Abstract
The invention relates to an antibiotic 16,511 R.P., which is a yellow amorphous powder of m. pt. 114 C. and which is an acid forming salts with bases, e.g. alkali metal salts it is soluble in methanol and ethanol, methylene chloride, chloroform, ethyl acetate, dimethylformamide, dimethylsulphoxide and acetone, less so in benzene and diethyl ether and practically insoluble in water, cyclohexane and hexane it has the elementary analysis C= = 65À8%, H=7À8%, O=22À7%, N=3À55% its ultraviolet spectrum (Fig. 1) shows absorption max. at 232 nm. (E1% 1 cm =682) and 350 nm max. at 232 nm. (E1% 1cm = 682) and 350 nm (E1% 1 cm. = 375) and minimum at 260 nm (E1% 1 cm. =130) its infra-red spectrum is shown in Fig. 3 [[alpha]] D 22 = - 121 2 (C=1À0) it contains components A (Rf value=0À1), B (Rf=0À2) also known as 18,051 R.P., C (Rf= 0À35) and D (Rf=0À60). Rf value is determined by thin layer chromatography using a Kieselgel H support and ethyl acetate as solvent. Antibiotic 18,051 R.P. is a yellow amorphous powder of m. pt. 112 C. having the elementary analysis C=64À7%, H=8À2%, O=23À75%, N=3À35% U.V. as in Fig. 6 (not shown) I.R. as in Fig. 8 (not shown), and [[alpha]]22 D = - 116À5 5À5 (C=0À73). Antibiotic 16,511 R.P. is produced by aerobically cultivating Streptomyces hygroscopicus var. tepidalitus NRRL 3237, Streptomyces lavendulae NRRL 3277, Streptomyces lavendulae NRRL 3278, or a 16,511 R.P.- producing mutant thereof. 16,511 R.P. may be isolated from the fermentation broth by (a) solvent extraction of the whole or clarified broth, (b) adsorption or strongly anionic exchange resins from a clarified broth, followed by elution with aqueous alcohol containing an electrolyte, e.g. sodium chloride. Purification may be by (a) precipitation from solution by a non-solvent such as hexane, (b) chromatography on alumina, silica gel or florisil, (c) treatment with carbon. 18,051 R.P. is separated from 16,511 R.P. by (A) chromatography on Kieselgel buffered at pH 4À0 with M/15 phosphate buffer, the chromatogram being developed with ethyl acetate (B) selective extraction with a methylene chloride-cyclohexane M/15 phosphate buffer at pH 9À0 (80: 20: 100 by volume). The antibiotics are effective against Gram positive micro-organisms. Pharmaceutical compositions, e.g. injections, comprise at least one of 16,511 R.P. and 18,051 R.P. or a non-toxic salt thereof and a carrier.
