte N-(PyrιdyI-2-carbonyl)glycine ist eine weitere
Möglichkeit.
Beispiel 1
3-Methoxypyridin-2-carbonsäure-N-((( 1 -butyloxy)carbonyl)methyl)amid
a) 3-Methoxypyridin-2-carbonsäure-Hydrochlorid
a 1 .) 4-Chlor-3-methoxy-2-methylpyridin- 1 -oxid
1 1 ,2 g (80,5 mmol) 3-Methoxy-2-methyl-4( 1 H)-pyridon wurden in 100 ml Phosphoroxychlorid 10 Stunden rückfließend erhitzt. Anschließend engte man ein, versetzte 2 ml mit je 30 ml Toluol, engte wiederum ein, nahm den Rückstand in 1 50 ml Wasser auf, brachte mit K2CO3 auf pH 1 1 , extrahierte mit Dichlormethan, wusch die organische Phase mit Wasser, trocknete und befreite vom Lösungsmittel. Aus dem hellbraunen Öl (9 g) wurden mit m-Chlorperbenzoesäure in Dichlormethan unter Standardbedingungen 8 g des Produktes erhalten, Fp. 88 bis 89°C (aus Petrolether).
a2.) 4-Chlor-2-hydroxymethyl-3-methoxypyridin
30 g (1 73 mmol) 4-Chlor-3-methoxy-2-methylpyridin-N-oxid wurden in 100 ml Eisessig gelöst, bei 80°C unter Rühren tropfenweise mit 1 50 ml Acetanhydrid versetzt und 2 Stunden bei 1 10°C gerührt. Dann wurde auf 80°C abgekühlt, 200 ml Methanol zugetropft, 1 5 Minuten zum Sieden erhitzt, nach Abkühlen im Vakuum eingeengt, der Rückstand in Methanol aufgenommen und in 300 ml 1 ,5 N methanolische Natronlauge einfließen lassen, 30 Minuten bei 20°C gerührt, im Vakuum eingeengt, der Rückstand in Wasser aufgenomen, dreimal mit Dichlormethan extrahiert, die organische Phase getrocknet, eingeengt und der Rückstand mit Petrolether zur Kristallisation gebracht. Man erhielt 23 g Produkt, Fp. 64 bis 66°C.
a3.) 4-Chlor-3-methoxypyridin-2-carbonsäure
Zu 24 g KOH, gelöst in 900 ml Wasser wurden bei 20° C 52,2 g (0,3 Mol) des vorstehenden Alkohols zugegeben und bei 50 bis 60°C jeweils bis zur Entfärbung portionsweise mit 70 g (0,44 Mol) Kaliumpermanganat versetzt. Nach 1 Stunde Rühren bei 50°C wurde heiß abgesaugt, dreimal mit heißem Wasser nachgewaschen, das Filtrat im Vakuum auf 300 ml eingeengt und unter Kühlung mit konzentrierter HCI auf pH 1 gebracht. Nach Absaugen und Trocknen erhielt man 50 g Produkt, Fp., 121 °C (n. Zers.).
a4.) 29 g (0, 1 5 Mol) der obigen 4-Chlorpyridincarbonsäure wurden in 500 ml Methanol/Tetrahydrofuran (1 : 1 ) mit Pd/C (10 %) in der Schüttelente hydriert. Nachdem 3, 1 I Wasserstoff aufgenommen worden waren, wurde vom Katalyator abgesaugt, das Filtrat eingeengt, der kristalline Rückstand mit Ethyiacetat behandelt, abgesaugt und getrocknet; Ausbeute 29 g, Fp. 1 70°C (unter Zers.) .
b) 3,8 g (20 mmol) 3-Methoxypyridin-2-carbonsäure-Hydrochlorid wurden in 500 ml wasserfreiem Dichlormethan suspendiert und bei 20°C unter Rühren mit 6, 1 g (20 mmol) Glycin-( 1 -butyl)ester-Tosylat (hergestellt aus Glycin, 1 -Butanol, p-Toluolsulfonsäure am Wasserabscheider mit Toluol), dann mit 7,5 ml (60 mmol) N-Ethylmorpholin, 3 g (22,5 mmol) 1 -Hydroxy-1 H-benztriazol und 8,5 g (20 mmol) N-Cyclohexyl-N'-(2- morpholinoethyl)-carbodiimid-methyl-p-toluolsulfonat (CMC) versetzt und 24 Stunden bei 20°C gerührt. Dann wurde vom Ungelösten abgesaugt, das Filtrat nacheinander mit wäßriger Na-bicarbonat-Lösung, mit 1 N wäßriger Salzsäure und mit Wasser ausgeschüttelt. Die organische Phase wurde getrocknet, im Vakuum eingeengt und der ölige Rückstand durch Anreiben zur Kristallisation gebracht. Man erhielt 1 , 76 g der farblosen Titelverbindung, Fp. 60 bis 62°C.
