Abstract

Die Erfindung betrifft DNS-Sequenzen, die für Proteine mit Laccase-Aktivität kodieren und eine der DNS-Sequenzen (SEQ ID NO:1, SEQ ID NO:2) umfassen. Die Erfindung betrifft ferner die Expression dieser DNS-Sequenzen, die durch die DNS-Sequenzen kodierte thermophilen Laccasen sowie deren Verwendung für die Delignifizierung von Zellstoff, die Depolymerisierung hochmolekularer Aggregate, das Deinking von Altpapier, die Polymerisierung aromatischer Verbindungen in Abwässern, vor allem von ligninhaltigen Abwässern aus der Zellstoffbleiche, die Oxidation von Farbstoffen, bzw. die Aktivierung von Farbstoffen zur Pigmentbildung, die Verwendung in der organischen Synthese bei Kopplungsreaktionen von aromatischen Verbindungen oder der Oxidation aromatischer Seitenketten.

Die Erfindung betrifft DNS-Sequenzen, die für Proteine mit Laccase-Aktivität kodieren, die Expression dieser DNS-Sequenzen, die durch die DNS-Sequenzen kodierten ther ophilen Laccasen sowie deren Verwendung.
Eine Enzymklasse, die für industrielle Anwendungen von großem Interesse ist, ist die Enzymklasse der Laccasen (p-Hydroxyphe- noloxidase, EC 1.10.3.2.). Laccasen gehören zur Proteinfamilie der sogenannten "blauen Kupferproteine" und enthalten in der Regel vier Kupferionen, die in drei, Typ 1 bis Typ 3 bezeich- neten Kupferzentren angeordnet sind. Laccasen zeichnen sich dadurch aus, daß es sich allgemein um sekretierte Proteine handelt und daß sie einen Glykosylierungsanteil von 10 bis 45 % des Molekulargewichts enthalten können. Laccasen besitzen eine sehr breite Substratspezifität für aromatische Verbindun- gen, die sie oxidieren. Die bei dieser Oxidation erzeugten
Elektronen werden verwendet um Sauerstoff zu reduzieren. Dabei entsteht Wasser. Die Funktion der Laccasen, die in Weißfäulepilzen vorkommen, besteht unter anderem im Ligninabbau. Daher stammt auch das Interesse, Laccasen bei der Papierherstellung zur Delignifizierung von Zellstoff einzusetzen.
Neben der Depolymerisierung von makromolekularen Verbindungen wie Lignin können Laccasen auch die Polymerisierung vor allem aromatischer Verbindungen katalysieren. Als Beispiel hierfür dient die Ligninbiosynthese in Pflanzen, bei der in Pflanzen vorkommende Laccasen beteiligt sind. Technische Anwendungsmöglichkeiten für Laccasen ergeben sich daher auch allgemein bei Polymerisierungsreaktionen aller Art, z.B. bei der Abwasserbehandlung. Die Verwendung von Laccasen ist auch in der organi- sehen chemischen Synthese bekannt, z.B. bei Kopplungsreaktionen oder der Seitenkettenoxidation von Aromaten. Für viele dieser potentiellen Anwendungen ist es jedoch von Nachteil, daß die meisten bekannten Laccasen mesophil sind, d. h. sie besitzen ein niedriges Temperaturoptimum und eine begrenzte Temperaturstabilität .
Voraussetzung für die technische Anwendung von Laccaseenzymen ist, daß sie kostengünstig hergestellt werden können. Dies ist im allgemeinen nur durch Verwendung rekombinant hergestellter Enzyme möglich. Verschiedene prokaryontische und eukaryontische Expressionssysteme stehen für die Proteinproduktion zur Verfügung. Beispiele prokaryontischer Expressionssysteme sind Escherichia coli und Bacillus subtilis. Eukaryontische Expressionssysteme mit breiter Anwendung sind Zellkultursysteme sowohl von Säugerzellen wie auch Insektenzellen sowie eukaryontische Mikroorganismen wie Hefen oder filamentöse Pilze.
In WO96/00290 werden fünf Laccasegene aus dem filamentösen Pilz der Unterklasse der Basidiomyceten, Polyporus pinsitus, beschrieben. Eines dieser Laccasegene (LCC1) wurde als rekom- binantes Protein hergestellt. Die thermophilen Eigenschaften dieses Enzyms wurden nicht weiter untersucht, die beschriebene Verwendung der LCC1 Laccase für Zwecke der Haarfärbung läßt darauf schließen, daß dieses Enzym einen mesophilen Charakter hat.
Die Herstellung einer rekombinanten Laccase mit thermophilen Eigenschaften aus dem filamentösen Pilz der Unterklasse der Deuteromyceten, Scytalidiu thermophilum wird in WO 95/33837 beschrieben. Von diesem Enzym ist nicht bekannt, ob es sich für die Zellstoffbleiche eignet.
Es wurde noch kein Verfahren für die Herstellung als rekombi- nantes Protein einer thermophilen Laccase aus einem filamentösen Pilz aus der Unterklasse der Basidiomyceten beschrieben.
In dem CA: AN 96-203142 werden verschiedene Charakteristika einer thermophilen Laccase offenbart. DNS oder Proteinsequenz dieses Proteins sind nicht genannt. Die Erfindung betrifft eine DNS-Sequenz, die für ein Protein mit Laccaseaktivität kodiert, dadurch gekennzeichnet, daß sie die DNS-Sequenz SEQ ID NO : 1 ab Position 76 bis einschließlich Position 1572 oder SEQ ID NO : 2 ab Position 76 bis einschließ- lieh Position 1572 oder eine DNS-Sequenz mit einer Sequenzhomologie größer als 80 % zu den genannten DNS-Sequenzen umfasst .
SEQ ID NO:l und SEQ ID NO : 2 geben von Position 1 bis Position 75 die DNS Sequenz kodierend für die Signalsequenz zur Sekretion des Proteins wieder. Diese Signalsequenz läßt sich gegen beliebige andere Signalsequenzen zur Proteinsekretion austauschen.
Die erfindungsgemäße DNS-Sequenz läßt sich beispielsweise durch Klonierung aus dem Basidiomycetenstamm Trametes versico- lor TV-1 (hinterlegt bei der DSMZ Deutschen Sammlung von Mikroorganismen und Zellkulturen GmbH, D-38124 Braunschweig unter der Nummer DSM 11523) erhalten. Dazu wird mittels an sich bekannter Methoden eine Genbank von Trametes versicolor TV-1 erstellt. Es kann sich dabei um eine cDNS- oder um eine genomische Genbank handeln.
Zur Isolierung der erfindungsgemäßen DNS-Sequenz in der Gen- bank werden DNS-Sonden verwendet, die laccasespezifische DNS- Sequenzen enthalten. Solche DNS-Sonden lassen sich beispielsweise mittels PCR-Reaktion unter Verwendung von DNS-Primern aus genomischer DNS von Trametes versicolor TV-1 gewinnen.
Als Primer werden degenerierte DNS Abschnitte einer Länge von vorzugsweise 14 bis 27 bp verwendet, deren Sequenz durch Sequenzvergleich bekannter Laccasegene festgelegt wird.
Die als Primer geeigneten DNS-Abschnitte erhält man vorzugs- weise durch Oligonukleotidsynthese der festgelegten DNS- Abschnitte. Die Isolation eines erfindungsgemäßen Laccasegens kann beispielsweise wie in den Beispielen 1 bis 5 beschrieben erfolgen.
Ein beispielsweise derart isoliertes Laccasegen läßt sich mittels dem Fachmann bekannter Techniken wie beispielsweise der site directed Mutagenese an jeweils gewünschter Position der Sequenz modifizieren. Daher ist auch eine DNS-Sequenz kodierend für ein Protein mit Laccaseaktivität umfassend eine DNS- Sequenz mit einer Sequenzhomologie größer als 80 % zu der DNS- Sequenz SEQ ID NO:l ab Position 76 bis einschließlich Position 1572 oder SEQ ID NO : 2 ab Position 76 bis einschließlich Position 1572 von der Erfindung umfasst .
Zur Expression der erfindungsgemäßen DNS wird diese in an sich bekannter Art und Weise in einem Expressionsvektor kloniert, dieser das Laccasegen enthaltende Expressionsvektor in einen Mikroorganismus eingebracht und in dem Mikroorganismus exprimiert .
Bei dem Expressionsvektor kann es sich um ein DNS-Konstrukt handeln, das in das Genom des Wirtsorganismus integriert und zusammen mit diesem repliziert wird. Alternativ kann es sich bei dem Expressionsvektor um ein autonom replizierendes DNS- Konstrukt handeln, das nicht in das Wirtsgenom integriert, wie z.B. ein Plasmid, ein artifizielles Chromosom oder ein vergleichbares extrachromosomales genetisches Element.
