Abstract
Η παρούσα εφεύρεση αναφέρεται σε χημική μέθοδο η οποία υπολογίζει την ισορροπία μεταξύ οξειδωτικών και αντιοξειδωτικών παραγόντων σε βιολογικά δείγματα και άλλα υλικά. Το παράδειγμα της εφαρμογής πουπεριγράφεται χρησιμοποιεί ως ουσία στόχο την 3,3΄,5,5΄- Τετραμεθυλοβενζδίνη (ΤΜΒ) και το αντίστοιχοέγχρωμο κατιόν της. Αντί όμως της ΤΜΒ μπορούν να χρησιμοποιηθούν και άλλες ουσίες οι οποίες μεταβάλλουν τις οπτικές ιδιότητες (απορροφητικότητα, φθορισμός, εκπομπή φωτεινής ή άλλης ακτινοβολίας) με την οξείδωση ή με την αναγωγή. Η εφεύρεση στηρίζεται σε δύο χημικές αντιδράσεις οι οποίες γίνονται ταυτόχρονα. Μία άμεση αντίδραση οξειδοαναγωγής η οποία έχει ως στόχο την ΤΜΒ και αναφέρεται στις αντιοξειδωτικές ουσίες και μία έμμεση, ένζυμη, αντίδραση η οποία έχει ως στόχο τα οξειδωτικά και ως υπόστρωμα το ίδιο το μόριο (ΤΜΒ). Κατά την άμεση αντίδραση οξειδοαναγωγής το κατιόν ΤΜΒ ανάγεται από τα αντιοξειδωτικά και αποχρωματίζεται. Με την έμμεση ενζυμική αντίδραση η ΤΜΒ μετατρέπεται στο έγχρωμο κατιόν ΤΜΒ. Έτσι η περίσσεια των αναγωγικών καταγράφεται με τη μείωση της έντασης του χρώματος ενώ η περίσσεια των οξειδωτικών με την αύξηση της έντασης του χρώματος του διαλύματος. Μετά την εξισορρόπηση της αντίδρασης και τη διακοπή της με την προσθήκη HC1 η ένταση του χρώματος μπορεί να υπολογισθείεύκολα με τη χρήση φωτόμετρου στα 450 nm (μήκος κύματος αναφοράς 620 ή 570 nm). Η τελική συγκέντρωση του κατιόντος ΤΜΒ αντιπροσωπεύει την ισορροπία μεταξύ οξειδωτικών αντιοξειδωτικών και μπορεί να συσχετισθεί με μία καμπύλη αναφοράς η οποία κατασκευάζεται από ανάλογες μετρήσεις που διεξάγονται σεδιαλύματα με γνωστές αναλογίες οξειδωτικού-αντιοξειδωτικού (0-100 της εκατό). Στο παράδειγμα που παρουσιάζεται χρησιμοποιούνται το υπεροξείδιο του υδρογόνου (ως αντιπροσωπευτικό των οξειδωτικών) καιτο ουρικό οξύ (ως αντιπροσωπευτικό των αντιοξειδωτικών). Η εφεύρεση μπορεί να είναι χρήσιμη για την εκτίμηση της ισορροπίας οξειδωτικών αντιοξειδωτικών σε βιολογικά δείγματα σε σχέση με το γήρας καινόσους που σχετίζονται με αυτό καθώς και σε μεταβολικά νοσήματα όπως ο διαβήτης. Επίσης μπορεί να είναι χρήσιμη για την εκτίμηση της θεραπείας με αντιοξειδωτικές ουσίες.
