Abstract

Die Erfindung betrifft ein Verfahren zur Herstellung einer aromatischen Carbonylverbindung durch Oxidation einer am Aromaten gebundenen Methyl- oder Methylengruppe, bei dem der Aromat in Gegenwart eines Oxidationsmittels und eines Sulfoxids umgesetzt wird wobei ein Sulfoxid für die Oxidation eines spezifischen Aromaten ausgewählt wird, dessen lonisationspotential in der Höhe vom lonisationspotential des zu oxidierenden Aromaten um maximal ± 0,25 eV abweicht.

Description

Verfahren zur Herstellung aromatischer Carbonylverbindunqen
[0001] Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung einer aromatischen Carbonylverbindung durch Oxidation einer am Aromaten gebundenen Methyl- oder Methylengruppe, bei dem der Aromat in Gegenwart eines Oxidationsmittels und eines Sulfoxids umgesetzt wird.
Technologischer Hintergrund und Stand der Technik
[0002] Aromatische Carbonylverbindungen (Aldehyde, Ketone) sind wichtige Intermediate bzw. Zielprodukte in der Zwischenprodukte- und Feinchemikalienindustrie. Neben klassischen Syntheseverfahren, zum Beispiel unter Nutzung von Benzalchloriden, werden umweltschonende Verfahren der direkten Oxidation von Alkylaromaten mit Luftsauerstoff industriell angewandt. Es sind Flüssigphasenverfahren, zum Beispiel zur Herstellung von Benzaldehyd, als auch Gasphasenverfahren, zum Beispiel zur Synthese von Methoxybenzaldehyd oder para-Chlorbenzaldehyd, bekannt (siehe US 4,054,607, EP 0 723 949). Diese Verfahren sind allerdings nicht allgemein anwendbar und leiden zum Teil unter niedrigen Aktivitäten, geringen Selektivitäten und der Bildung von Nebenprodukten. Insbesondere dann, wenn sich weitere Substituenten am aromatischen Ringsystem befinden, ist die Gefahr der Bildung von Nebenprodukten hoch.
[0003] Aus WO 2004/043891 ist ein katalytisches Oxidationsverfahren zur Herstellung von aliphatischen und aromatischen Carbonylverbindungen aus Ausgangsstoffen bekannt, die mindestens eine aliphatische und/oder aromatisch gebundene funktionelle Gruppe der Formel -[-CH(R)-X]n mit R = H, Alkyl oder Aryl und X = H aufweisen. Die Oxidation wird in Gegenwart eines Oxidationsmittels und eines in katalytischen Mengen zugesetzten Sulfoxids oder Sulfids durchgeführt. Die Sulfoxide bzw. Sulfide fungieren in oxidativer Umgebung als Sauerstoffüberträger. Es werden Dialkyl-, Alkylaryl- und Diarylsulfoxide sowie deren Gemische bzw. entsprechende Sulfide erwähnt. Es werden keine Angaben zu Umsatz, Selektivität und Raumzeitausbeute in Bezug auf Kombinationen bestimmter Alkylverbindungen mit bestimmten Sulfoxiden bzw. Sulfiden gemacht.
[0004] Ferner ist aus der Doktorarbeit von F. Antognoli (ETH Zürich, 2003) bekannt, dass für die Beurteilung der Reaktivität verschieden substituierter Aromaten das lonisationspotenzial herangezogen werden kann, lonisationspotenziale substituierter alkylaromatischer Verbindungen unterscheiden sich von dem des unsubstituierten Toluols signifikant und somit hat Art und Ort der Substitution Einfluss auf die Oxidationsgeschwindigkeit bzw. die Selektivität der Reaktion. Das lonisationspotenzial der Aromaten ist entscheidend für die Bildung von Radikalkationen R-H+*, des ersten Aktivierungsschrittes in Oxidationsreaktionen diesen Typs. Da nur die wenigsten einfachen Alkylaromaten bzw. entsprechende Oxidationsprodukte über tabellarisierte lonisationspotenziale verfügen, wurden für weitere Verbindungen quantenchemische Rechnungen durchgeführt.
