C. Análisis de la expresión de zcytoR17 mediante PCR y Northern
Anotar los tipos celulares y las condiciones de crecimiento que afectan a la expresión del receptor es un medio útil 5 de elucidar su función y predecir una fuente de ligando. Con este fin, mediante PCR se analizó una amplia variedad de tipos de tejido y de células. Se usó la polimerasa termoestable Advantage II™ (Clontech, La Jolla, CA) con los cebadores oligonucleotídicos ZC29,180 (SEC ID Nº 22) y ZC29,179 (SEC ID Nº 82) y 1-10 ng de los diversos moldes de ADNc indicados más adelante para 30 ciclos de amplificación de (94°C, 30 s.; 66°C, 20 s.; 68°C, 1 min. 30 s). Tras esto, el 20 % de cada reacción se pasó por geles de agarosa al 0,8 %, TAE/bromuro de etidio y se 10 visualizaron con luz UV. Después, las muestras se puntuaron en base a la intensidad de la señal. Véase la Tabla 14 a continuación.

5 B. Producción de citocinas por las líneas celulares epiteliales humanas normales cultivadas con zcytor17lig humano
Además de las líneas celulares epiteliales humanas, también se analizaron las celulares epiteliales bronquiales humanas normales (NHBE, Clonetics). Las células se sembraron en placas a una densidad de 1 x 105 células por pocillo en una placa de 24 pocillos y se cultivaron con reactivos de ensayo; 1000 ng/ml, 100 ng/ml and 10 ng/ml del
10 ligando de zcytor17 (A760F), 10 ng/ml de TNFa, 10 ng/ml de OSM, 10 ng/ml de IFNa, 10 ng/ml de TGFb o 10 ng/ml de linfotactina. Los sobrenadantes se recolectaron a las 24 y 48 horas y se analizó la presencia de citocinas: IL-6, IL8, MCP-1, MIP-1a, RANTES y Eotaxina. Las citocinas se analizaron como se ha descrito anteriormente. La producción de citocinas (pg/ml) para cada línea celular en las muestras de 48 horas se muestra a continuación en la Tabla 18.

Las células DU145 produjeron IL-6 en respuesta al ligando de zcytor17, de modo que se repiten los resultados anteriores del Ejemplo 43A.