Ein geeigneter Expressionsvektor sollte vorzugsweise folgende genetische Elemente enthalten:
Einen Promotor, der die Expression des Laccasegens in dem Wirtsorganismus vermittelt. Es sollte sich dabei vorzugsweise um einen starken Promotor handeln, damit eine hohe Expressi- onsleistung gewährleistet werden kann. Der Promotor ist vorzugsweise funktionell mit dem 5 ' -Ende des Laccasegens verknüpft . Vorzugsweise geeignete Promotoren sind ausgewählt aus der Gruppe tac-Promotor, Subtilisin-Promotor, GAL-Promotor, TAKA- Amylase-Promotor, Polyhedrin-Promotor, Glucoamylase-Promotor, gapDH-Promotor und Alkoholoxidase Promotor.
Vorzugsweise eignet sich der tac-Promotor für die Expression in E. coli, der Subtilisin-Promotor für die Expression in Bacillus, der GAL-Promotor für die Expression in der Hefe Sac- charomyces cerevisiae, der TAKA-Amylase-Promotor für die Ex- pression in Aspergillus niger, der Polyhedrin-Promotor für die Expression in Baculovirus-infizierten Insektenzellen, der Glucoamylase-Promotor aus Aspergillus niger oder der Alkoho- loxidase-Promotor aus der Hefe Pichia pastoris.
Insbesondere eignen sich der Glucoamylase-Promotor aus Aspergillus niger oder der Alkoholoxidase-Promotor aus der Hefe Pichia pastoris für die Expression der erfindungsgemäßen thermophilen Laccasen.
Ferner sollten vorzugsweise für den Wirtsorganismus passende
Signale für die Transkriptionstermination und in Eukaryonten zusätzlich Signale für die Polyadenylierung in dem Expressionsvektor enthalten und funktioneil mit dem 3 ' -Ende des Laccasegens verknüpft sein.
Vorzugsweise soll das exprimierte Protein durch den Wirtsorganismus in das Anzuchtsmedium sekretiert werden. Die Sekretion aus dem Wirtsorganismus wird durch eine N-terminale Signalsequenz vermittelt. Bei der Signalsequenz handelt es sich um die natürliche im Laccasegen enthaltene Signalsequenz oder um eine heterologe Signalsequenz, deren kodierende DNS im Expressions- vektor funktionell mit dem 5 ' -Ende des Laccasegens verknüpft ist .
Bevorzugt sind die Signalsequenzen folgender sekretierter Proteine: alpha-Cyclodextrin-Glucosyltransferase aus Klebsieila oxytoca, Subtilisin aus Bacillus subtilis, alpha-Faktor aus Saccharomyces cerevisiae, saure Phosphatase aus Pichia pastoris, alpha-Amylase aus Aspergillus niger, Glucoamylase aus Aspergillus niger bzw. aus Aspergillus awamori oder die natürlich im Laccasegen enthaltene Signalsequenz.
Insbesondere eignet sich die natürlich im Laccasegen enthaltene Signalsequenz sowie folgende heterologe Signalsequenzen: Die Signalsequenz der Glucoamylase aus Aspergillus niger, bzw. aus Aspergillus awamori, die Signalsequenz des alpha-Faktors aus Saccharomyces cerevisiae sowie der sauren Phosphatase aus Pichia pastoris.
Die Sekretion der Laccase kann auch durch die Expression eines Fusionsproteins erreicht werden, bei dem im Expressionsvektor ein Gen für ein sekretiertes Protein, bzw. für ein sekretier- tes Fragment dieses Proteins, funktioneil mit dem Laccasegen verknüpft ist. Insbesondere bevorzugt ist hier die Expression eines Fusionsproteins zwischen einem N-terminalen Fragment der Glucoamylase aus Aspergillus niger und der thermophilen Laccase .
Die Verknüpfungsstelle zwischen der Glucoamylase und der Laccase ist dabei vorzugsweise so gewählt, daß die Aminosäuresequenz dieser Verknüpfungsstelle als Erkennungsstelle für eine prozessierende Peptidase im Sekretionsapparat der Wirtszelle dient, wobei das exprimierte Fusionsprotein in vivo gespalten und die Laccase freigesetzt wird.
Des weiteren enthält der Expressionsvektor vorzugsweise das Gen für einen Selektionsmarker. Die durch das Gen kodierten Selektionsmarker können dem Wirtsorganismus entweder Resistenz gegen ein Antibiotikum verleihen oder einen Defekt im Wirtsorganismus komplementieren.
Bevorzugte Selektionsmarker sind Gene, die Resistenz gegen An- tibiotika wie Ampicillin, Kanamycin, Chloramphenicol, Tetracy- clin, Hygromycin, Zeocin oder Bialaphos verleihen. Als Selektionsmarker für die Komplementation von Wachstumsdefekten sind Gene wie amdS, pyrG, trpC, His4, niaD, argB, oder hygB bevorzugt .
Insbesondere eignen sich als Selektionsmarker das amdS und das pyrG Gen sowie das His4 Gen und das Resistenzgen für das Antibiotikum Zeocin.
Das Selektionsmarkergen kann dabei zusammen mit dem Laccasegen in einem DNS-Molekül enthalten sein oder beide Gene können ge- trennt in verschiedenen DNS-Molekülen enthalten sein. Im letzteren Fall wird der Wirtsorganismus mit beiden DNS-Molekülen gemeinsam cotransformiert .
Die Erfindung betrifft somit auch Expressionsvektoren die er- findungsgemäße DNS-Sequenzen enthalten.
Als Mikroorganismen zur Expression der erfindungsgemäßen Expressionsvektoren eignen sich vorzugsweise solche bakteriellen Ursprungs wie E. coli oder Bacillus subtilis, Mikroorganismen eukaryontisehen Ursprungs wie Hefen der Gattung Saccharomyces oder Pichia, oder aber filamentöse Pilze der Gattung Aspergillus, Trichoderma, Neurospora oder Schyzophillum oder eukaryontische Zellkulturen wie z. B. mit Baculoviren infizierte Insektenzellen .
Insbesondere eignen sich filamentöse Pilze der Gattung Aspergillus wie Aspergillus niger oder Aspergillus awamori oder Hefen wie Saccharomyces cerevisiae oder Pichia pastoris.
Die Proteine, die durch die erfindungsgemäße DNS kodiert werden haben folgende biochemischen Eigenschaften:
Die Proteine besitzen die enzymatische Aktivität von Laccasen. Das pH-Optimum der Enzymaktivität liegt im sauren Bereich und ist maximal bei pH 2.0, mit halbmaximaler Enzymaktivität bei pH 4.0.
Die Enzymstabilität bei 45°C und einem pH von 4.5 bis 6.0 beträgt bis zu einer Zeitdauer von 2 h 100 %. Die Enzymstabilitat bei 45°C und einem pH von 3.0 beträgt bis zu einer Zeitdauer von 1 h 50 %.
Das Temperaturoptimum beträgt bei einem pH von 4.5 70°C. Von 65 - 75°C beträgt die Enzymaktivität noch 80 % der maxima- len Aktivität.
Von 50 - 80°C beträgt die Enzyτnaktivität noch 50 % der maximalen Aktivität.
Die Enzymstabilität bei einem pH von 4.5 und Temperaturen bis zu 55°C beträgt bis zu einer Zeitdauer von 2 h 90 %. Die Enzymstabilitat bei einem pH von 4.5 und einer Temperatur von 65°C beträgt bis zu einer Zeitdauer von 1 h 50 %.
Die erfindungsgemäßen Proteine umfassen die Proteinsequenz SEQ ID NO: 3.
Das erfindungsgemäße Protein wird vorzugsweise durch Expression erfindungsgemäßer DNS-Sequenzen in einem vorstehend genannten Mikroorganismus hergestellt.
Vorzugsweise wird die DNS unter Verwendung eines der vorstehend genannten Expressionsvektoren im Mikroorganismus exprimiert .
Die Erfindung betrifft somit auch Mikroorganismen, die erfin- dungsgemäße DNS-Sequenzen oder erfindungsgemäße Expressionsvektoren enthalten.
Besonders bevorzugt werden Kombinationen von Mikroorganismen und Expressionssystemen verwendet, die auch die Sekretion des Proteins aus dem Mikroorganismus ermöglichen. Beispiele für solche bevorzugten Kombinationen sind:
Die Verwendung des Glucoamylase Promoters zur Expression der erfindungsgemäßen DNS-Sequenzen in Aspergillus niger oder As- pergillus awamori. Als Sekretionssignal in diesem Expressionssystem dient vorzugsweise die Signalsequenz der thermophilen Laccase selbst oder der Glucoamylaseanteil des Glucoamylase- Laccase-Fusionsproteins . Die Verwendung des Alkoholoxidasepromotors zur Expression der erfindungsgemäßen DNS Sequenzen in Pichia pastoris. Als Sekretionssignal in diesem Expressionssystem dient vorzugsweise die Signalsequenz der thermophilen Laccase selbst oder die Signal- sequenz des alpha-Faktors aus Saccharomyces cerevisiae oder aber die Signalsequenz der sauren Phosphatase aus Pichia pastoris .