[0005] Des weiteren sind in oben genannter Arbeit auch lonisationspotenziale von Lösungsmitteln, zum Beispiel auch Dimethylsulfoxid (DMSO) angegeben. Es wurde überraschend gefunden, dass in DMSO teils hohe Selektivitäten für die gewünschten Oxidationsprodukte auftraten. Weiterhin wurde festgestellt, dass Methylaromaten, welche in guten Ausbeuten in die Aldehyde oxidiert werden können, lonisationspotenziale besitzen, die unter dem von DMSO liegen. Methylaromaten mit höheren Oxidationspotenzialen als DMSO lassen sich dagegen nicht bzw. nur schlecht oxidieren.
[0006] Aufgabe der vorliegenden Erfindung ist es, Ausbeuten und Selektivitäten in der beschriebenen katalytischen Oxidation der Alkylaromaten zu aromatischen Aldehyden mit Hinsicht auf eine industrielle Anwendung weiter zu verbessern.
[0007] Die Erfindung geht aus von einem Verfahren zur Herstellung einer aromatischen Carbonylverbindung durch Oxidation einer am Aromaten gebundenen Methyl- oder Methylengruppe, bei dem der Aromat in Gegenwart eines Oxidationsmittels umgesetzt wird.
[0008] Der Erfindung liegt die Erkenntnis zugrunde, dass sich überraschenderweise die Ausbeute an Aldehyden und die Selektivität des Oxidationsverfahrens steigern lassen, wenn zusätzlich ein Sulfoxid anwesend ist, wobei die lonisationspotenziale des zu oxidierenden Aromaten und des Sulfoxids aneinander angeglichen sind. Wird eine maximale Abweichung der lonisationspotenziale von ± 0,25 eV eingehalten, so werden Ausbeute und Selektivität bezüglich der Aldehyde erheblich gesteigert. Im Vergleich zu Kombinationen mit höheren Abweichungen der lonisationspotenziale sind Selektivitätssteigerungen von 50-100 % möglich. Je näher die lonisationspotentiale von Sulfoxid und Aromat aneinander angeglichen sind, um so höher ist in der Regel die Ausbeute und Selektivität bezüglich der Aldehyde. Ohne sich auf eine Theorie festlegen zu wollen, kann vermutet werden, dass die Bildung von Radikalkationen im ersten Aktivierungsschritt der Oxidationsreaktion eine maßgebliche Rolle spielt.
[0009] Die Erfindung löst somit die obige Aufgabe durch ein Verfahren zur Oxidation von Alkylaromaten mit einem Oxidationsmittel in Anwesenheit eines Sulfoxids, wobei ein Sulfoxid für die Oxidation eines spezifischen Aromaten ausgewählt wird, dessen lonisationspotenzial der Höhe nach vom lonisationspotenzial des zu oxidierenden Aromaten um maximal ± 0,25 eV, vorzugsweise um maximal ± 0,2 eV, insbesondere um maximal ± 0,1. eV abweicht.

[00011] Sofern mehr als ein Substituent R1 vorhanden ist, also b = 2, 3 oder 4 ist, können die Substituenten R1 gleich oder verschieden sein, d.h. unabhängig voneinander gewählt werden.

[00015] Besonders bevorzugte Verbindungen der Formel I sind p,p'-Dimethyl- biphenyl, p,p'-Dimethyl-biphenylether, p-Methoxytoluol, 4-Methyl-2'-cyano- biphenyl, p-Xylol, p-Bromtoluol, p-Chlortoluol, Terephthalaldehyd und p- Cyanotoluol.
[00016] Besonders bevorzugte Verbindungen der Formel III sind 1 ,1'-Dinaphthyl- sulfoxid, Di-tert-butyl-sulfoxid, Phenyl-benzylsulfoxid, Naphthyl-methyl-sulfoxid, Diisopropyl-sulfoxid, Diphenylsulfoxid, Bis(p-chlorphenyl)-sulfoxid, Phenyl-methyl- sulfoxid, Diethylsulfoxid, Dimethylsulfoxid, Trifluormethyl-phenyl-sulfoxid, Bis(monofluormethyl)-sulfoxid, Trifluormethyl-methyl-sulfoxid und
Bis(trifluormethyl)-sulfoxid.
[00017] Als Oxidationsmittel können Peroxoverbindungen, wie Peroxodisulfat, Oxon, Wasserstoffperoxid, Alkylperoxide, Persäuren, sowie molekularer Sauerstoff verwendet werden. Bevorzugt ist Peroxodisulfat.
