Abstract
Die vorliegende Erfindung betrifft eine GDNF-kodierende DNA, Teile davon und GDNF-Varianten sowie gegen sie gerichtete Antikörper. Ferner betrifft die Erfindung die Verwendung der GDNF-kodierenden DNA, Teile davon und der GDNF-Varianten.
Description
GDNF-kodierende DNA, Teile davon und GDNF-Varianten
Die vorliegende Erfindung betrifft eine GDNF-kodierende DNA, Teile davon und GDNF-Varianten sowie gegen sie gerichtete Antikörper. Ferner betrifft die Erfindung die Verwendung der GDNF-kodierenden DNA, Teile davon und der GDNF-Varianten.
Neurotrophe Faktoren sind Proteine, die im Nervensystem oder in durch das Nervensystem innervierten Geweben vorliegen. Ihre Aufgabe liegt in der Versorgung von Glia- und Nervenzellen. Ferner fördern sie die Differenzierung von Neuronen. Ein von Gliazellen stammender neurotropher Faktor ist GDNF ("glial cell line- derived neurotrophic factor") . Dieser gehört zur TGF-ß ("transforming g rowth f acto r-ß " ) Su perf ami l ie . G D N F i st ei n etw a 30 kd gro ßer Differenzierungs-Faktor für die verschiedensten Neuronen. Dies sind z.B. dopaminerge Neuronen des Mittel hirns, spinal-motorische, kranial-sensorische und sympathische Neuronen wie auch noradrenerge Neuronen des Hinterhirns. Ferner fördert GDNF die Aufnahme von Dopamin durch das Mittelhirn. Des weiteren haben Versuche mit Parkinson aufweisenden Rhesusaffen gezeigt, daß eine intrazerebrale Verabreichung von GDNF eine funktionelle Wiederherstellung bewirken kann.
Dies läßt vermuten, daß über GDNF in neurodegenerative Erkrankungen, wie
Parkinson, amyotrophische Lateralsklerose und Alzheimer, eingegriffen werden kann. Ein solches Eingreifen könnte auf mehreren Ebenen, z.B. auf der Ebene der Expression des GDNF-Gens und der Prozessierung von GDNF, erfolgen. Bisher liegen allerdings über diese Ebene keine ausreichenden Erkenntnisse vor.
Der vorliegenden Erfindung liegt somit die Aufgabe zugrunde, ein Mittel bereitzustellen, mit dem die Regulation von GDNF auf molekularer Ebene untersucht und
ggfs. Wege aufgezeigt werden können, mit denen in diese Regulation eingegriffen werden kann.
Erfindungsgemäß wird dies durch die Gegenstände in den Patentansprüchen erreicht.
Gegenstand der vorliegenden Erfindung sind somit Mittel, mit denen kodierende und regulatorische Bereiche einer genomischen GDNF-DNA und ihre gegenseitigen Einflüsse untersucht werden können. Ferner sind es Mittel, mit denen die Expression, insbesondere verschiedene Expressionsformen, einer solchen
DNA, bestimmt werden können. Desweiteren sind es Mittel, die ein gezieltes Eingreifen in die Regulation dieser DNA ermöglichen können.
