Abstract
Procedimiento para el enriquecimiento de una glicoproteína que se distingue por a) una fracción de proteínas de 75 + _ 6%; b) una fracción de hidratos de carbono de 24,6 + _ 5,2%, de ellos hexosas 8,9 + _ , hexosamina acetilada en N 7,1 + _ 1,5% , fucosa 0,2% , ácido neurámico acetilado en N 8,4 + _ 1,5% , c) un coeficiente de sedimentación S(sub 20w) de 2,5 + _ 0,3 S ; d) un peso molecular determinado en la ultracentrífuga de 35000 + _ 5000; e) un punto isoeléctrico de pH 3,4 + _ 0,4 ; f) un coeficiente de extinción E (sup 1%, sub 1 cm) (280 nm) de 1,9 + _ 0,3 ; g) una movilidad electroforética en el margen entre [alfa (sub 1)-globulinas] y [alfa (sub 2)-globulinas; y h) una reacción inmunológica específica con un anticuerpo específicamente orientado contra la glicoproteína; caracterizado porque una solución de proteínas, en la que se puede reconocer inmunológicamente la glicoproteína, es sometida al menos a una de las siguientes medidas y se obtiene la fracción enriquecida respecto a la glicoproteína: a) adición de sales neutras hasta la separación de la glicoproteína y por precipitación; b) fraccionamiento en tamiz molecular y obtención de la fracción con un peso molecular de 25000 a 75000; c) adsorción de la glicoproteína en un cambiador de iones débilmente básico, y elución del mismo; d) adición de derivados solubles en agua de una base acridínica o quinoleínica, en el margen de pH 5-10, hasta una concentración final de aproximadamente 0,8%; e) tratamiento de la solución de glicoproteína con hidroxilapatito; f) electroforésis preparativa de zonas y obtención de la zona entre [alfa (sub 1)-globulinas y [alfa (sub 2)-globulinas; y g) tratamiento de la solución de proteínas con un inmuno-adsorbente.