Beispiel 2 3-Methoxypyridin-2-carbonsäure-N-((( 1 -octyloxy)carbonyl)methyl)amid
2,5 g (1 3 mmol) 3-Methoxypyridin-2-carbonsäure-Hydrochlorid, 5,5 ml (45 mmol) N-Ethylmorpholin, 2 g ( 1 5 mmol) 1 -Hydroxy- 1 H-benztriazol, 4,7 g ( 13 mmol) Glycinoctylester-Tosylat (am Wasserabscheider mit Toluol aus Glycin, 1 -Octanol und p-Toluolsulfonsäure hergestellt) und 6,3 g ( 1 5 mmol) CMC (vgl. Beispiel 1 ) wurden 48 Stunden in 350 wasserfreiem Dichlormethan gerührt. Nach analoger Aufarbeitung wie in Beispiel 1 ) wurde das Rohprodukt mit Dichlormethan (im Verlauf bis zu 2,5 % Methanol-Zusatz) an Kieselgel chromatographiert. Man erhielt 3,6 g der farblosen, öligen Titelverbindung, 1 H-NMR (CDCI3) : δ = 4,26 (d, CH2-Glycin).
Beispiel 3 3-Methoxypyridin-2-carbonsäure-N-((( 1 -dodecyloxy)carbonyl)methyl)amid
Analog Beispiel 1 wurden 2,8 g (1 5 mMol) 3-Methoxypyridin-2-carbonsäure- Hydrochlorid in 400 ml wasserfreiem Dichlormethan 24 Stunden mit 6,2 g (1 5 mMol) Glycin-( 1 -dodecyl)ester-Tosylat (am Wasserabscheider hergestellt aus 38 g (0,5 Mol) Glycin, 93,2 g (0,5 Mol) 1 -Dodekanol und 1 1 5 g (0,6 Mol) p-Toluolsulfonsäure; 140 g Produkt wurden mit Diethylether zur Kristallisation gebracht, Fp. ca. 106°C, sintern bei 80°C), 8,2 ml (60 mMol) N-Ethylmorpholin, 2 g ( 1 5 mMol) 1 -Hydroxy-1 H-benztriazol und 6,3 g (15 mMol) CMC bei 20°C gerührt. Wegen starker Emulsionsbildung wurde bei der Aufarbeitung in Anlehnung an Beispiel 1 Dichlormethan im Vakuum abdestilliert und der Rückstand mit Diethylether aufgenommen. Nach dem Trocknen der organischen Phase mit Magnesiumsulfat, Einengen im Vakuum, wurden 4,7 g des erhaltenen gelben Öls mit Ethylacetat an Kieselgel chromatographiert. Man erhielt 4 g eines hellgelben Öls, das nach 2 tägigem Stehen nach Anreiben mit dem Glasstab kristallisierte. 3,6 g der farblosen Titelverbindung, Fp. 47 bis 49°C, wurden isoliert.

Beispiel 14 3-Methoxypyridin-2-carbonsäure-N-(((hexadecyloxy)carbonyl)methyl)amid
5,7 g (30 mmol) 4-Chlor-3-methoxypyridin-2-carbonsäure und 14,2 g (30 mmol) Glycinhexadecylester-tosylat (Fp. ca. 90°C, hergestellt aus Glycin, 1 -Hexadecanol, p-Toluolsulfonsäure am Wasserabscheider mit Toluol) wurden in 300 ml Dichlormethan analog zu Beispiel 1 ) mit 7, 7 (60 mmol) N-Ethylmorpholin, 4,5 g (33 mmol) 1 -Hydroxy-1 H-benztriazol und 1 2,8 g (30 mmol) N-Cyclohexyl-N'-(2-morpholinoethyl)-carbodiimid-methyl-p- toluolsulfonat (CMC) versetzt und 24 Stunden gerührt. Dann wurde vom Ungelösten abgesaugt, das Filtrat mit wäßriger Na-bicarbonat-Lösung, mit Wasser und mit wäßriger Salzsäure ausgeschüttelt, die organische Phase
eingeengt, der Rückstand ( 14 g) in 500 ml Tetrahydrofuran/Methanol ( 1 : 1 ) gelöst, mit Pd/C ( 1 0 %) versetzt und an der Schüttelente hydriert. Nach beendeter Wasserstoff-Aufnahme wurde vom Katalysator abgesaugt, das Filtrat eingeengt und der Rückstand mit Ethylacetat an Kieselgel chromatographiert. Entsprechende Fraktionen wurden eingeengt und der Rückstand mit Diisopropylether zur Kristallisation gebracht. Man erhielt 2, 1 g der Titelverbindung als farblose Substanz, Fp. 63 bis 65 °C.
