Pharmazeutische Formulierung für die Herstellung von schnell zerfallenden Tabletten Beschreibung
Die vorliegende Erfindung betrifft pharmazeutische Formulierungen in Form von Granulaten für die Herstellung von schnell zerfallenden Tabletten, enthaltend nicht filmbildende Zucker oder Zuckeralkohole, filmbildende Zucker oder Zuckeralkohole, quervernetztes Polyvinylpyrrolidon und wasserunlösliche Polymere.
Im Mund schnell zerfallende und/oder sich schnell auflösende Tabletten gewinnen für die orale Applikation von Arzneistoffen immer größere Bedeutung. Solche Tabletten müssen innerhalb kurzer Zeit, am besten innerhalb von 30 Sekunden in der Mundhöhle zerfallen, angenehm schmecken und dürfen kein sandiges Gefühl hinterlassen. Ferner sollen sie einfach herstellbar sein, wobei die Direkttablettierung erhebliche Vorteile gegenüber der Feuchtgranulation bietet, und eine hohe mechanische Festigkeit besitzen, damit sie Verpackungsprozeduren, Transporte und auch das Herausdrücken aus Verpackungen unbeschadet überstehen.
Die bisher beschriebenen Produkte und Verfahren erfüllen diese Anforderungen nicht oder nur sehr unzureichend.
Schnell zerfallende Tabletten bestehen häufig aus Zucker und Zuckeralkoholen, Brausesyste- men, mikrokristalliner Cellulose und anderen nicht wasserlöslichen Füllstoffen wie Calcium- hydrogenphosphat, Cellulose-Derivaten, Maisstärke, oder Polypeptiden. Weiterhin kommen wasserlösliche Polymere, übliche Sprengmittel (quervernetztes PVP, Na- und Ca-Salz der quervernetzten Carboxymethylcellulose, Na-Salz der Carboxymethylstärke, niedrigsubstituierte Hydroxypropylcellulose L-HPC) und im wesentlichen anorganische wasserunlösliche Bestand- teile (Kieselsäuren, Silikate, anorganische Pigmente) zum Einsatz. Weiterhin können die Tabletten auch Tenside enthalten.
In der WO 2003/051338 ist eine direkttablettierbare und gut verpressbare Hilfsstoffformulierung, welche Mannitol und Sorbitol enthält, beschrieben. Zunächst wird durch Lösen von Mannitol und Sorbitol in Wasser und anschließender Sprühtrocknung (gewöhnliche Sprühtrocknung und SBD-Verfahren) eine Hilfsstoffvormischung hergestellt. Dieser coprozessierten Mischung kann zusätzlich Mannitol zugesetzt werden. Tabletten, welche zusätzlich Sprengmittel, Trennmittel, Pigment und einen Wirkstoff enthalten, sollen innerhalb von 60 Sekunden in der Mundhöhle zerfallen.
In der US 2002/0071864 A1 wird eine Tablette beschrieben, welche innerhalb von 60 Sekunden in der Mundhöhle zerfällt, und hauptsächlich aus einer physikalischen Mischung von sprühgetrocknetem Mannitol und einem grobkörnigen quervernetztem Polyvinylpyrrolidon sowie einer begrenzten Auswahl an Wirkstoffen formuliert ist. Diese Tabletten besitzen eine Bruchfestigkeit von ca. 40N und erzeugen ein unangenehmes, sandiges Mundgefühl.
Gemäß der US 6,696,085 B2 soll ein Methacrylsäure-Copolymer Typ C als Zerfallsmittel eingesetzt werden. Das Methacrylsäure-Copolymer Typ C ist ein magensaft-resistentes Polymer, welches im sauren pH-Bereich nicht löslich ist, im pH-Bereich von 7, wie er in der Mundhöhle vorliegt, aber wasserlöslich ist. Die Tabletten weisen neben einer niedrigen Bruchfestigkeit (<20N) eine hohe Friabilität (>7%) auf und beinhalten einen hohen Anteil im Bereich von
15Gew.-% eines grobkörnigen Sprengmittels. Sie besitzen folglich eine niedrige mechanische Festigkeit und erzeugen aufgrund des hohen Anteils an grobkörnigem Sprengmittel ein unangenehmes, sandiges Mundgefühl. EP 0839526 A2 beschreibt eine pharmazeutische Darreichungsform bestehend aus einem Wirkstoff, Erythritol, kristalliner Cellulose und einem Sprengmittel. Weiterhin wird Mannitol eingearbeitet und als Sprengmittel quervernetztes Polyvinylpyrrolidon verwendet, sodass eine physikalische Mischung entsteht. Die Tabletten sollen innerhalb von 60 Sekunden in der Mundhöhle zerfallen.
In der Anmeldung JP 2004-265216 wird eine im Mund innerhalb von 60 Sekunden zerfallende Tablette, bestehend aus einem Wirkstoff, einem wasserlöslichen Polyvinylalkohol- Polyethylenglykol-Copolymer, Zucker/Zuckeralkohol (Mannitol) und Sprengmittel, beschrieben. In der WO 2007071581 werden Zubereitungen beschrieben, die aus einem Zucker oder Zuckeralkohol und quervernetztem Polyvinylpyrrolidon bestehen, die mit einer wässrigen Dispersion eines wasserunlöslichen Polymers agglomeriert werden. Nachteilig bei diesen Zubereitungen ist, dass daraus hergestellte Tabletten bei Lagerung bei erhöhter Temperatur und erhöhter Feuchte erhebliche Verlängerungen der Zerfallszeit erfahren. Zudem benötigen die Zubereitun- gen relativ hohe Mengen an Schmiermittel in der Tablettierung und sie neigen zum Kleben an den Stempeln.