Die erfindungsgemäßen Proteine eignen sich für alle Anwendungen die für Laccasen bekannt sind. Insbesondere eignen sie sich für die Delignifizierung von Zellstoff bzw. die Depolyme- risierung von hochmolekularen Aggregaten. Weitere Verwendung finden Laccasen beim Deinking von Altpapier, der Polymerisie- rung von aromatischen Verbindungen bei der Abwasserreinigung, hier vor allem von ligninhaltigen Abwässern aus der Zellstoffbleiche oder in einer erweiterten Anwendung bei der Entgiftung von verseuchten Böden. Weitere Einsatzgebiete betreffen die Oxidation von Farbstoffen bzw. die Aktivierung von Farbstoffen durch Reaktion mit Vorstufenkomponenten unter Pigmentbildung. Einsatzgebiete in der organischen Synthese umfassen Kopplungsreaktionen von aromatischen Verbindungen oder die Substituen- tenoxidation von Aromaten, hier z. B. die Oxidation von Ben- zylalkoholen zu den entsprechenden Aldehyden unter Vermeidung der weiteren Oxidation zur Carbonsäure. Bei den o. g. Verwendungen kann die Laccase für sich allein genommen in die entsprechenden Reaktionen eingesetzt werden oder aber auch durch Kombination mit einem reaktionsverstärkenden Mediator. Beispiele für solche Mediatoren sind ABTS oder N-Hydroxybenzotriazol .
Die Erfindung betrifft somit auch die Verwendung eines erfindungsgemäßen Proteins für die Delignifizierung von Zellstoff, die Depolymerisierung hochmolekularer Aggregate, das Deinking von Altpapier, die Polymerisierung aromatischer Verbindungen in Abwässern, vor allem von ligninhaltigen Abwässern aus der Zellstoffbleiche, die Oxidation von Farbstoffen, bzw. die Aktivierung von Farbstoffen zur Pigmentbildung, die Verwendung in der organischen Synthese bei Kopplungsreaktionen von aromatischen Verbindungen oder der Oxidation aromatischer Seitenketten .
Die o. g. Verwendungen können durch Einsatz der erfindungsgemäßen Laccaseproteine für sich allein genommen oder aber durch Kombination mit einem reaktionsverstärkenden Mediator ausgeführt werden.

Die folgenden Beispiele dienen der weiteren Erläuterung der Erfindung. Die in den Beispielen verwendeten Standardmethoden zur Behandlung von DNS oder RNS, wie die Behandlung mit Re- striktionsendonukleasen, DNS Polymerasen, Reverser Transkrip- tase etc. sowie die Standardverfahren wie Transformation von Bakterien, Southern und Northern Analyse, DNS-Sequenzierung, radioaktive Markierung, Screening und PCR-Technologie wurden, wenn nicht anders angegeben, durchgeführt wie vom Hersteller der verwendeten Kits empfohlen oder, wenn keine Herstelleranleitung vorhanden war, entsprechend dem aus den Standardlehrbüchern bekannten Stand der Technik.
1. Beispiel: Herstellung einer cDNS-Bank aus Trametes versicolor TV-1
Es wurde der Stamm Trametes versicolor TV-1 verwendet. Mycel von Trametes versicolor wurde zuerst durch Anzucht für 7 Tage bei 28 °C auf Malzagarplatten (3 % Malz Extrakt, 0,3 % Pepton aus Sojabohnenmehl, 1,5 % Agar-Agar, pH 5,0) gewonnen. Von den Malzagarplatten wurden drei Stücke ausgestanzt und damit 100 ml steriles Malzextrakt Medium (3 % Malz Extrakt, 0,3 % Pepton aus Sojabohnenmehl, pH 5,0) in 500 ml Erlenmeyerkolben angeimpft. Die Kultur wurde unter Schütteln bei 100 rpm für 7 Tage bei 28°C inkubiert. Die auf diese Weise hergestellte Mycelsus- pension wurde über eine Porzellannutsche abgesaugt, mit 0,9 % Saline gewaschen und das Mycel in flüssigem Stickstoff gefroren und mit Mörser und Pistill zerkleinert. RNS wurde mit einem RNeasy-Kit (Qiagen) isoliert. Die Ausbeute aus 200 mg My- cel betrug 100 μg RNS.
600 μg RNS wurden verwendet, um mRNS zu isolieren. Dies erfolgte durch Chromatographie über Oligo-dT Sepharose (mRNA- Isolierungskit , Pharmacia) . Die Ausbeute an mRNS betrug 26 μg . 7,25 μg der isolierten mRNS wurden zur Synthese von cDNS verwendet. Dazu wurde ein cDNS-Synthesekit der Fa. Stratagene verwendet. Die cDNS wurde nach Auftrennung durch Agarose Gelelektrophorese fraktioniert in einen Größenbereich von 0,8 - 2,1 kb und einen Größenbereich von 2,1 - 5 kb. Die cDNS beider Fraktionen wurde aus der Agarose isoliert (Qiagen Gelextraktionskit) und zur Herstellung der cDNS-Bank verwendet. Die cDNS- Bank wurde in Lambda-Phagen hergestellt (Stratagene, ZAP Express Klonierungssystem) . Von der 0,8 - 2,1 kb Fraktion wurden 4 x 105 Phagen/μg Vektor-DNS erhalten. Von der 2,1 - 5 kb Fraktion wurden 1 x 10 Phagen/μg Vektor-DNS erhalten. Die erhaltenen Phagen wurden durch Infektion des E. coli Stammes XL-1 Blue MRF' (Stratagene) amplifiziert . 2 . Beispiel
Herstellung einer chromosomalen Genbank von Trametes versicolor
Mycel von Trametes versicolor TV-1 wurde hergestellt wie im 1. Beispiel beschrieben. Das Mycel wurde über eine Porzellannut- sche abgesaugt und mit 0,9% Saline gewaschen, anschließend in flüssigem Stickstoff gefroren, mit Mörser und Pistill zerklei- nert und in lg Portionen aufgeteilt. Je 1 g des zerriebenen Mycels wurden in einem sterilen Probengefäß aufgenommen und sofort mit 5 ml Extraktionslösung (0,1 M Tris-HCl, pH 8.0, 0,1 M EDTA, 0,25 M NaCl , 0,6 mg/ml Proteinase K) und 0 , 5 ml einer 10 % (w/v) Natriumlauroylsarcosinlösung versetzt. Nach Inkubation bei 50 °C für mindestens 2 h wurde das Gemisch mit 0,85 ml 5 M NaCl und 0 , 7 ml einer 10 % (w/v) CTAB-Lösung in 0,7 M NaCl versetzt und 30 min bei 65°C inkubiert. Nach Zugabe von 7 ml eines Chloroform-Isoamylalkoholgemisches (24:1) wurde der Ansatz geschüttelt und die beiden Phasen durch Zentrifuga- tion getrennt. Die wassrige Phase wurde entfernt und chromoso- male DNS durch Zugabe von 0,6 Volumenanteilen Isopropanol gefällt. Die gefällte DNS wurde anschließend über eine Säule (Qiagen Genomic Tip) gereinigt. Auf diese Weise konnten aus 16 g Mycel 0 , 5 mg chromosomale DNS isoliert werden.
Zur Herstellung der chromosomalen Genbank wurden 90 μg chromosomale DNS von Trametes versicolor TV-1 vollständig mit Eco RI geschnitten und durch Agarose Gelelektrophorese aufgetrennt . Die chromosomalen DNS-Fragmente wurden im Größenbereich von 2 - 4 kb und von 4 kb - 10 kb isoliert und jeweils in Lambda- Phagen kloniert (Stratagene, ZAP Express Klonierungssystem) . Von der 2 - 4 kb DNS-Fraktion wurden 1 x 10 Phagen/μg Vektor- DNS und von der 4 - 10 kb DNS-Fraktion wurden 5.4 x 104 Pha- gen/μg Vektor-DNS erhalten. Die Phagen wurden durch Infektion des E. coli Stammes XL-1 Blue MRF ' amplifiziert . 3 . Beispiel
Herstellung einer Laccase-spezifischen DNS-Sonde aus genomischer DNS von Trametes versicolor
Eine DNS-Sonde zur Isolierung von Laccasegenen wurde durch PCR-Amplifikation aus T. versicolor genomischer DNS mit degenerierten Primern erzeugt . Die degenerierten Primer wurden anhand eines Sequenzvergleichs bekannter Laccasegene konstruiert . Es wurden die Aminosäuresequenzen der in der EMBL Gen- datenbank enthaltenen Laccasegene von Neurospora crassa, Co- riolus hirsutus, Phlebia radiata, Agaricus bisporus und eines nicht näher charakterisierten filamentösen Pilzes aus der Unterklasse der Basidiomyceten verglichen. Durch den Sequenzvergleich konnten vier Peptide mit einer Länge von 5 bis 7 Ami- nosäuren identifiziert werden, die in allen Laccasen vollständig konserviert waren. Unter Berücksichtigung degenerierter Codons wurden diese Peptide in DNA zurück übersetzt, um degenerierte Primer herzustellen. Die Primer hatten die folgenden Sequenzen:

Primer C und D enthielten am 5 ' -Ende eine Barn HI Schnittstelle (unterstrichen) und daran angeschlossen jeweils die entsprechende degenerierte Laccasesequenz .