Beispiel 1 5
3-Methoxypyridin-2-carbonsäure-N-((ethyloxycarbonyl) methyl )amid-Hydrochlorid
a) 4-Chlor-3-methoxypyridin-2-carbonsäure-N-((ethyloxycarbonyl)methyl)- amid
Zur Herstellung wurden 4,7 g (25 mmol) 4-Chlor-3-methoxypyridin-2- carbonsäure in 200 ml wasserfreiem Dichlormethan suspendiert und bei 20°C unter Rühren nacheinander mit 3,5 g (25 mmol) Glycinethylester- Hydrochlorid, 6,4 ml (50 mmol) N-Ethylmorpholin, 3,8 g (28 mmol) 1 - Hydroxy-( 1 H)-benztriazol und 5, 1 5 g (25 mmol) N,N'- Dicyclohexylcarbodiimid versetzt und 20 Stunden bεi 20°C gerührt. Dann wurde vom Ungelösten abfiltriert, die organische Phase mit gesättigter, wäßriger Natriumcarbonat-Lösung geschüttelt, getrocknet, im Vakuum eingeengt, der Rückstand (6 g Öl) mit Ethylacetat an Kieselgel chromatographiert und 5,4 g öliges Produkt erhalten.
b) Die Titelverbindung wurde erhalten, indem die vorstehende Substanz mit Pd/C (10 %) in der Schüttelente hydriert wurde, Fp. 141 bis 142 °C (unter Gasentwicklung, aus Diethylether).
Beispiel 1 6
3-Methoxypyridin-2-carbonsäure-N-(((2-nonyloxy)carbonyl) methyl Jamid-Racemat
2,5 g ( 10 mmol) 3-Methoxypyridin-2-carbonsäure-N-(carboxymethyl)amid- Hydrochlorid (Fp. 1 57°C unter Gasentwicklung) wurden in 100 ml wasserfreiem Tetrahydrofüran suspendiert, mit 1 ,6 ml ( 1 2 mmol) Triethylamin, sodann unter Rühren tropfenweise mit 2,4 g Pivaloylchlorid, gelöst in wenig Tetrahydrofüran, versetzt (Temperaturanstieg auf 35-40°C). Nach 30 min wurde i.Vak. eingeengt, der rötlich gefärbte Rückstand in 100 ml wasserfreiem Tetrahydrofüran aufgenommen, mit 1 ,6 ml Triethylamin versetzt und sodann bei 20°C einer Lösung von Na-2-nonanolat in 2-Nonanol (hergestellt aus 30 ml 2-Nonanol und 0,8 g (20 mmol) NaH) hinzugegeben . Nach 1 h wurde i.Vak. eingeengt, der Rückstand mit Dichlormethan versetzt, mit 2N wäßriger Ammoniumchlorid-Lösung ausgeschüttelt, die organische Phase getrocknet, i.Vak. eingeengt und der Rückstand (9 g) mit Ethylacetat an Kieselgel chromatographiert. Man erhielt 1 , 1 g der öligen farblosen Titelverbindung, 1 H-NMR (DMSO): δ = 3,95 (d, CH2-Glycin).
Beispiel 1 7 3-Methoxypyridin-2-carbonsäure-N-(((4-heptyloxy)carbonyl)methyl)amid
2,5 g ( 10 mmol) 3-Methoxypyridin-2-carbonsäure-N-(carboxymethyl)amid- Hydrochlorid wurden wie in Beispiel 1 6 behandelt und sodann bei 20°C mit 140 ml einer Lösung von Na-4-heptanolat in 4-Heptanol (hergestellt aus 140 ml 4- Heptanol und 0,6 g (25 mmol) Natrium, Ultraschallbad) versetzt. Nach 30 Minuten wurde 1 Stunde auf 70 bis 80°C erhitzt, nach dem Abkühlen im Vakuum eingeengt, der Rückstand in Wasser aufgenommen, mit Dichlormethan extrahiert, die organische Phase im Vakuum eingeengt und an der Ölpumpe getrocknet. Das ölige Rohprodukt kristallisierte nach ca. 1 5 Stunden, Fp. 75 bis 78°C.