Aufgabe der vorliegenden Erfindung war es, im Mund schnell zerfallende Tabletten zu finden, die ein angenehmes Mundgefühl hinterlassen, mechanisch sehr stabil sind, bei Lagerung bei erhöhter Feuchte und Temperatur keine Zerfallszeitverlängerungen erfahren und in der Tablettierung keine Klebetendenzen zeigen.
Demgemäß wurde eine pharmazeutische Zubereitung gefunden, welche aus Granulaten, ent- haltend
a) 60 - 96 Gew.-% nicht filmbildende Zucker oder Zuckeralkohole,
b) 1 - 10 Gew.-% filmbildende Zucker oder Zuckeralkohole
c) 3 - 25 Gew.-% Sprengmittel,
d) 0 - 10 Gew.-% wasserunlösliche, filmbildende Polymere
e) 0 - 15 Gew.-% weitere pharmazeutisch übliche Hilfsstoffe, besteht, wobei die Summe der Komponenten a) bis e) 100 Gew.-% beträgt.
Weiterhin wurde ein Verfahren zur Herstellung solcher Zubereitungen in Form von Granulaten gefunden. Die Granulate eignen sich insbesondere für die Herstellung von im Mund schnell zerfallenden Tabletten.
Weiterhin wurden im Mund schnell zerfallende Tabletten, enthaltend solche Zubereitungen, gefunden. Die Tabletten zerfallen im Mund oder in wässrigem Milieu innerhalb von 40 Sekunden, bevorzugt innerhalb von 30 Sekunden, besonders bevorzugt innerhalb von 20 Sekunden.
Als filmbildend im erfindungsgemäßen Sinne wird ein Zucker oder Zuckeralkohol dann bezeichnet, wenn eine wässrige Lösung beim Abtrocknen bei 45°C einen filmähnlichen, glatten Belag ergibt. Nicht-filmbildende Zucker oder Zuckeralkohole zeigen nach dem Abtrocknen eine pul- verartige oder kristalline Struktur.
Die pharmazeutischen Zubereitungen enthalten als Komponente a) 60 - 96 Gew.-% mindestens eines nichtfilmbildenden Zuckers oder Zuckeralkohols, besonders bevorzugt 80 bis 90 Gew.-% nicht-filmbildende Zucker, Zuckeralkohole oder Mischungen davon. Als nicht filmbildende Zucker oder Zuckeralkohole eignen sich Mannitol, Erythritol, oder Xylitol. Die Zucker- oder Zuckeralkoholkomponenten sind bevorzugt feinteilig, mit mittleren Teilchengrößen von 5 bis 100 μιτι. bevorzugt 10 bis 80μιη. Gewünschtenfalls können die Teilchengrößen durch Mahlen eingestellt werden.
Bevorzugt werden Mannitol, Xylitol, Erythritol oder Mischungen davon eingesetzt. Ganz besonders bevorzugt ist Mannitol.
Als Komponente b) werden 1 - 10 Gew.-% filmbildende Zucker oder Zuckeralkohole, besonders bevorzugt 2 - 6 Gew.-% filmbildende Zucker oder Zuckeralkohole oder Mischungen davon ein- gesetzt. Zu den filmbildenden Zuckern oder Zuckeralkoholen zählen: Saccharose, Glucose, bspw. in Form von Glucosesirup, Fructose, Lactose, Trehalose, Sorbitol, Maltitol, Lactitol, Pala- tinit® (Isomalt) und Glucosesirup.
Zur Bestimmung der filmbildenden Eigenschaft werden zunächst bei 23°C 30,0 g einer 15 gew.- %igen Lösung des Zuckers oder Zuckeralkohols in Wasser angesetzt. Diese Lösung wird auf Glasplatten mit einer Vertiefung (Maße: 20 cm x 6,5 cm, Vertiefung 0,3 cm) ausgegossen und bei 45°C für 4 Stunden getrocknet und anschließend visuell beurteilt. Die Messung erfolgt in Umgebungsluft. Die Umgebungsluft weist hierbei bei 23°C eine relative Feuchte von 40% auf. Die Trocknung kann beispielsweise auf der Heizplatte eines Filmziehgerätes erfolgen. Als Film- ziehgerät eignet sich beispielsweise ein kommerzielles Gerät wie der Coatmaster 509 MC.
Als„filmbildend" werden die Produkte bezeichnet, die nach dem Trocknungsvorgang als klarer Film oder Belag vorliegen. Sind Kristalle sichtbar oder erscheint die Masse pulverförmig, werden die Substanzen als„nicht filmbildend" eingestuft. Demgemäß bilden beispielsweise einen klaren Film/Belag: Sorbitol, Lactitol Maltitol, Palatinit, Lactose, Saccharose, Trehalose oder Glucose. Kristalle oder eine pulverförmige Struktur bilden beispielsweise Mannitol, Xylitol oder Erythritol.
Als Komponente c) werden Sprengmittel in Mengen von 3 bis 25 Gew.-%, bevorzugt, 4 bis 20 Gew.-%, besonders bevorzugt 5 bis 15 Gew.-%, eingesetzt. Als Sprengmittel eignen sich quervernetzte Polyvinylpyrrolidone, Natrium-Croscarmellose, quervernetzte Na- Carboxymethylstärke oder L-Hydroxpropylcellulose (L-HPC). L-HPC ist eine niedrig substituierte Hydroxypropylcellulose mit einem Substitutionsgrad von 5 bis 16% (nach Trocknung von 1 h bei 105°C). Es können auch Mischungen dieser Sprengmittel eingesetzt werden.