[00018] Schließlich ist bevorzugt, wenn die lonisierungspotenziale der Sulfoxide sowie der zu oxidierenden Aromaten nach der B3LYP-(Becke-3-Parameter-Lee- Yang-Parr)-Methode bestimmt werden.
[00019] Molekülorbital-Berechnungen basieren auf der LCAO (linear combination of atomic orbitale) Methode. Ziel von quantenchemischen Molekülorbital-Verfahren ist stets die Ermittlung der Energie von Molekülen. Diese werden als Ensemble aus im Raum angeordneten Atomkernen und umgebenden Elektronen angesehen. Die Elektronen werden in quantenmechanischer Näherung durch Wellenfunktionen beschrieben. Diese Wellenfunktionen werden durch Basissätze (z.B. 3-21 G, 6-31 G*) angenähert, je größer der Basissatz, um so genauer ist die Berechnung. B3LYP/6-31G* gibt Methode und Basissatz an, mit denen die Berechnung durchgeführt werden soll. Die B3LYP-(Becke-3-Parameter-Lee-Yang- Parr)-Methode ist ein Hybrid-Dichtefunktional-Theohe-(DFT)-Verfahren, das für kleine Moleküle sehr gute Ergebnisse liefert.
[00020] In den nachfolgenden Tabellen 1 und 2 sind lonisierungspotenziale für die bevorzugten Aromaten und Sulfoxide zusammengefasst, die nach der B3LYP- Methode mit dem Basissatz 6-31 G* bestimmt wurden. Anzumerken ist, dass die in der NIST-Datenbank hinterlegten Werte für lonisationspotenziale nicht nach standardisierten Verfahren ermittelt wurden und daher fehlerbehaftet sein können. Zudem liegen sie nicht vollständig vor. lonisationspotenzial gemäß

Tabelle 3 nennt einige erfindungsgemäß bevorzugte Kombinationen.

[00022] Es hat sich überraschenderweise gezeigt, dass sich das Oxidationsverfahren mit hohen Ausbeuten und guter Selektivität durchführen lässt, wenn auf die in Tabelle 3 genannten Paare aus aromatischem Edukt und Sulfoxid zurückgegriffen wird.
[00023] Die Reaktion wird ansonsten in der dem Fachmann bekannten Weise durchgeführt. Temperatur, Lösungsmittel usw. werden an den zu oxidierenden Aromaten angepasst. Die Reaktion wird im Allgemeinen bei Temperaturen von 40- 100 0C, Molverhältnissen Aromat : Sulfoxid von 25 : 1 bis 1 : 1 vorzugsweise 10:1 bis 1 ,1 :1 , insbesondere 5:1 bis 1 ,2:1 , Aromat : Oxidationsmittel von 1 : 50 bis 1 : 1 und in einem Zeitraum von 1 bis 10 Stunden durchgeführt. Als Lösungsmittel werden stark polare Lösungsmittel wie Wasser, Acetonitril, Nitromethan, Essigsäure, DMF und deren Gemische, bevorzugt Wasser und Acetonitril verwendet.
[00024] Die folgenden Beispiel sollen die Erfindung näher veranschaulichen, ohne sie jedoch auf die konkret beschriebenen Ausführungsformen zu beschränken.
Beispiel 1 : Herstellung von p-Chlorbenzaldehyd aus p-Chlortoluol a) System p-Chlortoluol / Dimethylsulfoxid (nicht erfindungsgemäß)
[00025] In einen mit Argon gespülten, auf Reaktionstemperatur (700C) vorgewärmten 500 ml 3-Halskolben werden unter Rühren 5,06 g (40 mmol) 4- Chlortoluol, 90 ml Acetonitril und 2 ml (28,2 mmol) Dimethylsulfoxid (DMSO) gegeben. Dazu werden 187,3 mg (0,75 mmol) CuSO4 x 5H2O und 81 ,8 mg (0,30 mmol) FeSO4 x 7H2O gelöst in 15 ml Wasser hinzugefügt. Die Lösung wird 10 min. stark gerührt. Anschließend wird unter weiterem starken Rühren innerhalb 60 min. eine Lösung von 22 g Na2S2O8 (92,4 mmol) in 60 ml Wasser mittels einer Spritzenpumpe zugegeben. Die Reaktionslösung wird dann 3h weiter bei 70 0C gerührt. [00026] Nach Ende der Reaktion und Abkühlung auf Raumtemperatur wird Ethylacetat hinzugefügt, das zweiphasige Gemisch getrennt und die wässrige Phase erschöpfend mit weiterem Ethylacetat extrahiert.
