- a murine xenograft model is used for the purpose of investigating the inhibitory activity of SDF- la mutants on cancer metastasis. 10 week old female immunodeficient SCID mice are used for the in vivo experiments. They are kept in isolation care during the whole course of the experiments. The animals are left for a few days after delivery to settle down before the experiments started .
- LMD-231 cells are used in this xenograft model. They are cultured from lung metastasis of mice inoculated with MDA-MB-231. These cells are known to express high levels of CXCR4 and are routinely used in xenograft experiments. These adherent cells are cultured in complete Minimum Essential Medium and complete RPMI mixed in a 50:50 ratio and cell flasks are split in a ratio of 1:3. The cells are cultured at 37°C, 5% C0 2 in a humidified atmosphere.

Abstract
L'invention concerne de nouveaux polypeptides mutants à base de SDF-1 présentant une affinité de liaison aux GAG accrue et une activité GPCR réduite par comparaison avec la protéine SDF-1 de type sauvage. Ce polypeptide SDF-1 est modifié de manière à préserver sa structure, par substitution de la glutamine en position 60 par un acide aminé basique ou donneur d'électrons, et par substitution d'au moins un acide aminé supplémentaire en positions 29, 39, 49, 67 et/ou 68 selon la séquence SEQ ID No 1 par au moins un acide aminé basique ou donneur d'électrons, au moins un acide aminé parmi les 10 premiers acides aminés de la région N-terminale du polypeptide SDF-1 de type sauvage étant modifié par addition, délétion et/ou substitution d'au moins un acide aminé. L'invention concerne en outre des méthodes de production desdits mutants et leur utilisation pour la préparation de médicaments destinés au traitement du cancer.