Bevorzugte Sprengmittel sind quervernetzte Polyvinylpyrrolidone. Solche quervernetzten Polyvinylpyrrolidone sind wasserunlöslich (weniger als 1 Teil in 10000 Teilen Lösungsmittel), aber nicht filmbildend. Das quervernetzte Polyvinylpyrrolidon kann eine mittlere Teilchengröße (Volumenmittel) von 2 bis 60 μιη, bevorzugt 2 bis kleiner 50 μιη, besonders bevorzugt 2 bis kleiner 30 μιη aufweisen. Ganz besonders bevorzugt sind quervernetzte Polyvinylpyrrolidone mit einer Hydratationskapazität von größer 6,5 g/g. Hierbei erfolgt die Bestimmung der Hydratationskapazität nach folgender Methode:
2 g Polymer werden in ein Zentrifugenglas eingewogen und mit 40 ml Wasser 15 Minuten ausquellen lassen. Anschließend wird 15 Minuten bei 2000 U/Min zentrifugiert und die überstehen- de Flüssigkeit so vollständig wie möglich abgegossen. Die Messung erfolgt bei 20°C.
Hydratationskapazität = Auswaage-Tara
Einwaage Die hohe Hydratationskapazität des quervernetzten Polyvinylpyrrolidons führt in der Formulierung zu einem sehr schnellen Zerfall und ergibt ein besonders weiches Mundgefühl.
Als Komponente d) können optional wasserunlösliche filmbildende Polymere in Mengen von 0 bis 10 Gew.-%, bevorzugt 0 bis 6 Gew.-%, besonders bevorzugt 1 bis 5 Gew.-%, eingesetzt werden. Bevorzugt sind Polymere, die im pH-Bereich von 1 bis 14 unlöslich sind, also eine bei jedem pH-Wert pH-unabhängige Wasserunlöslichkeit aufweisen. Weiterhin eignen sich aber auch Polymere, die bei jedem pH-Wert im pH-Bereich von 6 bis 14 wasserunlöslich sind. Die Polymere sollen filmbildende Polymere sein. Filmbildend bedeutet in diesem Zusammenhang, dass die Polymere in wässriger Dispersion eine Mindestfilmbildetemperatur von -20 bis +150°C, bevorzugt 0 bis 100°C aufweisen.
Geeignete Polymere sind Polyvinylacetat, Ethylcellulose, Methylmethacrylat-Ethylacrylat- Copolymere, Ethylaci lat-Methylmethacrylat-Trimethylammoniumethylmethacrylat-Terpolymere, oder Butylmethaci lat-Methylmethacrylat-Dimethylaminoethylmethacrylat-Terpolymere
Die Acrylat-Methacrylat-Copolymere sind näher beschrieben in der Europäischen Pharmaco- poeia als Polyacrylate Dispersion 30%, in der USP als Ammonio Methacrylate Copolymer und in JPE als Aminoalkyl-Methacrylate Copolymer E.
Als bevorzugte Komponente d) kommt Ethylcellulose zum Einsatz. Geeignete Ethylcellulosen weisen üblicherweise einen Ethoxylgehalt von 45 bis 50% auf. Solche Ethylcellulosen und auch die Bestimmung des Ethoxylgehaltes sind in verschiedenen Arzneibüchern wie z.B. der USP 34 (201 1 ) beschrieben. Die Viskosität der Ethylcellulose gemessen als 5 gew.-%ige Lösung in To- luol/Ethanol 8:2 beträgt 3 bis 400 mPas, bevorzugt 4 bis 200 und besonders bevorzugt 5 bis 100 mPas. Die Messung erfolgt hierbei bei 25°C in einem Ubbelohde Viskosimeter.
Gewünschtenfalls können durch Zusatz von pharmazeutisch üblichen Hilfsstoffen (Komponen- ten e) in Mengen von 0 bis 15 Gew.-%, beispielsweise wie Säuerungsmitteln, Puffersubstanzen, Süßstoffen, Aromen, Geschmacksverstärkern und Farbstoffen Geschmack und Aussehen der aus den Formulierungen erhaltenen Tabletten weiter verbessert werden. Folgende Stoffe sind hierbei besonders geeignet: Citronensäure, Weinsäure, Ascorbinsäure, Natriumdihydro- genphosphat, Cyclamat, Saccharin-Na, Aspartam, Menthol, Pfefferminzaroma, Fruchtaromen, Vanillearoma, Glutamat, Riboflavin, Betacarotin, wasserlösliche Farbstoffe, feinteilige Farblacke.
Durch Zusatz von wasserlöslichen Polymeren kann das Mundgefühl durch Erhöhung der Weichheit und des Volumengefühls zusätzlich verbessert werden. Geeignete Stoffe sind zum Beispiel Polyvinylpyrrolidone, Vinylpyrrolidon-Vinylacetat-Copolymere, Polyvinylalkohole, Poly- vinylalkohol-Polyethylenglykol-Pfropfcopolymere, Polyethylenglykole, Ethylenglykol- Propylenglykol-Blockcopolymere, Hydroxypropylmethylcellulose, Hydroxypropylcellulose, Hyd- roxyethylcellulose, Carragenane, Pektine, Xanthane, Alginate. Weiterhin können als Komponenten e) auch Tenside zugegeben werden. Als Tenside eignen sich beispielsweise Natriumlaurylsulfat, Dioctylsulfosuccinat, alkoxilierte Sorbitanester wie Polysorbat 80, polyalkoxilierte Derivate von Rizinusöl oder hydriertem Rizinusöl, beispielsweise Cremophor® RH 40, alkoxilierte Fettsäuren, alkoxilierte Hydroxyfettsäuren, alkoxilierte Fettalkohole, Alkalisalze von Fettsäuren und Lecithine.
