Abstract
Célula de levadura que produce isobutanol, caracterizada por que la célula presenta un flujo de sustancias metabólico incrementado de piruvato a través de acetolactato, 2,3-dihidroxiisovalerato, 2-cetoisovalerato, isobutiraldehído para formar isobutanol, en donde todos los genes que codifican las enzimas que participan en esta reacción están sobre-expresados, y por que estos genes son homólogos a dicha célula de levadura, en donde Ilv2 con SEQ ID Nº 2 cataliza la reacción de acetolactato sintasa de piruvato para formar acetolactato, ilv5 con SEQ ID Nº 6 cataliza la reacción de acetohidroxiácido reductoisomerasa de acetolactato para formar 2,3-dihidroxiisovalerato, Ilv3 con SEQ ID Nº 8 cataliza la reacción de dihidroxiácido deshidratasa de 2,3-dihidroxiisovalerato para formar 2- cetoisovalerato, una 2-cetoácido descarboxilasa cataliza la reacción de 2-cetoisovalerato para formar isobutiraldehído y una alcohol deshidrogenasa cataliza la reacción de isobutiraldehído para formar isobutanol, en donde al menos uno de los promotores de estos genes es intercambiado por al menos un promotor más fuerte, o las secuencias de ácidos nucleicos de estos genes son transformadas en alelos optimizados en codones, caracterizada por que se sobre-expresan los genes ILV2, ILV5 e ILV3 sin la secuencia diana mitocondrial de las enzimas acetolactato sintasa, acetohidroxiácido reductoisomerasa y dihidroxiácido deshidratasa o se sobre expresan con una secuencia diana mitocondrial destruida e inactivada, y caracterizada porque, adicionalmente, se reduce o bien se suprime la expresión de los genes PCD1 con SEQ. ID. No. 13, PDC5 con SEQ. ID. No. 15 y PDC6 con SEQ. ID. No. 17 o bien la actividad de las enzimas codificadas.