[00027] Die vereinigten organischen Phasen werden gesammelt und über Na2SO4 getrocknet. Die Produktzusammensetzung wird mit GC bzw. HPLC bestimmt.
[00028] Die Ausbeute an p-Chlorbenzaldehyd beträgt 38 %. Der Unterschied in den lonisationspotenzialen beträgt 0,3 eV.
b) System p-Chlortoluol / (Methyl-phenyl)-Sulfoxid (erfindungsgemäß)
[00029] In einen mit Argon gespülten, auf Reaktionstemperatur (700C) vorgewärmten 500 ml 3-Halskolben werden unter Rühren 5,06 g (40 mmol) 4- Chlortoluol, 90 ml Acetonitril und 3,96 g (28,2 mmol) (Methyl-phenyl)-Sulfoxid gegeben. Dazu werden 187,3 mg (0,75 mmol) CuSO4 x 5H2O und 81 ,8 mg (0,30 mmol) FeSO4 x 7H2O gelöst in 15 ml Wasser hinzugefügt. Die Lösung wird 10 min. stark gerührt. Anschließend wird unter weiterem starken Rühren innerhalb 60 min. eine Lösung von 22 g Na2S2O8 (92,4 mmol) in 60 ml Wasser mittels einer Spritzenpumpe zugegeben. Die Reaktionslösung wird dann 3h weiter bei 70 0C gerührt.
[00030] Nach Ende der Reaktion und Abkühlung auf Raumtemperatur wird Ethylacetat hinzugefügt, das zweiphasige Gemisch getrennt und die wässrige Phase erschöpfend mit weiterem Ethylacetat extrahiert.
[00031] Die Ausbeute an p-Chlorbenzaldehyd beträgt 58 %. Gegenüber der Reaktion mit DMSO als Sulfoxid, liegt die Ausbeute damit um den Faktor 1 ,53 höher. Der Unterschied in den lonisationspotenzialen beträgt hier nur 0,1 eV. c) System p-Chlortoluol / Bis(p-Chlorphenyl)-Sulfoxid (erfindungsgemäß)
[00032] In einen mit Argon gespülten, auf Reaktionstemperatur (700C) vorgewärmten 500 ml 3-Halskolben werden unter Rühren 5,06 g (40 mmol) p- Chlortoluol, 90 ml Acetonitril und 7,64 g (28,2 mmol) Bis(p-chlorphenyl)-Sulfoxid gegeben. Dazu werden 187,3 mg (0,75 mmol) CuSO4 x 5H2O und 81 ,8 mg (0,30 mmol) FeSO4 x 7H2O gelöst in 15 ml Wasser hinzugefügt. Die Lösung wird 10 min. stark gerührt. Anschließend wird unter weiterem starken Rühren innerhalb 60 min. eine Lösung von 22 g Na2S2Os (92,4 mmol) in 60 ml Wasser mittels einer Spritzenpumpe zugegeben. Die Reaktionslösung wird dann 3h weiter bei 70 0C gerührt.
[00033] Nach Ende der Reaktion und Abkühlung auf Raumtemperatur wird Ethylacetat hinzugefügt, das zweiphasige Gemisch getrennt und die wässrige Phase erschöpfend mit weiterem Ethylacetat extrahiert.
[00034] Die Ausbeute an p-Chlorbenzaldehyd beträgt 50 %. Gegenüber der Reaktion mit DMSO als Sulfoxid, liegt die Ausbeute damit um den Faktor 1,32 höher. Der Unterschied in den lonisationspotenzialen beträgt hier 0,2 eV.
