Claims (13)
1. Izolirani ili pročišćeni polipeptid, naznačen time što ima aktivnost egzoglukanaze poboljšanu za najmanje 10 % na temperaturi od otprilike 35 °C, te izborno za najmanje 10 % na temperaturi od otprilike 50 °C, u odnosu na aktivnost egzoglukanaze referentnog proteina CBH1 sa slijedom SEQ ID NO: 2 na supstratu kojeg se bira između Avicel celuloze, PASC celuloze, reduciranog celodekstrinskog supstrata ili kromogenskog supstrata, poput pNP laktozida, gdje se navedeni polipeptid bira iz skupine koju čine:
i. aminokiselinski slijed kojeg se bira između SEQ ID NO: 8, SEQ ID NO: 4, SEQ ID NO: 16, SEQ ID NO: 6, SEQ ID NO: 10, SEQ ID NO: 12, SEQ ID NO: 18 i SEQ ID NO: 20, i
ii. aminokiselinski slijed koji ima, u odnosu na slijed SEQ ID NO: 4, SEQ ID NO: 6, SEQ ID NO: 12, SEQ ID NO: 18 ili SEQ ID NO: 20, postotak istovjetnosti od najmanje 99 %.
2. Pročišćena ili izolirana nukleinska kiselina, naznačena time što kodira najmanje jedan polipeptid u skladu s patentnim zahtjevom 1.
3. Pročišćena ili izolirana nukleinska kiselina u skladu s patentnim zahtjevom 2, koju se bira između sljedećih sljedova: SEQ ID NO: 7, SEQ ID NO: 3, SEQ ID NO: 15, SEQ ID NO: 5, SEQ ID NO: 9, SEQ ID NO: 11, SEQ ID NO: 17 i SEQ ID NO: 19.
4. Vektor, naznačen time što sadrži najmanje jednu nukleinsku kiselinu u skladu s bilo kojim od patentnih zahtjeva 2 ili 3.
5. Izolirana stanica domaćin, naznačena time što sadrži najmanje jedan polipeptid u skladu s patentnim zahtjevom 1, ili najmanje jednu nukleinsku kiselinu u skladu s bilo kojim od patentnih zahtjeva 2 ili 3, ili najmanje jedan vektor u skladu s patentnim zahtjevom 4.
6. Izolirana stanica domaćin u skladu s patentnim zahtjevom 5, naznačena time što je se bira između rodova Trichoderma, Aspergillus, Neurospora, Humicola, Penicillium, Fusarium, Thermomonospora, Myceliophthora, Chrysosporium, Bacillus, Pseudomonas, Escherichia, Clostridium, Cellulomonas, Streptomyces, Yarrowia, Pichia i Saccharomyces.
7. Izolirana stanica domaćin u skladu s bilo kojim od patentnih zahtjeva 5 ili 6, naznačena time što je se bira između vrsta Trichoderma reesei, Trichoderma viridae, Trichoderma koningii, Aspergillus niger, Aspergillus nidulans, Aspergillus wentii, Aspergillus oryzae, Aspergillus phoenicis, Neurospora crassa, Humicola grisae, Myceliophthora thermophila, Chrysosporium lucknowense, Penicillium pinophilum, Penicillium oxalicum, Escherichia coli, Clostridium acetobutylicum, Clostridium saccharolyticum, Clostridium benjerinckii, Clostridium butylicum, Pichia pastoris, Yarrowia lipolityca i Saccharomyces cerevisiae.
8. Upotreba navedenog polipeptida u skladu s patentnim zahtjevom 1 u hidrolizi celuloze.
9. Upotreba navedenog polipeptida u skladu s patentnim zahtjevom 1 u proizvodnji biogoriva.
10. Enzimski pripravak koji može djelovati na lignoceluloznu biomasu, gdje navedeni enzimski pripravak proizvode filamentozne gljivice i sadrži najmanje jedan polipeptid u skladu s patentnim zahtjevom 1.
11. Postupak proizvodnje biogoriva iz lignocelulozne biomase, naznačen time što se sastoji u sljedećim uzastopnim koracima:
– suspendiranju biomase koju treba hidrolizirati u vodenoj fazi;
– hidroliziranju lignocelulozne biomase u prisutnosti enzimskog pripravka u skladu s patentnim zahtjevom 10 kako bi se proizveo hidrolizat koji sadrži glukozu;
– fermentiranju glukoze iz hidrolizata kako bi se proizvela fermentacijska masa;
– odvajanju biogoriva iz fermentacijske mase.
12. Postupak proizvodnje biogoriva iz biomase, naznačen time što se sastoji u sljedećim uzastopnim koracima:
– suspendiranju biomase koju treba hidrolizirati u vodenoj fazi;
– istodobnom dodavanju enzimskog pripravka u skladu s patentnim zahtjevom 10 i fermentirajućeg organizma u suspenziju i fermentiranju smjese kako bi se proizvela fermentacijska masa;
– odvajanju biogoriva iz fermentacijske mase.
13. Postupak u skladu s bilo kojim od patentnih zahtjeva 11 ili 12, gdje je fermentirajući organizam stanica domaćin u skladu s bilo kojim od patentnih zahtjeva 5 do 7.
