Abstract
La présente invention concerne la préparation de composés de triazole-5-yl(pyridine-2-yl)benzamides substitués, ayant la structure I, sous forme libre ou sous forme de sel acceptable pour la dégradation ciblée de la kinase 1 régulant le signal d'apoptose (ASK 1) en tant que cible thérapeutique potentielle pour améliorer l'hépatopathie stéatosique non alcoolique (NAFLD)/la stéatohépatite non alcoolique (NASH). Plus particulièrement, l'invention concerne de petites molécules, R1, R2 et R3 étant tels que définis dans la description, aptes à une dégradation ciblée de ASK1, qui peuvent finalement être utilisées pour obtenir une santé humaine améliorée chez des patients atteints de fibrose hépatique. La présente invention représente le repère de cette technologie PROTAC pour une protéine cible thérapeutique potentielle ASK1 et apportera une percée dans la recherche NAFLD/NASH.
