Definitions

  • the present invention relates to new uses of the heat shock protein gp96.
  • Heat shock proteins are proteins that are upregulated in their expression during heat shock or in response to stress. On the one hand, they act as chaperones, ie they are involved in the folding process of proteins, and they also play a major role in the field of immunology in what is known as antigen processing.
  • APCs antigen-presenting cells
  • MHC major histocompatibility complex
  • the other presenting cells include, for example, dendritic cells, B cells (B lymphocytes) and macrophages, these are also referred to as professional APCs.
  • protein antigens are digested with the help of proteolytic enzymes to form peptides, which are then loaded onto the MHC molecules.
  • Peptides for MHC class I are primarily generated in the cytoplasm and bound to the MHC molecules in the endoplasmic reticulum.
  • Peptides for MHC class II are created by enzymatic digestion in lysosomes.
  • the loaded MHC molecules are then transported to the cell surface.
  • T cells recognize antigenic peptides together with these classic MHC molecules.
  • APCs can be activated by various reagents, for example by bacterial components such as lipopolysaccharide (LPS). Such components cause fever in mammals, which is why they are also called pyogens.
  • LPS lipopolysaccharide
  • Other activators include cytokines such as TNF-alpha, which is generally used in research today to activate APCs. It has long been known that the heat shock protein gp96 can mediate a tumor-specific immune response if it has been isolated from the tumor against which the response is to be directed.
  • a known tumor is induced in a mouse, this tumor is removed and gp96 is isolated from the tumor. If this gp96 is now injected as part of an immunotherapy of a mouse, which is also affected by the same tumor, it can be seen that the tumor is rejected. As part of an immunization, however, the mouse is injected with gp96 and then the tumor is implanted or induced. It turns out that the tumor cannot grow. gp96 from non-tumor tissue cannot mediate this immune response.
  • the tumor specificity is therefore not based on gp96 itself, but on small peptides (antigens) that are associated with gp96. These peptides provide the immune system with the information it needs to recognize the tumor. In today's model, gp96 is taken up by the antigen presenting cells (APCs).
  • APCs antigen presenting cells
  • the antigen peptide bound to gp96 is transferred to MHC molecules and presented on the surface in the context of these MHC molecules. This "cross presentation" enables the specific recognition of the antigen peptide by T cells, which are thereby activated.
  • Exogenous antigens include, for example, proteins, bacteria and apoptotic cells.
  • gp96 therefore serves as an antigen carrier.
  • other heat shock proteins such as hsp70, hsp90, hspllO and grpl70. Since the immunization with gp96 works successfully even when extremely small amounts are used, it has already been speculated in the literature that APCs are specific see receptors that bind and endocytate gp96 and other heat shock proteins on the surface.
  • the ER-internal heat shock protein gp96 has the best documented history regarding the induction of specific CTL (cytotoxic T lymphocyte) responses.
  • the sequence for human gp96 is e.g. from the publication by R.A. Mazzarella and M. Green (Erp99, an abundant, conserved glyco-protein of the endoplasmatic reticulum, is homologous to the 90- kDa heat shock protein (hsp90) and the 94-kDa glucose regulated protein (GRP94); J.Biol.Chem 262: 8875-8883 (1987)) and the sequence for mouse gp96 for example from the publication by R.G. Maki, L.J. Old and P.K. Srivastava (Human homologue of murine tu or reje ⁇ tion antigen gp96: 5 'regulatory and coding regions and relationship to stress-induced proteins; Proc. Natl. Acad. Sei. U.S.A. 87: 5658-5662 (1990)).
  • a method for treating or preventing cancer and infectious diseases with sensitized APCs is known from WO 97/10002.
  • the APCs are first sensitized in vitro with a complex of a heat shock protein and an attached antigen molecule and then administered to the patient in an effective amount.
  • gp96-FITC fluorescein isothiocyanate
  • the present invention is based on the object of providing at least one new use for gp96.
  • the object is achieved according to the invention in that gp96 molecules which do not carry any antigen peptides of interest are used for labeling and / or activating antigen-presenting cells (APCs).
  • APCs antigen-presenting cells
  • interesting antigen peptides refer to peptides that are not presented by the antigen-presenting cells (APCs) or that at least do not trigger an immune response.
  • APCs antigen-presenting cells
  • the gp96 molecules that do not carry any antigen peptides of interest are referred to as "unloaded" gp96 molecules.
  • gp96 can not only act as an antigen carrier, it is surprisingly also unloaded, ie not complexed with antigen peptides, capable of APCs, in particular dendritic cells (DCs) and B cells, so that they in turn are even better able to activate T cells.

