Abstract
Un proceso de concentración de moléculas de ácido nucleico (5a, 5b), en una superficie (201) de un campo sensor (103a) de un biochip, el proceso consta de los pasos siguientes: (a) proporcionar perlas magnéticas (1), con moléculas de transferencia (3a, 3b, 3c) inmovilizadas sobre ellas, que son capaces de hibridarse con una porción de las moléculas de ácido nucleico (5a, 5b), con lo cual forman complejos que contienen dichas perlas magnéticas (1), dichas moléculas de transferencia (3a, 3b, 3c) y dichas moléculas de ácido nucleico; proporcionar una superficie (201) de un campo sensor (103a, 103b, 1 10 03c), que constituye una parte de una superficie (101) de un biochip, dichas moléculas de captura (203) se inmovilizan sobre dicha superficie (201), dichas moléculas de captura (203) se hibridan de modo específico con una porción de las moléculas de ácido nucleico (5a, 5b), en el que las secuencias de ácido nucleico de las moléculas de captura (203) y las moléculas de transferencia (3a, 3b, 3c) se eligen de tal manera que la temperatura de fusión del híbrido entre una molécula de ácido nucleico (5a, 5b) y una molécula de transferencia (3a, 3b, 3c) sea inferior a la temperatura de fusión del híbrido entre una molécula de ácido nucleico (5a, 5b) y una molécula de captura (203); y proporcionar un generador de campo magnético (105); (b) incubar las perlas magnéticas (1) con las moléculas de ácido nucleico (5a, 5b), a una temperatura T1 que es inferior a la temperatura de fusión de los híbridos entre las moléculas de ácido nucleico (5a, 5b) y las moléculas de transferencia (3a, 3b), permitiendo de este modo que las moléculas de ácido nucleico (5a, 5b) se hibriden con las moléculas de transferencia (3a, 3b); (c) aplicar un campo magnético con el generador de campo magnético (105), con lo cual se mueven las moléculas de ácido nucleico (5a, 5b) unidas a las perlas magnéticas (1) a través del campo magnético hacia la superficie (101) del biochip, con lo cual se ponen en contacto las moléculas de ácido nucleico (5a, 5b) unidas a las perlas magnéticas (1) con una superficie (201) del campo sensor (103a, 103b); (d) aumentar la temperatura desde una temperatura T1 a una temperatura T2, que es superior a la temperatura de fusión de los híbridos de las moléculas de ácido nucleico (5a, 5b) y las moléculas de transferencia (3a, 3b) y que es inferior a la temperatura de fusión de los híbridos de las moléculas de ácido nucleico (5a, 5b) y las moléculas de captura (203), con lo cual se disuelven o separan las moléculas de ácido nucleico (5a) de las perlas magnéticas (1) y se permite la unión de por lo menos una porción de la molécula de ácido nucleico (5a, 5b) con la molécula de captura (203); (e) mover las perlas magnéticas (1) para separarlas de la superficie (101) del biochip por interrupción de la aplicación del campo magnético; (f) repetir los pasos de (b) a (e).