Weiterhin können zur weiteren Verbesserung des Zerfalls auch feinteilige Pigmente zugegeben werden, weil sie die inneren Grenzflächen erhöhen und somit Wasser schneller in die Tablette eindringen kann. Diese Pigmente wie Eisenoxide, Titandioxid, kolloidale oder gefällte Kieselsäure, Calciumcarbonate, Calciumphosphate müssen natürlich sehr feinteilig sein, ansonsten entsteht wiederum ein körniger Geschmack.
Die Herstellung der erfindungsgemäßen Formulierungen in Form von Granulaten kann durch Aufbau-Agglomeration in Mischern, Wirbelschichtgeräten, Schaufeltrocknern oder Sprühtürmen erfolgen. Dabei werden feste Ausgangsmaterialien und Granulierflüssigkeit zunächst miteinander in Verbindung gebracht und das feuchte Gut simultan oder anschliessend getrocknet. Ge- mäß der vorliegenden Erfindung wird als Granulierflüssigkeit eine wässrige Lösung der Komponente b), des filmbildenden Zuckers oder Zuckeralkohols, eingesetzt.
Bei der Agglomeration zu Granulaten in der Wirbelschicht wird eine eine wässrige Lösung des filmbildenden Zuckers oder Zuckeralkohols auf Pulvervorlage, die eine wirbelnde Mischung aus nicht-filmbildendem Zucker oder Zuckeralkohol und quervernetztem PVP sowie gegebenfalls weiteren Polymeren oder Hilfsstoffen darstellt, gesprüht, wodurch die feinen Teilchen agglomerieren.
Gemäß einer weiteren Ausführungsform der Erfindung wird in der wässrigen Granulierflüssig- keit, enthaltend die filmbildenden Zucker oder Zuckeralkohole ein Sprengmittel, vorzugsweise quervernetztes PVP, suspendiert.
Gemäß einer weiteren Ausführungsform der Erfindung wird in der wässrigen Granulierflüssigkeit, enthaltend die filmbildenden Zucker oder Zuckeralkohole, unlösliches filmbildendens Poly- mer suspendiert.
Gemäß einer weiteren Ausführungsform der Erfindung wird in der wässrigen Granulierflüssigkeit, enthaltend die filmbildenden Zucker oder Zuckeralkohole ein Sprengmittel, vorzugsweise quervernetztes PVP, und unlösliches filmbildendes Polymer suspendiert.
Bei der Herstellung in Sprühtürmen wird vorzugsweise die sogenannte FSD- oder SBD- Technologie (FSD: Fluidized spray drying; SBD: Spray bed drying) eingesetzt. Hierbei wird eine Lösung des nicht-filmbildenden Zuckers oder Zuckeralkohols in Wasser zunächst sprühgetrocknet, im unteren Teil des Sprühtrockners oder in einem angeschlossenen Wirbelbett erfolgt die Zugabe von quervernetztem PVP und die Eindüsung einer wässrigen Lösung des filmbildenden Zuckers oder Zuckeralkohols als Sprühlösung, wodurch die Teilchen agglomerieren. Feine Teilchen können ferner nochmals vor die Sprühdüse der Zucker oder Zuckeralkohollösung geblasen werden und zusätzlich agglomeriert werden. Es ist im Sprühturm, FSD oder SBD auch eine Prozeßführung ausgehend von der kristallinen Form des nicht-filmbildenden Zuckers oder nicht-filmbildenden Zuckeralkohols möglich. Dabei wird der kristalline Zucker oder Zuckeralkohol am Kopf des Sprühturms oder in den Feingut-Recyclingstrom zugegeben. Durch das Versprühen einer wässrigen Lösung des filmbildenden Zuckers oder Zuckeralkohols wird d er kristalline Feststoff des nicht-filmbildenden Zucker(alkohol)s im Turm zu Granulaten agglomeriert.
Die Sprühtrocknung des nicht-filmbildenden Zucker(alkohol)s erfolgt üblicherweise aus wässrigen Lösungen mit einer Konzentration von 10 bis 50 Gew.-%.
Der filmbildende Zuckeralkohol wird üblicherweise in Form von wässrigen Lösungen mit einer Konzentration von 1 bis 40 Gew.-% eingesetzt. Die Konzentration des filmbildenden Zuckeralkohols in der wässrigen Sprühlösung kann auch in zwei oder mehreren Verfahrensschritten variiert werden.
Für alle beschriebenen Ausführungsformen des Granulationsprozess ist entscheidend, dass die Prozessbedingungen so gewählt werden, dass eine relative Abluftfeuchte größer 85 %, bevorzugt größer 90 % und besonders bevorzugt größer 95 % bei einer Obergrenze von 99,5 %. resultiert. Die Einstellung einer relativen Abluftfeuchte von über 85 % erfolgt durch Anpassung der Sprührate, der Zulufttemperatur, der Zuluftmenge und der Feuchte der Zuluft. Die beiden Haupteinstellungsgrößen sind die Sprührate und die Zulufttemperatur. Eine Erhöhung der Sprührate sowie eine Erniedrigung der Zulufttemperatur führen zu einer Erhöhung der Abluftfeuchte.