Genomische DNS von T. versicolor wurde aus dem Mycel einer
Schüttelkolbenkultur isoliert wie im 2. Beispiel beschrieben. PCR-Amplifikationen wurden nach dem für den Fachmann geläufigen Stand der Technik durchgeführt. In einer ersten PCR-Reak- tion wurden 200 ng chromosomaler T. versicolor DNS in einer 100 μl PCR Reaktion eingesetzt, die darüberhinaus 1,25 U Taq Polymerase, 1,25 mM MgCl2, je 0,2 mM der vier dNTPs und jeweils 100 pmol der Primer A und B enthielt. Die weiteren Bedingungen zur spezifischen Amplifikation des gewünschten PCR- Produkts waren: 5 min bei 94°C, gefolgt von 7 Zyklen von 0,5 min bei 94°C, 1 min bei 40°C und 2,5 min bei 60°C sowie 30 Zyklen von 0,5 min bei 94°C, 1 min bei 50°C und 2,5 min bei 72 °C. 1 μl der ersten PCR-Reaktion wurden in einer zweiten PCR-Reaktion eingesetzt, die darüberhinaus 1,25 U Taq Polyme- rase, 1,25 mM MgCl2, je 0,2 mM der vier dNTPs und jeweils 100 pmol der Primer C und D enthielt . Die weiteren Bedingungen zur spezifischen Amplifikation des gewünschten PCR-Produkts waren: 5 min bei 94°C, gefolgt von 7 Zyklen von 0,5 min bei 94°C, 1 min bei 40°C und 2,5 min bei 60°C sowie 30 Zyklen von 0,5 min bei 94°C, 1 min bei 50°C und 2,5 min bei 72°C. Es wurde ein PCR-Produkt von ca. 1 , 1 kb erhalten. Das PCR-Produkt wurde durch Agarose Gelelektrophorese gereinigt, mit dem Restriktionsenzym Barn HI geschnitten und in einen mit Barn HI geschnit- tenen pUC18 Vektor kloniert und E. coli transformiert. Aus der Anzucht transformierter E. coli wurde das Plasmid isoliert. DNS-Sequenzanalyse vom 51- und 3 ' -Ende bestätigte, daß es sich bei dem klonierten DNS-Fragment um das Fragment eines Laccasegens handelte.
Zur Vorbereitung der DNS-Sonde für das Screening von Laccase- genen wurde das Laccase-spezifische PCR-Fragment durch Behandlung mit Bam HI ausgeschnitten, über Agarose-Elektrophorese isoliert und mit α- [ 32P] -dATP radioaktiv markiert ("Random
Priming" Kit, Boehringer Mannheim) . Freie Radioaktivität wurde durch Chromatographie über Sephadex G25 (Pharmacia) abgetrennt. Die spezifische Aktivität der radioaktiv markierten DNS-Sonde betrug 1 x 107 cpm/μg DNS.
4. Beispiel
Isolierung des cDNS-Gens einer Laccase aus Trametes versicolor
TV-1
Es wurde die im 1. Beispiel beschriebene cDNS Genbank von Trametes versicolor TV-1 verwendet. Das Screening nach Laccase cDNS-Genen wurde nach dem Stand der Technik durchgeführt. In einer ersten Screeningrunde wurden Zellen von E. coli XL-1 Blue MRF ' auf 10 Petrischalen zuerst kultiviert und dann mit 50000 Phagen der cDNS-Bank (0,8 - 2,1 kb Fraktion, siehe 1. Beispiel) pro Petrischale infiziert. Nach Inkubation bei 37°C über Nacht wurden die neu gebildeten Phagen auf Nylonfilter
(Stratagene) transferiert. Die Filter wurden dann entsprechend den Empfehlungen des Herstellers mit der radioaktiv markierten Laccase-spezifischen Sonde (siehe 3. Beispiel) hybridisiert. Die Hybridisierungstemperatur betrug 45°C in einem Hybridisie- rungspuffer mit 50% Formamidgehalt. Positive Klone wurden gepickt und durch Wiederholung des Screeningverfahrens gereinigt. Nach drei Runden der Vereinzelung wurden bei dem Screening 20 stark hybridisierende Phagenklone isoliert, die nach einer Vorschrift des Herstellers (Stratagene) durch "in vivo excision" in den pBK CMV Vektor (Stratagene) umkloniert wurden. Analyse der Klone durch Verdau mit Restriktionsendo- nucleasen und DNS-Sequenzierung ergab, daß zwei Laccasegene isoliert worden waren, die auf DNS-Ebene fast identisch waren. Die vollständige DNS-Sequenzierung der beiden Klone ergab, daß es sich um Allele eines Laccasegens handelte, deren Aminosäuresequenz identisch war. Die beiden Laccase cDNS-Gene erhielten die Bezeichnung Lac5.5 und Lac5.6. Entsprechend wurden die Plasmide mit den beiden Laccase cDNS-Genen pLac5.5 und pLac5.6 bezeichnet .
5. Beispiel
Isolierung des chromosomalen Gens einer Laccase aus Trametes versicolor TV-1
Das Screening nach chromosomalen Laccasegenen in der im 2. Beispiel beschriebenen chromosomalen Genbank von Trametes versicolor TV-1 (2 - 10 kb Fraktion, siehe 2. Beispiel) erfolgte analog dem im 4. Beispiel beschriebenen Screening nach cDNS- Klonen. Es wurde wiederum die im 3. Beispiel beschriebene, radioaktiv markierte Laccase-spezifische Sonde verwendet. Die Hybridisierungstemperatur betrug 45 °C in einem Hybridisie- rungspuffer mit 50% Formamidgehalt. Bei dem Screening wurden drei stark hybridisierende Phagenklone isoliert, die nach einer Vorschrift des Herstellers (Stratagene) durch "in vivo excision" in den pBK CMV Vektor (Stratagene) umkloniert wurden. Analyse mit Restriktionsendonucleasen führte zu dem Schluß, daß alle drei Klone identisch waren und eine Länge von ca. 7 kb hatten. Analyse durch DNS-Sequenzierung ergab, daß alle drei Klone dem chromosomalen Gen der Laccase Lac5.6 entsprachen. Es wurden der codierende Bereich eines Klons und jeweils ca. 1 kb flankierende Sequenz im 5 ' - und im 3 ' -Bereich sequenziert. (SEQ ID NO: 8)
6. Beispiel
Herstellung von DNS-Konstrukten für die Expression von Laccase
Lac5.5 in Aspergillus
cDNS der Trametes versicolor Laccase Lac5.5 wurde mit Expressionssignalen funktioneil verknüpft, die spezifisch für filamentöse Pilze aus der Familie Aspergillus sind. Folgende Genexpressionselemente aus Aspergillus wurden verwendet: a) Der Promotor für das Glucoamylase Gen (glaA) aus Aspergillus niger ( J.C. Verdoes, P.J. Punt , J.M. Schrickx, H.W. van Verseveld, A.H. Stouthamer und C.A.M.J.J. van den Hondel Transgenic Research 2 (1993), 84 - 92). b) Der glaA Promotor gefolgt von einem DNS-Fragment, das für die Signalsequenz und ein Bruchstück der reifen Glucoamylase kodiert. Daran angeknüpft wurde eine DNS-Sequenz, die für die Spaltstelle der KEX2-Protease kodiert (M.P. Broekhuij sen, I.E. Mattern, R. Contreras, J. R. Kinghorn, C.A.M.J.J. van den Hondel (1993), J. Biotechnol. 3., 135-145). c) der Transkriptionsterminator des trpC Gens aus Aspergillus nidulans (E. J. Mullaney, J. E. Hamer, M. M. Yelton und W. E. Timberlake (1985), Mol. Gen. Genet . 19 , 37-45). Für die funktioneile Verknüpfung der Lac5.5 cDNS mit den As- pergillus-Expressionssignalen war es jedoch zuerst notwendig, die 5'- und 3 ' -Bereiche des Lac5.5 cDNS Gens zu modifizieren.
A: Verknüpfung der Laccase Lac5.5 cDNS mit dem glaA Promotor: Für die weitere Bearbeitung wurde das Lac5.5 cDNS Gen in den Vektor pUC19 umkloniert . Dazu wurde das Lac5.5 cDNS Gen als 1,9 kb Eco Rl-Xba I Fragment aus dem im 1. Beispiel erhaltenen pBK CMV Vektor isoliert und in den mit Eco RI und Xba I vorgeschnittenen pUC19 Vektor subkloniert. Das dabei entstandene 4.6 kb Plasmid wurde pLac5 genannt .
Zur Modifizierung des Start-ATG Kodons des Lac5.5 cDNS Gens wurden die Primer E und F verwendet .