  • unloaded gp96 activates human and mouse APCs. This can be seen from the marker molecules that are upregulated on the surface when, for example, DCs are activated: CD86 (B7.2), MHC class II and (only with human DCs) CD83.

  • the cytokine IL-12 in particular is becoming increasingly important.

In addition to the expression of maturation markers on the cell surface, gp96 also induces the secretion of the inflammation-promoting cytokines IL-12 and TNF-alpha (FIG. 2B). • Example 5 Mature DCs express reduced levels of the gp96 receptor
  • DCs The maturation of DCs induces up-regulation of MHC class II, CD83 and CD86 molecules, which leads to increased T cell proliferation. In addition, once activated, the DCs are unable to continue receiving gp96-mediated stimuli. As shown in Figure 4, all CDllc positive mouse DCs bind gp96 but not ovalbumin. However, only immature DCs that express low levels of CD86 are able to bind gp96. Since gp96 is complexed with peptides from the cell from which it was isolated and DCs are able to cross-present these peptides on MHC class I molecules, mature DCs can no longer be expected to be able to Present gp96-associated peptides for T cells.
  • B cells were isolated from spleen cells from C57BL / 6 mice by negative magnetic depletion (MACS TM, Miltenyi Biotech) with the aid of an antibody against CD43, which had been conjugated to super-paramagnetic microbeads (Miltenyi Biotech).
  • CD43 is expressed by all spleen cells with the exception of resting, peripheral B cells.
  • the B cells are activated by adding 50 ⁇ g / ml gp96 to 300,000 B cells.
  • As a negative control traded B cells ("medium”), as a positive control 2 ⁇ g / ml lipopolysaccharide (LPS, Sigma-Aldrich).
  • heat-inactivated gp96 (20 min at 95 ° C, "gp96 cooked") is also tested. The evaluation is carried out either after 2 or 3 days in flow cytometry (FACSCalibur, Becton-Dickinson) by measuring the surface molecules CD45R / B220, CD86 and MHC class II with the aid of fluorescence-labeled antibodies (Becton-Dickinson Pharmigen). As shown in Figure 5, primary mouse B cells are activated by unloaded gp96. This is shown by the stronger expression of the surface molecules CD45R / B220, CD86 and MHC class II (H2-Ab) compared to the expression of these antigens in cells treated only with medium (“medium”). Heat-inactivated gp96 ("gp96 cooked"), like medium, has no activation effect, which means that it can be excluded that LPS contamination in the gp96 preparation is responsible for the observations.
  • FACSCalibur Becton-Dickinson
  • the invention provides the first proof that gp96 is not only a peptide carrier which directs the associated peptides to professional APCs, but is also a direct activator of the DCs and B cells. Gp96 could therefore act as a hazard signal when released from necrotic or stressed cells that deliver both non-specific and specific stimuli to the immune system.
  • the DCs lose the ability to incorporate new gp96-complexed peptides and gain the ability to communicate efficiently with T cells that are specific to the MHC / peptide complexes presented. This situation is very similar to that initially observed for the presentation of soluble antigens on MHC class II molecules, where it has been described that DCs are "locked" in a state of antigen presentation and are highly efficient in activating the T cells ,
  • This new feature of gp96 provides an additional, previously unknown explanation for its high immunogenicity and will allow its use in inducing specific immune responses to be improved in vivo.
