Abstract
Método para detectar la presencia de polimorfismos de DNA, estuche de ensayo y ácidos nucleicos recombinantes correspondientes. El método consiste en analizar DNA genómico humano, DNA genómico clonado o DNA genómico amplificado mediante el empleo de moléculas de oligonucleótidos como sondas-que hibridan con una porción del gen de la apolipoproteína E. En otra variante, la detección se realiza mediante la digestión del DNA con una o varias enzimas de restricción, o bien mediante la utilización de diferentes técnicas electroforéticas. El estuche de ensayo se caracteriza por una cantidad definida de una o más sondas de oligonucleótidos marcados, una cantidad definida de una o más enzimas de restricción y medios necesarios para la amplificación del DNA. El ácido nucleico recombinante a utilizar en el método contiene cualquier fragmento del promotor del gen de la apolipoproteína E que incluye una serie de nucleótidos determinados del mencionado gen. La invención es aplicable a la diagnosis y terapia de la enferme dad de Alzheimer, enfermedad coronaria arteriosclerótica y otros síndromes clínicos asociados con la apolipoproteína E.