Mit den Primern E und F wurde im 5 ' -Bereich des Lac5.5 cDNS Gens ein 188 bp großes Fragment durch eine PCR-Reaktion ampli- fiziert. Mit den Primern G und H wurde im 3 ' -Bereich des
Lac5.5 cDNS Gens ein 110 bp großes Fragment amplifiziert . Ein 100 μl PCR-Ansatz enthielt jeweils 10 ng Lac5.5 cDNS (im pBK CMV-Vektor) , 0,5 U Tth Polymerase, 1,25 mM MgCl2, je 0,2 mM der vier dNTPs und je 140 pmol des Primerpaares E und F, bzw. des Primerpaares G und H. Die PCR-Reaktion wurde unter folgenden Bedingungen durchgeführt: 5 min bei 94°C, gefolgt von 30 Zyklen von 1 min bei 94°C, 2 min bei 50°C und 1 min bei 72°C und schließlich 7 min bei 72 °C. Das DNS-Fragment aus der PCR-Reaktion mit den Primern E und F wurde zuerst mit Eco RI und Xma I geschnitten und anschließend durch Gelelektrophorese gereinigt und in den Vektor pLac5 kloniert, der vorher mit Eco RI und Xma I geschnitten worden war. Dabei entstand der Vektor pLac51, bei dem am ATG-Codon des Translationsstarts eine Bsp HI Schnittstelle eingeführt wurde .
Das DNS-Fragment aus der PCR-Reaktion mit den Primern G und H wurde zuerst mit Eco RI und Xba I geschnitten und anschließend durch Gelelektrophorese gereinigt und in einen mit Eco RI und Xba I vorgeschnittenen pUC19 Vektor kloniert. Aus dem dabei entstanden Plasmid pLT5 wurde das ca. 100 bp große Insert mit Bbs I und Xba I herausgeschnitten. Das Bbs I - Xba I Fragment wurde schließlich in den mit Bbs I und Xba I geschnittenen Vektor pLac51 kloniert. Dadurch entstand der Vektor pLac513, der am 3 ' -Ende des Laccase cDNS Gens eine neue Afl II Schnittstelle enthielt.
Das cDNS Gen für die Trametes versicolor Laccase Lac5.5 mit den modifizierten 5 ' - und 3 ' -Bereichen wurde isoliert durch Partialverdau des Plasmids pLac513 mit Bsp HI. Dabei entstand ein 2,6 kb Fragment, das den kodierenden Bereich des Lac5.5 cDNS Gens und ca. 1 , 1 kb des pUC19 Vektors enthielt. Dieses
Fragment wurde in einem zweiten Schritt mit Afl II geschnitten und das dabei entstandene 1,5 kb große Lac5.5 cDNS Fragment isoliert. Dieses Fragment wurde in das 7,4 kb große Afl II - Nco I Fragment des Vektors pAN52-12 ligiert, der ein 4,0 kb großes Fragment des glaA Promotors aus Aspergillus niger, ein 0,7 kb großes Fragment des trpC Transkriptionsterminators aus Aspergillus nidulans und das 2,7 kb große Fragment des pUC18 Vektors enthielt. Der dabei entstandene 8,8 kb große Vektor wurde pANlacl genannt. In pANlacl war die glaA Promotorregion über eine Nco I - Bsp HI Kreuzung funktioneil mit dem Translationsstart ATG-Kodon des Laccase cDNS Gens verknüpft. B: Verknüpfung der Laccase Lac5.5 cDNS mit dem glaA Promotor über einen Austausch der N-terminalen Signalseguenz gegen ein Fragment der Glucoamylase
Für die Modifizierung des für den N-Terminus der reifen Laccase Lac5.5 kodierenden Bereichs des cDNS Gens wurden die Primer I und F verwendet .
Primer I: 5 ' -CCGGAATTCGATATCCAAGCGCGGGATCGGGCCTGTGCTCGAC-3 ' (SEQ ID NO: 13)
Unterstrichen in Primer I ist eine Eco RV Schnittstelle. Zur Modifizierung des 3 ' -Bereiches des Lac5.5 cDNS Gens wurden die Primer G und H verwendet (siehe Abschnitt A dieses Beispiels) .
Mit den Primern I und F wurde im 5 ' -Bereich des Lac5.5 cDNS Gens ein 110 bp großes Fragment durch eine PCR-Reaktion ampli- fiziert. Mit den Primern G und H wurde im 3 ' -Bereich des Lac5.5 cDNS Gens ein 110 bp großes Fragment amplifiziert . Die PCR-Reaktionen wurden, wie in Abschnitt A dieses Beispiels beschrieben, durchgeführt.
Das DNS-Fragment aus der PCR-Reaktion mit den Primern I und F wurde zuerst mit Eco RI und Xma I geschnitten und anschließend durch Gelelektrophorese gereinigt und in den Vektor pLac5 kloniert, der vorher mit Eco RI und Xmal geschnitten worden war. Dabei entstand der Vektor pLac52, bei dem im 5 ' -Bereich vor dem Kodon für die erste Aminosäure des reifen Laccaseproteins eine Eco RV Schnittstelle gefolgt von den Kodons für die Aminosäuren der Sequenz Ile Ser Lys Arg (Seq ID NO: 14) eingeführt worden waren. Diese Sequenz ist eine Erkennungsstelle für die KEX2 Protease . Der 3 ' -Bereich des Laccase cDNS Gens im Vektor pLac52 wurde modifiziert wie für den Vektor pLac51 be- schrieben. Aus dem Plasmid pLT5 wurde das ca. 100 bp große
Insert aus der PCR-Reaktion mit den Primern G und H mit Bbs I und Xba I herausgeschnitten und isoliert. Das Bbs I - Xba I Fragment wurde schließlich in den mit Bbs I und Xba I geschnittenen Vektor pLac52 kloniert. Dadurch entstand der Vektor pLac523, der am 3 ' -Ende des Laccase cDNS Gens eine neue Afl II Schnittstelle enthielt.
Das cDNS Gen für die Trametes versicolor Laccase Lac5.5 mit den modifizierten 5 ' - und 3 ' -Bereichen wurde gewonnen, indem das Plasmid pLac523 mit Eco RV und Afl II geschnitten und das dabei entstandene 1,5 kb große Lac5.5 cDNS Fragment nach Aga- rose-Gelelektrophorese isoliert worden war. Dieses Fragment wurde in das 9,3 kb große Afl II - Eco RV Fragment des Vektors pAN56-9 ligiert. Vektor pAN56-9 enthielt ein 4 , 0 kb großes Fragment des glaA Promotors aus Aspergillus niger, gefolgt von einem 2,0 kb großen Fragment, das für ein Fragment der Aspergillus niger glaA Glucoamylase kodiert, einem kurzen DNS-Ab- schnitt, der für vier Aminosäuren der KEX2 -Spaltstelle kodiert, ein 0,7 kb großes Fragment des trpC Transkriptionsterminators aus Aspergillus nidulans und das 2 , 7 kb große Fragment des pUC18 Vektors. Die Eco RV und Afl II Schnittstellen waren 3' zur KEX2-Spaltstelle angeordnet. Nach Transformation in E. coli erhielt man einen 10,8 kb großen Vektor, der pANlac2 genannt wurde. In pANlac2 war der kodierende Bereich des cDNS Gens für die reife Laccase Lac5.5 so mit dem kodierenden Bereich des Glucoamylasegens verknüpft, daß bei der Expression zuerst ein Fusionsprotein aus einem Bruchstück der Glucoamylase inklusive Signalsequenz, der Erkennungssequenz für die KEX2 Protease und der vollständigen Laccase Lac5.5 entstand. Bei der Sekretion wird dieses Fusionsprotein durch die KEX2 Protease gespalten und die reife Laccase in den Kulturüberstand sekretiert.
C: Einbau der amdS und pyrG Selektionsmarker in die Vektoren pANlacl und pANlac2
pANlacl und pANlac2 wurden linearisiert durch Verdau von je 5 μg der Vektoren über Nacht mit Not I. Darauf folgte eine Behandlung mit alkalischer Phosphatase aus Kälberdarm, Extraktion mit Phenol/Chloroform und Fällung mit Ethanol . Die Gene für die Selektionsmarker amdS (Acetamidase) und pyrG (Orotidin-5 ' -Monophosphat-Decarboxylase) waren auf dem Plasmid pAN52-ll enthalten, aus dem sie nach Verdau mit Not I als ein 6,4 kb großes Fragment isoliert werden konnten. Je 0,2 μg der linearisierten und Phosphatase-behandelten Vektoren pANlacl und pANlac2 wurden mit 0,6 μg des isolierten Not I Fragments ligiert und damit E. coli JM109 transformiert. Von Ampicillin- resistenten Kolonien aus diesen Transformationen wurde die Plasmid-DNS präpariert, mit Not I geschnitten und durch Agarose Gelelektrophorese analysiert. 6 von 8 analysierten Klonen aus der Transformation mit dem Vektor pANlacl und 5 von 7 analysierten Klonen aus der Transformation mit dem Vektor pANlac2 enthielten das 6.4 kb Not I Fragment. Die mit den Selektions- markergenen ausgestatteten Vektoren wurden als pANlaclS (Größe 15,3 kb, Fig.l) und pANlac2S (Größe 17,2 kb, Fig.2) bezeich- net . 3 der 6 erhaltenen pANlaclS Klone enthielten das Not I Fragment in der gewünschten, in Fig.l angegebenen Orientierung, 1 der 5 erhaltenen pANlac2S Klone enthielt das Not I Fragment in der gewünschten, in Fig. 2 angegebenen Orientierung.