Die vorliegende Erfindung beruht auf den Erkenntnissen des Anmelders über die Struktur und die transkriptioneile Regulation einer genomischen GDNF-DNA. Der
Anmelder hat eine solche DNA im humanen Genom am Genlocus 5p 1 2-p1 3.1 identifiziert. Er hat diese DNA auf dem PAC Klon 24B 1 2 isoliert und charakterisiert (vgl. Fig. 1 ). Ferner hat er die Sequenz für die kodierenden und für die transkriptioneile Regulation wichtigen Bereiche der GDNF-DNA aufgeklärt (vgl. Fig. 2) . Die DNA enthält drei Exons, die für eine 3.6 kb große cDNA kodieren, sowie große 5'- und 3'-untranslatierte Regionen (UTRs) . Die 3'-UTR enthält ein polymorphes AGG-" repeat" , das allerdings in Patienten mit neurodegenerativen Erkrankungen, wie Parkinson, idiopathisches Parkinson- Syndrom, ALS, Alzheimer, spinozerebellare Ataxie oder dopa-responsive Dystonie, nicht in klinisch relevanter Häufung vorliegt. Die Transkription der
GDNF-DNA wird durch einen eine TATA-Box enthaltenden Promotor bewirkt, der vor Exon 1 liegt. Ein zweiter Promotor befindet sich vor Exon 2. Der erste Promotor kann durch verschiedene Substanzen in seiner Promotoraktivität verstärkt werden. Solche Substanzen sind z.B. der Entzündungsmediator Tetradecanoyl- 1 2-phorbol-acetat (TPA) , der Fibroblasten-Wachstumsfaktor 2
(FGF2bFGFf), cAMP wie auch das Differenzierungsmittel Retinoesäure (RA). Diese Substanzen spielen eine wichtige Rolle in Entwicklungsprozessen der Zelle
wie auch in anti-apoptotischen, durch Verletzungen induzierten Signalwegen. Des weiteren hat der Anmelder DNAs gefunden, die für GDNF-Varianten kodieren. Eine dieser DNAs umfaßt die Exons 1 , 2 und 3. Eine andere DNA umfaßt die Exons 1 , 2 und 3, wobei Exon 2 eine 78 bp große Deletion aufweist. Eine weitere DNA umfaßt die Exons 1 und 3. Die Sequenzen dieser DNAs und der entsprechenden GDNF-Varianten sind in Fig. 3 angegeben. Der Anmelder hat erkannt, daß Exon 2 für eine sekretierbare Form von GDNF notwendig ist, während Exon 3 für eine intrazelluläre Form ausreicht. Auch hat er erkannt, daß Exon 1 nicht translatiert wird.
Erfindungsgemäß werden die Erkenntnisse des Anmelders genutzt, eine GDNF- kodierende DNA bereitzustellen, umfassend die Sequenz von Fig. 2 oder eine hiervon durch ein oder mehrere Basenpaare unterschiedliche Sequenz, wobei letztere Sequenz mit der DNA von Fig. 2 hybridisiert und keine GDNF-cDNA ist. Die DNA von Fig. 2 wurde als E.coli 24B1 2 bei der DSMZ (Deutsche Sammlung von Mikroorganismen und Zellkulturen) am 20. März 1 998 unter DSM 1 2063 hinterlegt.
Der Ausdruck "eine durch ein oder mehrere Basenpaare unterschiedliche Se- quenz" umfaßt jegliche für GDNF-kodierende Sequenz, die mit der DNA von Fig.
2 hybridisiert und keine GDNF-cDNA ist. Die Sequenz kann sich von der DNA von Fig. 2 durch Additionen, Deletionen, Substitutionen und/oder Inversionen vo n ei n od er m e h rere n Basen paa ren u nterscheid e n . Der Ausd ruc k "Hybridisierung" weist auf eine Hybridisierung unter üblichen Bedingungen, insbesondere bei 20°C unter dem Schmelzpunkt der Sequenz hin. Der Ausdruck
"keine GDNF-cDNA" weist darauf hin, daß die Sequenz Elemente umfaßt, die in einer cDNA fehlen. Solche Elemente sind z.B. Intron- oder transkriptioneile Regulations-Sequenzen.
Ein weiterer Gegenstand der vorliegenden Erfindung ist eine DNA mit Promotor-
Aktivität, umfassend die in Fig. 2 als Promotorfragment 1 angegebene Sequenz oder eine hiervon durch ein oder mehrere Basenpaare unterschiedliche Sequenz,
wobei letztere Sequenz mit der ersteren hybridisiert.
Der Ausdruck "eine durch ein oder mehrere Basenpaare unterschiedliche Sequenz" umfaßt jegliche Sequenz, die mit der in Fig. 2 als Promotorfragment angegebenen Sequenz hybridisiert und eine Promotor-Aktivität aufweist. Die
Sequenz kann sich von der in Fig. 2 als Promotorfragment angegebenen Sequenz durch Additionen, Deletionen, Substitutionen und/oder Inversionen von ein oder mehreren Basenpaaren unterscheiden. Hinsichtlich des Ausdrucks "Hybridisierung " wird auf vorstehende Ausführungen entsprechend verwiesen. Der Ausdruck "Promotor-Aktivität" weist darauf hin, daß übliche Verfahren zum
Nachweis einer Promotor-Aktivität verwendet werden können. Beispielsweise kann ein Reporter-Gen, z.B. ein Luciferase-Gen, an das 3'-Ende der auf ihre Promotor-Aktivität hin zu testenden Sequenz ligiert werden. Das erhaltene DNA- Molekül kann in Zellen transfiziert und die Expression des Luciferase-Gens bestimmt werden, wodurch die Promotor-Aktivität nachgewiesen wird.