Patentansprüche:

Verbindungen der allgemeinen Formel I
in welcher
R1 , R2 und R3 Wasserstoff, R4 (CrC6)-Alkyl,
A eine -CH2-Gruppe bedeutet, in der ein Wasserstoff durch eine Methylgruppe ersetzt sein kann, und
B -CO2G bedeutet, wobei
G den Rest eines Alkohols GOH bedeutet, einschließlich der physiologisch wirksamen Salze.
2. Verbindungen der Formel I gemäß Anspruch 1 , in denen
R1 , R2 und R3 Wasserstoff,
R4 (CrC6)-Alkyl,
A eine -CH2-Gruppe bedeutet, in der ein Wasserstoff durch eine Methylgruppe ersetzt sein kann, und
B -CO2G bedeutet, wobei
G einen verzweigten oder unverzweigten oder cyclischen (C1-C20)- Alkyl-Rest, oder einen verzweigten oder unverzweigten oder cyclischen (C2-C20)-Alkenyl-Rest, einen entsprechenden (C2-C20)- Alkinyl-Rest, einen entsprechenden (C4-C20)-Alkeninyl-Rest oder einen Retinyl-Rest bedeutet, wobei die Reste jeweils eine oder mehrere Mehrfachbindungen enthalten können, oder einen
Phenylalkyl-Rest bedeutet, wobei die vorstehenden Reste insbesondere einen oder mehrere Substituenten aus der Reihe Hydroxy, Halogen, Cyano, Trifluormethyl, Carboxy, (C1 -C1 2)-Alkyl, (C3-C8)-Cycloalkyl, (C5-C8)-Cycloalkenyl, (CrC1 2)-Alkoxy, (C1-C12)-Alkoxy-(C1-C1 2)-alkyl, (CrC1 2)-Alkoxy-(CrC12)-alkoxy, Phenyl-(CrC4)-alkyloxy, (CrC8)-Hydroxyalkyl, (CrC1 2)- Alkylcarbonyloxy, (C3-C8)-Cycloalkylcarbonyloxy, Benzoyloxy, Phenyl-(C1 -C4)-alkylcarbonyloxy enthalten, einschließlich der physiologisch wirksamen Salze.
3. Verbindungen der Formel I gemäß den Ansprüchen 1 oder 2, in denen
R1 , R2 und R3 Wasserstoff,
R* (CrC4)-Alkyl,
A eine -CH2-Gruppe bedeutet, in der ein Wasserstoffatom durch eine Methylgruppe ersetzt sein kann, und
B -CO2G bedeutet, wobei
G einen verzweigten oder unverzweigten oder cyclischen aliphatischen (Cj-C^J-Alkyl-Rest, einen (C3-C8)-Cycloalkyl-(C1 -C8)- alkylrest, einen verzweigten oder unverzweigten (C2-C1 8)- Alkenylrest, wie z.B. einen Geranyl-, Farnesyl- oder einen Retinyl- Rest, oder einen entsprechenden (C2-C1 8)-Alkιnylrest, einen Benzyl-, Phenethyl-, Phenylpropyl- oder Phenylbutylrest bedeutet,
wobei die vorstehenden Reste einen Substituenten aus der Reihe Hydroxy, (CrC4)-Alkoxy, (CrC6)-Alkylcarbonyloxy, (C3-C8)- Cycloalkylcarbonyloxy, Benzoyloxy oder Phenyl-(C1 -C4)- alkylcarbonyloxy, enthalten, einschließlich der physiologisch wirksamen Salze.
4. Verbindungen der Formel I gemäß den Ansprüchen 1 bis 3, in denen
R1 , R2 und R3 Wasserstoff,
R4 (CrC4)-Alkyl,
A eine -CH2-Gruppe bedeutet, in der ein Wasserstoffatom durch eine Methylgruppe ersetzt sein kann, und
B -CO2G bedeutet, wobei
G einen verzweigten oder unverzweigten oder cyclischen aliphatischen (C1-C1 8)-Alkyl-Rest, einen (C3-C8)-Cycloalkyl-(C1-C4)- alkylrest, einen verzweigten oder unverzweigten (C2-C1 8)- Alkenylrest, einen Benzyl-, Phenethyl-, Phenylpropyl- oder Phenylbutylrest, bedeutet, einschließlich der physiologisch wirksamen Salze.
5. Verbindungen der Formel I gemäß den Ansprüchen 1 bis 4, in denen
R1 , R2 und R3 Wasserstoff,
R4 Methyl,
A eine -CH2-Gruppe bedeutet, in der ein Wasserstoff durch eine
Methylgruppe ersetzt sein kann, und B -CO2G bedeutet, wobei
G einen verzweigten oder unverzweigten (C1 -C18)-Alkyl-oder
(C2-C 1 8)-Alkenyl-Rest, bedeutet, einschließlich der physiologischen wirksamen Salze.