Die Sprührate kann im Bereich von 3 g bis 35 g pro min pro kg Pulvervorlage gewählt werden, vorzugsweise 5 g bis 30 g pro min pro kg Pulvervorlage.
Die Zulufttemperaturen betragen 20 bis 80°C, vorzugsweise 30 bis 60°C.
Die Ablufttemperaturen betragen 10 bis 60°C, vorzugsweise 10 bis 40°C und besonders bevorzugt 15 bis 30°C.
Bevorzugt erfolgt die Granulation bei einer Sprührate von 5 g bis 30 g pro min pro kg Pulvervor- läge in Kombination mit einer Zulufttemperatur von 30 bis 60°C und einer Ablufttemperatur von 10 bis 40°C
Besonders bevorzugt erfolgt die Granulation bei einer Sprührate von 6 g bis 18 g pro min pro kg Pulvervorlage in Kombination mit einer Zulufttemperatur von 35 bis 50°C und einer Ablufttemperatur von 15 bis 25°C.
Die Zuluftfeuchte kann bei allen genannten Ausführungsformen bis zu 99,5 % r.F. betragen.
Für den Granulationsprozess kann es sich als günstig erweisen, einen mehrstufigen Sprühpro- zess zu fahren. Hierbei wird die Konzentration der wässrigen Sprühlösung des filmbildenden Zuckers oder Zuckeralkohols in mehreren Schritten variiert. Zu Anfang wird die Konzentration sehr niedrig gehalten - sie kann sogar 0 Gew.-% betragen (Einsprühen von Wasser) -, um die vorgelegten Materialien zu befeuchten und eine hohe Abluftfeuchte einzustellen. Hierbei setzt noch keine Agglomeration zu Granulaten ein. Danach wird die Konzentration der wässrigen Sprühlösung des filmbildenden Zuckers oder Zuckeralkohols erhöht, wodurch die vorgelegten Materialien zu Granulaten agglomerieren. In den einzelnen Stufen können die Prozeßbedingungen im Rahmen der vorgegeben Grenzen variiert werden.
Bevorzugt ist ein zweistufiger Prozess, bei dem die Sprührate in der ersten Stufe das 1 ,5- bis 3- fache der Sprührate der zweiten Stufe beträgt. Besonders bevorzugt ist ein zweistufiger Prozess wie vorstehend beschrieben, bei dem in der ersten Stufe reines Wasser als Sprühlösung eingesetzt wird. Das Massenverhältnis der Sprühlösungen der ersten und der zweiten Stufe kann von 1 zu 5 bis 5 zu 1 , bevorzugt 1 zu 2 bis 2 zu 1 , variiert werden.
Die Granulatgröße kann über die Menge des filmbildenden Zuckers oder Zuckeralkohols im Produkt gesteuert werden. Je mehr dieses Stoffes eingebracht wird, umso größer werden die Granulate.
Als weitere Anpassungsparameter für die Granulatgröße kann die Feinheit des Sprühtröpfchens der Bindemittellösung (einstellbar über den Zerstäubungsgasdruck), die Düsengeometrie und Abstand der Düse zum Produktbett angesehen werden. Je feiner und einheitlicher gesprüht wird, desto feiner und einheitlicher resultieren die Granulate. Je weiter die Düse vom Produkt- bett entfernt ist, desto schlechter ist das Agglomerationsverhalten. Üblicherweise liegt der Zerstäubungsgasdruck im Bereich von 0,5 bis 5 bar, bevorzugt 1 ,0 bis 3,0 bar. Als Düsen werden in der Regel Zweistoffdüsen eingesetzt. Die Düsendurchmesser liegen üblicherweise im Bereich von 0,5 bis 4,0 mm, bevorzugt 0,8 bis 3,0 mm. Der Abstand der Düsen vom Produktbett richtet sich nach der Größe der Zerstäubungsvorrichtung. Welche Abstände im Einzelfall geeig- net sind, kann vom Fachmann durch einige übliche Versuche ermittelt werden.
Nach Beendigung des Sprühprozesses wird das feuchte Gut abgetrocknet. Hierbei wird die Zu- lufttemperatur erhöht, um den Trocknungsprozess zu beschleunigen. Übliche Temperaturen liegen hierbei bei 30 - 100°C. Während der Trocknungsphase kann es vorteilhaft sein, die Zuluftmenge zu reduzieren und damit einem Abrieb der Granulate durch eine hohe mechanische Belastung vorzubeugen.
Bei der Herstellung in einem Schaufeltrockner werden die vorgelegten Materialien mit einer Lösung des filmbildenden Zuckers oder Zuckeralkohols in Wasser besprüht und simultan oder anschließend abgetrocknet.
Weiterhin können die Granulate auch in einem Mischer durch eine kontinuierlich geführte Mi- schaggregationhergestellt werden. Eine solche kontinuierlich geführte Form der Mischaggregation ist die sogenannte„Schugi-Granulation". Dabei werden in einem kontinuierlich arbeitenden vertikal angeordneten Hochgeschwindigkeitsmischer feste Ausgangsmaterialien und die den filmbildenden Zucker oder Zuckeralkohol enthaltende Granulierflüssigkeit intensiv miteinander vermischt (siehe auch M. Bohnet,„Mechanische Verfahrenstechnik", Wiley VCH Verlag, Weinheim 2004, S. 198 ff.)