d) System p-Chiortoluol / (Trifluormethyl-phenyl)-Sulfoxid (Vergleich)
[00035] In einen mit Argon gespülten, auf Reaktionstemperatur (700C) vorgewärmten 500 ml 3-Halskolben werden unter Rühren 5,06 g (40 mmol) 4- Chlortoluol, 90 ml Acetonitril und 5,48 g (28,2 mmol) (Trifluormethyl-phenyl-)- Sulfoxid gegeben. Dazu werden 164,8 mg (0,66 mmol) CuSO4 x 5H2O und 60 mg FeSO4 x 7H2O (0,22 mmol) gelöst in 15 ml Wasser hinzugefügt. Die Lösung wird 10 min. stark gerührt. Anschließend wird unter weiterem starken Rühren innerhalb 60 min. eine Lösung von 22 g Na2S2O8 (92,4 mmol) in 60 ml Wasser mittels einer Spritzenpumpe zugegeben. Die Reaktionslösung wird dann 3h weiter bei 70 °C gerührt. [00036] Nach Ende der Reaktion und Abkühlung auf Raumtemperatur wird Ethylacetat hinzugefügt, das zweiphasige Gemisch getrennt und die wässrige Phase erschöpfend mit weiterem Ethylacetat extrahiert.
[00037] Die Ausbeute an p-Chlorbenzaldehyd beträgt 14 %. Gegenüber der Reaktion mit DMSO als Sulfoxid, liegt die Ausbeute nur bei einem Faktor von 0,37. Der Unterschied in den lonisationspotenzialen beträgt hier 0,6 eV.
Beispiel 2: Herstellung von p-Brombenzaldehyd aus p-Bromtoluol a) System p-Bromtoluol / Dimethylsulfoxid (nicht erfindungsgemäß)
[00038] In einen mit Argon gespülten, auf Reaktionstemperatur (7O0C) vorgewärmten 500 ml 3-Halskolben werden unter Rühren 6,84 g (40 mmol) 4- Bromtoluol, 80 ml Acetonitril und 1 ,8 ml (25,4 mmol) Dimethylsulfoxid (DMSO) gegeben. Dazu werden 149,8 (0,60 mmol) CuSO4 x 5H2O und 95,5 mg (0,35 mmol) FeSO4 x 7H2O gelöst in 15 ml Wasser hinzugefügt. Die Lösung wird 10 min. stark gerührt. Anschließend wird unter weiterem starken Rühren innerhalb 60 min. eine Lösung von 21 g Na2S2Os (88,2 mmol) in 70 ml Wasser mittels einer Spritzenpumpe zugegeben. Die Reaktionslösung wird dann 1 h weiter bei 70 °C gerührt.
[00039] Nach Ende der Reaktion und Abkühlung auf Raumtemperatur wird Methylenchlorid hinzugefügt, das zweiphasige Gemisch getrennt und die wässrige Phase erschöpfend mit weiterem Methylenchlorid extrahiert.
[00040] Die Ausbeute an p-Brombenzaldehyd beträgt 34 %. Der Unterschied in den lonisationspotenzialen beträgt 0,4 eV.
b) System p-Bromtoluol / Diphenylsulfoxid (erfindungsgemäß)
[00041] In einen mit Argon gespülten, auf Reaktionstemperatur (70°C) vorgewärmten 500 ml 3-Halskolben werden unter Rühren 6,84 g (40 mmol) 4- Bromtoluol, 80 ml Acetonitril und 5,14 g (25,4 mmol) Diphenylsulfoxid (IP=8,2/8,6) gegeben. Dazu werden 149,8 (0,60 mmol) CuSO4 x 5H2O und 95,5 mg (0,35 mmol) FeSO4 x 7H2O gelöst in 15 ml Wasser hinzugefügt. Die Lösung wird 10 min. stark gerührt. Anschließend wird unter weiterem starken Rühren innerhalb 60 min. eine Lösung von 21 g Na2S2Os (88,2 mmol) in 70 ml Wasser mittels einer Spritzenpumpe zugegeben. Die Reaktionslösung wird dann 1 h weiter bei 70 °C gerührt.
[00042] Nach Ende der Reaktion und Abkühlung auf Raumtemperatur wird Methylenchlorid hinzugefügt, das zweiphasige Gemisch getrennt und die wässrige Phase erschöpfend mit weiterem Methylenchlorid extrahiert.
