Abstract

Un método para rellenar un lecho de medios de una columna de cromatografía a través de la cual se hace fluir un fluido, teniendo dicha columna un ajustador y medios para determinar la posición de dicho ajustador dentro de dicha columna, comprendiendo dicho método: (i) introducir en un sistema de relleno de columnas el tipo de medios, la velocidad del ajustador y la altura de la columna terminal a través de menús o entradas de teclado. (ii) realizar un proceso de rellenado a velocidad constante de una columna con la velocidad del ajustador deseada facilitada. (iii) completar el proceso de rellenado cuando la altura de la columna alcance la altura de la columna terminal introducida en el sistema. en el que la altura óptima de la columna introducida en el sistema se determina empíricamente rellenando varias columnas que utilizan una velocidad constante o un algoritmo de presión constante; en el que el algoritmo de velocidad constante comprende: accionar dicho ajustador a una velocidad constante para comprimir dicho lecho de medios; determinar la fuerza de entrada de columna ejercida por el fluido en una entrada por medio de un sensor de presión colocado en dicha entrada; determinar la fuerza total, que es la suma de dicha fuerza de entrada de columna y la fuerza de compresión del medio, mediante una célula de carga; y continuar el movimiento de dicho ajustador mientras que la relación de dicha fuerza de entrada de columna con dicha fuerza total sea más grande que un nivel predeterminado, y cesar el movimiento de dicho ajustador cuando dicha relación proporcional descienda por debajo de dicho nivel predeterminado, en donde dicho nivel predeterminado es una relación deseada de dicha fuerza de entrada de columna con dicha fuerza total y es en función del tipo de medios; y en el que el algoritmo de presión constante comprende: accionar dicho ajustador a una velocidad creciente hasta que la presión hidráulica alcance un nivel predeterminado; disminuir la velocidad de dicho ajustador para mantener dicha presión hidráulica a dicho nivel predeterminado; alcanzar un estado estable en donde la velocidad y la presión hidráulica permanezcan constantes; almacenar la velocidad en dicho valor de estado estable; incrementar la velocidad de dicho ajustador para mantener dicha presión hidráulica a dicho nivel predeterminado después de alcanzar dicho valor de estado estable; comparar la velocidad actual con dicha velocidad almacenada; y continuar incrementando la velocidad de dicho ajustador para mantener dicha presión hidráulica si dicha velocidad actual no es más grande que dicha velocidad almacenada por una cantidad predeterminada y cesar el movimiento de dicho ajustador cuando dicha velocidad actual sea más grande que dicha velocidad almacenada por dicha cantidad predeterminada.