Die so erhaltenen Granulate weisen eine mittlere Teilchengröße (Volumenmittel) von 100 - 600 μιη, bevorzugt 120 - 500 μιη und besonders bevorzugt von 140 - 400 μιη auf.
Der filmbildende Zucker oder Zuckeralkohol dient dabei als Agglomerationsmittel, um die feinen nicht-filmbildenden Zucker oder Zuckeralkoholkristalle und die Teilchen von quervernetztem PVP zu agglomerieren. Die erfindungsgemäßen Granulate können mit geringen Mengen an Schmiermitteln tablettiert werden, wodurch sich keine signifikante Beeinflussung der Zerfallszeit ergibt. Ferner kleben die erfindungsgemäßen Granulate nicht an den Stempeln, wodurch höhere Tablettiergeschwindigkeiten und kürzere Tablettierprozesse erzielt werden. Die erfindungsgemäßen Formulierungen eignen sich vorzugsweise für die Herstellung von im Mund schnell zerfallenden Tabletten.
Die erfindungsgemäßen Formulierungen können vorteilhaft auch für die Herstellung von Tabletten verwendet werden, die vor der Anwendung in einem Glas Wasser zerfallen gelassen wer- den. Auch die Herstellung von Tabletten, die intakt geschluckt werden, ist natürlich möglich.
Für die Herstellung von Tabletten können die üblichen Verfahren verwendet werden, wobei die Direkttablettierung und die Walzenkompaktierung besondere Vorteile bieten. Aufgrund der besonderen Eigenschaften der erfindungsgemäßen Formulierungen werden in der Regel nur Wirkstoff, erfindungsgemäße Formulierung und ein Schmiermittel benötigt. Die Tablettenrezeptur ist somit sehr einfach, sehr reproduzierbar und das Verfahren leicht zu validieren.
Überraschenderweise wurde gefunden, dass ein filmbildender Zucker oder Zuckeralkohol bei entsprechender Prozesssteuerung als sehr wirksames Bindemittel fungieren kann, das darüber hinaus den Zerfall von Tabletten erheblich beschleunigt. Auf diese Weise ist die Herstellung von Granulaten möglich, die weder ein wasserlösliches Polymer noch ein wasserunlösliches Polymer als Bindemittel enthalten. Die Verwendung von wasserlöslichen Polymeren führt nämlich zu deutlich längeren Zerfallszeiten von Tabletten und die Verwendung von wasserunlöslichen Polymeren ergibt zwar schnell zerfallende Tabletten, die aber bei Lagerung bei erhöhter Tempera- tur und/oder erhöhter Feuchte erhebliche Zerfallszeitverlängerungen aufweisen.
Die Zerfallszeit der erfindungsgemäßen Zubereitungen wird durch erhöhte Temperatur und Feuchte nicht beeinflusst.
Ferner besitzen die erfindungsgemäßen Zubereitungen außergewöhnlich gute Fließfähigkeiten und Verpressbarkeiten, die zu mechanisch sehr stabilen Tabletten führen. Die Bruchfestigkeit der mit Hilfe der erfindungsgemäßen pharmazeutischen Formulierungen erzeugten Tabletten beträgt > 50 N. Häufig liegen die Bruchfestigkeiten oberhalb von 80 N, selbst unter Verwendung schwer pressbarer Wirkstoffe. Die Friabilitäten betragen < 0,2%. Beschädigungen beim üblichen Tablettenhandling treten somit nicht auf.
Aufgrund des feinen quervernetzten Polyvinylpyrrolidons zeigen die Tabletten nahezu keine Veränderungen der Tablettenoberfläche bei feuchter Lagerung. Im Gegensatz zu grobem quervernetzten Polyvinylpyrrolidon findet sich keine Pickelbildung durch stark aufgequollene Teilchen. Die erfindungsgemäßen Formulierungen sind damit bei Lagerung sehr stabil und behal- ten ihr ansprechendes Aussehen.
Die Herstellung der Granulate erfolgt wie erwähnt bei Abluftfeuchten von mindestens 85% r.F. Solche Abluftfeuchten sind völlig unüblich, da konventionelle Rezepturen bei solchen Abluftfeuchten völlig zusammenklumpen und nicht mehr verwirbelt und prozessiert werden können. Überraschenderweise wurde gefunden, dass unterhalb von 85% r.F. keine zufriedenstellende Granulation stattfindet, aber oberhalb von 85% sehr gleichmäßige Granulate entstehen und der Prozeß problemlos gefahren werden kann.

Prüfung auf Filmbildung:
Zur Bestimmung der filmbildenden Eigenschaft wurden zunächst bei 23°C 30,0 g einer 15 gew.- %igen Lösung des Zuckers oder Zuckeralkohols in Wasser angesetzt. Diese Lösung wurde auf Glasplatten mit einer Vertiefung (Maße: 20 cm x 6,5 cm, Vertiefung 0,3 cm) ausgegossen und bei 45°C für 4 Stunden getrocknet und anschließend visuell beurteilt. Die Umgebungsluft wies hierbei bei 23°C eine relative Feuchte von 40% auf. Die Trocknung erfolgte auf einer Heizplatte des Filmziehgerätes Coatmaster 509 MC (Fa. Erichsen).
Die im Folgenden aufgeführten Teilchengrößenbestimmerungen wurden mittels eines Malvern Mastersizers durchgeführt. Die Beispiele A - H zeigen den agglomerierenden Effekt von filmbildender Zuckern oder Zuckeralkoholen.
