Abstract

1,237,268. Proteinaceous reagent. MILES LABORATORIES Inc. 11 Oct., 1968 [1 Nov., 1967], No. 48249/68. Heading B1X. [Also in Divisions C3 and C6] A proteinaceous reagent is prepared by a process comprising (a) treating an antigen, antibody or enzyme with either (i) a bis-diazotised aromatic compound capable of coupling with protein, or (ii) a linking compound (defined below), (b) treating protein-surfaced insoluble particles with whichever of compounds (i) or (ii) is not used in step (a), and (c) reacting the two treated materials together so that the unreacted diazo groups of compound (i) couple with the linking compound (ii) to form the proteinaceous reagent. Compound (i) may be bis-diazotised benzidine, dianisidine, or 4, 4', diphenylamine. The linking compound (ii) is a compound reactive with proteinaceous material and containing an aromatic radical which is more susceptible to electrophilic substitution than any aromatic material occurring in protein. Compounds (ii) exemplified are pyrrole-2-carboxylic acid azide, thiophene-2-carboxylic acid azide, 2-furoic acid with N-ethyl-5-phenylisoxazolium-3'- sulphonate (Woodward's reagent K) as activator, 2-thiophenesulphonyl chloride; others are mentioned. The protein-surfaced insoluble particles may be red blood cells (opossum, rat, rabbit, guinea pig, human, goat, sheep, chicken, alligator, turtle), microbial cells (preferably Brucella abortus, Escherichia coli (exemplified), Bacillus substilis, Bacillus pumilis, Lactobacillus kichmannii, Pseudomonas fragi, or Saccharonyees cerevisiae), or 0.1 to 10 microns size particles of for example polystyrene coated with gelatin. Antigens &c exemplified are myoglobin, HCG, haemoglobin, BSA, bovine gamma globulin, insulin; others are mentioned. Unit pack capsules may be prepared each carrying in different areas the lyophilized residues of a drop of a suspension of the reagent and of a dilution of the appropriate antiserum.