Ein weiterer Gegenstand der vorliegenden Erfindung ist eine DNA, die sich zum Nachweis von GDNF-Sequenzen eignet. Insbesondere eignet sich die DNA als Primer bzw. Primer-Paar für ein PCR-Verfahren. Eine solche DNA umfaßt eine der in Fig. 4 angegebenen Sequenzen oder eine hiervon durch ein oder mehrere
Basen unterschiedliche Sequenz, wobei letztere mit der komplementären Sequenz der in Fig. 4 angegebenen DNA hybridisiert.
Der Ausdruck "eine durch ein oder mehrere Basen unterschiedliche Sequenz" umfaßt jegliche Sequenz, die mit der komplementären Sequenz der in Fig. 4 angegebenen DNA hybridisiert. Die Sequenz kann sich von der DNA von Fig. 4 durch Additionen, Deletionen, Substitutionen und/oder Inversionen von ein oder mehreren Basen unterscheiden. Hinsichtlich des Ausdrucks "Hybridisierung" wird auf vorstehende Ausführungen entsprechend verwiesen.
Ein weiterer Gegenstand der vorliegenden Erfindung ist eine Nukleinsäure, die für eine GDNF-Variante kodiert. Die Nukleinsäure kann eine RNA oder eine DNA,
z.B. eine cDNA, sein. Bevorzugt ist eine DNA, die folgendes umfaßt:
(a) Die DNA von Fig. 3 oder eine hiervon durch ein oder mehrere Basenpaare unterschiedliche DNA, wobei letztere DNA mit der DNA von Fig. 3 hybridisiert und Exon- 1 und/oder Exon 2-Sequenzen von GDNF aufweist, oder
(b) eine mit der DNA von (a) über den degenerierten genetischen Code verwandte DNA.
Der Ausdruck "eine durch ein oder mehrere Basenpaare unterschiedliche DNA" umfaßt jegliche für GDNF-kodierende DNA, die mit der DNA von Fig. 3 hybridisiert und Exon- 1 und/oder Exon 2-Sequenzen von GDNF aufweist. Die DNA kann sich von der DNA von Fig. 3 durch Additionen, Deletionen, Substitutionen und/oder Inversionen von ein oder mehreren Basenpaaren unterscheiden . Hinsichtlich des Ausdrucks " Hybrid isierung " wird auf vorstehende Ausführungen entsprechend verwiesen.
Ein weiterer Gegenstand der vorliegenden Erfindung ist eine GDNF-Variante. Eine solche umfaßt durch Exon 2 und Exon 3 kodierte Aminosäuren. Insbesondere umfaßt eine erfindungsgemäße GDNF-Variante die Aminosäuresequenz von Fig. 3 (a) bzw. (b) oder eine hiervon durch ein oder mehrere Aminosäuren unterschiedliche Aminosäuresequenz, wobei die DNA-Sequenz der letzteren Aminosäuresequenz mit der DNA von Fig. 3 (a) bzw. (b) hybridisiert und Exon 2- und Exon 3-Sequenzen aufweist.
Der Ausdruck "eine durch ein oder mehrere Aminosäuren unterschiedliche Aminosäuresequenz" umfaßt jegliche GDNF-Aminosäuresequenz, deren DNA- Sequenz mit der DNA von Fig. 3 (a) bzw. (b) hybridisiert und Exon 2 und Exon 3-Sequenzen aufweist. Die DNA-Sequenz kann sich von der DNA von Fig. 3 (a) bzw. (b) durch Additionen, Deletionen, Substitutionen und/oder Inversionen von ein oder mehreren Basenpaaren unterscheiden. Hinsichtlich des Ausdrucks
"Hybridisierung " wird auf vorstehende Ausführungen entsprechend verwiesen.
Ein weiterer Gegenstand der vorliegenden Erfindung ist eine Kombination aus einer GDNF-Variante und einem Rezeptor, an den die GDNF-Variante bindet. Der Ausdruck "GDNF-Variante" umfaßt eine vorstehende GDNF-Variante. Ferner umfaßt er eine GDNF-Variante, die keine durch Exon 2 kodierten Aminosäuren aufweist.