Nicht-filmbildender Zucker/Zuckeralkohol und quervernetztes PVP sowie weitere Hilfsstoffe wurden im Wirbelbett vorgelegt und mit einer 3 gew.-%igen wässrigen Lösung des filmbilden- den Zuckers oder Zuckeralkohols, die in den Wirbelschichtgranulator (Firma Glatt,GPCG 3.1 ) mittels Topspray-Verfahren eingesprüht wurden, agglomeriert.

Die Teilchengröße wurde mittels Lichtbeugung ermittelt und als mittlere Teilchengröße (Volumenmittel) der D(4,3)-Wert herangezogen.
Die so hergestellten Granulate wurden mit 2,0 Gew.-% Schmiermittel (Natriumstearylfumarat) in einem Turbulamischer für 5 min gemischt. Anschließend wurden diese Mischungen auf einer voll instrumentierten Rundläufertablettenpresse (Korsch XL 100/8) bei einer Umdrehungsge- schwindigkeit von 40 Upm tablettiert. Die Rundläufertablettenpresse war mit 8 Stempeln (10 mm, biplan, facettiert) ausgestattet. Das Tablettengewicht wurde auf 300 mg eingestellt. Die Press kraft wurde so eingestellt, dass die Bruchfestigkeit der Tabletten 50 N betrug.
Die Tabletten wurden hinsichtlich Bruchfestigkeit (Tablettentester HT-TMB-CI-12 F, Fa. Krae- mer), Friabilität (Roche-Friabilator, Erweka) und Zerfallszeit in Phosphatpuffer pH 7,2 (Zerfallstester ZT 74, Erweka) untersucht.

Die Herstellung erfolgte analog Beispiel B, allerdings wurde statt einer Sorbitollösung eine Xyli- tollösung verwendet.
Es erfolgte nahezu keine Agglomeration. Die mittlere Teilchengröße lag bei 97μιτι und der Bö- schungswinkel bei 44°. Eine Tablettierung war aufgrund der schlechten Fließfähigkeit nicht möglich.
Beispiel K (Vergleichsbeispiel zu B)
Agglomeration bei einer relativen Feuchte von 70% (weniger als 85%)
Die Herstellung erfolgte analog Beispiel B, allerdings wurde die Sprührate so eingestellt, dass eine relative Abluftfeuchte von 70% resultierte.
Es erfolgte nahezu keine Agglomeration. Die mittlere Teilchengröße lag bei 89μιη und der Böschungswinkel bei 43°. Eine Tablettierung war aufgrund der schlechten Fließfähigkeit nicht möglich.

Die Herstellung erfolgte analog Beispiel H der oben zitierten Offfenlegungsschrift.
Die Zerfallszeit der Tabletten direkt nach Herstellung betrug 30s. Anschließend wurden die Tab- letten bei 40°C/75% r.F. und bei 60°C 4 Wochen gelagert. Dabei stieg die Zerfallszeit auf 180s bzw. 204s an.
Beispiele M - P
Beispiele M bis P zeigen die Eignung eines schnell zerfallenden Hilfsstoffes in einer Wirkstoffformulierung. Der schnell zerfallende Hilfsstoff wird durch Agglomeration in der Wirbelschicht analog Beispiel C hergestellt. Das so hergestellte Direkttablettierungsmittel wurde mit Wirkstoff und 0,5 bis 1 ,0 Gew.-% Schmiermittel (Mg-Stearat) abgemischt und anschließend auf einer Rundläufertablet- tenpresse (Korsch PH 100/6) zu Tabletten mit 50 N Bruchfestigkeit verpresst. Die Tabletten wurden hinsichtlich Bruchfestigkeit (Tablettentester HT-TMB-CI-12 F, Fa. Krae- mer), Friabilität (Roche-Friabilator, Erweka) und Zerfallszeit in Phosphatpuffer pH 7,2 (Zerfallstester ZT 74, Erweka) untersucht.

Die hergestellten Granulate wiesen folgende Eigenschaften auf:
Die Teilchengröße wurde mittels Lichtbeugung ermittelt und als mittlere Teilchengröße (Volu- menmittel) der D(4,3)-Wert herangezogen.
Claims
1 . Pharmazeutische Formulierung in Form von Granulaten enthaltend
a) 60 - 96 Gew.-% nicht filmbildende Zucker oder Zuckeralkohole,
b) 1 - 10 Gew.-% filmbildende Zucker oder Zuckeralkohole
c) 3 - 25 Gew.-% Sprengmittel,
d) 0 - 10 Gew.-% wasserunlösliche, filmbildende Polymere
e) 0 - 15 Gew.-% weitere pharmazeutisch übliche Hilfsstoffe
wobei die Summe der Komponenten a) bis e) 100 Gew.-% beträgt.
2. Formulierung nach Anspruch 1 dadurch gekennzeichnet, dass die mittlere Teilchengröße der Granulate 100 μιη bis 600 μιη beträgt.
3. Formulierung nach Anspruch 1 oder 2, enthaltend als nicht filmbildender Zuckeralkohol Mannit, Erythritol oder Xylitol oder Mischungen davon.
4. Formulierung nach Anspruch 1 oder 2, enthaltend als filmbildender Zuckeralkohol Sorbi- tol, Lactitol, Isomalt oder Maltitol oder Mischungen davon.
5. Formulierung nach einem der Ansprüche 1 bis 3, enthaltend als Sprengmittel ein quer- venetztes Polyvinylpyrrolidon mit einer mittleren Teilchengröße kleiner 50 μιη.
