Definitions
- the invention relates to a device and method for the creation and maintenance of a personal cell bank, in particular for genetic analysis by DNA analysis with the features of the preamble of claim 1.
- saliva samples obtained from suspects.
- these are smears from the cheek mucosa, which remove the cell material from the mucous membrane using cotton swabs and convey it into a sample vessel.
- the cell material is isolated in the laboratory and subjected to DNA analysis using suitable preparation methods.
- the "genetic fingerprint" obtained in this way is used for comparative examinations if sperm samples or traces of blood could be secured at the crime scene.
- Cell material should be obtained from an authorized agency (pharmacies or doctor's offices). However, the archiving of a device with cell content should remain in private hands or in institutions that are interested and authorized in this procedure.
- the device should take in a cellular material (e.g. oral mucosal cells), while at the same time a preservation of the material should take place in the system, which is permanent, i.e. provides cells capable of analysis even after years have passed.
- a cellular material e.g. oral mucosal cells
- the shape of a check card 1, shown in FIGS. 1 and 3, is chosen as the base carrier.
- a perforation 3 is attached with a 0 of 20mm, which serves to hold a flat capsule 4, shown in Fig.2 and 1, which is clicked into the opening 3 with edge support.
- the flat capsule container 4 is made of a gas- and water-impermeable plastic and consists of two half-shells 5, 6, which can be referred to as upper and lower parts 5, 6.
- the shelled lower part 5 has one
- Indentation 7 from 4mm and has a circumferential, approximately 3mm recessed edge zone 8 on the outside, which has a width of 2mm.
- the lower shell edge 10 penetrating the card 1 projects beyond the upper side 2 and 2-r-m of the card.
- the shell edge 10 is provided with a circumferential groove 11 on the outside just above the card surface 2.
- the shell-like upper part 6 of the capsule 4 is designed in diameter such that it overlaps the lower shell edge 10 flush with a rounded edge 12.
- a circumferential bead 13 lying to the side in the upper part edge 12 is designed such that when the lower part 5 is pressed together with the upper part 6, this can be pressed into the circumferential groove 11 of the lower shell edge 10 by means of compressive force.
- sample material is to be obtained from the capsule 4 for analysis, this is only possible if the right edge of the card 14, shown in FIG. 2, is separated and the capsule 4 is blown up from the side by lifting force.
- the capsule 4 is to be used as a separate material carrier (without a data card).
- round data carriers adapted to the capsule shape 4 are to be made available, which are to be applied to the top and bottom of the capsule.
- This named embodiment can be used when an identity check of newborn children is desired.
- a drop of blood is applied to a fleece backing for newborns. After drying, this is fixed with a drop of methanol and, after repeated drying, placed in the capsule and clipped in.
- the device according to the invention is intended to be designed as a double chamber for this purpose in order to be able to introduce separate blood samples from mother and child into separate subjects. If a child is confused, samples brought in in this way can be used for genetic analysis at any time.
- the capsule or double capsule is designed so that the circumferential groove in the clipped state between the lower and Upper part (this results as a recess when using the card) can be used as a hanging aid, so that the capsule can be attached to the baby's name ribbon with a thread.
- the capsule surface can always be used as a data carrier.
- a separate capsule can also be used for jewelry or souvenir purposes, whereby the material training can be designed variably.
- the user determines the content of the capsule for this purely private use. This can be liquid or a solid. in the
- the swab technique was therefore adapted to the device 1 according to the invention by means of a cotton swab on the handle or also by means of round fleece disks which are grasped with anatomical tweezers.
- the mucous membrane of the tub is wiped off with cotton swabs.
- 80% alcohol is placed in the lower part 5 of the capsule 4 with a dropper, so that it is filled up to half is.
- the material-carrying swab is swiveled out in the alcoholic liquid, cell material then being transferred into the solution.
- the capsule 4 is closed immediately. If a nonwoven disk (the diameter corresponds to the inside diameter of the capsule 4) is used for cell extraction, it is inserted into the capsule with the adhering cells and drizzled with alcohol. After closing the capsule 4, the material also lies in a moist chamber in this technique and is permanently preserved. Hair root material can also be introduced into the capsule 4 consisting of the lower and upper part 5, 6. Although dry storage is common, alcohol is also recommended.
- the cell material adhering to nonwoven disks can also be deposited in a shallow recess in the card carrier.
- the recess is adapted to the diameter and thickness of the disc.
- the cell-carrying nonwoven disk is inserted so that the cell layer comes to lie downwards.
- the material carrier is covered with a polymer mixture, e.g. PVA / cellulose, sprayed.
- the material thus enclosed, including cells or hair, is permanently preserved after the polymers have dried on and can be used for gene analysis studies using a solvent, e.g. Alcohol to be washed out of the polymer layer.
- the card 15 provided as a data and capsule carrier in the device 1 according to the invention is also intended to be integrated
- Cell chamber 16 are manufactured by injection molding.
- a lower card 17 and an upper card 18 are produced in the manufacturing process and are joined together by an ultrasonic welding process.
- a groove 19 30 mm long and 3 mm wide and 1.5 mm deep will be provided. If both card halves 17.18 are joined together, there is a congruent chamber. With a card thickness of 1.5 mm, this channel, with its curvature, projects only 1.5 mm above the card surface and bottom surface 2.20.
- the chamber On the top of the card 2, the chamber has an inlet and an outlet opening. If a drop of liquid is attached to an opening (inlet opening), it is drawn into the chamber 16 by capillary force. If the chamber openings are welded or sealed on both sides, the task results in a closed vessel in which a cell sample from a suspension can be enclosed and made available for later DNA analysis.
- this chamber 21 can be designed as a half-channel, the volume of the socket being reduced by half.
- the injection molding card 15 is on the front 2 again as
- Disk and on the back can be used as an advertising medium.
- This device can be used advantageously if e.g. the oral mucosa cells of cotton wool are to be transferred into an alcoholic medium.
- the chamber is then filled directly from the sample container using a pipette.
- the donor will also decide whether his photo should be placed on the front 2.
- Cards equipped in this way can also have medical data, such as blood group (rhesus factor), medical measure to be initiated in an emergency, for example in the case of diabetes, etc.
- the entire surface of the card can be covered with a layer of cellulose, so that in addition to the personal data and the genetic material, a fingerprint of the person can also be applied to this.
- This embodiment of the card dispenses with depressions and hole cutouts, because the cell material is also printed directly on the surface and fixed by means of a polymer overspray.
- the carrier card in this embodiment is covered with a self-adhesive transparent film immediately after the material applications over the entire visible surface. The cells are recovered for genetic analysis by cutting out the carrier field, pulling off the cover film and washing off the cell material, e.g. with an alcoholic solution.