Eine erfindungsgemäße DNA kann als solche oder in Kombination mit jeglicher anderen DNA vorliegen. Beispielsweise kann eine erfindungsgemäße, eine
Pro m oto r-Aktivität auf weisend e D N A i n Ko m bi n atio n m it ei ne r zu transkribierenden DNA, z.B. einem Reporter-Gen oder einem Struktur-Gen, vorliegen. Eine solche Kombination kann in üblichen Vektoren erfolgen. Für eine Kombination mit einem Reporter-Gen, z.B. einem Luciferase-Gen, bieten sich Vektoren, wie pXP1 und pAH 1409, an. Diese können dann zur Transfektion von
Zellen, wie NIH3T3 und 293-Zellen, verwendet werden. Mit der Kombination aus einer erfindungsgemäßen, eine Promotor-Aktivität aufweisenden DNA und einem Reporter-Gen ist es möglich, die Promotor-Aktivität unter den verschiedensten Bedingungen zu testen. Damit können Substanzen gefunden werden, mit denen die Promotor-Aktivität und somit die Regulation der Expression einer GDNF-DNA beeinflußt werden können. Solche Substanzen können aktivierend bzw . inhibierend sein.
Ferner kann eine erfindungsgemäße, für eine GDNF-Variante kodierende DNA in einem Expressionsvektor vorliegen. Beispiele solcher sind dem Fachmann bekannt. Im Falle eines Expressionsvektors für E. coli sind dies z.B. pGEMEX, pUC-Derivate, pGEX-2T, pET3b und pQE-8. Für die Expression in Hefe sind z.B. pY100 und Ycpad l zu nennen, während für die Expression in tierischen Zellen z.B. pKCR, pEFBOS, cDM8, pCEV4, pBC140 und Rep 7 anzugeben sind. Für die Expression in Insektenzellen eignet sich besonders der Bacculovirus-Expressions- vektor pAcSGHisNT-A.
Der Fachmann kennt geeignete Zellen, um die erfindungsgemäße, in einem Expressionsvektor vorliegende DNA zu exprimieren. Beispiele solcher Zellen umfassen die E.coli-Stämme HB101 , DH 1 , x1 776, JM 101 , JM 1 09, BL21 und SG 1 3009, den Hefe-Stamm Saccharomyces cerevisiae und die tierischen Zellen L, NIH 3T3 , FM3A, CHO, COS, Vero, HeLa und HEK293 sowie die
Insektenzellen sf9.
Der Fachmann weiß, in welcher Weise die erfindungsgemäße DNA in einen Expressionsvektor inseriert werden muß. Ihm ist auch bekannt, daß diese DNA in Verbindung mit einer für ein anderes Protein bzw. Peptid kodierenden DNA inseriert werden kann, so daß die erfindungsgemäße DNA in Form eines Fusionsproteins exprimiert werden kann.
Des weiteren kennt der Fachmann Bedingungen, transformierte bzw. trans- fizierte Zellen zu kultivieren. Auch sind ihm Verfahren bekannt, das durch die erfindungsgemäße DNA exprimierte Protein bzw. Fusionsprotein zu isolieren und zu reinigen.
Ein weiterer Gegenstand der vorliegenden Erfindung ist ein gegen ein vorstehen- des Protein bzw. Fusionsprotein gerichteter Antikörper. Ein solcher Antikörper kann durch übliche Verfahren hergestellt werden. Er kann polyklonal bzw. monoklonal sein. Zu seiner Herstellung ist es günstig, Tiere, insbesondere Kaninchen, Hühner oder Ratten für einen polyklonalen und Mäuse oder Ratten für einen monoklonalen Antikörper, mit einem vorstehenden (Fusions)protein oder Fragmenten davon zu immunisieren. Weitere "Booster" der Tiere können mit dem gleichen (Fusions)protein oder Fragmenten davon erfolgen. Der polyklonale Antikörper kann dann aus dem Serum bzw. Eigelb der Tiere erhalten werden. Für den monoklonalen Antikörper werden Milzzellen der Tiere mit Myelomzellen fusioniert.