6. Verbindungen der Formel I gemäß den Ansprüchen 1 bis 5, in denen
R1 , R2 und R3 Wasserstoff,
R4 Methyl,
A eine -CH2-Gruppe, und
B -CO2G bedeutet, wobei
G einen verzweigten oder unverzweigten (C C^J-Alkyl- oder (C2-C18)-Alkenyl-Rest bedeutet, einschließlich der physiologisch wirksamen Salze.
7. Verbindungen der Formel I gemäß den Ansprüchen 1 bis 6, in denen
R1 , R2 und R3 Wasserstoff,
R4 Methyl,
A eine -CH2-Gruppe,
B -CO2G bedeutet, wobei
G einen linearen (C, -C18)-Alkyl-Rest bedeutet, einschließlich der physiologisch wirksamen Salze.
8. Verfahren zur Herstellung von Verbindungen der Formel I gemäß den Ansprüchen 1 bis 7, in denen A eine -CH2-Gruppe, in der ein Wasserstoff durch eine Methylgruppe ersetzt sein kann, und B CO2G bedeuten, dadurch gekennzeichnet, daß i i .) Pyridin-2-carbonsäuren der Formel II (R5 = H) mit den Aminoestern
Formel lll oder deren Salze zu den Amidestern der Formel I umgesetzt werden; oder i2.) Pyridin-2-carbonsäureester der Formel II (R5 = (CrC1 6)-Alkyl) unter den Bedingungen der Aminolyse zu den Verbindungen der
Formel I umgesetzt werden; oder
ii) die Verbindungen der Formel IV mit einem Alkohol GOH verestert werden; oder
iii) die Verbindungen der Formel V mit R X alkyliert werden, worin X eine Abgangsgruppe, insbesondere Halogen, -OSO2Me, -OSO2Phenyl u.a. bedeutet
Verbindungen der Formel I gemäß den Ansprüchen 1 bis 7 zur Hemmung
der Prolyl-4-hydroxylase in vivo.
10. Verbindungen der Formel I gemäß den Ansprüchen 1 bis 7 zur Hemmung der Kollagenbiosynthese.
1 1 . Verbindungen der Formel I gemäß den Ansprüchen 1 bis 7 zur Vermeidung/Verringerung der Ablagerungen von Bindegewebe (Kollagenen) in den erkrankten menschlichen Organen (Fibröse).
1 2. Verbindungen der Formel I gemäß den Ansprüchen 1 bis 7 zur Anwendung bei fibrotischen Erkrankungen.
1 3. Verbindungen der Formel I gemäß Anspruch 1 2 zur Anwendung bei fibrotischen Erkrankungen der Lunge, der Leber, der Niere, des Herzens, des Auges und der Haut und bei Artherosklerose.
14. Verbindungen der Formel I gemäß den Ansprüchen 1 2 und 1 3, wobei die Anwendung lokal und/oder systemisch erfolgt.
1 5. Verbindungen der Formel I gemäß Anspruch 14, zur lokalen Anwendung zur Vermeidung/Verringerung von Vernarbungen nach chirurgischen Eingriffen am menschlichen Körper.
1 6. Verbindungen der Formel I gemäß den Ansprüchen 9 bis 1 4 zui lokalen Anwendung bei erhöhter Bildung von Bindegewebe (Fibröse) in der Haut.
1 7. Verbindungen der Formel I gemäß den Ansprüchen 9 bis 14 zur lokalen Anwendung (inhalativ) bei erhöhter Bildung von Bindegewebe (Fibröse) in der Lunge.
1 8. Verbindungen der Formel I gemäß den Ansprüchen 9 bis 14 zur lokalen
Anwendung bei erhöhter Bildung von Bindegewebe (Fibröse) am Auge.
1 9. Verbindungen der Formel I gemäß den Ansprüchen 9 bis 14 zur lokalen Anwendung am Auge zur nachoperativen Behandlung des Glaukoms.
20. Verbindungen der Formel I gemäß den Ansprüchen 9 bis 14 zur lokalen Anwendung bei strahleninduzierter oder durch Chemotherapie induzierter Fibröse, insbesondere Fibrösen der Lunge.
21 . Arzneimittel, enthaltend eine oder mehrere Verbindungen der Formel I gemäß den Ansprüchen 1 bis 8.
22. Verwendung der Verbindungen zur Hemmung der Prolyl-4-hydroxylase und bei fibrotischen Erkrankungen.
