Sustancia de actividad apoptótica, caracterizada por que comprende al menos una porción peptídica con una de las secuencias de aminoácidos representadas en las SEQ ID nºs 12, 18 y 19 e induce la apoptosis.

EJEMPLO 5: Identificación de péptidos hexámeros de actividad apoptótica con ayuda del procedimiento de acuerdo con la invención
Las células se cultivan como se describe en el Ejemplo 1. Las células se siembran en los respectivos recipientes de 5 cultivo (p. ej. placa de 24 pocillos/0,5 ml por cavidad) y, tras alcanzar la confluencia completa, se emplean para el ensayo. Las células se abastecen con medio nuevo:
(a)
Medio de cultivo reciente [tasa base de la apoptosis],
(b)
Medio reciente con 1 µg/ml de TSP-1 [sustancia inductora dela apoptosis; control],
(1) Medio (b) + péptido de SEQ ID NO: 12, 1 mM 10 (r) Medio (b) + péptido de SEQ ID NO: 18, 1 mM
(s) Medio (b) + péptido de SEQ ID NO: 19, 1 mM
Después de 24 h de incubación bajo condiciones de cultivo (Ejemplo 1) se fijan las células, se tiñen con DAPI y se examinan morfológicamente bajo el microscopio de fluorescencia o se examinan por citometría de flujo. Se determinan las células apoptóticas y el número total de células y se calcula el índice de la apoptosis (porcentaje de
15 células apoptóticas). En la Tabla 2 se indican los datos de 3 experimentos independientes con el dato de los valores medios y la desviación estándar. Péptidos de acción inhibidora presentan, de acuerdo con la invención, un índice de inhibición positivo, mientras que péptidos inductores presentan, de acuerdo con la invención, un índice de inhibición negativo.