Ein weiterer Gegenstand der vorliegenden Erfindung ist ein Kit. Ein solcher umfaßt eine oder mehrere der folgenden Komponenten:
(a) eine erfindungsgemäße DNA,
(b) eine erfindungsgemäße GDNF-Variante,
(c) ein erfindungsgemäßer Antikörper, sowie
(d) übliche Hilfsstoffe, wie Träger, Puffer, Lösungsmittel, Kontrollen, etc.
Von den einzelnen Komponenten können jeweils ein oder mehrere Vertreter vorliegen . Hinsichtlich der einzelnen Ausdrücke wird auf vorstehende Ausführungen verwiesen. Diese gelten hier entsprechend.
Die vorliegende Erfindung ermöglicht es, die Regulation von GDNF auf molekularer Ebene zu untersuchen. Mit einem erfindungsgemäßen Antikörper kann eine GDNF-Variante nachgewiesen werden. Es kann eine Beziehung der GDNF-Variante zu Geweben und/oder Funktionen hergestellt werden. Ferner kann mit einer erfindungsgemäßen GDNF-Variante ein gegen dieses Protein gerichteter Autoantikörper nachgewiesen werden. Beide Nachweise können durch übliche Verfahren, insbesondere einen Western Blot, einen ELISA, eine Immunpräzipitation oder durch Immunfluoreszenz, erfolgen. Desweiteren kann mit einer erfindungsgemäßen Nukleinsäure, insbesondere einer DNA und hiervon abgeleiteten Primern, die Organisation und die Expression des für GDNF kodierenden Gens nachgewiesen werden. Dieser Nachweis kann in üblicher
Weise, insbesondere in einem Southern Blot oder über "in situ" Hybridisierung, erfolgen.
Darüberhinaus eignet sich die vorliegende Erfindung, Maßnahmen für und gegen das Vorliegen von GDNF in Personen zu ergreifen. Mit einem erfindungsgemäßen
Antikörper kann eine GDNF-Variante inhibiert werden. Andererseits kann mit einer erfindungsgemäßen GDNF-Variante, insbesondere nach Kopplung an ein vom Körper nicht als fremd angesehenes Protein, z.B. Transferrin oder BSA, die Menge der GDNF-Variante in Personen erhöht werden. Entsprechendes kann auch mit einer erfindungsgemäßen Nukleinsäure, insbesondere einer DNA, erreicht werden, die unter die Kontrolle eines in bestimmten Geweben, z.B. Gehirn, induzierbaren Promotors gestellt wird und nach ihrer Expression zur
Bereitstellung einer GDNF-Variante in diesen Geweben führt. Darüberhinaus kann eine erfindungsgemäße Nukleinsäure, insbesondere eine DNA, auch zur Inhibierung von GDNF genutzt werden. Hierzu wird die Nukleinsäure, z.B. als Basis für die Erstellung von Anti-Sinn-Oligonukleotiden zur Expressions- Inhibierung des für GDNF kodierenden Gens verwendet. Somit stellt die vorliegende Erfindung Mittel dar, neurodegenerative Erkrankungen, wie Parkinson, amyotrophische Lateralsklerose und Alzheimer, besser zu diagnostizieren und therapeutisch eingreifen können.
Kurze Beschreibung der Zeichnungen.
Fig. 1 zeigt die Organisation einer genomischen GDNF-DNA, wobei in A) eine grobe und in B) eine detaillierte Restriktionskarte angegeben ist. Exons sind als Boxen angegeben, wobei ausgefüllte Teile dieser kodierende Bereiche und nicht-ausgefüllte Teile UTRs sind. Ferner ist die Lage des AGG-"repeat" und der Poly A-Sequenzen angegeben.
Fig.2 zeigt die Sequenzen der Exons 1, 2 und 3 einer genomischen GDNF-DNA. Ferner sind Sequenzen der Introns, des
Promotorfragments 1, des AGG-"repeat" und der Poly A- Sequenzen angegeben.
Fig.3 zeigt die DNA- und Aminosäuresequenzen für GDNF- Varianten, In Fig. 3(a) umfaßt die Variante auf der DNA-Ebene die Exons 1, 2 und 3, in Fig.3(b) die Exons 1, 2 und 3, wobei Exon 2 eine 78 bp große Deletion aufweist, und in Fig.3(c) die Exons 1 und 3. Auf der Proteinebene umfaßt die GDNF-Variante jeweils nicht den durch Exon 1 kodierten Bereich.
Fig.4 zeigt Primer-Paare, die sich für eine PCR-Reaktion zum Nachweis von GDNF-Sequenzen eignen.
