Abstract

PROCEDIMIENTO PARA PREPARAR UNA VACUNA DE VIRUS VIVOS DE LA PERITONITIS INFECCIOSA DE LOS FELINOS (PIF). CONSISTENTE EN AISLAR EL VIRUS DE LA PIF DE LOS ORGANOS O TEJIDOS DE ANIMALES INFECTADOS CON EL MISMO, DESAGREGANDO LOS ORGANOS O TEJIDOS EN CELULAS AISLADAS O UTILIZANDOLOS COMO CULTIVOS DE ORGANO. EL VIRUS SE REPRODUCE EN CELULAS OBTENIDAS DIRECTAMENTE DE TEJIDOS DE GATO Y EN LINEAS CELULARES CONTINUAS OBTENIDAS PRIMITIVAMENTE DEL FELINO. EL VIRUS SE REPRODUCE A UNA TEMPERATURA COMPRENDIDAD ENTRE APROXIMADAMENTE 30 Y 40 C, EN CELULAS QUE SE DESARROLLAN EN UNA MULTITUD DE MEDIOS DE CULTIVO. DESPUES DE COMPROBAR QUE EL VIRUS SE HA MULTIPLICADO EN LOS CULTIVOS DE CELULAS NORMALES, LOS CULTIVOS CELULARES PUEDEN SER CONGELADOS Y DESCONGELADOS UNA O MAS VECES, Y EL VIRUS EXISTENTE EN EL FLUIDO QUE SOBRENADA HACERSE PASAR A OTROS CULTIVOS DE CELULAS NORMALES, PUDIENDO PASARSE DE MODO SERIADO SOBRE LINEAS CELULARES ESTABLES POR LO MENOS 80 VECES SIN EXHIBIR CAMBIO, AUMENTANDO EL CONTENIDO DE VIRUS DEL FLUIDO CON CADA PASO CELULAR SERIADO, HASTA ALCANZAR UNA CONCENTRACION MAXIMA. LAS VACUNAS SE PREPARAN DILUYENDO SUSPENSIONES DE VIRUS DE LA PIF, Y TAMBIEN COMO VACUNAS POLIVALENTES CON OTROS VIRUS CONOCIDOS DE FELINOS.
