Abstract
Un procedimiento para la preparación de una enterotoxina termolábil aislada de E. coli, con un peso molecular de 10.000-13.000, que contiene 90% de proteína, 2% de hexosa y nada de ácido 2 - cetodesoxioctónico y que no presenta actividad en el ensayo de lisato de límulo; cuyo procedimiento consiste en: a) desarrollar un cultivo de E.coli en un medio de cultivo adecuado; b) aislar la proteína del medio de cultivo por precipitación con (NH4)2SO4 para formar un concentrado crudo; c) cromatografiar la solución de concentrado cruso de proteína sobre agarosa y eluir con un tapón adecuado, tal como Tris 0,02M pH 7,2, bicarbonato amónico pH 7,2 o acetato amónico pH 7,2 y después d) cromatografiar la solución resultante sobre hidroxilapatito y eluir con fosfato sódico 0,01M, pH 7,0; recuperar el producto purificado así obtenido.