Fig. 5 zeigt die Expression einer GDNF-Variante, welche die durch die
Exon 2 und 3 kodierten Bereiche umfaßt. Die GDNF-Variante wird in das Medium sekretiert.
Fig. 6 zeigt die Promotor-Aktivität des Promotorfragments 1 von Fig. 2 und ihre Verstärkung durch Substanzen.
Die vorliegende Erfindung wird durch die nachstehenden Beispiele erläutert.
Beispiel 1 : Expression einer für eine GDNF-Variante kodierenden DNA
Es wird eine erfindungsgemäße für eine GDNF-Variante kodierende DNA exprimiert, welche die Exons 1 , 2 und 3 umfaßt. Hierzu wird aus 293-Zellen Gesamt-RNA isoliert und einem RT-PCR-Verfahren unterzogen. Als Primer-Paar für das PCR-Verfahren werden die nachstehenden Primer verwendet, die eine Xhol-Restriktionsstelle aufweisen:

GDX.r: 5'-GAGCTCGAGTCAGATACATCCACACC-3'
Die erhaltene cDNA wird einer Xhol-Spaltung unterzogen und in den mit Xhol- geöffneten Expressionsvektor pBC140 bzw. Rep 7 inseriert. Es wird das Expressionsplasmid pBC1 40-GDNF bzw. Rep 7-GDNF erhalten.
Dieses wird zur Transfektion von tierischen Zellen verwendet, wobei 2 μg des Expressionsplasmids 1 , 5 x 1 06 Zellen zugegeben werden. Die Zellen werden in konditioniertem Medium kultiviert. 24 Stunden bzw. 48 Stunden nach Transfektion werden die Zellen geerntet und das Medium gesammelt. Die Zellen werden einer Ultraschall-Behandlung unterzogen, wodurch Zellysate gewonnen werden. Diese werden zusammen mit dem Medium und GDNF-Kontrolle einer 1 2
%igen Polyacrylamid-Gelelektrophorese unterzogen, der anschließend ein Blot- Verfahren folgt. Nach diesem wird die Nitrocellulosemembran über Nacht bei 4°C mit einem Kaninchen-Antikörper (Sta.Cruz) inkubiert, der den durch Exon 3 kodierten Bereich von GDNF erkennt. Die Antikörperbindung wird dann durch 2stündige Inkubation bei 4°C mit einem markierten Ziege-anti-Kaninchen-
Antikörper (Tropix) sichtbar gemacht (vgl. Fig. 5) .
Es zeigt sich, daß eine erfindungsgemäße, für eine GDNF-Variante kodierende DNA exprimiert werden kann. Ferner wird deutlich, daß eine die Exons 1 , 2 und 3 umfassende GDNF-DNA für eine GDNF-Variante kodiert, die von der Zelle sekretiert wird.
Beispiel 2: Nachweis von Promotor-Aktivität einer erfindungsgemäßen DNA
Es wird der Vektor pAH 1409 verwendet. Dieser enthält ein Reporter-Gen, nämlich ein Luciferase-Gen. Am 5'-Ende des Luciferase-Gens befindet sich ein Polylinker, in den das 1 , 1 kb große EcoRI/Pstl-Fragment (Promotorfragment 1 von Fig. 2) inseriert wird. Es wird das Expressionsplasmid pAH 1 409-GDNF erhalten.
Dieses wird zur Transfektion von tierischen Zellen verwendet, wobei 2 μg von pAH1409-GDNF 1 ,5 x 106 Zellen zugegeben werden. Ferner wird in einem Verhältnis von 1 :5 ein DNA-Konstrukt cotransfiziert, das für GFP ("Green Fluorescent Protein") kodiert. Als Kontrolle wird pAH 1 409 in Parallelversuchen transfiziert. Die Zellen werden unbehandelt bzw. mit TPA ( 1 00 ng/ml) , 8-Brom- cAMP ( 1 mM), Retinoesäure (RA 1 0//M), bFGF ( 1 0 ng/ml) oder IFNg ( 1000 Enzymeinheiten) behandelt. 44-48 Stunden nach Transfektion werden die Zellen geerntet und es wird ein Luciferase-Nachweis durchgeführt (vgl. Fig. 6) .
