LA PROTEINA CODIFICADA POR LA ORF-5 DEL VIRUS DEL SINDROME REPRODUCTIVO Y RESPIRATORIO PORCINO SE PREPARA Y PURIFICA TRAS SU EXPRESION EN CELULAS EUCARIOTAS. PARA ELLO, LA SECUENCIA CODIFICANTE DE DICHA PROTEINA SE OBTIENE POR PCR, CLONADA EN EL VECTOR PGEM-T, SUBCLONADA EN EL VECTOR PRSET QUE CONFIERE A LA SECUENCIA ANTERIOR LA CODIFICANTE PARA UN TAG DE HISTIDINAS, Y NUEVAMENTE SUBCLONADA EN EL VECTOR DE EXPRESION PTM-1 PARA SU EXPRESION COMO PROTEINA DE FUSION, SIRVIENDO A SU VEZ DE BASE PARA LA SINTESIS DE UN VIRUS VACCINIA RECOMBINANTE PARA SU USO MEDIANTE PROTOCOLOS DE DOBLE INFECCION CON UN VIRUS VACCINIA QUE ESPESA LA POLIMERASA DEL FAGO T7 EN CELULAS EUCARIOTAS. LA PROTEINA ASI EXPRESADA, ES PURIFICADA MEDIANTE SU UNION A UNA RESINA DE NIQUEL ACETONITRILO, DEBIDO A LA AFINIDAD DE LAS HISTIDINAS POR DICHO METAL.
