Abstract

παρούσα ευρεσιτεχνία σχετίζεται με τη χρήση των ανταγωνιστών του υπότυπου CRH-R1 του κυτταρικού υποδοχέα της Εκλυτικής Ορμόνης της Κορτικοτροπίνης (corticotropine-releasing hormone, CRH) καθώς και/ή των αγωνιστών του υπότυπου CRH-R2του ανωτέρω υποδοχέα στη θεραπεία των φλεγμονωδών νόσων μέσω της ρύθμισης της ενεργοποίησης, πολλαπλασιασμού, διαφοροποίησης, ή/και απόπτωσης των μονοκύτταρων/μακροφάγων. Με τον όρο σύστημα του CRH προσδιορίζουμε τις φυσικές και συνθετικές ουσίες, αγωνιστές και ανταγωνιστές των CRH και Urocortin (Ucn) για τους κυτταρικούς υποδοχείς CRH-R1 και CRH-R2 και των υπότυπων τους καθώς και τη δεσμευτική πρωτείνη της CRH (CRH-building protein, BP), ένα ψευδο-υποδοχέα της CRH. Η ευρεσιτεχνία εστιάζει στις φαρμακολογικέςπαρεμβάσεις για τη βελτιστοποίηση της θεραπείας φλεγμονωδών νόσων με τη χρήση του συστήματος της CRH στον έλεγχο των μονοκύτταρων μακροφάγων τα οποία παίζουν καθοριστικό ρόλο στην έναρξη και διατήρηση της φλεγμονής μέσω της παραγωγής προ-γλεγμονωδώνκυτταροκινών, όπως π.χ. οι Ιντερλευκίνη 1 (IL)-1, η IL-6 και ο παράγων νέκρωσης των όγκων (tumor necrosis factor (TNF)-alpha). Με τον όρο φλεγμονή προσδιορίζουμε την απάντηση του οργανισμού σε επιβλαβείς ενδογενείς και εξωγενείς διεγέρσεις που προκαλούν ιστική βλάβη. Η φλεγμονή είναι μία αμυντική αντίδραση του ξενιστή που μπορεί όμως να καταλήξει και στη βλάβη του αμυνόμενου οργανισμού. Η ευρεσιτεχνία παρέχει επίσης μεθόδους για την in vitro και in vivo εκτίμηση των φυσικών και συνθετικών τροποποιητών του συστήματος της CRH ως παραγόντων ελέγχου των μονοκύτταρων/ μακροφάγων. ΰ