[00043] Die Ausbeute an p-Brombenzaldehyd beträgt 66 %. Gegenüber der Reaktion mit DMSO als Sulfoxid, liegt die Ausbeute damit um den Faktor 1,94 höher. Der Unterschied in den lonisationspotenzialen beträgt hier 0,2 eV.
c) System p-Bromtoluol / Di(tert.-butyl)-Sulfoxid (Vergleich)
[00044] In einen mit Argon gespülten, auf Reaktionstemperatur (700C) vorgewärmten 500 ml 3-Halskolben werden unter Rühren 6,84 g (40 mmol) 4- Bromtoluol, 80 ml Acetonitril und 4,12 g (25,4 mmol) Di(tert.-butyl)-Sulfoxid (IP=7,9/8,2) gegeben. Dazu werden 149,8 (0,60 mmol) CuSO4 x 5H2O und 95,5 mg (0,35 mmol) FeSO4 x 7H2O gelöst in 15 ml Wasser hinzugefügt. Die Lösung wird 10 min. stark gerührt. Anschließend wird unter weiterem starken Rühren innerhalb 60 min. eine Lösung von 21 g Na2S2O8 (88,2 mmol) in 70 ml Wasser mittels einer Spritzenpumpe zugegeben. Die Reaktionslösung wird dann 1 h weiter bei 70 0C gerührt.
[00045] Nach Ende der Reaktion und Abkühlung auf Raumtemperatur wird Methylenchlorid hinzugefügt, das zweiphasige Gemisch getrennt und die wässrige Phase erschöpfend mit weiterem Methylenchlorid extrahiert. [00046] Die Ausbeute an p-Brombenzaldehyd beträgt 15 %. Gegenüber der Reaktion mit DMSO als Sulfoxid liegt die Ausbeute nur bei einem Faktor von 0,44. Der Unterschied in den lonisationspotenzialen beträgt hier 0,5 eV.

Claims

Patentansprüche
1. Verfahren zur Herstellung einer aromatischen Carbonylverbindung durch Oxidation einer am Aromaten gebundenen Methyl- oder Methylengruppe, bei dem der Aromat in Gegenwart eines Oxidationsmittels und eines Sulfoxids umgesetzt wird, dadurch gekennzeichnet, dass eine Kombination aus Sulfoxid und Aromat ausgewählt wird, deren lonisationspotentiale, berechnet nach Becke-3-Parameter-Lee-Yang-Parr Methode, nicht mehr als ± 0,25 eV voneinander abweichen.
2. Verfahren nach Anspruch 1 , bei dem der Aromat eine Verbindung der Formel (I)
ist , wobei a = 1 - 3 und b = 0 - 4 ist;
A für ein ein- oder mehrkerniges aromatisches Ringsystem steht, das auch Heteroatome enthalten kann und
R1 für CrCi5-Alkyl, Halogen, Halogenalkyl, Alkoxy, Carboxy, Alkoxycarbonyl, Cyano, Amino, Amido, Sulfonyl, unsubstituiertes oder substituiertes Phenyl sowie unsubstituiertes oder substituiertes Phenoxy steht.
3. Verfahren nach Anspruch 2, bei dem A für eine aromatische Gruppe der Formel (II) steht
C(6+4m)-nNnH(6+2m)-x-y-n (H) mit m = 0 - 2 und n = 0 - 2 sowie x = 1 - 3 und y = 1 - 4, wobei (x + y) < ((6+4m)-n) ist.
4. Verfahren nach einem der vorhergehenden Ansprüche, bei dem das Sulfoxid eine Verbindung der Formel (III) ist
R2 - SO - R3 (III) wobei R2 und R3 unabhängig voneinander für substituierte oder unsubstituierte Reste gewählt aus der Gruppe umfassend Alkyl, Aryl, Naphthyl und Biphenyl stehen.
5. Verfahren nach Anspruch 4, bei dem R2 oder R3 einen oder mehrere Substituenten aus der Gruppe Alkyl, Halogen, Halogenalkyl, Methoxy, Carboxy, Alkoxycarbonyl, Cyano, Amino, Amido, Sulfonyl, Phenyl und Phenoxy tragen.
6. Verfahren nach einem der vorhergehenden Ansprüche, bei dem ein Sulfoxid ausgewählt wird, dessen lonisationspotential der Höhe nach vom lonisationspotential des zu oxidierenden Aromaten um nicht mehr als ± 0,2 eV, insbesondere um nicht mehr als ± 0,1. eV abweicht.