6. Formulierung nach einem der Ansprüche 1 bis 5, enthaltend als Sprengmittel ein quer- vernetztes Polyvinylpyrrolidon mit einer Hydratationskapazität größer 6,5 g/g.
7. Formulierung nach einem der Ansprüche 1 bis 6, wobei als weitere pharmazeutisch übliche Stoffe Säuerungsmittel, Süßstoffe, Aromen, Geschmacksverstärker, Farbstoffe, Bindemittel, Verdickungsmittel, Tenside und feinteilige Pigmente verwendet werden.
8. Formulierung nach einem der Ansprüche 1 bis 7, enthaltend Granulate aus a) 70 - 93 Gew.-% nicht filmbildenden Zuckern oder Zuckeralkoholen,
b) 2 - 8 Gew.-% filmbildenden Zuckern oder Zuckeralkoholen
c) 4 - 20 Gew.-%Sprengmittel,
d) 0 - 6 Gew.-% wasserunlöslichen, filmbildende Polymeren
e) 0 - 15 Gew.-% weitere pharmazeutisch üblichen Hilfsstoffen.
9. Formulierung nach einem der Ansprüche 1 bis 8, enthaltend Agglomerate aus
80 - 90 Gew.-% nicht filmbildenden Zuckern oder Zuckeralkoholen, 2 - 6 Gew.-% filmbildenden Zuckern oder Zuckeralkoholen
Gd/Ya 08.02.201 1 0 Fig/0 Seq c) 5 - 15 Gew.-% quervernetztem Polyvinylpyrrolidon,
d) 1 - 5 Gew.-% wasserunlöslichen, filmbildenden Polymeren
e) 0 - 15 Gew.-% weiteren pharmazeutisch üblichen Hilfsstoffen.
10. Tabletten, erhalten unter Verwendung einer pharmazeutischen Formulierung nach einem der Ansprüche 1 bis 9, wobei die Tabletten in wässrigem Milieu Zerfallszeiten von kleiner 30 Sekunden aufweisen.
1 1 . Tabletten nach Anspruch 10, wobei die Tabletten eine Bruchfestigkeit größer 50 N aufweisen.
12. Tabletten nach Anspruch 10 oder 1 1 , enthaltend 20 bis 99 Gew.-%, bezogen auf das gesamte Tablettengewicht, einer pharmazeutischen Formulierung gemäß einem der Ansprüche 1 bis 9.
13. Tabletten nach einem der Ansprüche 10 bis 12, enthaltend weitere Hilfsstoffe.
14. Verfahren zur Herstellung einer pharmazeutischen Formulierung gemäß einem der Ansprüche 1 bis 9, dadurch gekennzeichnet, dass nicht filmbildende Zucker- oder Zuckeralkoholteilchen und quervernetztes Polyvinylpyrrolidon mit einer wässrigen Lösung des filmbildenden Zuckers oder Zuckeralkohols agglomeriert werden.
15. Verfahren nach Anspruch 16, dadurch gekennzeichnet, dass nicht-filmbildenden Zuckeroder Zuckeralkoholteilchen mit einer wässrigen Lösung des filmbildenden Zuckers oder Zuckeralkohols, ddie zusätzlich quervernetztes Polyvinylpyrrolidon suspendiert enthält, agglomeriert werden.

Verfahren nach Anspruch 15, dadurch gekennzeichnet, dass nicht-filmbildenden Zuckeroder Zuckeralkoholteilchen mit einer wässrigen Lösung des filmbildenden Zuckers oder Zuckeralkohols, die zusätzlich wasserunlösliches Polymer und quervernetztes Polyvinylpyrrolidon suspendiert enthält, agglomeriert werden.
17. Verfahren nach einem der Ansprüche 14 bis 16, dadurch gekennzeichnet, dass die Agglomeration in einem Wirbelschichtgranulator, einem Mischer, einem Schaufeltrockner oder einem Sprühturm erfolgt.
18. Verfahren nach einem der Ansprüche 14 bis 17, dadurch gekennzeichnet, dass die Agglomeration bei einer relativen Abluftfeuchte von größer 85% durchgeführt wird.
19. Verfahren nach einem der Ansprüche 14 bis 18, dadurch gekennzeichnet, dass die Agglomeration bei einer relativen Abluftfeuchte von größer 90% durchgeführt wird.
20. Verfahren nach einem der Ansprüche 14 bis 19, dadurch gekennzeichnet, dass die Agglomeration bei einer relativen Abluftfeuchte von größer 95% durchgeführt wird.
21 . Verfahren nach einem der Ansprüche 14 bis 20, dadurch gekennzeichnet, dass die Granulation zweistufig durchgeführt wird und in der ersten Stufe Wasser als Granulierflüssigkeit auf die Pulvervorlage aufgesprüht wird.
22. Verfahren nach einem der Ansprüche 14 bis 21 , dadurch gekennzeichnet, dass die Granulation bei einer Sprührate von 5 g bis 30 g pro min pro kg Pulvervorlage in Kombination mit einer Zulufttemperatur von 30 bis 60°C und einer Ablufttemperatur von 10 bis 40°C durchgeführt wird.
23. Verfahren nach einem der Ansprüche 14 bis 21 , dadurch gekennzeichnet, dass die Granulation bei einer Sprührate von 6 g bis 18 g pro min pro kg Pulvervorlage in Kombination mit einer Zulufttemperatur von 35 bis 50°C und einer Ablufttemperatur von 15 bis 25°C durchgeführt wird.