Es zeigt sich, daß eine erfindungsgemäße, das Promotorfragment 1 von Fig. 2 umfassende DNA eine Promotor-Aktivität aufweist und diese durch Substanzen verstärkt werden kann.
Beispiel 3: Herstellung und Reinigung einer erfindungsgemäßen GDNF-Variante
Die amplifizierte DNA von Beispiel 1 wird mit Xhol gespalten und in den mit Xhol gespaltenen Expressionsvektors pQE-8 (Qiagen) inseriert. Es wird das Expres- sionsplasmid pQ/GDNF erhalten. Ein solches kodiert für ein Fusionsprotein aus 6 Histidin-Resten (N-Terminuspartner) und dem erfindungsgemäßen GDNF von Fig. 3 (a) (C-Terminuspartner) . pQ/GDNF wird zur Transformation von E.coli SG 1 3009(vgl. Gottesman, S. et al. , J. Bacteriol. 1 48, ( 1 981 ), 265-273) verwen- det. Die Bakterien werden in einem LB-Medium mit 100μg/ml Ampiciliin und
25 yg/ml Kanamycin kultiviert und 4 h mit 60μM Isopropyl-ß-D-Thiogalactopyra- nosid (IPTG) induziert. Durch Zugabe von 6 M Guanidinhydrochlorid wird eine Lyse der Bakterien erreicht, anschließend wird mit dem Lysat eine Chromatographie (Ni-NTA-Resin) in Gegenwart von 8 M Harnstoff entsprechend der Angaben des Herstellers (Qiagen) des Chromatographie-Materials durchgeführt.
Das gebundene Fusionsprotein wird in einem Puffer mit pH 3,5 eluiert. Nach seiner Neutralisierung wird das Fusionsprotein einer 1 8 % SDS-Polyacrylamid- Gelelektrophorese unterworfen und mit Coomassie-Blau angefärbt (vgl. Thomas, J.O. und Kornberg, R.D., J.Mol.Biol. 149 ( 1 975), 709-733) .
Es zeigt sich, daß ein erfindungsgemäßes (Fusions)protein in hochreiner Form hergestellt werden kann.
Beispiel 4: Herstellung und Nachweis eines erfindungsgemäßen Antikörpers
Ein erfindungsgemäßes Fusionsprotein von Beispiel 3 wird einer 1 8 % SDS- Polyacrylamid-Gelelektrophorese unterzogen. Nach Anfärbung des Gels mit 4 M Natriumacetat wird eine ca. 30 kD Bande aus dem Gel herausgeschnitten und in Phosphat gepufferter Kochsalzlösung inkubiert. Gel-Stücke werden sedimentiert, bevor die Proteinkonzentration des Überstandes durch eine SDS-Polyacrylamid-
Gelelektrophorese, der eine Coomassie-Blau-Färbung folgt, bestimmt wird. Mit dem Gel-gereinigten Fusionsprotein werden Tiere wie folgt immunisiert:

Pro Immunisierung werden 35 μg Gel-gereinigtes Fusionsprotein in 0,7 ml PBS und 0,7 ml komplettem bzw. inkomplettem Freund's Adjuvans eingesetzt. Tag O: 1 . Immunisierung (komplettes Freund's Adjuvans)

Das Serum des Kaninchens wird im Immunoblot getestet. Hierzu wird ein erfindungsgemäßes Fusionsprotein von Beispiel 1 einer SDS-Polyacrylamid- Gelelektrophorese unterzogen und auf ein Nitrocellulosefilter übertragen (vgl. Khyse-Andersen, J. , J. Biochem. Biophys. Meth. 1 0, (1 984), 203-209). Die
Western Blot-Analyse wurde wie in Bock, C.-T. et al., Virus Genes 8, ( 1 994), 21 5-229, beschrieben, durchgeführt. Hierzu wird das Nitrocellulosefilter eine Stunde bei 37°C mit einem ersten Antikörper inkubiert. Dieser Antikörper ist das Serum des Kaninchens ( 1 : 10000 in PBS). Nach mehreren Waschschritten mit PBS wird das Nitrocellulosefilter mit einem zweiten Antikörper inkubiert. Dieser
Antikörper ist ein mit alkalischer Phosphatase gekoppelter monoklonaler Ziege Anti-Kaninchen-lgG-Antikörper (Dianova) ( 1 : 5000) in PBS. Nach 30-minütiger Inkubation bei 37 °C folgen mehrere Waschschritte mit PBS und anschließend die alkalische Phosphatase-Nachweisreaktion mit Entwicklerlösung (36μM . 5' Bromo-4-chloro-3-indolyiphosphat, 400μM Nitroblau-tetrazolium, 100mM Tris-
HCI, pH 9.5, 1 00 mM NaCI, 5 mM MgCI2) bei Raumtemperatur, bis Banden sichtbar werden.
