Claims (15)

1. Postupak priprave biblioteke sekvenciranja, naznačen time što se postupak sastoji u: (a) dodavanju sredstva koje se veže na kromatin u uzorak koji sadrži nukleinsku kiselinu; (b) izoliranju kromatina vezanog na navedeno sredstvo; (c) dodavanju transpozaze izoliranom kromatinu iz koraka (b); (d) izoliranju nukleinske kiseline iz kromatina; i (e) dobivanju biblioteke sekvenciranja.
2. Postupak kartiranja molekulskih međudjelovanja koja uključuju nukleinsku kiselinu, naznačen time što se postupak sastoji u: (a) dodavanju sredstva koje se veže na kromatin u uzorak koji sadrži nukleinsku kiselinu; (b) izoliranju kromatina vezanog na navedeno sredstvo; (c) dodavanju transpozaze izoliranom kromatinu iz koraka (b); (d) izoliranju nukleinske kiseline iz kromatina; (e) amplificiranju nukleinske kiseline; (f) sekvenciranju amplificirane nukleinske kiseline; i (g) identificiranju molekulskih međudjelovanja.
3. Postupak u skladu s patentnim zahtjevima 1 ili 2, naznačen time što je sredstvo protutijelo.
4. Postupak u skladu s patentnim zahtjevima 1 do 3, naznačen time što se uzorak koji sadrži nukleinsku kiselinu pripravlja (i) uzgajanjem i prikupljanjem stanica; (ii) fiksiranjem stanica; (iii) liziranjem stanica, a time i dobivanjem prvog uzorka koji sadrži nukleinsku kiselinu; i (iv) sonificiranjem prvog uzorka, a time i dobivanje drugog uzorka koji sadrži nukleinsku kiselinu, gdje se navedeni drugi uzorak upotrebljava u postupku u skladu s patentnim zahtjevima 1 ili 2.
5. Postupak u skladu s patentnim zahtjevima 1 do 4, naznačen time što postupak dodatno uključuje korak prekidanja veza za umrežavanje unesenih tijekom fiksiranja stanica.
6. Postupak u skladu s bilo kojim od patentnih zahtjeva 1 do 5, naznačen time što je nukleinska kiselina DNA.
7. Postupak u skladu s bilo kojim od patentnih zahtjeva 4 do 6, naznačen time što stanice sadrže komplekse nukleinska kiselina-protein.
8. Postupak u skladu s patentnim zahtjevom 7, naznačen time što su stanice ljudske stanice, životinjske stanice, bakterijske stanice, stanice kvasca, arhejske stanice, biljne stanice ili virusi.
9. Postupak u skladu s patentnim zahtjevom 8, naznačen time što ljudske ili životinjske stanice su bolesne stanice ili nebolesne stanice ili stanice dobivene iz bolesnog ili nebolesnog tkiva i/ili što ljudske ili životinjske stanice su stanice raka, imunosne stanice, krvne stanice ili matične stanice.
10. Postupak u skladu s patentnim zahtjevom 9, naznačen time što je rak čvrsti rak ili krvni rak, po mogućnosti što je krvni rak leukemija ili što je čvrsti rak tumor.
11. Postupak u skladu s patentnim zahtjevom 4, naznačen time što korak (ii) se sastoji u dodavanju kemijske tvari i/ili fizičkog sredstva i/ili što korak (iv) se sastoji u sonificiranju dok većina fragmenata nukleinske kiseline ne bude duga 20-5000, po mogućnosti 200-300, parova baza.
12. Postupak u skladu s patentnim zahtjevom 1 ili 2, naznačen time što se protutijelo specifično veže na histone, transkripcijski čimbenici ili proteini koji se vežu na histone i/ili transkripcijske čimbenike.
13. Postupak u skladu s patentnim zahtjevom 12, naznačen time što su proteini koji se vežu na histone i/ili transkripcijske čimbenike proteini za remodeliranje nukleinskih kiselina ili enzimi za modificiranje kromatina, po mogućnosti što je histon H3.3, H2A.Z, CENP-A, H3.2, H3.3A, H3.3B, H4 ili H3.1, po mogućnosti što je histon modificirani histon, gdje je modificiranje metiliranje, acetiliranje, propioniliranje, butiriliranje, krotoniliranje, 2-hidroksiizobutiriliranje, maloniliranje, sukciniliranje i/ili riboziliranje, po mogućnosti što je modificirani histon H3K4me1/2/3, H2BK5me1, H3K27me1/2/3, H3K9me1/2/3, H4K20me1, H3K79mel, H3K36me3, H2AK5ac, H2AK9ac, H2BK5ac, H2BK12ac, H2BK20ac, H2BK120ac, H3K4ac, H3K9ac, H3K14ac, H3K18ac, H3K23ac, H3K27ac, H3K36ac, H4K5ac, H4K8ac, H4K12ac, H4K16ac, H4K91ac, H2Aub ili H2Bub.
14. Postupak u skladu s patentnim zahtjevima 1 ili 2, naznačen time što je sredstvo kemijska tvar, po mogućnosti što je kemijska tvar lijek, po mogućnosti što je kemijska tvar biotinilirana.
15. Postupak u skladu s patentnim zahtjevima 1 ili 2, naznačen time što transpozaza sadrži nasumične sljedove DNA kao biljege ili definirane sljedove DNA kao biljege, po mogućnosti što je transpozaza transpozaza Tn5.
