Abstract

Procedimiento para la obtención de polímeros naturales y/o modificados con una secuencia definida de componentes nucleotídicos y/o no nucleotídicos, que comprende los siguientes pasos: a) unión de un cebador de la formula general p(dNp)m o p(rNp)m, pudiendo faltar, en caso dado, el grupo p en la posición 3¿ del cebador, con un soporte de la fórmula general (rM)ndN o (rM)ndU-rN, en los que p, dN, rN, m, rM, n y dU tienen los siguientes significados: p un grupo fosfato y/o fosforotioato y/o fosfonato, como preferentemente metilfosfonato y/o H-fosfonato, y/o fosforoamidato y/o formacetal y/o fosforoditioato y/o boranofosfato y/o fosforotriéster, dN un desoxiribonucleósido natural y/o modificado con bases y/o azúcares, y/o un grupo de enlace no nucleosídico apto para la unión, rN un ribonucleósido natural y/o modificado con bases y/o azúcares, y/o un grupo de enlace no nucleosídico apto para la unión, m un número entero de 1 o más, rM un ribonucleósido de purina y/o pirimidina, n un número entero de 1 o más. dU una desoxiuridina, pudiendo comprender el cebador de la fórmula general p(dNp)m o p(rNp)m también restos mixtos de dN y/o rN, con ayuda de una ARN-ligasa en presencia de cationes metálicos divalentes, no poseyendo el soporte ningún grupo protector en la posición 3¿; b) desprotección del producto de unión, en el caso de empleo del soporte de la fórmula general (rM)ndN mediante eliminación del grupo protector (rM)n en presencia de una RNasa, o bien eliminación del grupo protector (rM)ndU, en el caso de empleo del soporte de la fórmula general (rM)ndU-rN con ayuda de una uracil-ADN-glicosilasa y una endonucleasa apurínica, o eliminación de los citados grupos protectores en el intervalo alcalino; c) transferencia de un grupo terminal natural y/o modificado, de la fórmula general p definida anteriormente, al grupo 5¿-hidroxilo del producto de unión con ayuda de una polinucleótido-quinasa; d) en caso dado repetición de los pasos a)-c), y; e) en caso dado purificación del producto de unión obtenido en el paso c).