7. Beispiel
Transformation von Aspergillus
Für die Transformation wurden die Stämme Aspergillus niger ABI.13 (pyrG") (W. van Hartingsveldt , I.E. Mattern, C.M.J. van Zeijl, P.H. Pouwels und C.A.M.J.J. van den Hondel (1987) Mol. Gen. Genet . 206, 71 - 75) und Aspergillus awamori (Stamm ATCC 11358) verwendet. Die Transformation von Aspergillus wurde nach dem Stand der Technik durchgeführt (P. J. Punt und C.A.J.J. van den Hondel (1992) , Meth. Enzymology 216, 447-457)
Protoplasten von Aspergillus wurden durch Behandlung des My- cels mit Novozyτn 234 gewonnen. In einem sterilen Erlenmeyer- kolben wurde Mycelium eines Kolbens in 15 ml einer frisch her- gestellten und sterilfiltrierten Lösung des lytischen Enzymgemisches Novozym 234 (Novo Nordisk) in OM-Medium (0,27 M Calci- umchlorid, 0,6 M NaCl) suspendiert. Das in der Enzymlösung re- suspendierte Mycelium wurde bei geringer Drehzahl (80 rpm) für 1 bis 3 h bei 30°C inkubiert. Während der Inkubation wurde die Bildung der Protoplasten im Mikroskop beobachtet. Frei bewegliche Protoplasten waren üblicherweise nach 1 h zu sehen. Nachdem eine große Zahl frei beweglicher Protoplasten erhalten worden war, wurden sie durch Filtration über Miracloth (Cal- biochem) in einem Glasfilter von restlichem Mycelium abgetrennt und sorgfältig mit STC-Medium (1,2 M Sorbitol, 50 mM Calciumchlorid, 35 mM NaCl, 10 mM Tris-HCl, pH 7,5) gewaschen. Protoplasten wurden durch Zentrifugation der Suspension in ei- nem sterilen Probengefäß isoliert (2000 rpm, 4°C, 10 min) und zweimal mit STC-Medium gewaschen. Die Konzentration von Protoplasten wurde unter dem Mikroskop in einer Zählkammer bestimmt. Die Protoplastensuspension wurde für Experimente zur Protoplastenregenerierung oder für Transformationen auf eine Konzentration von 1 x 10" Protoplasten/ml eingestellt.
Protoplasten von Aspergillus niger und Aspergillus awamori wurden mit den Plasmiden pANlaclS und pANlac2S transformiert. Beide Plasmide enthielten als Selektionsmarker das pyrG Gen (kodiert für die Orotidin 5 '-Phosphat Decarboxylase) und das amdS Gen (kodiert für Acetamidase) . In Inkubationsgefäßen von 12 ml Volumen wurden 0,1 ml Aliquots der Protoplasten mit 10 μg Plasmid-DNS versetzt und 25 min auf Eis inkubiert. Danach wurde langsam und unter wiederholtem mischen 1,25 ml einer 60 % PEG4000 Lösung (60 % PEG4000, 50 mM Calciumchlorid, 10 mM
Tris-HCl, pH 7,5) zum Transformationsansatz gegeben. Nach weiteren 20 min Inkubation bei 20°C wurden die Reaktionsgefäße mit STC-Medium aufgefüllt, gemischt und 10 min bei 4°C zentri- fugiert . Die Pellets wurden resuspendiert und auf mit Sorbitol osmotisch stabilisiertem, selektivem Medium ausplattiert. Die Platten wurden bei 30°C für 7 Tage inkubiert und auf Wachstum von Kolonien überprüft. Die Transformationsrate aus mehreren Experimenten betrug für Aspergillus niger 1 - 5 Transforman- ten/μg Plasmid DNS, für Aspergillus awamori 0,1 - 0,5 Trans- for anten/μg Plasmid DNS.
Transformanten wurden durch überimpfen auf Selektivplatten mit Acetamid gereinigt. Transformanten mit hoher Kopienzahl wurden durch plattieren auf Selektivplatten mit Acrylamid identifiziert. Acrylamid ist im Gegensatz zu Acetamid ein schlechtes Substrat für das vom amdS Gen kodierte Enzym Acetamidase und kann nur bei Transformanten mit hoher Kopienzahl des amdS Gens das Wachstum unterstützen. Transformanten, die Laccaseenzym funktioneil exprimieren können, wurden identifiziert, indem sie zuerst auf Platten mit Maltodextrin, einem Induktor für die Expression vom glaA Promotor, ausplattiert und nach zwei Tagen Wachstum bei 28°C mit ABTS-Agar (0,1 % ABTS, 1 % Agaro- se, in Mclllvainepuffer, pH 4,5) überschichtet wurden. Laccase exprimierende Transformanten wurden durch Bildung eines grünen Hofes angezeigt. Von Transformanten, die sowohl auf Acryla- midplatten gutes Wachstum zeigten als auch im Aktivitätstest mit ABTS positiv reagierten, wurden Sporensuspensionen hergestellt.
8. Beispiel
Expression der Trametes versicolor Laccase Lac5.5 in
Aspergillus Die Expression von Laccase Lac5.5 in Aspergillus wurde im
Schüttelkolbenmaßstab untersucht. Es wurde folgendes Anzuchtsmedium verwendet : Eine 5 % (w/v) Lösung von Maltodextrin in Leitungswasser wurde 20 min autoklaviert . Dann wurden zu 500 ml dieses Grundmediums 1 ml IM MgSulfat, 0,5 ml 1000 x Spuren- elementlösung, 10 ml 50 x Asp A Lösung und 5 ml 10 % (w/v) Ca- saminosäuren zugegeben. Die 1000 x Spurenelementlösung hatte folgende Zusammensetzung: In 80 ml H20 waren gelöst 2,2 g ZnS04 x 7 H20, 1,1g H3B03, 0,5 g MnCl2 x 4 H20, 0,5 g FeS04 x 7 H20, 0,17 g CoCl2 x 5 H20, 0,16 g CuS04 x 5 H20, 0,15 g Na2Mo04 x 2 H20 und 5 g EDTA. Die 50 x AspA Lösung hatte die Zusammensetzung: In 500 ml H20 waren gelöst 150 g NaN03 , 13 g KC1, 38 g KH2P04, pH 5,5 mit 10 M KOH eingestellt. Dem Medium wurde zusätzlich CuS04 x 5 H 0 in einer Endkonzentration von 0,5 mM zugesetzt.
Für Expressionsexperimente wurden 50 ml des Anzuchtsmediums in einem 300 ml Erlenmeyerkolben mit 1 x 10 Sporen/ml inokuliert. Die Anzucht erfolgte in einem Schüttelinkubator bei 30°C und 300 rpm. Während einer Woche wurden täglich Proben entnommen und die Laccaseaktivität im Kulturüberstand bestimmt. Die maximale Laccaseaktivität wurde zwischen 60 und 100 h Wachstum beobachtet und betrug zwischen 0,5 und 2,5 U/ml . Die Laccaseaktivität wurde colorimetrisch mit dem
Substrat ABTS (0,1 mM im Test) in Mclllvaine Puffer pH 4 , 5 und bei 37 °C bestimmt. Mclllvaine Puffer wurde hergestellt, indem man eine 0,1 M Zitronensäurelösung mit 0,2 M Na2HP04 -Lösung auf pH 4,5 titrierte. Die Extinktionszunähme wurde bei 420 nm (Extinktionskoeffizient von ABTS bei 420 nm: 3,6 x 104 1 x Mol x cm ) gemessen. 1 U Laccaseaktivität wurde definiert als 1 μmol ABTS-Substratumsatz/min.
9. Beispiel Expression der Trametes versicolor Laccase Lac5.5 in Pichia pastoris
Es wurde ein kommerziell erhältliches Expressionssystem der Fa. Invitrogen mit den dazugehörenden Expressionsvektoren (pPIC3 und pPIC9) und Pichia pastoris Stämmen (GS115 und KM71) verwendet. Die Pichia pastoris Stämme GS115 und KM71 sind Hi- stidin auxotroph. Die Expressionsvektoren enthielten Promotor und Terminator des Alkoholoxidasegens A0X1 aus Pichia pastoris. Zwischen diese beiden genetischen Regulationselementen wurde das cDNS Gen der Trametes versicolor Laccase Lac5.5 kloniert. Der Vektor pPIC9 enthielt, hinter dem AOX1 Promotor angeordnet, eine DNS-Sequenz, die für die Signalsequenz des al- pha-Faktor Proteins aus Saccharomyces cerevisiae kodiert, gefolgt von einem kurzen DNS-Abschnitt , der für die Erkennungs- sequenz Glu-Lys-Arg-Glu-Ala-Glu-Ala (Seq ID NO: 15) kodiert.