Es zeigt sich, daß erfindungsgemäße, polyklonale Antikörper hergestellt werden können.
Immunisierungsprotokoll für polyklonale Antikörper im Huhn
Pro Immunisierung werden 40μg Gel-gereinigtes Fusionsprotein in 0,8 ml PBS und 0,8 ml komplettem bzw. inkomplettem Freund's Adjuvans eingesetzt.

Pro Immunisierung werden 1 2μg Gel-gereinigtes Fusionsprotein in 0,25 ml PBS und 0,25 ml komplettem bzw. inkomplettem Freund's Adjuvans eingesetzt; bei der 4. Immunisierung ist das Fusionsprotein in 0,5 ml (ohne Adjuvans) gelöst.

Überstände von Hybridomen werden im Western Blot getestet. Erfindungsgemäße, monoklonale Antikörper werden nachgewiesen.
Es zeigt sich, daß eine erfindungsgemäße DNA eine Promotor-Aktivität aufweist und diese durch Substanzen verstärkt werden kann.
Claims
1 . GDNF-kodierende DNA, umfassend die Sequenz von Fig. 2 oder eine hiervon durch ein oder mehrere Basenpaare unterschiedliche Sequenz, wobei letztere Sequenz mit der DNA von Fig. 2 hybridisiert und keine GDNF-cDNA ist.
2. DNA mit Promotor-Aktivität, umfassend die in Fig. 2 als Promotorfragment 1 angegebene Sequenz oder eine hiervon durch ein oder mehrere Basenpaare unterschiedliche Sequenz, wobei letztere Sequenz mit der ersteren hybridisiert.
3. DNA nach Anspruch 2, wobei die DNA in Kombination mit einer zu transkribierenden DNA vorliegt.
4. DNA nach Anspruch 3, wobei die zu transkribierende DNA ein Reporter- Gen ist.
5. DNA, geeignet zum Nachweis von GDNF-Sequenzen, umfassend eine der in Fig. 4 angegebenen Sequenzen oder eine hiervon durch ein oder mehrere Basen unterschiedliche Sequenz, wobei letztere mit der komplementä- ren Sequenz der in Fig. 4 angegebenen DNA hybridisiert.
6. DNA, kodierend für eine GDNF-Variante, umfassend:
(a) Die DNA von Fig. 3 oder eine hiervon durch ein oder mehrere Basenpaare unterschiedliche DNA, wobei letztere DNA mit der DNA von Fig. 3 hybridisiert und Exon-1 und/oder Exon 2-Sequenzen von
GDNF aufweist, oder
(b) eine mit der DNA von (a) über den degenerierten genetischen Code verwandte DNA.
7. Expressionsplasmid, umfassend die DNA nach Anspruch 6.
8. Transformante, enthaltend das Expressionsplasmid nach Anspruch 7.
9. GDNF-Variante, umfassend die Aminosäuresequenz von Fig. 3 (a) bzw. (b) oder eine hiervon durch ein oder mehrere Aminosäuren unterschiedliche Aminosäuresequenz, wobei die DNA-Sequenz der letzteren Aminosäuresequenz mit der DNA von Fig. 3 (a) bzw. (b) hybridisiert und Exon 2- und Exon 3-Sequenzen aufweist.
10. Antikörper, gerichtet gegen die GDNF-Variante nach Anspruch 9.
1 1 . Verwendung der DNA nach einem der Ansprüche 1 -6 oder der GDNF- Variante nach Anspruch 9 zur Untersuchung der Regulation von GDNF.
12. Verwendung der DNA nach einem der Ansprüche 1 -6 oder der GDNF- Variante nach Anspruch 9 zur Diagnose und/oder Therapie von neurodegenerativen Erkrankungen.
1 3. Verwendung nach Anspruch 1 2, wobei die neurodegenerativen Erkrankungen Parkinson, amyothrophische Lateralsklerose und Alzheimer umfassen.