Für die Selektion in E. coli enthielten die Vektoren das Ampi- cillin Resistenzgen. Für die Selektion in Pichia pastoris enthielten die Vektoren das HIS4 Gen aus Pichia pastoris.
A: Konstruktion eines Laccase Lac5.5 Expressionsvektors mit der Signalsequenz von Laccase Lac5.5 Zur Amplifikation des Laccasegens wurde das Plasmid pLac5.5 und die Primer K und L verwendet. Primer F und G hatten folgende Sequenz :

Primer K enthielt die Sequenz für eine Eco RI Schnittstelle (unterstrichen) und daran angeschlossen die DNS-Sequenz für die ersten sieben Aminosäuren der Laccase Lac5.5 Signalsequenz. Primer L enthielt die Sequenz für eine Not I Schnittstelle (unterstrichen) und daran angeschlossen, in komplemen- tärer und reverser Orientierung, das Translationsstopcodon und die DNS-Sequenz für die letzten 7 Aminosäuren der Laccase Lac5.5.
PCR-Amplifikationen wurden nach dem für den Fachmann geläufi- gen Stand der Technik durchgeführt. 20 ng pLac5.5 DNS wurden in einer 50 μl PCR Reaktion eingesetzt, die darüberhinaus 0,5 U Vent Polymerase, 1 mM MgCl2, je 0,2 mM der vier dNTPs und jeweils 100 pmol der Primer K und L enthielt. Die weiteren Bedingungen zur spezifischen Amplifikation des gewünschten PCR-Produkts waren: 5 min bei 94 °C, gefolgt von 25 Zyklen von 1 min bei 94°C, 1 min bei 55°C und 2 min bei 72°C. Es wurde das erwartete PCR-Fragment der Größe von 1,5 kb erhalten. Das PCR-Fragment wurde durch Agarose-Gelelektrophorese gereinigt, mit den Restriktionsenzymen Eco RI und Not I geschnitten, mit Ethanol gefällt und in H20 aufgenommen. Der Vektor pPIC3 wurde mit Eco RI und Not I geschnitten, durch Agarose-Gelelektrophorese gereinigt, isoliert und mit alkalischer Phosphatase behandelt. Es folgte eine Extraktion mit Phenol/Chloroform (Verhältnis 3:1) und eine Ethanolfällung. In den so vorbereiteten pPIC3 -Vektor wurde das durch PCR-Amplifikation hergestellte
DNS-Fragment der Laccase Lac5.5 ligiert und damit E. coli Top 10F' Zellen (Invitrogen) transformiert. Von Ampicillin-resi- stenten Klonen wurde die Plasmid DNS isoliert und das klonierte 1,5 kb Insert durch Restriktionsverdau mit Eco RI und Not I identifiziert. Von 12 untersuchten Klonen waren 9 positiv. Der so erhaltene Vektor wurde mit dem Namen pL512 (Fig.3) bezeichnet.
B: Konstruktion eines Laccase Lac5.5 Expressionsvektors mit der Signalsequenz des alpha-Faktors aus Saccharomyces cerevisiae
Zur Amplifikation des Laccasegens wurde das Plasmid pLac5.5 und die Primer L und M verwendet . Primer M hatte folgende Sequenz :
Primer M: 5 ' -ACTCGAGAATTCGGGATCGGGCCTGTGCTCGACCTCACG-3 ' (SEQ ID NO: 18)
Primer M enthielt die Sequenz für eine Eco RI Schnittstelle (unterstrichen) und daran angeschlossen die DNS-Sequenz für die ersten neun Aminosäuren des vermuteten N-terminus der pro- zessierten Laccase Lac5.5. Der N-terminus der prozessierten
Laccase Lac5.5 war aus dem Sequenzvergleich mit anderen Laccasen abgeleitet worden.
Die Herstellung des DNS-Fragments für die prozessierte Form der Laccase Lac5.5 erfolgte, wie in Abschnitt A dieses Beispiels beschrieben, durch PCR-Amplifikation mit pLac5.5 cDNS und den Primern L und M. Der Vektor pPIC9 wurde mit Eco RI und Not I geschnitten und wie der in Abschnitt A dieses Beispiels beschriebene pPIC3 Vektor vorbereitet, mit dem durch PCR- Amplifikation hergestellten DNS-Fragment der Laccase Lac5.5 ligiert und damit E. coli Top 10F' Zellen (Invitrogen) transformiert. Von Ampicillin-resistenten Klonen wurde die Plasmid DNS isoliert und das klonierte 1,5 kb Insert durch Restriktionsverdau mit Eco RI und Not I identifiziert. Von 12 unter- suchten Klonen waren 3 positiv. Der so erhaltene Vektor wurde mit dem Namen pL532 (Fig.4) bezeichnet.
C: Transformation von Pichia pastoris Pichia pastoris Stämme GS115 bzw. KM71 wurden zuerst in 5 ml YPD-Medium (1 % Hefeextrakt, 2 % Pepton, 2 % Dextrose) bei 30 °C über Nacht kultiviert. Mit je 0,2 ml dieser Vorkultur wurden zwei Hauptkulturen von je 250 ml YPD-Medium inokuliert und wiederum bei 30°C über Nacht bis zu einer optischen Dichte (OD600 nm) von 1,3 - 1,5 kultiviert. Die Hefezellen einer 250 ml Hauptkultur wurden daraufhin abzentrifugiert (5 min 1500 x g) , zweimal mit je 200 ml H20 und einmal mit 10 ml 1 M Sorbi- toi gewaschen und schließlich in 0,5 ml 1 M Sorbitol aufgenommen .
Plasmid DNS der Vektoren pL512 oder pL532 wurden entweder mit Stu I oder mit Nsi I linearisiert , mit Ethanol gefällt und in H20 in einer Konzentration von lμg DNS/μl aufgenommen.
Ein Transformationsansatz enthielt 80 μl Pichia pastoris Zellen und 10 μg linearisierte Vektor-DNS. Transformation erfolgte durch Elektroporation bei 1500 V, 25 μF und 200 Ohm (BioRad Gene Pulser) . Die Entladungszeit betrug ca. 4,2 msec. Der Transformationsansatz wurde mit 1 ml 1 M Sorbitol versetzt, 30 min auf Eis inkubiert und anschließend in Aliquots zu je 0,3 ml auf MD-Platten ohne Histidin (1,34 % YNB, Hefe Stickstoff- base, 4 x 10 % Biotin, 1 % Dextrose, 1,5 % Agar) ausplattiert . Transformanten erschienen nach 3 - 5 Tagen Inkubation bei 30°C. Transformanten wurden durch zweimaliges Ausstreichen auf MD-Platten gereinigt. Laccaseproduzenten wurden auf MM-In- dikatorplatten identifiziert. MM- Indikatorplatten enthielten 1,34 % YNB, 4 x 10"5 % Biotin, 0,5 % Methanol, 1,5 % Agar, 1 mM ABTS und 0 , 1 mM CuSulfat. Der Induktor Methanol wurde im Deckel der Petrischalen vorgelegt und täglich erneuert, um eine Versorgung der Kolonien mit Methanol durch Diffusion zu gewährleisten. Laccaseproduzenten erzeugten nach 2 - 3 Tagen Inkubation bei 30°C einen grünen Hof.
E: Expression in Schüttelkolben:
50 ml BMGY Medium (1 % Hefeextrakt, 2 % Pepton, 0,1 M K-Phos- phat, pH 6,0, 1,34 % YNB, 4 x 10"5 % Biotin, 1 % Glycerin) wurden mit einer Laccase produzierenden Pichia pastoris Trans- formante inokuliert und 48 h bei 28°C und 300 rpm auf einem Schüttler kultiviert. Die Zellen dieser Vorkultur wurden durch Zentrifugation (10 min 1500 x g) isoliert und in 10 ml Haupt- kulturmedium (MMY, 1 % Hefeextrakt, 2 % Pepton, 1,34 % YNB, 4 x 10 % Biotin, 3 % Methanol) suspendiert. MMY-Medium wurde mit 0 , 5 mM CuSulfat Supplementiert und die Hauptkultur bei Raumtemperatur und 300 rpm auf einem Schüttler weiterkultiviert . In Abständen von 24 h wurde die Hauptkultur mit Metha- nol (0,3 ml/10 ml Medium) supplementiert . Die Produktion der rekombinanten Laccase Lac5.5 setzte nach 24 h ein. Produkti- onsraten bis zu 4 U/ml nach 190 h nach Start der Hauptkultur wurden erreicht .
10. Beispiel
Isolierung von rekombinanter Laccase Lac5.5
Rekombinante Laccase Lac5.5 wurde durch Anzucht transformierter Aspergillusstämme im Schüttelkolben gewonnen, wie im 8. Beispiel beschrieben. Laccase Lac5.5 enthaltende Kulturüberstände wurden durch Querstromfiltration konzentriert. Dazu wurden Sartocon Micro Filtrationsmodule (Sartorius) mit einer Ausschlußgrenze von 30 kD verwendet. Konzentrierte Laccase Lac5.5 wurde anschließend lyophilisiert und in 20 mM Na-Phos- phat , pH 6.0 gelöst. Die Aktivität der konzentrierten rekombinanten Laccase Lac5.5 betrug 18,6 U/ml.
Nach Querstromfiltration konzentrierte Laccase wurde gegen 20 mM BisTris-HCl, pH 6.5 dialysiert . Danach wurde eine Leitfä- higkeit von 1,5 mS/cm gemessen. Dialysierte Laccase wurde anschließend über eine Säule von DEAE Sepharose (Pharmacia) , äquilibriert in 20 mM BisTris-HCl (Auftragspuffer), pH 6.5, chromatographiert . Unter diesen Bedingungen bindet die Laccase an die DEAE Sepharose. Bei der Elution mit einem linearen Gra- dienten von 0 bis 0,5 M NaCl in Auftragspuffer wurde die Laccaseaktivität bei einer Salzkonzentration von 0,15 M NaCl wiedergefunden. Die Laccasefraktion von der DEAE Sepharosesäule wurde mit Ammoniumsulfat bis zu einer Sättigung von 20 % sowie mit Na-Acetat, pH 4.5 (Endkonzentration 20 mM) versetzt und pH 4,5 mit Essigsäure eingestellt. Die so vorbereitete Laccase- fraktion wurde dann über eine Säule von Phenylsepharose (Pharmacia) , äquilibriert in Auftragspuffer (20 mM Na-Acetat, pH 4,5, 20 % gesättigtes Ammoniumsulfat), chromatographiert . Unter diesen Bedingungen bindet die Laccase an die Phenylsepha- rose. Bei der Elution mit einem linearen Gradienten von 20 - 0 % gesättigtem Ammoniumsulfat, in 20 mM Na-Acetat, pH 4,5 wurde die Laccaseaktivität bei einer Ammoniumsulfatsättigung von 16 % wiedergefunden. Die Laccasefraktion wurde gegen 20 mM BisTris-HCl, pH 6.5 dialysiert und durch Bindung an DEAE Sepharose und anschließender Stufenelution mit 0,3 M NaCl konzentriert. Bindung und Elution mit 0,3 M NaCl erfolgten jeweils in 20 mM BisTris-HCl, pH 6.5. Die Ausbeute an Laccaseaktivität bezogen auf das Startmaterial betrug 20 %. Die isolierte Laccase wurde durch N-terminale Aminosäuresequenzierung analysiert. Die dabei bestimmte Sequenz,
Gly Ile Gly Pro Val Leu Asp Leu Thr Ile Ser Arg Ala Val (SEQ ID NO:19)
befand sich in Übereinstimmung mit dem aus der cDNS-Sequenz und Homologievergleichen abgeleiteten N-terminus der reifen Laccase Lac5.5.
11. Beispiel
Biochemische Eigenschaften von rekombinanter Laccase Lac5.5
Es wurden das pH- und Temperaturoptimum sowie die pH- und Tem- peraturstabilität von rekombinanter Laccase Lac5.5, in Aspergillus hergestellt wie im 8. Beispiel beschrieben, untersucht. Für diese Experimente wurde rekombinante Laccase Lac5.5 zuerst über Sephadex G25 (Pharmacia, PDIO-Säulen) in Mclllvaine-Puf- fer, pH 4.5 umgepuffert.
A: pH-Optimum:
Es wurden für die in Fig.5 angegebenen pH-Werte jeweils die entsprechenden Puffer durch geeignetes mischen von ungepufferten Na-Citrat- und Na-Phosphatlösungen hergestellt. Für rekombinante Laccase Lac5.5 wurde anschließend beim jeweiligen pH-Wert die Laccaseaktivität bei 37°C bestimmt. Wie aus Fig.5 hervorgeht, hat die Laccase Lac5.5 für das Substrat ABTS ein Aktivitätsoptimum im stark sauren Bereich.
B: pH-Stabilität:
Laccase Lac5.5 wurde in Mclllvaine Puffer bei pH 3 , 0 , 4,5 und
6,0 vorinkubiert (Temperatur 37°C) . Nach 0, 30, 60 und 120 min wurden jeweils Aliquots entnommen, in Mclllvaine Puffer mit pH 4,5 verdünnt und die Laccaseaktivität bei 37°C bestimmt. Bei pH 4,5 und 6,0 wurde die Laccaseaktivität durch die Vorbehandlung nicht beeinträchtigt. Bei pH 3 , 0 betrug die Halbwertszeit der Laccase zwischen 60 und 120 min. (Fig.6)
C: Temperaturoptimum:
Die Laccaseaktivität der rekombinanten Laccase Lac5.5 wurde bei den in Fig.7 angegebenen Temperaturen bestimmt. Die Laccaseaktivität wurde mit dem ABTS-Test in Mclllvaine-Puffer, pH 4,5 bestimmt. Überraschenderweise wurde dabei festgestellt, daß das Temperaturoptimum der Laccase Lac5.5 bei 70°C liegt. Die Temperaturen, bei der noch halbmaximale Laccaseaktivität gemessen wurde, liegen bei 50 °C und 80°C.
D: Temperaturstabilität:
Laccase Lac5.5 wurde in Mclllvaine Puffer pH 4,5 bei 45, 55 und 65°C vorinkubiert. Nach 0, 30, 60 und 120 min wurden jeweils Aliquots entnommen, in Mclllvaine Puffer mit pH 4,5 verdünnt und die Laccaseaktivität bei 37°C bestimmt. Bei 45°C wurde die Laccaseaktivität durch die Vorbehandlung nicht beeinträchtigt. Bei 55°C wurden nach 120 min Vorinkubation noch 80 % Restaktivität gemessen. Bei 65°C betrug die Halbwertszeit der Laccase 60 min. (Fig.8) . SEQUENZPROTOKOLL

Claims

Patentansprüche
1. DNS-Sequenz, die für ein Protein mit Laccaseaktivität kodiert, dadurch gekennzeichnet, daß sie die DNS-Sequenz SEQ ID N0:1 ab Position 76 bis einschließlich Position 1572 oder SEQ ID NO: 2 ab Position 76 bis einschließlich Position 1572 oder eine DNS-Sequenz mit einer Sequenzhomologie größer als 80 % zu den genannten DNS-Sequenzen umfasst.
2. Expressionsvektor, der eine DNS-Sequenz gemäß Anspruch 1 enthält .
3. Expressionsvektor nach Anspruch 2, dadurch gekennzeichnet, daß er zusätzlich enthält: einen Promotor, der die Expression des Laccasegens in einem Wirtsorganismus vermittelt sowie für den Wirtsorganismus passende Signale für die Transkrip- tionstermination, die funktioneil mit dem 3 ' -Ende der DNS-Sequenz gemäß Anspruch 1 verknüpft sind.
4.Expressionsvektor nach Anspruch 3, dadurch gekennzeichnet, daß der Promotor ausgewählt ist aus der Gruppe tac-Promotor, Subtilisin-Promotor, GAL-Promotor, TAKA-Amylase-Promotor, Polyhedrin-Promotor, Glucoamylase Promotor, GAPDH-Promotor und Alkoholoxidase-Promotor.
5. Expressionsvektor nach Anspruch 3 oder 4, dadurch gekennzeichnet, daß er zusätzlich eine N-terminale Signalsequenz enthält .

Expressionsvektor nach Anspruch 5, dadurch gekennzeichnet, daß die N-terminale Signalsequenz die natürliche im Laccasegen enthaltene Signalsequenz ist oder ausgewählt ist aus der Gruppe der Signalsequenzen der folgenden sekretierten Proteine: alpha-Cyclodextrin-Glucosyltransferase aus Klebsieila oxytoca, Subtilisin aus Bacillus subtilis, alpha-Faktor aus Saccharomyces cerevisiae, saure Phosphatase aus Pichia pastoris, alpha- Amylase aus Aspergillus niger oder Glucoamylase aus Aspergillus niger bzw. aus Aspergillus awamori.
7. Expressionsvektor nach Anspruch 5, dadurch gekennzeichnet, daß ein Gen für ein sekretiertes Protein, bzw. ein Genabschnitt für sekretiertes Fragment eines Proteins, funktioneil mit dem Laccasegen verknüpft ist .
8. Mikroorganismenstamm, der eine DNS-Sequenz gemäß Anspruch 1 oder einen Expressionsvektor gemäß einem der Ansprüche 2 bis 7 enthält .
9. Protein umfassend die Proteinsequenz SEQ ID NO : 3
10. Verwendung des Proteins gemäß Anspruch 9 für die Deligni- fizierung von Zellstoff, die Depolymerisierung hochmolekularer Aggregate, das Deinking von Altpapier, die Polymerisierung aromatischer Verbindungen in Abwässern, vor allem von ligninhaltigen Abwässern aus der Zellstoffbleiche, die Oxidation von Farbstoffen, bzw. die Aktivierung von Farbstoffen zur Pigmentbildung, die Verwendung in der organischen Synthese bei Kopp- lungsreaktionen von aromatischen Verbindungen oder der Oxidation aromatischer Seitenketten.
